[Study of caryotypes in 52 cases of chronic lymphocytic B-cell leukemia].

The karyotypes of 103 cases of B cell chronic lymphocytic leukemia were studied following stimulation or establishment of continuous cell lines using the Epstein-Barr virus. In 52 patients metaphases suitable for cytogenetic analysis were obtained; 30 revealed normal karyotypes and 22 abnormal karyotypes. The most frequently encountered abnormalities were 6 cases of extra chromosome 12, 4 cases of structured aberrations concerning chromosome 14, including 2 t(11;14), 4 translocations concerning chromosome 11, and 3 cases of extra chromosome 3. No relationship appears to exist between the abnor

Chromosome studies on acute nonlymphocytic leukaemia in children.

Cytogenetic studies have been carried out on 17 children with acute nonlymphocytic leukaemia (ANLL). Of the 16 patients analysed at diagnosis, 11 had acquired clonal chromosome abnormalities. Four out of seven cases with acute myeloid leukaemia (M2) had 8;21 translocations, two of which were variants. Comparisons with other data on ANLL confirmed the association between the 8;21 translocation and the younger age groups. There are indications that the Netherlands may be a high incidence area for this translocation. Differences in the type of chromosome anomalies between childhood and adult ANLL

'Expanded' Prader-Willi syndrome in a boy with an unusual 15q chromosome deletion.

A male infant showed features of the Prader-Willi syndrome (including profound hypotonia, cryptorchidism, and mildly dysmorphic facial appearance) but also had additional multiple malformations (congenital heart disease, unilateral renal malmigration, and bifid uvula). A deletion of the long arm of chromosome 15, larger than that usually demonstrated in children with Prader-Willi syndrome, was found. The cytogenetic findings suggest that the infant's hypotonia and cryptorchidism are explicable on the basis of the portion of the deletion usually associated with Prader-Willi syndrome (q11 to q12

Radiogenic male breast cancer with in vitro sensitivity to ionizing radiation and bleomycin.

A cytogenetically normal man with gynecomastia and a family history of diverse cancers developed adenocarcinoma of the breast 30 years following thymic irradiation. In vitro experiments measuring colony-forming ability of cultured skin fibroblasts from family members implied that the patient had a small but significant increase in sensitivity to ionizing radiation, and a moderate increase in sensitivity to bleomycin, a radiomimetic drug. Enhanced radiosensitivity of fibroblasts from the patient's mother, and bleomycin sensitivity of fibroblasts from the sister suggested, but did not prove, tha

Application of conventional and FPG staining for the analysis of chromosome aberrations induced by low levels of dose in human lymphocytes.

Human peripheral lymphocytes were irradiated with 0.05-0.5 Gy of 220 keV X-rays. After application of either a conventional or the fluorescence plus Giemsa (FPG) staining technique, the dose response for dicentrics and acentrics was studied. The analysis of exclusively first-division cells (M1), carried out by the FPG method, revealed significantly higher aberration yields as compared with the results of the conventional method. The data from M1 cells support the assumption of a linear dose response for both dicentrics and acentrics. The results are discussed with regard to the application of

A cytogenetic screening of 1868 mentally diseases criminals is performed. 5 man with abnormal karyotypes were found: 2 with the chromosome set 47,XXY; 1--with mosaicism 46,XY/47,XXY and 2--with karyotype 47,XYY. Total frequency of chromosome abnormalities was found to be 0.27% (XXY karyotype--0.16%, XYY--0.11%). Among tall mentally diseased criminals the frequency of XXY set was found to be 0.94%, and of XYY--1.89%. So among tall mentally diseased criminals the frequency of these abnormalities is higher. The frequency of karyotype abnormalities among mentally diseased criminals was not found t

[Congenital gastrointestinal diseases with manifestations in adulthood].

A genetic background is discussed in many disorders of the gastrointestinal tract with a disposition in addition to environmental factors. The pathophysiology of most hereditary diseases is unknown although the mode of inheritance is established. Biochemical analysis may show molecular defects or inborn lack of enzymes, cytogenetic studies may reveal chromosomal abnormalities. The knowledge of genetic factors in gastrointestinal disorders may contribute to the early detection of persons afflicted but not yet symptomatic, in some rare syndromes genetic counseling may become mandatory. Finally,

Meiotic consequences of an intrachromosomal insertion of chromosome No 1: a family pedigree.

In three generations of the proband's patrilineal relatives, 14 subjects were found to be carriers of a "shift" insertional chromosome No. 1 (46XX or XY, ins(1)(p32q25q31)). The proband and three female relatives, who were mild to moderate mental retardates with minor congenital anomalies, were trisomic for the insertional segment, (1)q25q31. Another subject, who was a markedly immature female abortus with congenital abnormalities, was found to be monosomic for this same chromosomal segment. The cytogenetic evidence suggests that each of these unbalanced recombinant progeny was the result of a

Chromosomal abnormalities in angio-immunoblastic lymphadenopathy.

Chromosomal studies have been performed in 2 patients with angio-immunoblastic lymphadenopathy. In both the cases the presence of abnormal cell lines characterized by marker chromosomes has been detected. Application of banding techniques allowed to detect the structural composition of the marker chromosome in 1 of the cases and to show a clonal evolutive pattern of the rearranged chromosomal set; In the same patient a consistent Y loss observed in the major fraction of the investigated metaphases did not appear to be related to any defined rearrangement of the karyotype. An equine cryptorchid with testicular and ovarian tissues.

Cytogenetic and histological studies were carried out on an intersex horse which was diagnosed clinically as a cryptorchid. Surgery confirmed the horse to be a bilateral abdominal cryptorchid and histological examination revealed ovarian tissue associated with the left epididymis. Chromosome analysis of cultured cells from testicular tissue, ovarian tissue and skin revealed 64-XX and 64-XY make-up, the left gonad containing a greater preponderance of XX cells over XY cells. The external characteristics and behaviour of the horse were indistinguishable from that of a "routine" cryptorchid. Othe

The numerous facets of 1q21+ in multiple myeloma: Pathogenesis, clinicopathological features, prognosis and clinical progress (Review).

Multiple myeloma (MM) is a malignant neoplasm characterized by the clonal proliferation of abnormal plasma cells (PCs) in the bone marrow and recurrent cytogenetic abnormalities. The incidence of MM worldwide is on the rise. 1q21+ has been found in ~30-40% of newly diagnosed MM (NDMM) patients.1q21+ is associated with the pathophysiological mechanisms of disease progression and drug resistance in MM. In the present review, the pathogenesis and clinicopathological features of MM patients with 1q21+ were studied, the key data of 1q21+ on the prognosis of MM patients were summarized, and the clin

[Prognosis and immune biomarkers of multiple myeloma].

Multiple myeloma (MM) is a highly heterogeneous hematological malignancy. The complex cytogenetic changes, immune cell infiltration and genetic mutations in MM jointly promote the proliferation, survival and drug resistance of MM cells. Therefore, understanding the tumor biological characteristics of MM and exploring the prognosis and immune biomarkers of MM are crucial for improving MM patient outcomes. Immune biomarkers, such as neutrophil/lymphocyte ratio (NLR), lymphocyte/monocyte ratio (LMR), and platelet/monocyte ratio (PLR), are of great significance for risk stratification and evaluati

[Nouveaux traitements du myélome multiple].

NEW THERAPIES IN MULTIPLE MYELOMA Multiple myeloma is the second most common hemopathy in Western countries. The development of the therapeutic arsenal has significantly improved patients' prognosis. In the early 2000s, autograft, the addition of immunomodulatory drugs and proteasome inhibitors have undoubtedly changed the face of the disease. In the last decade, a new chapter has opened, targeted therapies led by anti-CD38 and more recently the introduction of CAR T-cells and bispecific antibodies. Because of the survival improvement, quality of life is now a major concern as well as personal

B-cell acute lymphoblastic leukaemia: recent discoveries in molecular pathology, their prognostic significance, and a review of the current classification.

Acute lymphoblastic leukaemia (ALL) remains a leading cause of non-traumatic death in children, and the majority of adults diagnosed will succumb to the disease. Recent advances in molecular biology and bioinformatics have enabled more detailed genomic analysis and a better understanding of the molecular biology of ALL. A number of recurrent genomic drivers have recently been described, which not only aid in diagnosis and prognostication, but also may offer opportunities for specific therapeutic targeting. The present review summarises B-ALL genomic pathology at diagnosis, including lesions de

Trisomy 8-positive Polycythemia Vera Complicated with Intestinal Behçet's-like Disease: A New Perspective for a Clinical Approach.

Behçet's disease (BD) is a multisystem inflammatory disease of unknown origin. It rarely but occasionally occurs together with myelodysplastic syndrome and primary myelofibrosis. Trisomy 8 is one of the most common cytogenetic abnormalities in myeloid neoplasms; however, the association of BD with polycythemia vera (PV) and trisomy 8 has not been reported. A 70-year-old woman, diagnosed with PV and treated with hydroxyurea, had bloody stool due to multiple ulcers in the ileocecal region. Considering the lack of a response to treatment and other features, we suspected complication with intestin

Genetics of Equine Reproductive Diseases.

Reproductive disorders are genetically heterogeneous and complex; available genetic tests are limited to chromosome analysis and 1 susceptibility gene. Cytogenetic analysis should be the first test to confirm or rule out chromosomal aberrations. No causative genes/mutations are known. The only available genetic test for stallion subfertility is based on a susceptibility gene FKBP6. The ongoing progress in equine genomics will improve the status of genetic testing. However, because subfertile phenotypes do not facilitate collection of large numbers of samples or pedigrees, and clinical causes o

Acquired Aplastic Anemia as a Clonal Disorder of Hematopoietic Stem Cells.

Aplastic anemia is rare disorder presenting with bone marrow failure syndrome due to autoimmune destruction of early hematopoietic stem cells (HSCs) and stem cell progenitors. Recent advances in newer genomic sequencing and other molecular techniques have contributed to a better understanding of the pathogenesis of aplastic anemia with respect to the inflammaging, somatic mutations, cytogenetic abnormalities and defective telomerase functions of HSCs. These have been summarized in this review and may be helpful in differentiating aplastic anemia from hypocellular myelodysplastic syndrome. Furt

Tetraploidy acute myeloid leukaemia after chromosome 16 inversion.

Our patient is a 36-year-old man referred by his general physician to the Department of Hematology because of mild neutropenia in a routine analysis at work. There was no history of previous diseases, and examination was normal. Blood investigations confirmed the neutropenia and showed elongation of prothrombin time. A bone marrow examination was performed revealing about 10% of myeloblasts on the aspirate smears. A cytogenetic study showed chromosome 16 inversion in all of these cells and tetraploidy only in some of them, which were extremely large in size. According to the revised WHO classi

[A case of 63,X/64,XX mosaicism in a subfertile pony mare].

The present case report describes a 6-year old subfertile pony mare, which became pregnant after the eleventh artificial insemination. The examination of the ovaries and the uterus did not reveal any abnormal clinical findings and the mare showed a regular oestrous cycle. Based on cytogenetic and molecular genetic analyses it became possible to elucidate the observed subfertility. The mosaic karyotype of the mare consisted of 63,X (20%) and 64,XX (80%) cells. A PCR analysis failed to amplify sequences from the equine SRY gene. The observed classic 63,X/64,XX mosaicism is a plausible explanatio
