Aerobic bacterial flora of biotic and abiotic compartments of a hyperendemic Zoonotic Cutaneous Leishmaniasis (ZCL) focus.

BACKGROUND: Identification of the microflora of the sand fly gut and the environmental distribution of these bacteria are important components for paratransgenic control of Leishmania transmission by sand flies. METHODS: Biotic and abiotic bacterial communities of four compartments of a hyper-endemic focus of Zoonotic Cutaneous Leishmaniasis (ZCL) were investigated using 16S ribosomal DNA sequencing and phylogenetic tree construction. These compartments include Phlebotomus papatasi's gut, skin and intestinal tract of great gerbil Rhombomys opimus, the gerbil nest supplies, and plant food sour

Mitochondrial genome evolution in species belonging to the Phialocephala fortinii s.l. - Acephala applanata species complex.

BACKGROUND: Mitochondrial (mt) markers are successfully applied in evolutionary biology and systematics because mt genomes often evolve faster than the nuclear genomes. In addition, they allow robust phylogenetic analysis based on conserved proteins of the oxidative phosphorylation system. In the present study we sequenced and annotated the complete mt genome of P. subalpina, a member of the Phialocephala fortinii s.l. - Acephala applanata species complex (PAC). PAC belongs to the Helotiales, which is one of the most diverse groups of ascomycetes including more than 2,000 species. The gene ord

Proteomic analysis of mouse thymoma EL4 cells treated with bis(tri-n-butyltin)oxide (TBTO).

Here, we report the results of proteomic analysis of the mouse thymoma EL4 cell line exposed to bis(tri-n-butylin)oxide (TBTO), an immunotoxic organotin compound. The objective of the work was to examine whether TBTO affects the expression of proteins in this cell line and to compare the differentially expressed proteins with the corresponding mRNA expression data. The identified proteins were quantified using a label-free quantitative method based on counting the observed peptides as an index of protein abundance. The calculation of the ratio of peptides obtained from exposed and control samp

Trichinella zimbabwensis n.sp. (Nematoda), a new non-encapsulated species from crocodiles (Crocodylus niloticus) in Zimbabwe also infecting mammals.

Since 1995, Trichinella larvae have been detected in 39.5% of farmed crocodiles (Crocodylus niloticus) in Zimbabwe. Morphological, biological, biochemical and molecular studies carried out on one isolate from a farmed crocodile in 2001 support the conclusion that this parasite belongs to a new species, which has been named Trichinella zimbabwensis n.sp. This species, whose larvae are non-encapsulated in host muscles, infects both reptiles and mammals. The morphology of adults and larvae is similar to that of Trichinella papuae. Adults of T. zimbabwensis cross in both directions with adults of

A simplified procedure for the subtractive cDNA cloning of photoassimilate-responding genes: isolation of cDNAs encoding a new class of pathogenesis-related proteins.

Transgenic tobacco plants (ppa-1) constitutively expressing Escherichia coli pyrophosphatase behind the 35S CaMV promoter accumulate high levels of soluble sugars in their leaves [27]. These plants were considered a tool to study adaptation of leaves to photoassimilate accumulation at the molecular level. By differential hybridization of a subtractive library enriched for transcripts present in the transgenic plants 12 different cDNAs were isolated. By sequence analysis four cDNAs could be identified as 1-aminocyclopropane-1-carboxylate-oxidase and as three different pathogenesis-related prote

Short-term clinical outcomes of nasoduodenal tube versus feeding jejunostomy after minimally invasive esophagectomy: a retrospective cohort study with exploratory microbiome analysis.

BACKGROUND: Esophagectomy remains an effective treatment for esophageal cancer but is associated with severe postoperative complications, in which nutritional status plays a critical role. The optimal route for enteral feeding remains uncertain. To address this uncertainty, we compared the short-term clinical outcomes of the two approaches and exploratorily analyzed the differences in gut microbiota. METHODS: This retrospective cohort study enrolled patients who underwent minimally invasive esophagectomy (MIE) for esophageal squamous cell carcinoma (ESCC) between July 2018 and June 2021. Prim

Renal tissue microbiota and metabolite profiling reveal dysregulated signatures in diabetic kidney disease.

INTRODUCTION: Diabetic kidney disease (DKD) has become the main cause of end-stage renal disease in China. Mounting evidence links microecological disorders to DKD progression. However, the composition and the functional characteristics of the renal microecology in DKD patients remain poorly defined. This study characterized renal tissue microbiota and metabolite profiles in DKD patients, aiming to provide insights for novel diagnostic strategies for DKD. METHODS: Renal tissue microbiome was analyzed in 42 DKD patients and 10 controls via 16S ribosomal DNA sequencing. Renal metabolomics was p

MRPL35 Suppresses Autophagy-Mediated Ferroptosis in Hepatocellular Carcinoma via the AMPK/mTOR Axis.

INTRODUCTION: Ferroptosis and autophagy are recently identified forms of non-apoptotic regulated cell death with strong therapeutic implications for hepatocellular carcinoma (HCC). Mitochondrial ribosomal protein L35 (MRPL35), a mitochondria- specific component of mitoribosomes, has been proposed as a potential therapeutic target for liver cancer. The study focused on autophagy-dependent ferroptosis in HCC, with an emphasis on the functional significance of MRPL35. METHODS: Bioinformatics approaches, RT-qPCR, along with western blotting, were used to determine MRPL35 expression in HCC. CCK-8,

Genome-Guided TaqMan qPCR for Detection of Xanthomonas albilineans and Quantitative Interpretation of Pathogen Abundance in Sugarcane Leaf Scald.

Sugarcane leaf scald, caused by Xanthomonas albilineans, is difficult to diagnose because latent infections often precede symptom development, allowing infected planting material to disseminate the pathogen unnoticed. Although quantitative PCR (qPCR) has substantially improved pathogen detection, most available assays target conserved ribosomal regions, which can limit species-level discrimination and offer little guidance for interpreting pathogen abundance. Comparative genomic analysis identified Xal_000736 as a candidate species-specific target for assay development. A TaqMan qPCR assay tar

Clinical utility of 16S ribosomal ribonucleic acid gene polymerase chain reaction versus tissue culture in shoulder periprosthetic joint infections.

BACKGROUND: Diagnosing shoulder periprosthetic joint infection remains challenging, particularly in the setting of indolent organisms. Broad-range 16S ribosomal ribonucleic acid (RNA) gene polymerase chain reaction (PCR) with sequencing has been proposed as an adjunct to culture; however, its diagnostic performance in revision shoulder arthroplasty using periprosthetic tissue remains incompletely defined. METHODS: Single center retrospective study of revision shoulder arthroplasty cases in which both intraoperative periprosthetic tissue cultures and broad-range 16S rRNA gene PCR with sequenci

Color polymorphism, taxonomic confusion and cryptic diversity in the kukri snake Oligodon albocinctus (Cantor, 1839) (Squamata: Colubridae).

The kukri snake Oligodon albocinctus (Cantor, 1839) is a widespread element of the Indo-Himalayan and Indo-Burmese snake fauna with two distinct color patterns. The first and more typical morphotype has light bands with black edges, while individuals of the second morphotype have brown blotches or narrow crossbars. The polychromatic nature of this species has led to considerable taxonomic confusion between O. albocinctus and members of Oligodon occurring in sympatry or parapatry. In this study, we investigate patterns of genetic and morphological variation in O. albocinctus and explore whether

Off-grid field-deployable molecular diagnostic platform for malaria surveillance.

BACKGROUND: Malaria, a major global health concern, continues to cause substantial morbidity and mortality, particularly in tropical regions. Traditional malaria diagnostic methods such as microscopy and quantitative polymerase chain reaction (qPCR) are effective but face challenges in field settings because of their requirement for laboratories with specialized equipment and trained personnel. This study presents the development and validation of a portable, cost-effective, field-deployable real-time qPCR platform for detecting Plasmodium species. METHODS: Field-compatible DNA isolation was

Morphological and molecular characterization of Sarcocystis cooperii n. sp. and related Sarcocystis species in three Colombian avian hosts.

BACKGROUND AND AIM: Apicomplexan parasites of the genus Sarcocystis exhibit complex two-host life cycles involving birds as both intermediate hosts (IH) and definitive hosts (DH). Despite Colombia's exceptional avian diversity, knowledge of Sarcocystis species in its wild birds remains limited. This study aimed to characterize the morphology and genetic identity of Sarcocystis species infecting three Colombian birds, the Great Kiskadee (Pitangus sulphuratus), American black vulture (Coragyps atratus), and Andean condor (Vultur gryphus). MATERIALS AND METHODS: Muscle samples from the three spe

A cross-sectional analysis identifies a low prevalence of Plasmodium ovale species  infections in symptomatic and asymptomatic individuals in Kilifi county, Kenya.

BACKGROUND: The focus on P. falciparum diagnosis has led to an underestimation of the global burden of malaria resulting from neglected Plasmodium species. However, there is still scarce data on the prevalence of P. ovale species (spp) globally. To address this knowledge gap, data collected from cross-sectional studies in Kilifi county were used to: 1) determine the prevalence of P. ovale spp infections; and 2) determine the sensitivity of different diagnostic assays in detecting P. ovale spp infections. METHODS: A total of 531 individuals were sampled across three study sites in Kilifi Count

Characterization of the intestinal microbiota in MSM with HIV infection.

BACKGROUND: HIV-infected persons demonstrate notable disturbances in their intestinal microbiota; however, the impact of intestinal microbiota on HIV susceptibility in men who have sex with men (MSM), as well as the effects of HIV and antiretroviral therapy (ART) on their gut microbiota, remains under active study. Thus, our research focuses on clarifying the distinctions in intestinal microbiota composition among uninfected MSM and non-MSM healthy controls, investigating the alterations in early-stage intestinal microbial communities following HIV infection, and assessing how ART affects the

REVISION OF SANGUINICOLA PLEHN, 1905 WITH REDESCRIPTION OF SANGUINICOLA VOLGENSIS (RAŠÍN, 1929) MCINTOSH, 1934, DESCRIPTION OF A NEW SPECIES, PROPOSAL OF A NEW GENUS, AND PHYLOGENETIC ANALYSIS.

Sanguinicola Plehn, 1905 comprises 26 species that collectively infect fishes from 8 orders (Cypriniformes, Characiformes, Siluriformes, Esociformes, Salmoniformes, Labriformes, Centrarchiformes, and Perciformes). Its revision is warranted because several species assigned to the genus could represent new genera, nucleotide sequences are wanting, many species have incomplete descriptions, and types for most species are missing or of poor quality. Herein, we emend Sanguinicola based on morphology and the first nucleotide-based phylogenetic analysis that includes multiple sequences from morpholog
