Osteoarthritis (OA) is a progressive disease characterized by cartilage degradation, subchondral bone remodeling, and osteophyte formation, leading to pain and functional decline. Animal models are crucial for studying injury mechanisms, biomechanical changes, and potential OA treatment. The bipedal turkey model shows promise as a valuable large-animal model due to its arthroscopic feasibility and relevance to traumatic knee injury research. However, the underlying kinematic mechanisms remain poorly understood. This study aimed to elucidate these mechanisms through biomechanical analysis of th

PURPOSE: Progressive Supranuclear Palsy (PSP) is an atypical parkinsonian syndrome characterized by significant postural instability and gait difficulties (PIGD). While brain cholinergic deficits are documented in PSP, their role in the pathophysiology of PIGD is an area of active research. This cross-sectional study aimed to elucidate relationships between regional cholinergic denervation, assessed by [18F]FEOBV PET, and PIGD severity in PSP patients.
METHODS: Nineteen subjects characterized clinically as PSP (twelve males, seven females; mean age of 69.47 ± 6.46 years [range 55-79]). Based

Accurate assessment of joint kinematics is fundamental to biomechanics, rehabilitation, and sports science. Although optical motion capture (OMC) remains the laboratory reference standard for biomechanical validation, its cost, infrastructure requirements, and limited applicability outside controlled environments restrict its broader use. Wearable inertial measurement units (IMUs) and vision-based markerless systems have consequently emerged as complementary alternatives, offering portability, reduced subject preparation, and applicability in ecological settings. Their rapid development, howev

BACKGROUND: White-matter abnormalities are common in phenylketonuria (PKU) and are typically attributed to metabolic hypomyelination or intramyelinic edema. Immune-mediated CNS involvement co-occurring with PKU has rarely been described in adults. CASE: An 18-year-old male university freshman diagnosed with classical PKU presented with a 3-week history characterized by progressive bilateral lower-limb weakness and gait instability. Cervical spinal magnetic resonance imaging (MRI) revealed intramedullary lesions, indicating possible immune-mediated CNS involvement overlapping PKU. Cerebrospinal

BACKGROUND: Acute vertigo and dizziness are among the most frequent complaints presenting to the emergency department (ED). If acute-onset and persistent vertigo/dizziness (lasting > 24 h) are accompanied by motion intolerance, gait imbalance, nausea/vomitus, and (usually also) nystagmus, this refers to the acute vestibular syndrome (AVS). We aimed at collecting epidemiologic data about the frequency of AVS etiologies for improving the diagnostic workup.
METHODS: We conducted a Prospero-registered systematic literature search (MEDLINE, Embase - in June 2024) retrieving information on AVS's cl

The G-protein-coupled receptor kinase-interacting protein 1, GIT1, is a multifunctional scaffold protein that plays key roles in the regulation of actin cytoskeletal dynamics, focal adhesion assembly, membrane trafficking, and intracellular signaling. In mice, loss of Git1 function causes a microcephaly-like phenotype characterized by a smaller brain due to reduced neuronal cell size, accompanied by behavioral deficits, altered gait, and impairment in motor coordination, learning and memory. To date, variants in GIT1 have not been definitely linked to human disease. By applying a combined geno

Thiamine (vitamin B1) deficiency is a reversible, yet potentially fatal condition that affects both the central and peripheral nervous systems. Although commonly linked to chronic alcoholism, non-alcoholic causes, including malabsorption syndromes, are increasingly being recognized. We report the case of a 22-year-old woman who presented with progressive lower limb weakness, gait disturbances, and confusional symptoms, following a history of intermittent diarrhea and vomiting. Neurological examination revealed ascending motor deficits, areflexia, cerebellar syndrome, and internuclear ophthalmo

BACKGROUND: A two-year-old female patient was referred to our clinic due to speech and gait disturbances, strabismus, vacant staring, truncal hypotonia, and spasticity in the extremities. The patient had a history of a complicated delivery resulting in perinatal asphyxia. Electrocardiogram (ECG), echocardiogram (ECHO) and abdominal ultrasound findings reported no abnormalities. Previously performed spinal muscular atrophy (SMA) test, chromosomal microarray and karyotype analyses yielded normal results. On clinical examination, facial dysmorphic features included: prominently low-set ears, stra

Spinocerebellar ataxia type 27B (SCA27B) is a recently identified autosomal dominant form of late-onset cerebellar ataxia (LOCA) caused by a guanine-adenine-adenine (GAA) > 250 intronic repeat expansion in the fibroblast growth factor 14 (FGF14) gene. Recent studies suggest that SCA27B may account for 10%-60% of undiagnosed patients with LOCA. Age at onset ranges from 40s to 70s, and clinical features include slowly progressive pancerebellar syndrome with prominent gait ataxia and cerebellar oculomotor abnormalities. Positive family history may be absent, particularly given the characteristica

BackgroundMultiple sclerosis (MS) is an autoimmune, inflammatory, progressive neurological disease. Individuals with MS frequently experience muscle weakness, limited walking endurance, and impaired balance, which collectively reduce functional independence and quality of life. Remotely supervised transcranial direct current stimulation (RS-tDCS) has emerged as a potential adjunct neuromodulation technique to enhance neuroplasticity and improve motor outcomes. However, the effectiveness of RS-tDCS on motor performance in MS remains insufficiently understood.ObjectiveTo systematically evaluate

INTRODUCTION: Temperature sensitivity affects many persons with multiple sclerosis (pwMS). Heat sensitivity describes a temporary symptom worsening in response to heat exposure. Cooling methods may counteract this and improve quality of life, but their real-world awareness and usage are poorly understood.
OBJECTIVE: To assess the frequency of temperature sensitivity among pwMS and examine awareness and usage of cooling methods.
METHODS: This cross-sectional analysis included 760 participants from the Swiss MS Registry. Data from a survey on temperature sensitivity and cooling methods were an

Most often, gait biomechanics is studied during straight-ahead walking. However, real-life walking imposes various lateral maneuvers people must navigate. Such maneuvers challenge people's lateral balance and can induce falls. Determining how people regulate their stepping movements during such complex walking tasks is therefore essential. Here, 24 adults (12F/12M; Age 25.8±3.5yrs) walked on wide or narrow virtual paths that were either straight, slowly-winding, or quickly-winding. From each trial, we computed time series of participants' step widths and their lateral body positions relative t

BACKGROUND: The alignment of the axis in ankle-foot orthoses is essential for ensuring optimal biomechanical function and comfort for the user. Correct alignment reduces orthosis displacement, minimizes skin irritation and joint stress, and improves overall performance. This study explores the effects of varying joint axis positions on ankle-foot orthoses functionality and shaft-to-leg movement.
METHODS: Six healthy adult males (mean age: 35 ± 12 years) participated, walking with bilateral ankle foot orthoses configured in five joint axis positions and a shod condition. A 3D motion analysis s

BACKGROUND: Controlling and stabilizing the body are crucial for walking on an inclined path. Healthy individuals adjust their gait to improve stability by reducing stride length and increasing cadence. Incline influences anteroposterior stability; however, its effects on mediolateral stability are inconclusive.
RESEARCH QUESTION: What are the effects of walking inclination and speed on mediolateral stability using foot placement quantified by explained variance (EV)?
METHODS: Fifteen healthy young adults walked on a treadmill under four speed (moderate 0.83 m/s, fast 1.0 m/s) and grade (0 a
