Typing of clinical and environmental strains of Aeromonas spp. using two PCR based methods and whole cell protein analysis.

Two PCR based typing methods i.e. random amplified polymorphic DNA analysis (RAPD) and enterobacterial repetitive intergenic consensus sequence (ERIC)-PCR were evaluated for typing of 42 Aeromonas isolates from clinical and environmental sources and whole cell protein (WCP) profiles were analyzed. Both RAPD and ERIC-PCR showed a high level of genetic diversity. Numerical index of the discriminatory (D) values were 0.94 and 0.96 (>0.90) for RAPD and ERIC-PCR, respectively. No correlation in banding pattern and evidence of genetic similarity was found between Aeromonas isolates from environmenta

Functional characterization of Cr-CATHs: Novel antimicrobial peptides from the coastal bird Chroicocephalus ridibundus.

The escalating global threat of antimicrobial resistance (AMR) and chronic biofilm-associated infections underscores the urgent need for novel therapeutic agents. Antimicrobial peptides (AMPs) offer a promising alternative due to their potent activity, broad-spectrum efficacy, and low resistance induction. In this study, we identified three novel cathelicidin-like peptides-Cr-CATH-1 (Cr1), Cr-CATH-2 (Cr2), and Cr-CATH-3 (Cr3)-from Chroicocephalus ridibundus. These peptides were systematically evaluated for their physicochemical properties, antimicrobial activity, bactericidal kinetics, and eff

Characterization of Lipidated New Delhi Metallo-β-lactamase Using Synthetic Nanodiscs.

Carbapenem-resistant bacteria present a serious challenge to current treatment methods for bacterial infections. Particularly concerning are metallo-carbapenemases, a subset of plasmid-borne β-lactamases, active against nearly all β-lactam antibiotics and insusceptible to available β-lactamase inhibitors. The most widespread metallo-carbapenemase is New Delhi metallo-β-lactamase (NDM). NDM is unique among all known metallo-β-lactamases due to a post-translational lipidation that anchors it to the outer membrane. Most β-lactamases, including all other metallo-carbapenemases, exist as soluble pr

In-silico approach of identifying novel therapeutic targets against Yersinia pestis using pan and subtractive genomic analysis.

The magnitude of human affliction brought about by bacterial infections has been on the rise since the mid-5th century. Yersinia pestis is one such notable, gram-negative bacterium that inflicted havoc around the globe three times throughout different millenniums by causing deadly plagues. Despite the unremitting efforts by scientists, different strains of Yersinia pestis are still affecting the populations in various parts of the world by growing resistant to existing antimicrobial agents owing to their overuse. The current scenario, therefore, calls for new therapeutics to further combat the

Computational identification of potential lead molecules targeting rho receptor of Neisseria gonorrhoeae.

Gonorrhea, one of the sexually transmitted disease caused by a gram negative diplococcus bacteria Neisseria gonorrhoeae. Rho protein is indispensable for bacterial viability due to its versatile functions in physiology apart from RNA dependent transcription termination. Based on conserved function and wider role in several cellular processes, inhibitors specifically targeting Rho proteins are largely in use these days to treat various bacterial infections. In this study, three dimensional structure of Rho protein was modeled using the template protein from E. coli and further the optimized mod

The presence of colistin resistance gene mcr-1 and -3 in ESBL producing Escherichia coli isolated from food in Ho Chi Minh City, Vietnam.

Colistin is indicated for the treatment of multidrug-resistant gram-negative bacterial infections. However, the spread of colistin-resistant bacteria harbouring an mcr gene has become a serious concern. This study investigated local foods in Vietnam for contamination with colistin-resistant bacteria. A total of 261 extended-spectrum β-lactamase (ESBL)- and AmpC-producing Escherichia coli isolates from 330 meat and seafood products were analysed for colistin susceptibility and the presence of mcr genes. Approximately, 24% (62/261) of ESBL- or AmpC-producing E. coli isolates showed colistin resi…

A Coming of Age Story: Chlamydia in the Post-Genetic Era.

Chlamydia spp. are ubiquitous, obligate, intracellular Gram-negative bacterial pathogens that undergo a unique biphasic developmental cycle transitioning between the infectious, extracellular elementary body and the replicative, intracellular reticulate body. The primary Chlamydia species associated with human disease are C. trachomatis, which is the leading cause of both reportable bacterial sexually transmitted infections and preventable blindness, and C. pneumoniae, which infects the respiratory tract and is associated with cardiovascular disease. Collectively, these pathogens are a signifi

Helicobacter pylori lipopolysaccharide activity in human peripheral blood mononuclear leukocyte cultures.

Helicobacter pylori (H. pylori) have been recognized as a major cause of chronic gastritis, gastric and duodenal ulcers and gastric cancer. Macrophages are the targets of lipopolysaccharide (LPS), which is a constituent of the outer membrane of Gram-negative rods. In this study we focused on a potential role of macrophages in the proliferation of human peripheral blood mononuclear leukocytes (PBML) in the milieu of H. pylori LPS and standard E. coli LPS. First, we found that H. pylori and E. coli LPS induced proliferation of total PBML (tPBML) from 5 out 21 healthy blood donors (LPS responders

Antimicrobial activity and spectrum of rifaximin, a new topical rifamycin derivative.

Rifaximin, a rifamycin derivative, was evaluated in vitro to assess its spectrum and potency against a wide variety of bacteria, yeasts, viruses, and parasites. High concentrations of rifaximin were often used to reflect topically achieved levels since this compound is poorly absorbed by oral route. Like rifampin, rifaximin possessed best activity against Staphylococcus spp. (MIC50 < or = 0.015 microgram/ml), Streptococcus spp. (MIC50s, < or = 0.03-0.12 microgram/ml), Enterococcus spp. (MIC50s, 0.25-2 micrograms/ml), Bacillus cereus (MIC50, 0.06 microgram/ml), Moraxella catarrhalis (MIC50, < o

Implications of endotoxins in wound healing: a narrative review.

Bacterial toxins are thought to play a role in delayed wound healing in critically colonised and infected wounds. Endotoxins are released from Gram-negative bacteria when they are lysed by host phagocytic cells during an immune response, or by antimicrobial agents, potentially leading to a detrimental effect on the host tissues. Endotoxins can affect all aspects of the wound healing process, leading to delayed healing and contributing to wound chronicity. Release of endotoxins by bacteria can also have serious systemic effects (for example, septic shock) that can lead to high levels of patient

Interferon-γ-dependent control of Anaplasma phagocytophilum by murine neutrophil granulocytes.

BACKGROUND: Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that is transmitted by ticks of the Ixodes ricinus complex. It replicates in neutrophils and elicits febrile disease in humans and animals. Because of its striking tropism for neutrophils, A. phagocytophilum has been used as a model organism to study the immune response against obligate intracellular pathogens. In mice, the control of A. phagocytophilum in the early phase of infection is dependent on natural killer cell-derived interferon-γ (IFN-γ). In contrast, the final elimination strictly requires CD4

Host Langerin (CD207) is a receptor for Yersinia pestis phagocytosis and promotes dissemination.

Yersinia pestis is a Gram-negative bacterium that causes plague. After Y. pestis overcomes the skin barrier, it encounters antigen-presenting cells (APCs), such as Langerhans and dendritic cells. They transport the bacteria from the skin to the lymph nodes. However, the molecular mechanisms involved in bacterial transmission are unclear. Langerhans cells (LCs) express Langerin (CD207), a calcium-dependent (C-type) lectin. Furthermore, Y. pestis possesses exposed core oligosaccharides. In this study, we show that Y. pestis invades LCs and Langerin-expressing transfectants. However, when the bac

Emerging therapies and diagnostic innovations for multidrug-resistant infections in kidney allograft recipients: Challenges and future directions.

Multidrug-resistant (MDR) bacterial infections are a major cause of morbidity, mortality, and graft loss in kidney transplant recipients, who are particularly vulnerable due to intensive immunosuppression, frequent healthcare exposure, and recurrent antibiotic use. The rising prevalence of resistance among key uropathogens and bloodstream isolates has outpaced the development and uptake of transplant-specific diagnostic and therapeutic strategies. This mini-review synthesizes contemporary evidence on MDR gram-negative and gram-positive infections in kidney allograft recipients, focusing on ext

Detection of tetracycline resistance genes by PCR methods.

Rapid, accurate, and sensitive determination of antibiotic resistance profiles of various human and animal pathogens becomes a vital prerequisite for successful therapeutic intervention in the face of the increased occurrences of drug-resistant bacterial infections. The current methods, which are dependent on cultivation of pathogens and phenotypic expression of antibiotic resistance, usually require excessive time, special microbiological equipment, and qualified personnel. However, even with all these requisites, for example, no bacteria can be grown from more than 80% of all clinical sample
