Adenoviruses encoding HPRT correct biochemical abnormalities of HPRT-deficient cells and allow their survival in negative selection medium.

The Lesch-Nyhan syndrome is an X-linked disorder caused by a virtually complete absence of the key enzyme of purine recycling, hypoxanthine-guanine phosphoribosyltransferase (HPRT). It is characterized by uric acid overproduction and severe neurological dysfunction. No treatment is yet available for the latter symptoms. A possible long-term solution is gene therapy, and recombinant adenoviruses have been proposed as vectors for gene transfer into postmitotic neuronal cells. We have constructed an adenoviral vector expressing the human HPRT cDNA under the transcriptional control of a short huma

Pseudohypoparathyroidism type 1b in pregnancy.

Rheumatoid fibroblast-like synoviocytes (RFLS) have an important role in the inflammatory pathogenesis of rheumatoid arthritis (RA). Toll-like receptor 3 (TLR3) is upregulated in RFLS; its activation leads to the production of interferon-β (IFN-β), a type I IFN. IFN-stimulated gene 56 (ISG56) is induced by IFN and is involved in innate immune responses; however, its role in RA remains unknown. Therefore, the purpose of this study was to investigate the role of TLR3-induced ISG56 in human RFLS. RFLS were treated with polyinosinic-polycytidylic acid (poly I:C), which served as a TLR3 ligand. ISG

Assessment of cell compatibility and antibacterial activity of nano-ZrO2/carbon fibre-reinforced PEEK bioactive composite: an in vitro study.

BACKGROUND: The peri-implant seal established between a dental implant abutment and the surrounding gingival soft tissue, serves as the first line of defence against the oral environment. It is a critical determinant of the long-term stability and success of an osseointegrated implant. Carbon-fibre-reinforced polyetheretherketone (CFR-PEEK) is a promising alternative for metallic surgical implants. This in vitro study determined the optimal zirconia nanostructure on CFR-PEEK surfaces to enhance the response of human gingival fibroblasts (HGFs) and its antibacterial properties, thereby facilita

Targeted Next Generation Sequencing Revealed a Novel Homozygous Loss-of-Function Mutation in ILDR1 Gene Causes Autosomal Recessive Nonsyndromic Sensorineural Hearing Loss in a Chinese Family.

Hereditary hearing impairment is one of the major and common birth defects in Chinese population. Non-syndromic sensorineural hearing loss (NSHL) is the most common types of hereditary hearing impairment. Genotypically and phenotypically NSHL is extremely heterogenous and follow either autosomal dominant or autosomal recessive or X-linked mode of inheritance. Presently, 127 genes have been identified to be associated with both syndromic and (NSHL). Here, we studied a Chinese family with moderate and profound hearing impairment. The proband is a 30-year old Chinese man. The proband was born wit

Genetic Susceptibility to Chronic Kidney Disease - Some More Pieces for the Heritability Puzzle.

Chronic kidney disease (CKD) is a major global health problem with an increasing prevalence partly driven by aging population structure. Both genomic and environmental factors contribute to this complex heterogeneous disease. CKD heritability is estimated to be high (30-75%). Genome-wide association studies (GWAS) and GWAS meta-analyses have identified several genetic loci associated with CKD, including variants in UMOD, SHROOM3, solute carriers, and E3 ubiquitin ligases. However, these genetic markers do not account for all the susceptibility to CKD, and the causal pathways remain incompletel

Anophthalmia and microphthalmia.

Anophthalmia and microphthalmia describe, respectively, the absence of an eye and the presence of a small eye within the orbit. The combined birth prevalence of these conditions is up to 30 per 100,000 population, with microphthalmia reported in up to 11% of blind children. High-resolution cranial imaging, post-mortem examination and genetic studies suggest that these conditions represent a phenotypic continuum. Both anophthalmia and microphthalmia may occur in isolation or as part of a syndrome, as in one-third of cases. Anophthalmia/microphthalmia have complex aetiology with chromosomal, mon

Liver X receptor (LXR) signaling in macrophage inflammation and cardiometabolic disease.

Cardiometabolic diseases profoundly remodel tissue immune landscapes, characterized by the accumulation of diverse macrophage subtypes. Among these, lipid-associated macrophages (LAMs) have emerged as an important subset that is frequently observed in cardiometabolically affected tissues. LAMs are defined by a transcriptional profile linked to lipid metabolism and processing. In parallel, the nuclear receptor liver X receptor (LXR), is a key transcription factor that regulates cholesterol efflux, fatty-acid synthesis, as well as inflammation pathways. Through these pathways, LXR agonism has be

Unraveling a Rare Complication: BCG-associated Brain Abscess in Wiskott-aldrich Syndrome.

Wiskott-aldrich syndrome (WAS) is a rare X-linked primary immunodeficiency characterized by thrombocytopenia, eczema, and recurrent infections. Live vaccines, such as Bacillus Calmette-Guérin (BCG), routinely administered in tuberculosis-endemic regions, may cause severe complications in undiagnosed patients. We report a 2-year-old boy with genetically confirmed WAS who developed a rare BCG-related brain abscess. The patient initially presented with neonatal thrombocytopenia and eczema, followed by recurrent respiratory and gastrointestinal infections. Whole exome sequencing identified a patho

mRNA Sequencing to Identify Aberrant Splicing in X-linked Alport Syndrome.

INTRODUCTION: X-linked Alport syndrome (XLAS) is a well-known monogenetic kidney disease caused by pathogenic variants in the COL4A5 gene. Routine analysis of exons and direct flanking regions fails to identify a pathogenic variant in 10% to 20% of patients with XLAS. METHODS: We evaluated 11 selected patients with clinical features of XLAS, in whom routine analysis failed to identify a pathogenic variant. In 2 patients a variant of unknown significance was detected in the intronic splice site regions. We used mRNA analysis from fibroblasts or urine-derived podocyte-lineage cells to establish

Distinct Pathogenic Mechanisms of Two Novel NHS Mutations Identified in Chinese Han Families With Nance-Horan Syndrome.

Nance-Horan syndrome (NHS) is a rare X-linked genetic disorder characterized by congenital cataracts, dental anomalies, and neurodevelopmental impairments, caused by mutations in the NHS gene. In this study, two novel NHS mutations, c.3847C>T and c.2519_2520del, were identified in two unrelated Chinese Han families, and their pathogenic molecular mechanisms were elucidated. Functional analyses revealed that the c.3847C>T mutation exerts a dual pathogenic effect: It disrupts extracellular matrix homeostasis by downregulating COL4A1 and upregulating MMP9, and it triggers oxidative stress, eviden

Early differential impact of MeCP2 mutations on functional networks in Rett syndrome patient-derived human cortical organoids.

Human cerebral organoids derived from induced pluripotent stem cells can recapture early developmental processes and reveal changes involving neurodevelopmental disorders. Mutations in the X-linked methyl-CpG binding protein 2 (MECP2) gene are associated with Rett syndrome, and disease severity varies depending on the location and type of mutation. Here, we focused on neuronal activity in Rett syndrome patient-derived organoids, analyzing two types of MECP2 mutations-a missense mutation (R306C) and a truncating mutation (V247X)-using calcium imaging with three-photon microscopy. Compared to is

The p. Arg1800His Mutation of Factor VIII Results in Moderate Hemophilia A Due to Defective Processing and Stability of Functional Protein.

Haemophilia A (HA) is a rare congenital, recessive X-linked bleeding disorders caused by the lack or deficiency of clotting factor VIII (FVIII). Missense variants, which can alter protein structure, conformation and immunogenicity, are found in nearly all individuals with mild or moderate hemophilia A but in fewer than 20% of those with the severe form. Here, we identified a 26-year-old man with a hemizygous F8 mutation (NM_000132.4(F8):c.5399 G > A (p. Arg1800His)) through direct sequencing of all exons of the F8 gene, which is established to cause a lack of FVIII:c (%) in circulating blood a

Expanding the Phenotypic Spectrum of Raynaud-Claes Syndrome: A Rett-like Presentation with Two New Cases.

Pathogenic variants in the CLCN4 gene are associated with a rare X-linked neurodevelopmental disorder, Raynaud-Claes syndrome, characterized by intellectual disability, epilepsy, language impairment, motor deficits, stereotypies, and structural brain abnormalities. Although heterozygous females are often considered to be only mildly affected, severe phenotypes have also been reported, and the clinical presentation shows considerable heterogeneity. The present study aims to summarize the current knowledge on CLCN4-related neurodevelopmental disorders through a review of the available literature

X-Linked Anhidrotic Ectodermal Dysplasia in A 19-Year-Old Male: A Classic Phenotype.

X-linked anhidrotic/hypohidrotic ectodermal dysplasia (XLHED), also known as Christ-Siemens-Touraine syndrome, is a rare genetic disorder characterized by the abnormal development of ectodermal structures, primarily affecting sweat glands, hair, and teeth. It results from mutations in the Ectodysplasin A (EDA) gene. Clinically, the classic triad of hypohidrosis, hypotrichosis, and hypodontia or anodontia forms the diagnostic hallmark. Herein, we report a case of a 19-year-old male who presented generalized dryness of skin, heat intolerance, and inability to sweat since early childhood. On exam

Dynactin knockdown leads to synuclein aggregation by blocking autophagy in a zebrafish model of Parkinson's disease.

Axons of dopaminergic neurons projecting from substantia nigra to striatum are severely affected in the early stage of Parkinson's disease (PD), with axonal degeneration preceding the loss of cell bodies. Our previous study indicated that the dysfunctional retrograde axonal transport could lead to the death of dopaminergic neurons resulting in PD (10.1111/j.1471-4159.2008.05526.x). However, dynein, as the main molecule involved in retrograde axonal transport, was not affected. This study aimed to verify the hypothesis that dynactin rather than dynein may be one of the key factors in the retrog

In Vivo Phage Display for the Identification of Muscle Homing Peptides to Improve the Delivery of Phosphorodiamidate Morpholino Oligomers for Duchenne Muscular Dystrophy Therapy.

The severe X-linked degenerative neuromuscular disease Duchenne muscular dystrophy (DMD) is caused by the loss of dystrophin through reading frame disruptive mutations in the DMD gene. Dystrophin protein is crucial for the stability of the muscle. Targeting specific exons with antisense oligonucleotides (ASO) will prevent inclusion of the exon during pre-mRNA splicing, which can restore the reading frame, facilitating the production of partially functional dystrophin proteins. For DMD, four ASOs of the phosphorodiamidate morpholino oligomer (PMOs) chemistry are FDA approved. It is anticipated
