The SynMuvA lin-15A licenses natural transdifferentiation by antagonizing identity safeguarding mechanisms.

The mechanisms that restrict or enable latent cellular plasticity have attracted growing interest over the past decade, with important implications for cancer and regenerative therapies. However, the diversity of both pro- and anti-plasticity mechanisms remains incompletely understood. Here, we identify the THAP domain gene lin-15A as a novel factor involved in the natural rectal-to-neuronal Y-to-PDA transdifferentiation in Caenorhabditis elegans. Our genetic analyses show that lin-15A acts in parallel to previously described plasticity factors. Null mutant suggests that, rather than driving t

PIWI proximity proteome reveals Set1-mediated piRNA biogenesis for transposon silencing at telomeres.

Silencing complexes formed by PIWI-clade Argonaute (Ago) proteins and PIWI-interacting RNAs (piRNAs) are essential guardians of genome integrity, restricting the activities of transposable elements (TEs) in the animal germline. However, our understanding of PIWI-piRNA-directed TE silencing remains incomplete. Here, we systemically characterize the proximity proteome of the PIWI members Piwi, Aubergine (Aub), and Ago3 in the germline of Drosophila ovaries. Functional screening identifies previously uncharacterized factors involved in TE silencing, including the H3K4me3 writer and transcriptiona

Maternal obesity imprints methylation marks in oocytes to drive intergenerational metabolic dysfunction.

Maternal obesity predisposes offspring to metabolic disease, yet how oocytes transmit this risk remains unclear. Here, we show that maternal obesity induces locus-specific DNA methylation changes in oocytes to drive intergenerational metabolic dysfunction in a tissue-specific and sex-specific manner. Among these, a subset of metabolic genes, notably Hnf1α, Thra and Pdk4, exhibited persistent intergenerational methylation and transcriptional dysregulation. Although these methylation marks were erased during embryogenesis, H3K36me2 enrichment was retained at the loci, suggesting a potential chro

SUMOylation and palmitoylation in metastatic prostate cancer: A compartment-based framework for nuclear and membrane adaptation.

Metastatic prostate cancer remains lethal despite treatment intensification, in part because tumor cells can adopt rapid and reversible states that are not fully explained by genomic alterations. Post-translational modifications provide a dynamic regulatory layer by changing protein activity, localization, stability, and molecular interactions. This Review evaluates SUMOylation and reversible S-palmitoylation as compartment-biased systems that regulate complementary dimensions of metastatic adaptation. SUMOylation has a comparatively mature mechanistic foundation in androgen receptor transcrip

Cell-Autonomous AR Dependence in Luminal Prostatic Epithelium Governs Survival and Lineage Plasticity.

Prostate cancer resembles differentiated secretory luminal cells and shows cell-autonomous dependence on androgen receptor (AR) signaling, yet normal luminal cells are often considered dependent on paracrine stromal AR signaling. To resolve this, we conditionally deleted Ar in luminal acinar cells in vivo . Ar -deleted luminal cells persisted short-term, in contrast to the rapid regression observed after castration, but were impaired in regeneration and progressively lost. Their depletion was accompanied by replacement through basal-to-luminal differentiation of AR intact basal cells. Transcri

Clinicopathological and molecular characteristics of small cell carcinoma of the ovary, hypercalcemic type: three case reports.

Small cell carcinoma of the ovary, hypercalcemic type (SCCOHT), is a rare and aggressive ovarian cancer that affects populations ranging from children to young adults. It is usually associated with germline and somatic variants of SMARCA4, which encodes the SMARCA4 protein (BRG1), as well as SMARCA2 (BRM) deficiency, both subunits of the switch/sucrose-nonfermenting (SWI/SNF) chromatin remodeling complex. Here, we report the clinical, morphological, immunohistochemical, and molecular characteristics of three cases. All three patients showed abnormally elevated preoperative CA-125 levels and hy

C-Jun-activated S100A10 promotes malignant progression via in diminishing ubiquitin-dependent degradation of vimentin in gastric cancer.

Gastric cancer remains a significant global health challenge. Lymph node metastasis (LNM) has been identified to be relevant to the prognosis of gastric cancer (GC). However, its mechanisms of progression and metastasis are still not fully understood. Recently, S100A10 has been verified to be aberrantly expressed in various cancers. In this study, we found that S100A10 expression was significantly upregulated in metastatic GC tissues. S100A10 remarkably accelerated tumor metastasis and growth. Mechanistically, dual-luciferase assay and chromatin immunoprecipitation revealed that c-Jun could bi

Selenoprotein H Functions as a PPARα Coactivator to Link Selenium Homeostasis to Hepatic Lipid Metabolism and Protect against Steatohepatitis.

Selenium is an essential trace element whose dysregulation is associated with diverse disease risks; however, its specific role in hepatic metabolism remains poorly defined. Here we delineate a novel selenium-selenoprotein H (SELENOH)-PPARα signaling axis that is critical for hepatic lipid homeostasis. We first uncovered a global impairment of selenoprotein translation as a key feature of metabolic dysfunction-associated steatohepatitis (MASH) in human patients and mouse models. Both dietary selenium supplementation and genetically rescuing selenoprotein biosynthesis attenuated MASH pathology,

The mechanism and treatment of protein acylation modification in cardiovascular disease: a narrative review.

Cardiovascular diseases are the leading causes of death and disability worldwide. Their initiation and progression involve multiple complex processes, including metabolic disorders, oxidative stress, inflammatory responses, and cell death. Protein acylation, a rapidly advancing field in post-translational modification research, dynamically regulates protein function, chromatin status, and metabolic signaling networks by covalently attaching various acyl groups to specific protein sites, thereby serving as a key molecular mechanism linking cellular metabolic states to cardiovascular pathology.

Targeting POLRMT-driven epigenetic remodeling of Wnt/β-catenin to eradicate colorectal cancer stem cell proliferation.

Colorectal cancer stem cells (CSCs) drive tumor progression through poorly understood metabolic-epigenetic crosstalk. Here, we identify mitochondrial RNA polymerase POLRMT as a key link connecting mitochondrial transcription to CSC maintenance. Clinically, POLRMT is overexpressed in colorectal cancer (CRC) tissues and correlates with poor prognosis. Genetic ablation or pharmacological inhibition of POLRMT suppresses CSC self-renewal and tumorigenicity across cell line-derived CSCs, CRC organoids, and xenograft models. Mechanistically, POLRMT deficiency triggers mitochondrial dysfunction, which

A reference genome for cultured Drosophila ovarian somatic cells enables studies of transposon and piRNA biology.

BACKGROUND: Accurate reconstruction of repetitive genomic regions is essential to understand small RNA-mediated transposon silencing. In Drosophila ovarian somatic cells (OSCs), a primary model for nuclear PIWI-piRNA biology, reliance on the D. melanogaster reference (dm6) obscures cell-line-specific transposon insertions and the architecture of piRNA source loci. RESULTS: We generated a highly contiguous de novo assembly of the OSC genome that reveals a transposon landscape diverging substantially from dm6. The assembly fully resolves the ∼700 kb flamenco locus, the principal piRNA cluster i

Functional dynamics of lncRNAs and miRNAs in cancer metabolism and neuroinflammation.

Non-coding RNAs (ncRNAs) influence gene expression and diverse physiological and pathological processes. Long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) play roles in cancer and neurodegenerative diseases. LncRNAs, such as HOTAIR, MALAT1, and NEAT1, regulate chromatin remodeling through histone modification. Dysregulated expression leads to oncogenesis and neuronal dysfunction. Conversely, miRNAs such as miR-21 and miR-155, as post-transcriptional regulators, act in oncogenic signaling and neuroinflammation. MiR-21 targets PTEN and PDCD4, leading to aberrant activation of the PI3K/AKT an

Mitochondrial metabolism and epigenetic crosstalk drive SASP.

Senescent cells promote tissue dysfunction in part through the senescence-associated secretory phenotype (SASP)1. Cytosolic mitochondrial nucleic acids activate innate immune signalling to initiate this inflammatory programme2,3. Here we show that mitochondrial metabolism provides a second layer of control that enables execution of the inflammatory programme. In senescent cells, the mitochondrial pyruvate-citrate-acetyl-CoA axis is upregulated, increasing the availability of acetyl-CoA to support histone acetylation at SASP genes. Whereas mitochondrial DNA-driven signalling activates inflammat

Bioelectric regulation of stem cell fate: From the plasma membrane to organelles.

Stem cell identity has traditionally been viewed through the lens of transcriptional and metabolic programs. Recent work has revealed an additional layer of complexity: the plasma membrane and intracellular organelles maintain compartment-specific membrane potentials, pH gradients, and ion dynamics that can influence stemness and fate. In this review, we synthesize recent advances identified across diverse stem cell populations to highlight convergent and divergent bioelectric mechanisms driving cell fate decisions. We explore how plasma membrane mechanosensors convert physical cues into inter

Impact of Iron Speciation on the Cytotoxicity and Gene Expression Profile of Biofortified Hericium erinaceus in Human Colorectal Adenocarcinoma.

Colorectal cancer (CRC) is a major global health threat, necessitating the development of functional foods with chemopreventive potential. This study aimed to evaluate the modulation of the anticancer activity of Hericium erinaceus with different iron forms (FeCl3, FeSO4, and FeHBED) as it modulates its anticancer activity against HT-29 cells. The extracts were subjected to simulated in vitro digestion and analyzed for cytotoxicity (MTT), antioxidant capacity (DPPH), and morphological changes, alongside high-throughput RT-qPCR profiling of 92 cancer-related genes. The results demonstrated that

Uncovering Spatiotemporal and Functional Dynamics of Long Non-coding RNAs During Alzheimer's Progression in the Human Brain at Single-Cell Resolution.

Increasing prevalence of Alzheimer's disease, driven by population aging, highlights the need to investigate its underlying molecular mechanisms. Within this context, long non-coding RNAs (lncRNAs) have emerged as a key regulatory layer. To advance the understanding of lncRNA dysregulation and function during Alzheimer's disease progression, we reanalyzed publicly available single-nucleus RNA sequencing (snRNA-seq) datasets. The selected transcriptomic datasets were integrated and subjected to differential expression and genomic co-localization correlation analyses to infer putative cis-regula

Tau aggregates cause reactivation of transposable DNA elements, leading to Z-RNA-ZBP1-mediated neuronal death.

Once tau aggregates are formed, their neurotoxicity significantly contributes to neuronal death and cognitive decline in tauopathies, with Alzheimer's disease being the most well-known example. Despite its central pathogenic role, however, effective therapeutic strategies targeting the neurotoxicity of tau remain poor. Here we demonstrate the pathogenic role of neuronal cell death in tau-related neurodegeneration (PS19 mouse model). Tau-expressing neurons undergo cell death through Z-DNA-binding protein 1 (ZBP1) activation triggered by endogenous Z-RNAs. These Z-RNAs are derived from reactivat

Role of paraptosis in cancer: Molecular mechanisms and therapeutic potentials.

Paraptosis represents a mode of cell death, which is separate from classical apoptosis. Classical apoptotic pathways are caspase-dependent, accompanied by cell shrinkage, chromatin condensation, and apoptotic body formation. In contrast, paraptosis is characterized by cytoplasmic vacuolization without typical apoptotic nuclear alterations, and its signaling operates independently of the caspase system. Unlike apoptosis, paraptosis primarily depends on the mitogen-activated protein kinase (MAPK) signaling pathway for its mediation, highlighting its unique regulatory framework separate from clas

Distinct transcription factors control tissue adaptation and effector function in infant and adult memory T cells.

Early life is essential for establishing memory T cells, which rapidly populate mucosal sites during infancy, although these nascent memory T cells are less protective than their adult counterparts. Here we used single-cell RNA sequencing of resting and CD3+CD28 antibody-stimulated T cells from lymphoid and mucosal tissues of infant (2-9 months) and adult (40-63 years) organ donors to investigate age-dependent mechanisms for functional regulation of human memory T cells. Lung Cancer Chemoresistance: Insights into the Regulatory Role of lncRNAs.

Lung cancer remains the leading cause of cancer-related mortality worldwide, with chemoresistance being a major challenge in its treatment. Long non-coding RNAs (lncRNAs), a class of transcripts longer than 200 nucleotides without protein-coding potential, have emerged as critical regulators in tumour progression and drug resistance. This review systematically summarizes the multifaceted mechanisms through which lncRNAs influence chemoresistance across major lung cancer subtypes, particularly non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC), including regulation of cell pro
