Spirometra mansoni is a zoonotic tapeworm with a global distribution; however, its population-level genetic structure and phylogeographic patterns remain poorly understood. Robust phylogeographic inference is essential to understand diversification and the geographical structuring of its mitochondrial diversity. We analysed three datasets of newly generated and publicly available mitochondrial cox1 sequences differing in length: a short-fragment alignment (303 bp; n = 319), a full-length dataset (1556 bp; n = 101) and a length-matched subset extracted from the full-length sequences (303 bp; n

BACKGROUND: Elevated intracranial pressure (ICP) is a common postnatal complication in premature infants, particularly those with very low birth weight, and it is associated with hemodynamic impairments. Continuous monitoring of cerebral blood flow (CBF) and oxygenation may enable early detection and inform clinical management. We hypothesized that non-invasive, bedside optical spectroscopy measurements of CBF and oxygenation are sensitive to abrupt increases in ICP.
METHODS: A hybrid optical system combining broadband near-infrared spectroscopy (bNIRS) and diffuse correlation spectroscopy (D

Direct visualization of HIV-1 nuclear import through the nuclear pore complex (NPC) presents a technical challenge due to the rarity of this process. To enable systematic investigation, we developed a robust in situ system that mimics HIV-1 nuclear import in a near-native context using isolated HIV-1 virus like particles (VLP) cores and permeabilized CD4 + T lymphocyte (CEM) cells. This approach supports docking and translocation of abundant viral cores through nuclear pores into the nucleus. For high-resolution visualization, we implemented an integrated correlative approach to guide cryo-foc

Chilling stress induces photosystem I (PSI) photoinhibition in various plants, severely impairing their growth. However, the mechanisms suppressing chilling-induced PSI photoinhibition remain unclear. To identify factors preventing PSI photoinhibition, we compared two cucumber cultivars with different susceptibilities to PSI photoinhibition and chilling stress tolerance. Chilling stress caused uncoupling of thylakoid membrane in both cultivars, as previously reported. However, in the chilling-sensitive cultivar, electron efflux from ferredoxin (Fd) was more restricted under chilling stress, re

Doxorubicin (DOX) is a clinically approved chemotherapeutic agent that exerts its cytotoxicity through DNA intercalation and topoisomerase II inhibition. However, its redox-based mechanism remains underexplored. Here, it is revealed that DOX specifically binds to the heme center of cytochrome P450 (CYP450) enzymes via its quinone moiety, enabling electron transfer that catalyzes molecular oxygen into superoxide anions (O2 •-). To enhance this process, mesoporous polydopamine (mPDA) is introduced as an electron-rich scaffold, amplifying electron flow to DOX. Furthermore, L-arginine (L-Arg) is c

The COX4I1 is responsible for encoding a crucial component of cytochrome c oxidase, integral to electron transport in the mitochondrial respiratory chain. Mutations in COX4I1 can result in a rare autosomal recessive disorder characterized by growth retardation, slow weight gain, microcephaly, and potentially, hematologic symptoms such as Fanconi anemia or neurological impairments including developmental regression and severe epilepsy. In this study, we report the first case of COX4I1 deficiency in China, identified in a 6-year-old boy. The patient exhibited developmental regression, epilepsy,

Mutations in the mitochondrial respiratory complex I accessory subunit NADH:ubiquinone oxidoreductase subunit S4 (ndufs4) can cause the mitochondrial disease Leigh syndrome, which may be associated with vision loss. We previously demonstrated that mice with global deletion of ndufs4 exhibited impaired in vivo photoreceptor light responses prior to the early death of the mice around postnatal day 50. However, ex vivo electrophysiology recordings performed on retinas from ndufs4-/- mice were normal, suggesting that the in vivo phenotype may reflect altered homeostasis of the extracellular enviro

Mitochondria play a central role in muscle metabolism and function. A unique family of iron-sulfur proteins, termed CDGSH Iron Sulfur Domain-containing (CISD/NEET) proteins, support mitochondrial function in skeletal muscles. The abundance of these proteins declines during aging leading to muscle degeneration. Although the function of the outer mitochondrial CISD/NEET proteins, CISD1/mitoNEET and CISD2/NAF-1, has been defined in skeletal muscle cells, the role of the inner mitochondrial CISD protein, CISD3/MiNT, is currently unknown. Here, we show that CISD3 deficiency in mice results in muscl

The neotropical fish genus Astyanax (Characidae) and its associated helminths migrated northward from South America following the Great American Biotic Interchange (GABI): ca. 150 Astyanax spp. are found throughout South and Central America, up to the Mexico-USA border. Most characids are distributed south of the Trans-Mexican Volcanic Belt (TMVB), which bisects the country and represents a major transition zone between the neotropical and nearctic realms. Here, we characterize parasites of the monogenean genus Gyrodactylus infecting Astyanax spp. in Mexico: Astyanax aeneus south of the TMBV,

Lipid storage myopathies are considered inborn errors of metabolism affecting the fatty acid metabolism and leading to accumulation of lipid droplets in the cytoplasm of muscle fibers. Specific diagnosis is based on investigation of organic aids in urine, acylcarnitines in blood and genetic testing. An acquired lipid storage myopathy in patients treated with the antidepressant drug sertraline, a serotonin reuptake inhibitor, has recently emerged as a new tentative differential diagnosis. We analyzed the muscle biopsy tissue in a group of 11 adult patients with muscle weakness and lipid storage

Glycolytic and mitochondrial oxidative metabolism, which are two major energy sources in tumors, are potential targets in cancer treatment. Metabolic reprogramming from glycolysis to mitochondrial oxidative metabolism and vice versa is an adaptive strategy with which tumor cells obtain energy to survive and thrive under the compromised conditions of glycolysis and mitochondrial respiration. Developing highly potent, nontoxic, and tumor-selective oxidative phosphorylation (OXPHOS) inhibitors may help advance therapeutic targeting of mitochondrial drugs in cancer. The FDA-approved antimalarial d

Fasciola gigantica is a major pathogen that causes fasciolosis in Africa. A recent study in Uganda demonstrated that Fasciola flukes were present in 65.7% of slaughtered cattle. However, molecular identification of Fasciola species has not yet been performed in the country. In the present study, 292 Fasciola flukes were collected from Kampala and Gulu, Uganda. The samples were identified as F. gigantica using a multiplex polymerase chain reaction (PCR) assay for phosphoenolpyruvate carboxykinase (pepck) and a PCR-restriction fragment length polymorphism (RFLP) assay for DNA polymerase delta (p

Isolated complex I deficiency is the most common cause of pediatric mitochondrial disease. Exome sequencing (ES) has revealed many complex I causative genes. However, there are limitations associated with identifying causative genes by ES analysis. In this study, we performed multiomics analysis to reveal the causal variants. We here report two cases with mitochondrial complex I deficiency. In both cases, ES identified a novel c.580G>A (p.Glu194Lys) variant in NDUFV2. One case additionally harbored c.427C>T (p.Arg143*), but no other variants were observed in the other case. RNA sequencing show

Gaeumannomyces tritici, an ascomycete soilborne fungus, causes a devastating root disease in wheat. Carabrone, a botanical bicyclic sesquiterpenic lactone, is a promising fungicidal agent that can effectively control G. tritici. However, the mechanism of action of carabrone against G. tritici remains largely unclear. Here, we used immunogold for subcellular localization of carabrone and the results showed that carabrone is subcellularly localized in the mitochondria of G. tritici. We then explored the functional analysis of genes GtCytc1 , GtCytb, and GtIsp of the mitochondrial respiratory cha
