Effects of essential fatty acid deficiency on enterohepatic circulation of bile salts in mice.

Essential fatty acid (EFA) deficiency in mice has been associated with increased bile production, which is mainly determined by the enterohepatic circulation (EHC) of bile salts. To establish the mechanism underlying the increased bile production, we characterized in detail the EHC of bile salts in EFA-deficient mice using stable isotope technique, without interrupting the normal EHC. Farnesoid X receptor (FXR) has been proposed as an important regulator of bile salt synthesis and homeostasis. In Fxr(-/-) mice we additionally investigated to what extent alterations in bile production during EF

Developmental programming: differential effects of prenatal exposure to bisphenol-A or methoxychlor on reproductive function.

Increased occurrence of reproductive disorders has raised concerns regarding the impact of endocrine-disrupting chemicals on reproductive health, especially when such exposure occurs during fetal life. Prenatal testosterone (T) treatment leads to growth retardation, postnatal hypergonadotropism, compromised estradiol-positive feedback, polycystic ovaries, and infertility in the adult. Prenatal dihydrotestosterone treatment failed to affect ovarian morphology or estradiol-positive feedback, suggesting that effects of prenatal T may be facilitated via conversion of T to estradiol, thus raising c

Interactions between leptin and the human sympathetic nervous system.

Results from animal experimentation suggest a 2-way interaction between leptin and the sympathetic nervous system, with leptin causing sympathetic activation and conversely, with the sympathetic system exercising regulatory feedback inhibition over leptin release. We have now tested this hypothesis in humans. In the absence of results from leptin infusions, to test for sympathetic stimulation of leptin release, we sought a quantitative naturalistic linkage of sympathetic activity with leptin plasma concentration across a broad range of leptin values in men of widely differing adiposity. Fetal ethanol exposure alters pituitary-adrenal sensitivity to dexamethasone suppression.

The present study investigated the hypothesis that a deficit in feedback inhibition of the hypothalamic-pituitary-adrenal (HPA) axis may underlie the hormonal hyperresponsiveness seen in fetal ethanol-exposed rats. Male and female Sprague-Dawley rats from prenatal ethanol (E), pair-fed (PF) and ad lib-fed control (C) treatment groups were tested in adulthood. The effects of dexamethasone (DEX) blockade on basal and stress corticosterone (CORT) levels and stress adrenocorticotropin (ACTH) levels were examined over a 36-h period. Stress CORT and ACTH levels after DEX administration at the trough

Large language model-guided high-throughput screening for accelerated discovery of Pt-based high-entropy electrocatalysts.

Pt-based high-entropy alloy (Pt-HEA) catalysts show significant potential for the hydrogen evolution reaction (HER), yet their vast compositional space challenges conventional trial-and-error approaches. Here, we propose a large language model (LLM)-guided closed-loop framework for high-throughput discovery to accelerate the rational design and optimization of Pt-HEA catalysts. The framework first extracts structured knowledge from extensive literature and incorporates retrieval-augmented generation (RAG) to develop an LLM assistant tailored to HEA electrocatalysis research. The assistant tran

Enhancing engineering competencies through Outcome-based Education: A mixed-methods study in an intelligent control course.

Bridging the gap between theory and practice remains a persistent challenge in engineering education, especially in complex, algorithm-heavy courses like intelligent control. In such contexts, students often know the abstract concepts but struggle to turn them into workable engineering solutions. Outcome-Based Education (OBE), which focuses on what students can do, offers a promising way forward. However, detailed evidence on how well OBE works at the individual course level in technically demanding fields is still limited. In this quasi-experimental, mixed-methods study, we redesigned an unde

Modulation of human primary motor cortex plasticity during proactive and retroactive interference between motor practice and paired associative stimulation.

Proactive and retroactive interference have been widely demonstrated behaviorally, but their neurophysiological effects have rarely been compared within the same experimental framework. This study compared the primary motor cortex (M1) plasticity responses when motor practice (MP) was performed before or after paired associative stimulation (PAS25) and explored whether these responses were accompanied by changes in intracortical inhibition. A within-subject design was employed. A right-hand precision grip task with real-time electromyographic feedback was used to examine practice-related refin

The robotic PIANO: a critical narrative review of surgeon-controlled operative polyphony, human factors, and robotic training.

Robotic surgery can concentrate control of several technical functions at the console, but such concentration does not itself confer clinical independence, reduce dependence on the perioperative team, or improve patient outcomes. We define surgeon-controlled operative polyphony as the phase-bounded coordination of multiple partially independent technical control streams by one console operator within a distributed perioperative system. This critical narrative review and conceptual synthesis asks which technical functions can be concentrated coherently, where human-factors and team dependencies

From Visual Haptics to Force-Controlled Robotic Surgery: Preliminary Clinical Experience Using the da Vinci 5 System in Colorectal Surgery-Case Series Study.

BACKGROUND: The absence of haptic feedback has been a fundamental limitation of robotic surgery. The da Vinci 5 (dV5) system is the first da Vinci platform to incorporate true force feedback (FFB), enabling real-time transmission of instrument-tip forces to the surgeon. However, the quantitative impact of FFB sensitivity settings remains unclear. This study represents the first Japanese clinical evaluation of dV5 FFB in colorectal surgery. METHODS: We conducted a case-series analysis of 10 consecutive robotic colorectal cancer resections performed using the daV5 system by a single Japan Socie

The Nutri-Exposome Intelligence Framework: Integrating Multi-Omics, Machine Learning, and Digital Nutrition for Precision Chronic Disease Prevention.

Background/Objectives: Precision nutrition is moving beyond population-based guidance and isolated gene-diet interactions toward integrative models of dietary response. However, current approaches remain fragmented across nutrigenomics, microbiome research, multi-omics profiling, digital health, and machine learning. This review proposes the Nutri-Exposome Intelligence Framework as a conceptual, data science-driven model for integrating cumulative dietary, environmental, microbial, molecular, clinical, and digital exposures for precision chronic disease prevention. Methods: This conceptual rev

What Do We Actually Measure? The Epistemic Limits of Surrogate Markers in Chronic Disease.

BACKGROUND: Contemporary chronic disease evaluation relies heavily on surrogate markers such as LDL cholesterol, hemoglobin A1c, blood pressure thresholds, weight and body mass index. Although these measures are reproducible and supported by population level outcome data, they do not directly quantify the biologic processes that drive patient level instability. This mismatch creates an epistemic gap between what surrogate markers measure and what clinicians believe they measure. OBJECTIVE: To examine the epistemic status of surrogate markers, contrast them with biologic disease activity, and
