Integrative analysis of m6A-related genetic variants and single-cell RNA sequencing identifies HLA-DQA1/HLA-DQB1-associated immune signatures in CAR-T-treated diffuse large B-cell lymphoma.

Diffuse large B-cell lymphoma (DLBCL) shows heterogeneous responses to CAR-T therapy, but the links among m6A-related genetic variation, immune-cell states, and treatment response remain incompletely defined. We integrated FinnGen GWAS summary statistics, RMDisease V2.0 m6A-SNP annotations, eQTL annotation, and single-cell RNA-seq data from CAR-T-treated refractory B-cell lymphoma patients. Eleven DLBCL-associated m6A-SNPs were identified using a suggestive association threshold, and nine had eQTL signals in the HaploReg-based annotation. After distinguishing m6A annotation genes from eQTL tar

SynFrag: Synthetic Accessibility Predictor Based on Fragment Assembly Generation in Drug Discovery.

AI-driven molecular generation encounters a "generation-synthesis gap": most computationally designed molecules cannot be synthesized in laboratories, limiting AI-assisted drug design (AIDD) applications. Current approaches to assess synthetic accessibility (SA) include computer-aided synthesis planning (CASP) tools that perform retrosynthetic searches and machine learning-based SA prediction models that provide rapid scoring. CASP tools are computationally expensive for high-throughput screening, while existing SA prediction models may lack chemical synthesis logic or exhibit variable perform

Machine learning-based prediction of paracetamol solubility and CO₂ density in supercritical systems using artificial rabbits optimization.

The accurate prediction of solubility and solvent properties in supercritical CO₂ systems remains a critical challenge in pharmaceutical process design due to the nonlinear and coupled effects of temperature and pressure. This study proposes a novel artificial intelligence-based modeling framework for predicting solvent density and paracetamol mole fraction under supercritical conditions using temperature and pressure as input variables. Three regression models-Multilayer Perceptron (MLP), Support Vector Regression (SVR), and Tweedie Regression (TDR)-were developed and systematically optimized

Early warning signs: Applications of in vivo NMR spectroscopy to understand the impacts of industrial effluents.

Industrial effluents represent one of the most significant threats to water quality. To prevent any serious and sometimes irreversible damage to freshwater ecosystems, an improved understanding of how effluents impact aquatic environments is necessary. Environmental metabolomics is an ideal approach as it enables the identification of the impacts of a stressor on a model organism at the molecular level. In vivo 2D NMR is a powerful tool to understand responses to environmental stress directly in living organisms. Here, a framework to best obtain, process, and interpret in vivo NMR data is intr

Neuromuscular electrical stimulation improves stability and function in chronic ankle instability: a systematic review and meta-analysis.

BACKGROUND: Chronic ankle instability (CAI) commonly develops following lateral ankle sprains and is associated with recurrent sprains, persistent pain, and neuromuscular deficits. While neuromuscular electrical stimulation (NMES) has shown promising results in enhancing muscle strength and neuromuscular control, its specific effects on CAI remain insufficiently investigated. METHOD: A comprehensive search of electronic databases was conducted to identify randomized controlled trials (RCTs) evaluating NMES interventions in individuals with CAI. Studies were included if they reported outcomes r

ScGraphTrans: Pathway-Guided Graph Learning and Domain Adaptation for Cell Type Annotation in Single-Cell RNA-seq.

The tumor microenvironment (TME) is a complex ecosystem in which intercellular communication regulates tumor progression and therapeutic response. Yet inferring cell-cell interactions from non-spatial scRNA-seq remains challenging due to incomplete ligand-receptor databases and inaccurate cell type annotations. Here, we propose scGraphTrans, a graph neural network framework that integrates functional state pseudo-labels, graph structure learning, and graph domain adaptation to improve both cell type annotation and communication inference. Pathway activity scores across 14 cancer-relevant proce

Circulating tumor DNA in gastrointestinal cancers: promise, pitfalls, and the path forward.

Circulating tumor DNA (ctDNA) has emerged as a powerful prognostic biomarker in gastrointestinal (GI) oncology, with the strongest evidence in colorectal cancer. Postoperative ctDNA positivity identifies patients at high risk of recurrence, and serial clearance patterns further refine risk. However, prognostic validity has not consistently translated into treatment-selection utility. In stage II colon cancer, DYNAMIC supported chemotherapy de-escalation without compromising recurrence-free survival, whereas DYNAMIC-III showed that escalation in ctDNA-positive stage III disease did not improve

Impact of cryopreservation on immune cell subset frequencies in human peripheral blood mononuclear cells following assessment of monoclonal antibodies to CD39 and P2X7.

Peripheral blood mononuclear cells (PBMCs) are commonly cryopreserved for immune cell profiling analyses, like flow cytometry. Cryopreservation of PBMCs may affect the recovery of immune cell subsets or the presence of cell-surface molecules, potentially skewing results compared to freshly isolated PBMCs. The ectonucleotidase CD39 promotes immunosuppression through the hydrolysis of extracellular nucleotides such as adenosine 5'-triphosphate (ATP) to limit P2X7 activation and downstream immune responses. Because of this, CD39 and P2X7 are emerging as potential biomarkers in cancer and other di
