Strain-level dissection of the contribution of the gut microbiome to human metabolic disease.

The gut microbiota has been linked with metabolic diseases in humans, but demonstration of causality remains a challenge. The gut microbiota, as a complex microbial ecosystem, consists of hundreds of individual bacterial species, each of which contains many strains with high genetic diversity. Recent advances in genomic and metabolomic technologies are facilitating strain-level dissection of the contribution of the gut microbiome to metabolic diseases. Interventional studies and correlation analysis between variations in the microbiome and metabolome, captured by longitudinal sampling, can lea

Exploring the effect of dentition, dental decay and familiality on oral health using metabolomics.

As a proof of principle, we used an untargeted global metabolic profiling of saliva to understand the biochemical processes associated with dental decay, dentition (primary and secondary tooth eruption) and familiality in a sample of 25 sibling pairs. Pairs were selected to represent four different combinations of dentition and tooth health: (1) both siblings with primary teeth and no decay (n=5); (2) both siblings with primary teeth and discordant for dental decay (n=6); (3) both siblings with primary teeth and dental decay (n=4); and (4) one sibling with primary teeth the other with mixed de

Current status of vitamin D signaling and its therapeutic applications.

Vitamin D and in particular its biologically most active metabolite, 1α,25-dihydroxyvitamin D₃ (1α,25(OH)₂D₃), are central endocrine molecules that influence many aspects of human physiology, which are not only the well-known calcium and phosphorus up-take and transport controlling bone formation, but also the control of immune functions and of cellular growth and differentiation. Basically all actions of 1α,25(OH)₂D₃ are mediated by the transcription factor vitamin D receptor (VDR). The crystal structure of the VDR and detailed knowledge on its molecular interactions with the ligand provide s

Role of Mycobacterium tuberculosis pknD in the pathogenesis of central nervous system tuberculosis.

BACKGROUND: Central nervous system disease is the most serious form of tuberculosis, and is associated with high mortality and severe neurological sequelae. Though recent clinical reports suggest an association of distinct Mycobacterium tuberculosis strains with central nervous system disease, the microbial virulence factors required have not been described previously. RESULTS: We screened 398 unique M. tuberculosis mutants in guinea pigs to identify genes required for central nervous system tuberculosis. We found M. tuberculosis pknD (Rv0931c) to be required for central nervous system diseas

Sarcoidosis--scientific progress and clinical challenges.

Sarcoidosis is an uncommon systemic inflammatory disorder characterized by noncaseating granulomatous inflammation that most commonly affects the lungs, intrathoracic lymph nodes, eyes and skin. One-third or more of patients with sarcoidosis have chronic, unremitting inflammation with progressive organ impairment. Findings of family and genetic studies indicate a genetic susceptibility to sarcoidosis, with genes in the MHC region having a dominant role. Immunologic hallmarks of the disease include highly polarized expression of cytokines produced by type 1 T helper cells and tumor necrosis fac

The genetic organization and evolution of the broad host range mercury resistance plasmid pSB102 isolated from a microbial population residing in the rhizosphere of alfalfa.

Employing the biparental exogenous plasmid isolation method, conjugative plasmids conferring mercury resistance were isolated from the microbial community of the rhizosphere of field grown alfalfa plants. Five different plasmids were identified, designated pSB101-pSB105. One of the plasmids, pSB102, displayed broad host range (bhr) properties for plasmid replication and transfer unrelated to the known incompatibility (Inc) groups of bhr plasmids IncP-1, IncW, IncN and IncA/C. Nucleotide sequence analysis of plasmid pSB102 revealed a size of 55 578 bp. The transfer region of pSB102 was predicte

Antifungal Activity of Methyl 2-Methylbutyrate Against Botrytis cinerea and the Induced Physiological, Transcriptional, and Metabolic Responses.

Gray mold, caused by the necrotrophic fungus Botrytis cinerea, is a devastating disease responsible for substantial global crop losses. The escalating threat of fungicide resistance and environmental concerns necessitates the development of eco-compatible control alternatives. In this study, we investigated the antifungal efficacy of methyl 2-methylbutyrate (M2M), a microbial volatile organic compound (mVOC), against B. cinerea, and elucidated the underlying physiological, transcriptomic, and metabolomic responses. Our results demonstrated that M2M significantly inhibited mycelial growth in a

Metabolic engineering strategies for sustainable de novo isobutylamine production by Corynebacterium glutamicum.

BACKGROUND: Short-chain primary amines are a versatile group of molecules in biological systems, derived from the decarboxylation of amino acids or the transamination of carbonyl-containing molecules. Isobutylamine, the decarboxylation product of L-valine, has applications in chemical synthesis of pharmaceuticals, herbicides, fungicides and specialized chemicals, where it is especially interesting as a substitute for petrochemical compounds. Since the chemical synthesis of short-chain primary amines is hazardous and environmentally unsustainable, we established an efficient biosynthetic approa

Exogenous nitrogen mitigation of cadmium stress in Pinellia ternata is associated with the regulation of arginine and proline metabolism.

Cadmium (Cd) contamination is a major constraint limiting the yield and quality of medicinal plants. In this study, the effects of exogenous nitrogen on alleviating Cd toxicity in Pinellia ternata seedlings were investigated, and the underlying mechanisms were explored using integrated physiological, transcriptomic, untargeted metabolomic, and rhizosphere microbial analyses. Nitrogen supplementation (150 mg N kg-¹ supplied as urea) mitigated Cd-induced growth inhibition, increasing root length by approximately 31% on days 6 and 9 and partially improving leaf area by day 9. Nitrogen supplementa

Metagenomic insights into potential horizontal transfer of resistance/virulence genes in gut microbiota from patients with Crohn disease.

BACKGROUND: Unraveling the potential horizontal transfer of resistance genes/virulence genes (RGs/VGs) in gut microbiota from patients with Crohn disease (CD) is an interesting but poorly characterized issue. METHODS: Quantitative assessment was performed to estimate the relative abundance and diversity of RGs/VGs/mobile genetic elements (MGEs). Differential analysis was applied to identify the CD-specific enriched genetic subtypes. A species-RGs/VGs/MGEs association network was constructed to explore possible co-occurrence patterns of these genetic elements across potential microbial hosts.

Gut microbial resistance and metabolism of selective serotonin reuptake inhibitors drive multidrug resistance and contribute to antidepressant tachyphylaxis.

Selective serotonin reuptake inhibitors (SSRIs) are among the most widely used antidepressant medications in modern clinical practice. However, the underlying mechanisms of the rapid onset of tolerance to antidepressants (antidepressant tachyphylaxis [ADT]) led by SSRIs are not well-investigated. To clarify the roles of the interaction between gut microbes and SSRIs in the development of ADT and other adverse effects, we isolated 4 sertraline (STR; an SSRI)-resistant microbial strains, one of which was identified as uropathogenic Escherichia coli (E coli G5100a). Distinct host-pathogen and microbiome responses of aoudad (Ammotragus lervia) and bighorn sheep (Ovis canadensis) following exposure to Mycoplasma ovipneumoniae with or without co-exposure to leukotoxigenic Pasteurellaceae.

BACKGROUND: Pathogens can shape their host communities over various timescales. The potential role of host–pathogen co-evolution in driving contemporary shifts in disease ecology is becoming increasingly important as host species emerge and persist outside their native ranges. In North America, Mycoplasma ovipneumoniae can cause fatal pneumonia epizootics in native bighorn sheep (Ovis canadensis), whereas introduced free-ranging sympatric aoudad (Ammotragus lervia) typically act as asymptomatic reservoirs. We describe the responses of two hosts with different biogeographical histories to share

Environmental transition navigates phenotype switching, affecting the virulence and multidrug-resistant profile of pathogenic Morganella morganii.

The bacteria's ability to respond to environmental changes is critical for their survival. This allows them to form intricate communities, withstand stress, and initiate virulence responses in hosts during infection, a phenomenon known as phenotypic switching. In this study, we investigated the role of shaking conditions on phenotype switch in multidrug-resistant and pathogenic Morganella morganii both under in vitro and in vivo conditions. The results demonstrate that M. morganii grown in non-shaking conditions, possibly causing low fluid shear, developed floccules or cellular aggregates, and

Differences and fingerprints of ESBL-producing E. coli from chicken faeces.

E. coli is an important source of β-lactam resistance. Extended Spectrum β-Lactamase (ESBL) producing E. coli and their phylogenetic transmission are an important problem for health. On the other hand, MALDI-TOF MS allows sensitive and specific applications in food safety and many clinical studies, and it is also a method approved by the Food and Drug Administration (FDA) for microbial identification. We thought that the protein phenotypic character of the bacteria might provide data on resistance. This study aims to investigate β-lactamases in E. coli and to examine the differences in protein
