Possible contribution of dopaminergic receptors in the anteroventral third ventricular region to hyperosmolality-induced vasopressin secretion in conscious rats.

We have reported previously that regions encompassing the cerebral ventricle may contain dopamine receptors responsible for facilitatory roles in the osmotic release of vasopressin in conscious rats. In order to explore the location of these receptors, we injected (0.5 mul) the dopamine antagonist haloperidol (13.3 nmol) or dopamine (26.4 nmol) topically into the anteroventral third ventricular region or the paraventricular nucleus of rats, and their effects on the levels of plasma vasopressin and its controlling factors were examined in the presence or absence of an osmotic stimulus. The effe

Release to the cerebral ventricles of substances with possible transmitter function in the caudate nucleus.

1. One caudate nucleus of the anaesthetized cat was superfused by perfusing the anterior horn of one lateral cerebral ventricle. The perfusates were examined for their content in acetylcholine (ACh), dopamine, homovanillic acid (HVA) and 5-hydroxytryptamine (5-HT), at rest and after a variety of stimuli.2. When prostigmine was added to the perfusion fluid, ACh appeared in the effluent; its concentration tended to rise in the course of an experiment. Various afferent stimuli, all of which caused evoked responses recorded from the contra-lateral caudate nucleus, increased the ACh content of the

Reward Deficiency Syndrome (RDS): A Common Neurogenetic Trait/State of All Addictions: Is this the new DSM?

Steven Hyman, former director of the National Institute of Mental Health (2012), argued that neuroscience research in psychiatry frequently inherits the DSM's assumption that disorders are discrete entities, even though empirical boundaries between conditions are often porous. In response, Hyman and colleagues advanced the Research Domain Criteria (RDoC), which organizes psychopathology around core neurobiological domains that cut across diagnoses. In a conceptually similar direction, Blum (1995) introduced Reward Deficiency Syndrome (RDS) as a transdiagnostic construct intended to unify subst

Neurochemical effects of chronic dietary and repeated high-level acute exposure to chlorpyrifos in rats.

Very little is known about the effects of chronic exposure to relatively low levels of anticholinesterase insecticides or how the effects of chronic exposure compare to those of higher, intermittent exposure. To that end, adult male rats were fed an anticholinesterase insecticide, chlorpyrifos (CPF), for 1 year at three levels of dietary exposure: 0, 1, or 5 mg/kg/day (0+oil, 1+oil, and 5+oil). In addition, half of each of these groups also received a bolus dosage of CPF in corn oil ("spiked" animals; 60 mg/kg initially and 45 mg/kg thereafter) every 2 months (0+CPF, 1+CPF, 5+CPF). Animals wer

[Efficacy of atypical antipsychotics in depressive syndromes].

Depression is a frequent symptom in psychiatry, either isolated (major depression) or entangled with other psychiatric symptoms (psychotic depression, depression of bipolar disorders). Many antidepressant drugs are available with different pharmacological profiles from different classes: tricyclic antidepressants, monoamine oxydase inhibitors, selective serotonin reuptake inhibitors (SSRI). However, there are some limitations with these drugs because there is a long delay before relief for symptoms, some patients with major depression are resistant to treatment, there is a risk to induce manic

Hallucinations, Delusions and Impulse Control Disorders in Parkinson Disease.

Psychotic and compulsive symptoms in Parkinson disease are highly prevalent and associated with poor outcomes and greater caregiver burden. When acute, delirium should be ruled out or treated accordingly. When chronic, comorbid systemic illnesses, dementia, and psychiatric disorders should be considered. Reduction and discontinuation of anticholinergics, amantadine, dopamine agonists, and levodopa as tolerated, as well as adjunctive clozapine or quetiapine are frequently effective to manage Parkinson disease psychosis. Pimavanserin appears effective but is not widely available, and more experi

Efficacy of Octolig chromatography as a means of removal of aqueous antibiotics given to premature babies.

Reliable information indicated that a half-million prematurely born babies are routinely given antibiotics without evidence of infection. This study is based on a concern that such antibiotics might go into hospital waste systems and lead to antibiotic-resistant bacteria. We tested the removal of selected antibiotics by Octolig, a commercially available supported chelating agent using column chromatography. Five (Caffeine, Ampicillin, Furosemide, Dopamine, and Vancomycin) were selected for study. Ampicillin and Furosemide were quantitatively removed by column chromatography. Dopamine, Caffeine

Psychosis and concurrent impulse control disorder in Parkinson's disease: A review based on a case report.

Psychosis, impulse control disorders (e.g., pathological gambling and hypersexuality) and repetitive behaviors such as punding are known psychiatric complications of Parkinson's disease (PD). Impulsive, compulsive and repetitive behaviors are strongly associated with dopamine-replacement therapy. We present the case of a 58-year-old man with PD and a myriad of psychiatric symptoms. Concurrent psychosis, punding and pathological gambling developed more than six years after the introduction of pramipexole and ceased shortly after the addition of quetiapine and discontinuation of pramipexole. Impulsive and compulsive behaviors in Parkinson's disease.

Antiparkinson therapy can be the primary cause of a range of nonmotor symptoms that include a set of complex disinhibitory psychomotor pathologies and are linked by their repetitive, reward or incentive-based natures. These behaviors relate to aberrant or excessive dopamine receptor stimulation and encompass impulse control disorders (ICDs), punding, and the dopamine dysregulation syndrome (DDS). Common ICDs include pathological gambling, hypersexuality, compulsive eating, and compulsive buying. This review focuses on the phenomenology, epidemiology, and methods to identify and rate these diso

Serotonergic decompensation in the akinetic expression of Parkinson's disease.

The current diagnostic and treatment paradigm for the akinetic form of Parkinson's disease asserts that the majority of symptoms and treatment phenomena arise from a dysfunctional dopaminergic system. Recent studies have attempted to determine the roles of other biogenic amine neurotransmitters such as serotonin and norepinephrine. Metabolic breakdown product studies of Parkinsonian and non-Parkinsonian cerebrospinal fluid (CSF) samples indicate significant differences in the levels of serotonin and norepinephrine in addition to the levels of dopamine. Such changes may suggest that current pha

Reversal of phencyclidine effects by a group II metabotropic glutamate receptor agonist in rats.

Glutamatergic abnormalities have been associated with several psychiatric disorders, including schizophrenia and addiction. Group II metabotropic glutamate receptors were targeted to normalize glutamatergic disruptions associated with an animal model of schizophrenia, the phencyclidine model. An agonist of this group of receptors, at a dose that was without effects on spontaneous activity and corticolimbic dopamine neurotransmission, attenuated the disruptive effects of phencyclidine on working memory, stereotypy, locomotion, and cortical glutamate efflux. This behavioral reversal occurred in

The unique efficacy of clozapine is attributable to muscarinic receptor agonism.

Clozapine is superior to all other antipsychotic drugs for treatment-resistant schizophrenia, but the molecular underpinnings for this have remained elusive for decades. Recently, we proposed a cholinergic mechanism, based on the distinct pharmacology of clozapine in the parasympathetic branch of the autonomic nervous system, a hypothesis strengthened by the translation of a new cholinergic-based antipsychotic drug into clinical practice. A recent article has challenged the cholinergic hypothesis and instead proposed a mechanism involving the action of clozapine at noradrenaline receptors of t

Indirect spectrophotometric micro-determination of catecholamine drugs in dosage forms using safranin O dye and N-bromosuccinimide.

OBJECTIVES: The present work aims to develop a simple, sensitive, and fast spectrophotometric method for the determination catecholamine drugs namely, adrenaline, methyldopa, and dopamine in pure and dosage forms. METHOD: The proposed method is based on the oxidation of the drug compounds by an excess amount of the oxidizing agent N-bromosuccinimide, the remainder of which bleaches the safranin O dye which is measured at 535nm. RESULTS: The estimation limits were 0.1-16, 0.5-9, and 0.5-18μg/mL, with molar absorptivity values of 2.84×104, 5.32×104, and 3.06×104L/mol.cm with good accuracy and

Evaluating pimavanserin tartrate as a treatment in Parkinson's disease.

INTRODUCTION: Pimavanserin is an efficacious, atypical neuroleptic. It ameliorates the frequency and severity of hallucinations and delusions in patients with Parkinson's disease with psychosis. Most antipsychotic drugs directly block dopamine receptors and thus worsen motor behavior. In contrast, pimavanserin reduces the activity of excitatory serotonin receptor subtypes. They stimulate dopaminergic pathways in the mesolimbic system. As a result, onset of psychosis may occur particularly in patients on a chronic dopamine-substituting treatment regimen, like in Parkinson's disease. AREAS COVE

A hypothalamus-habenula circuit regulates psychomotor responses induced by cocaine.

Administration of cocaine increases synaptic dopamine levels by blocking dopamine reuptake and leads to increased locomotor activity and compulsive drug-seeking behaviour. It has been suggested that the lateral hypothalamus (LH) or lateral habenula (LHb) is involved in drug-seeking behaviours. To explore the role of the LH and the LHb in cocaine-induced psychomotor responses, we tested whether modulation of the LH or the LH-LHb circuit affects cocaine-induced locomotion. Cocaine-induced locomotor activity and dopamine release were suppressed by the activation of the LH with 2-[2,6-difluoro-4-[

Neuroinflammatory Response in Reward-Associated Psychostimulants and Opioids: A Review.

Substance abuse is one of the significant problems in social and public health worldwide. Vast numbers of evidence illustrate that motivational and reinforcing impacts of addictive drugs are primarily attributed to their ability to change dopamine signaling in the reward circuit. However, the roles of classic neurotransmitters, especially dopamine and neuromodulators, monoamines, and neuropeptides, in reinforcing characteristics of abused drugs have been extensively investigated. It has recently been revealed that central immune signaling includes cascades of chemokines and proinflammatory cyt

Tardive Dyskinesia in Older Persons Taking Antipsychotics.

Tardive dyskinesia (TD) is a hyperkinetic movement disorder caused by the use of dopamine receptor-blocking agents (DRBAs), a category of medications that includes first- and second-generation antipsychotics (APs) and agents such as metoclopramide that are used for the treatment of nausea and gastrointestinal dysmotility. While TD can affect people of all ages, older age is associated with increased risk of TD and also with the emergence of TD occurring after shorter treatment durations and lower dosages of DRBAs. TD is characterized by involuntary movements that include the face, limbs, and t

Management of giant prolactinoma causing craniocervical instability: illustrative case.

BACKGROUND: Giant prolactinomas (>4 cm) are a rare entity, constituting less than 1% of all pituitary tumors. Diagnosis can usually be achieved through endocrinological analysis, but biopsy may be considered when trying to differentiate between invasive nonfunctioning pituitary adenomas and primary clival tumors such as chordomas. OBSERVATIONS: The authors presented a rare case of a giant prolactinoma causing significant clival and occipital condyle erosion, which led to craniocervical instability. They provided a review of the multimodal management. Management involved medical therapy with d

The neurobiology of impulse control disorders in Parkinson's disease: from neurotransmitters to neural networks.

Impulse control disorders (ICD) are common neuropsychiatric disorders that can arise in Parkinson's disease (PD) patients after commencing dopamine replacement therapy. Approximately 15% of all patients develop these disorders and many more exhibit subclinical symptoms of impulsivity. ICD is thought to develop due to an interaction between the use of dopaminergic medication and an as yet unknown neurobiological vulnerability that either pre-existed before PD onset (possibly genetic) or is associated with neural alterations due to the PD pathology. This review discusses genes, neurotransmitters

Antipsychotic-like effect of the muscarinic acetylcholine receptor agonist BuTAC in non-human primates.

Cholinergic, muscarinic receptor agonists exhibit functional dopamine antagonism and muscarinic receptors have been suggested as possible future targets for the treatment of schizophrenia and drug abuse. The muscarinic ligand (5R,6R)-6-(3-butylthio-1,2,5-thiadiazol-4-yl)-1-azabicyclo[3.2.1]octane (BuTAC) exhibits high affinity for muscarinic receptors with no or substantially less affinity for a large number of other receptors and binding sites, including the dopamine receptors and the dopamine transporter. In the present study, we wanted to examine the possible antipsychotic-like effects of B
