Evaluation of islet heme oxygenase-CO and nitric oxide synthase-NO pathways during acute endotoxemia.

We investigated, by a combined in vivo and in vitro approach, the temporal changes of islet nitric oxide synthase (NOS)-derived nitric oxide (NO) and heme oxygenase (HO)-derived carbon monoxide (CO) production in relation to insulin and glucagon secretion during acute endotoxemia induced by lipopolysaccharide (LPS) in mice. Basal plasma glucagon, islet cAMP and cGMP content after in vitro incubation, the insulin response to glucose in vivo and in vitro, and the insulin and glucagon responses to the adenylate cyclase activator forskolin were greatly increased after LPS. Immunoblots demonstrated

Glycogen accumulation in rat pancreatic islets: in vitro experiments.

Under conditions of sustained hyperglycemia, glycogen accumulates in pancreatic islets, but not so in acinar pancreatic cells. Advantage conceivably could be taken from such a situation in the perspective of the noninvasive imaging of the endocrine pancreas. The present experiments aim, therefore, at characterizing the time course for glycogen accumulation in pancreatic islets cultured at a high concentration (30 mM) of D-glucose in the presence of tracer amounts of either D-[U-14C]glucose or 2-deoxy-2-[18F]fluoro-D-glucose. The 14C-labeled glycogen content of the cultured islets increased wit

Mechanisms of reduced and compensatory growth.

Growth is an integrated process, resulting from the response of cells dependent on the endocrine status and nutrient availability. During feed restriction, the production and secretion of growth hormone (GH) by the pituitary gland are enhanced, but the number of GH receptors decreases. Changes of GH binding proteins induce GH resistance and are followed by reduced insulin-like growth factor-I (IGF-I) secretion. On the other hand, high circulating levels of GH enhance the mobilization of fatty acids, which are used to support energy requirements. Thus, when feed restriction in growing animals i

Management of bone metastases.

Metastatic bone disease develops as a result of the many interactions between tumor cells and bone cells. This leads to disruption of normal bone metabolism, with the increased osteoclast activity seen in most, if not all, tumor types providing a rational target for treatment. The clinical course of metastatic bone disease in multiple myeloma, breast and prostate cancers is relatively long, with patients experiencing sequential skeletal complications over a period of several years. These include bone pain, fractures, hypercalcemia, and spinal cord compression, all of which may profoundly impai

[Relationship between corticotropin and arginine vasopressin in endocrine diseases].

VASOPRESSIN: The action of vasopressin (AVP) is not limited to regulating water excretion but also plays an essential role in regulating the corticotropic axis during stress. PHYSIOLOGY: Vasopressin is synthesized in the hypothalamus and stored in the posterior pituitary. It acts on 3 types of receptors (RV). RV1a are vascular receptors mediating the vasoconstrictor and glycogenolytic effects of the hormone. Anterior pituitary RV1b or V3 mediate stimulating effects on the corticotropic axis. Renal RV2 regulate water and urea excretion. Hypothetical extrarenal RV2 would be responsible for the v

Pituitary hyperplasia.

Comprehensive article summarizing more than 25 years of experience with pituitary hyperplasia in surgical material. Morphologic forms of hyperplasia--diffuse and nodular--are defined and, for comparison, the normal morphology, frequency and intraglandular distribution of cell types are briefly reviewed. All cell types can give rise to hyperplasia, although their frequency, extent and clinical importance widely vary. Somatotroph hyperplasia is rare; it is limited to cases of GHRH overproduction by extrapituitary endocrine neoplasms and sporadic examples of gigantism. Prolactin cells display the

Pancreatic agenesis attributable to a single nucleotide deletion in the human IPF1 gene coding sequence.

The homeodomain protein IPF1 (also known as IDX1, STF1 and PDX1; see Methods) is critical for development of the pancreas in mice and is a key factor for the regulation of the insulin gene in the beta-cells of the endocrine pancreas. Targeted disruption of the Ipf1 gene encoding IPF1 in transgenic mice results in a failure of the pancreas to develop (pancreatic agenesis). Here, we report the identification of a single nucleotide deletion within codon 63 of the human IPF1 gene (13q12.1) in a patient with pancreatic agenesis. The patient is homozygous for the point deletion, whereas both parents

Cystic glucagonoma with loss of heterozygosity on chromosome 11 in multiple endocrine neoplasia type 1.

A 52-year-old man with a past history of a pituitary adenoma and hyperparathyroidism due to a parathyroid adenoma was admitted because of a solitary tumour of the pancreas revealed by ultrasonography. His family history was unremarkable. Plasma glucagon levels were slightly elevated (280 ng/l, normal range, 40-180 ng/l) with decreased plasma amino acid levels. Plasma glucagon levels disclosed an exaggerated response during an arginine infusion test and paradoxical elevation during a 75 g oral glucose tolerance test. Endoscopic ultrasonography revealed a monolocular cystic mass of about 3 cm in

Cystic urogenital anomalies in ferrets (Mustela putorius furo).

Single or multiple semispherical to bilobulated fluid-filled cystic structures of variable size were observed on the dorsal aspects of the urinary bladder of four male and two female ferrets (Mustela putorius furo). All ferrets had been neutered. On physical examination, the cysts were palpated as caudal abdominal masses. Three of the six ferrets presented with dysuria, and two ferrets had signs compatible with endocrine dysfunction. Adrenal cortical hyperplasia or neoplasia were observed in all of the five ferrets examined. Sex hormones assayed in one of the six ferrets revealed elevated leve

Isolation and characterization of a chicken homolog of the c-ret proto-oncogene.

The c-ret proto-oncogene encodes a receptor tyrosine kinase that plays important roles in human disease and in normal mammalian development. Mutations in the human RET gene are associated with multiple endocrine neoplasia syndromes and Hirschsprung's disease in humans, while targeted mutagenesis of murine c-ret resulted in severe developmental abnormalities affecting the excretory and peripheral nervous systems. To examine the evolutionary conservation of the ret protein sequence and its developmental expression pattern, we isolated and sequenced cDNA clones of chicken c-ret and examined its e

Patterns of innervation of vasoactive intestinal polypeptide, neuropeptide Y, and gastrin-releasing peptide immunoreactive nerves in the feline pancreas.

In this study, we performed a detailed analysis of the immunoreactive (IR) patterns and tissue distribution of vasoactive intestinal polypeptide (VIP), neuropeptide Y (NPY), and gastrin-releasing peptide (GRP) in the feline pancreas by means of immunohistochemical and radioimmunological techniques. Immunoreactivity for each peptide is localized to varicose nerve fibers distributed throughout the exocrine and endocrine pancreas, with some differences in the density and pattern of fiber distribution. In the acinar and stromal compartments, VIP-IR processes have a higher density than NPY- and GRP

The role of the growth plate in longitudinal bone growth.

The epiphyseal growth plate is the main site of longitudinal growth of the long bones. At this site, cartilage is formed by the proliferation and hypertrophy of cells and synthesis of the typical extracellular matrix. The formed cartilage is then calcified, degraded, and replaced by osseous tissue. Proliferation and differentiation of cartilage cells (i.e., chondrocytes) as studied mostly in culture, is regulated by various endocrine, paracrine, and autocrine agents such as growth hormone, insulin-like growth factor-I (IGF-I), transforming growth factor (TGE-beta), and vitamin D metabolites (1

Effect of gastrin-releasing peptide on the secretion of mouse islet hormones in vitro.

Collagenase-isolated mouse islets were incubated with gastrin-releasing peptide (GRP). At 5.6 mmol glucose/l. 10 nmol GRP/l increased the release of insulin (by 50%) and glucagon (by twofold), decreased the release of pancreatic polypeptide (by 35%), but did not significantly affect the release of somatostatin. At 16.7 mmol glucose/l, 10 nmol GRP/l increased glucagon release (by fivefold) and decreased pancreatic polypeptide release (by 46%), without significantly altering insulin and somatostatin release. GRP (200 nmol/l) did not affect insulin release by perifused mouse islets at 2.8 mmol gl

Interleukin-1 suppresses follicle-stimulating hormone-induced estradiol secretion from cultured ovarian granulosa cells.

The effect of recombinant human interleukin-1 beta (IL-1 beta) on the follicle stimulating hormone-(FSH) induced secretion of estradiol was investigated using cultured granulosa cells obtained from immature rats with diethylstilbestrol implants. Estradiol secretion was significantly reduced by IL-1 beta in cultures containing FSH and either 10(-7) or 10(-8) M androstenedione as a substrate for estradiol synthesis. However, the inhibition of FSH-induced estradiol secretion by IL-1 beta was more apparent in the presence of 10(-8) M as compared to 10(-7) M androstenedione. IL-1 beta suppressed es

Milk signalling in the pathogenesis of type 2 diabetes.

The presented hypothesis identifies milk consumption as an environmental risk factor of Western diet promoting type 2 diabetes (T2D). Milk, commonly regarded as a valuable nutrient, exerts important endocrine functions as an insulinotropic, anabolic and mitogenic signalling system supporting neonatal growth and development. The presented hypothesis substantiates milk's physiological role as a signalling system for pancreatic β-cell proliferation by milk's ability to increase prolactin-, growth hormone and incretin-signalling. The proposed mechanism of milk-induced postnatal β-cell mass expansi

Endometrial receptivity failure in assisted reproduction: immune-cellular niches, mechanistic evidence, and translational boundaries.

Endometrial receptivity failure and recurrent implantation failure (RIF) are clinically heterogeneous outcomes shaped by embryo competence, uterine anatomy, endocrine support, immune adaptation, and the timing of embryo-endometrial synchrony. This Review focuses on how immune-cellular niches, cell-specific and spatiotemporal mechanisms, disease-modified receptivity states, and evidence-to-translation boundaries should be interpreted in assisted reproduction. Single-cell, spatial, and multi-omics studies increasingly resolve abnormalities in stromal, sepithelial, vascular, perivascular, and imm

3D bioprinted connections in Liver‒Pancreas crosstalk: Lessons learned and future directions.

The complexity of metabolic crosstalk between the liver and pancreas, which controls glucose and homeostasis levels, has been intriguing for models based on conventional in vitro systems. However, the 3D bioprinting path has created an incredible platform of multicellular and spatially concerted microtissues that mimic the interaction of the pancreas. This discovery contributes important insights into metabolic organization, regenerative strategies, and disease mechanisms. Therefore, our review delves into recent advancements in 3D bioprinted crosstalk between liver and pancreas constructs, wi

Triphenyl phosphate exposure at physiologically/environmentally relevant concentrations promotes malignant progression of breast cancer: Insights from metabolomics and lipidomics.

Triphenyl phosphate (TPHP) is one of the main alternatives to brominated flame retardants and a typical environmental endocrine disruptor. Although the in vitro tumor-promoting effect of TPHP has been partially emphasized in previous studies, its impact on breast cancer, particularly at physiological/environmental relevant concentrations, had received inadequate attention. To investigate the pro-breast cancer effects of TPHP, both three-dimensional cancer cell spheroids models and chronic murine exposure models were established. Spheroid growth analysis, bioluminescent imaging, and multi-omics

Hemoglobinopathies and diabetes mellitus: from pathophysiology to clinical practice.

Hemoglobinopathies, primarily β-thalassemia and sickle cell disease (SCD), represent a major global health burden. With extended lifespans, secondary diabetes mellitus (DM) has emerged as a frequent endocrine complication. This review examines the underlying pathophysiology, diagnostic pitfalls, and therapeutic strategies for this distinct population. A narrative review was performed using PubMed database, including English full-text articles published up to July 2026. The pathogenesis of DM in hemoglobinopathies involves a duality of iron-mediated β-cell apoptosis and hemolysis-driven hepatic

The Myokine Landscape: Sources, Signaling, and Context-Dependent Actions.

In addition to its contractile role, skeletal muscle has important secretory functions and communicates with local and distant tissues through myokines, a heterogeneous group of cytokines, chemokines, growth factors, peptides, and extracellular-matrix-associated proteins released by muscle cells. Their production is regulated by contraction, mechanical loading, energy availability, hypoxia, inflammation, injury, and mitochondrial or metabolic stress. Through autocrine, paracrine, and endocrine mechanisms, myokines influence satellite-cell activity, myogenesis, protein turnover, muscle mass, me
