A potential approach for gene therapy targeting hepatoma using a liver-specific promoter on a retroviral vector.

Recent technological advances made in molecular biology and in vitro culture of human and other mammalian cells have led to broad medical and scientific acceptance of the feasibility of gene therapy for genetic diseases. Cancer might practically be one of the attractive targets for such therapy. For the treatment of cancer, it is important to manipulate the gene of interest such that it is expressed solely in cancer cells. We have developed a tissue-specific gene expression system, based on a tissue-specific promoter on a retroviral vector. A murine ecotropic retroviral vector was constructed

Genomic imprinting in hereditary glomus tumours: evidence for new genetic theory.

A study based on fifteen pedigrees showed that familial glomus tumours are inherited almost exclusively via the paternal line, a finding inconsistent with autosomal dominant transmission. The results can be explained in terms of the genomic imprinting hypothesis--the maternally derived gene is inactivated during female oogenesis and can be reactivated only during spermatogenesis. Genomic imprinting may have considerable implications for genetic counselling with respect to glomus tumours and also for the understanding of other hereditary diseases.

Gene knockdown studies revealed CCDC50 as a candidate gene in mantle cell lymphoma and chronic lymphocytic leukemia.

The two B-cell non-Hodgkin's lymphoma entities, chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), show recurrent chromosomal gains of 3q25-q29, 12q13-q14 and 18q21-q22. The pathomechanisms affected by these aberrations are not understood. The aim of this study was to identify genes, located within these gained regions, which control cell death and cell survival of MCL and CLL cancer cells. Blood samples collected from 18 patients with CLL and 6 patients with MCL, as well as 6 cell lines representing both malignancies were analyzed by gene expression profiling. By a comparison

Specific patterns of oncogene activation in transplacentally induced tumors.

Transplacental exposure of rats to a single dose of the direct acting carcinogen methylnitrosourea (MNU) results in the induction of a variety of neoplasias of neuroectodermal, epithelial, and mesenchymal origin. Molecular analysis of the oncogenes present in these tumors revealed a striking degree of tissue specificity. neu oncogenes were found to be reproducibly activated in tumors derived from the peripheral nervous system (PNS), but not in those arising from the central nervous system (CNS). No ras oncogenes were found in either PNS- or CNS-derived tumors. However, Ha-ras oncogenes were de

Silencing of lncRNA HOTAIR enhances doxorubicin sensitivity and induces apoptosis in diffuse large B-cell lymphoma cells.

Diffuse large B-cell lymphoma (DLBCL) is an aggressive malignancy characterized by a poor prognosis, mainly caused by late diagnosis and the absence of effective treatment options. Malignant transformation of B-cells emphasizes the necessity of novel biomarkers for early diagnosis and prognosis. Dysregulation of long non-coding RNAs (lncRNAs), fundamental regulators in biological pathways, is linked to the development of different cancer types, including DLBCL. In this study, we assessed lncRNA HOTAIR expression in DLBCL and its association with clinicopathological features while investigating

[Activation of oncogenes by chromosomal translocation in hematopoietic tumors].

Specific chromosomal translocations are one of the cytogenetic characteristics of hematopoietic malignancies. The application of molecular biological techniques has led to important insight into the molecular consequences, accompanied by the chromosomal translocations. Specific oncogenes and related genes activated by the translocations have been identified at the translocation breakpoints. Translocated oncogenes are deregulated by their juxtaposition to enhancers of Ig or TCR genes, resulting in their overexpressions in some lymphoid tumors. Alternatively, they are activated by gene fusion, r

Enhancer collapse dictates PKHD1 silencing and centrosome-mediated chromosomal instability and metastasis in clear cell renal cell carcinoma.

Chromosomal instability (CIN) drives clear cell renal cell carcinoma (ccRCC) progression, yet its upstream triggers remain elusive. To identify key drivers governing renal genomic integrity, we integrated machine learning-driven screening with Mendelian randomization analysis to prioritize candidates associated with both CIN and clinical outcomes. This computational pipeline identified Polycystic Kidney and Hepatic Disease 1 (PKHD1) as a top candidate and a crucial guardian of the renal genome. Through single-cell and spatial transcriptomics, combined with functional assays in ccRCC cell lines

Identification of LncRNAs scaffolding a chromatin-associated gene regulatory complex in breast cancer cells comprising ERα, DOT1L and menin.

In Estrogen Receptor (ER)-positive Breast Cancer (BC), the development of resistance to endocrine therapies remains a significant clinical challenge, often linked to transcriptional and epigenetic deregulations. The main hallmark in this neoplasm, the nuclear receptor ERα, drives oncogenesis by recruiting multiple coregulators, including the histone methyltransferase DOT1L and the scaffold protein menin, forming a pro-tumorigenic machinery. However, the precise mechanism of this complex assembly is not fully understood. Our study investigates the molecular basis of ERα-DOT1L-menin functional a

CRISPR/Cas9-mediated miR-21 editing in high-grade urothelial carcinoma cells and its biological effects.

BACKGROUND: Urothelial carcinoma, the predominant form of bladder cancer, represents a global public health challenge due to its high rates of recurrence and progression. At the molecular level, microRNA-21 (miR-21) has been characterized as an "oncomir" because of its ability to negatively regulate tumor suppressor genes, thereby promoting tumor survival and progression. In this context, the CRISPR/Cas9 system has emerged as a precise genome-editing tool. OBJECTIVE: To investigate the biological effects of miR-21 modulation using CRISPR/Cas9-mediated genome editing in the T24 high-grade inva

Dual role and ceRNA networks of miR-146a in lung cancer: regulating proliferation, metastasis, and chemoresistance.

Lung cancer persists as the predominant cause of cancer-related mortality globally. MicroRNAs, a class of short non-coding RNAs, are recognized as critical regulators in the pathogenesis of lung cancer, functioning either as oncogenes or tumor suppressors. Among these, miR-146a has been identified as a significant modulator in non-small-cell lung cancer, exerting influence over cellular proliferation, resistance to chemotherapy, apoptosis, and metastatic potential. Notably, miR-146a displays a dualistic role, acting as either a tumor suppressor or an oncogene depending on the molecular mechani

A fishnet between nasopharyngeal carcinoma and resistance: the competing endogenous RNA network.

Chemotherapy and radiotherapy (chemo-/radiotherapy) have advanced as the main treatment modalities for nasopharyngeal carcinoma (NPC), improving patient survival rates. However, chemo-/radiotherapy resistance in NPC cells has emerged as a key factor contributing to poor prognosis. Recently, competing endogenous RNA networks (ceRNETs) have garnered attention for their potential clinical value in studying chemo-/radiotherapy resistance. In this review, we aimed to explore the molecular mechanisms of ceRNA-related molecules, including circular RNA (circRNA), long non-coding RNA (lncRNA), microRNA
