Intratumoural plasma cells mediate a pro-inflammatory response to kinase inhibition and successful immune checkpoint blockade therapy in malignant peripheral nerve sheath tumours.

BACKGROUND: The role of intratumoural plasma cells in immune checkpoint blockade (ICB) therapy has never been tested although their presence is linked with improved response and survival in people with cancer. Malignant peripheral nerve sheath tumours (MPNSTs) are deadly sarcomas with minimal responsiveness to ICB therapies. Strikingly, drugs inhibiting cyclin-dependent kinases 4/6 (CDK4/6) and MEK sensitise de novo MPNSTs to immunotherapy targeting programmed death-ligand 1 (PD-L1), which correlates with increased intratumoural plasma cells. Here, we tested if plasma cells mediate MPNST respo

A modular γδ TCR-T platform combining KRAS pMHC targeting with re-dosable mRNA engager redirection.

Solid tumors often evade TCR-engineered αβ T cells when antigen expression varies or when the restricting Human Leukocyte Antigen (HLA) allele is lost. γδ T cells, in contrast, detect cellular dysregulation through non-peptide/Major Histocompatibility Complex (MHC) cues, including phosphoantigens and stress ligands, and can be developed as allogeneic therapies. Although intratumoral γδ T cell signatures are associated with improved outcome across cancers, γδ recognition itself is broad and still selected within the thymus just as αβ T cell receptors (TCRs) are. It does not, however, anchor spe

Structural Modulation and Binding of HLA-DQ8 by Cysteine-to-Serine Mutated Insulin Peptide: Insights from Molecular Dynamics Simulations.

Type 1 diabetes (T1D) is driven by autoreactive CD4+ T-cell responses to pancreatic beta cell antigens presented by disease-associated human leucocyte antigen (HLA) class II molecules. However, the molecular mechanisms by which subtle antigenic modifications promote pathogenic immunity remain incompletely defined. Recent immunopeptidomic studies have identified a cysteine-to-serine substitution at position 19 of the insulin B chain, referred to as InsC19S, as a microenvironment-driven neoepitope that can be presented by HLA class II molecules, including HLA-DQ8, and is recognized by diabetogen

A new humanised TCR transgenic mouse model to study T cells reactive to citrullinated tenascin C, a relevant autoantigen in rheumatoid arthritis.

OBJECTIVES: Autoreactive T cells recognising citrullinated antigens, although rare and difficult to study, are implicated in rheumatoid arthritis (RA). To allow functional studies and manipulation of such T cells, we have generated and characterised transgenic mice expressing a T-cell receptor (TCR) cloned from a patient with RA and reactive with a prominent target, citrullinated tenascin C (citTNC). METHODS: A humanised TCR transgenic (hTCR-tg) mouse recognising the citrullinated tenascin C22 (citTNC22) antigen in an HLA-DRB1*04:01 (HLA-DR4) restricted manner was developed by genetically eng

Single-cell expression quantitative trait loci-based Mendelian randomization identifies MAN1A2 in naive CD4+ T cells as a candidate therapeutic target for osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is a leading cause of chronic pain and disability worldwide, with growing evidence implicating immune dysregulation in its pathogenesis. However, the specific peripheral immune cell populations contributing causally to OA risk remain poorly characterized. METHODS: We performed a multi-stage two-sample Mendelian randomization (MR) study integrating single-cell cis-expression quantitative trait loci (cis-eQTLs) for 14 peripheral immune cell types from the OneK1K cohort with two independent OA GWAS datasets - the Million Veteran Program for discovery and the UK Bi

Paeoniflorin prevents acute graft-versus-host disease while preserving graft-versus-tumor effects.

BACKGROUND: Acute graft-versus-host disease (aGVHD) remains a major complication after allogeneic hematopoietic stem cell transplantation (allo-HSCT). This study investigated the preventive effects of paeoniflorin (PF), a natural monoterpene glycoside, on aGVHD. METHODS: Candidate herb-derived compounds against aGVHD were screened using SymMap, HERB, ADMETlab, and InflamNat, followed by molecular docking. The preventive effects of PF were evaluated in major histocompatibility complex (MHC)-mismatched murine aGVHD models. An entire-process paeoniflorin (EP-PF) regimen, involving donor pretreat

Formulation Screening and Characterization of PLGA-Based Injectable In Situ Gel Loaded with Progesterone.

Objective: Conventional progesterone (P4) formulations suffer from low bioavailability, severe local irritation, and poor patient adherence due to P4's poor aqueous solubility. This work aimed to develop and screen a biodegradable PLGA/NMP (Poly(lactic-co-glycolic acid)/N-methyl-2-pyrrolidone) injectable in situ gel for sustained P4 delivery to overcome these clinical limitations. Significance: Commercial oral, vaginal, and oil-based intramuscular P4 preparations cannot maintain stable long-term drug exposure, and they cause injection-site pain/inflammation. The screened in situ depot system r

Molecular dynamics simulation of a novel multi-epitope vaccine design against Salmonella enterica paratyphi A using a computational approach.

Salmonella enterica serovar Paratyphi A is a significant pathogen responsible for enteric fever, particularly in developing countries, contributing to high morbidity and mortality. Current vaccines predominantly target S. Typhi, leaving a critical gap in protection against Paratyphi A strains. The absence of an effective vaccine for Paratyphi A is a significant public health concern, given its potential for outbreaks and long-term healthcare burdens. Designing a targeted vaccine is essential to reduce the incidence of enteric fever and improve global health outcomes. This study focuses on deve

Chemotherapy elevates cell surface PD-L1 and MHC-I expression in apoptotic gastric cancer cells.

BACKGROUND: The programmed cell death-ligand 1 (PD-L1) combined positive score is used as a patient selection tool and predictive factor for anti-programmed cell death-1 (PD-1)/PD-L1 therapy in gastric cancer. However, the expression of PD-L1 and major histocompatibility complex class I (MHC-I) can be affected by conventional treatment approaches. OBJECTIVE: In this study, we examined the effects of chemotherapy on surface PD-L1 and surface MHC-I expression in living and apoptotic gastric cancer cells. AGS (moderately differentiated) and SNU-1 (poorly differentiated) cells were treated 5-Fluo

Stress-NRF2 response axis polarizes tumor macrophages and undermines immunotherapy.

BACKGROUND: Tumor-associated macrophages (TAMs) can switch between immune-activating and cancer-promoting states; yet, the stress pathways that lock them into procancerous states remain obscure. Here we defined the role of transcription factor NRF2 as a mediator of procancerous macrophages. METHODS: We combined spatial transcriptomics, single-cell RNA sequencing, three-dimensional (3D) cell culture and in vivo tumor models to explore how NRF2 activation status in tumor-associated macrophages modifies responses to immunotherapy. RESULTS: In MC38 colon tumors, repeated anti-CD40 or radiotherap

The aryl hydrocarbon receptor inhibits antigen presentation to promote progression of pancreatic ductal adenocarcinoma.

BACKGROUND: The aryl hydrocarbon receptor (AhR) plays a pivotal role in modulating immune responses and influencing tumor development by detecting metabolites derived from tryptophan breakdown. In patients suffering from pancreatic ductal adenocarcinoma (PDAC), elevated levels of AhR are strongly correlated with poor clinical outcomes. Despite this, the cell-autonomous functions of AhR in pancreatic tumor cells, particularly its role in modulating anti-tumor immunity within the tumor microenvironment, remain poorly characterized and require further investigation. METHODS: CRISPR-Cas9 technolo

Structural Homology and Electrostatic Potential Comparisons of Epitope Pair Candidates for Molecular Mimicry Triggering of Type 1 Diabetes Mellitus.

BACKGROUND: Molecular mimicry, where foreign and self-peptides contain similar epitopes, can induce autoimmune responses. Identifying potential molecular mimics and studying their properties is key to understanding the onset of autoimmune diseases such as type 1 diabetes mellitus (T1DM). Previous work identified pairs of infectious epitopes (EINF) and T1DM epitopes (ET1D) that demonstrated sequence homology; however, structural homology was not considered. Correlating sequence homology with structural properties is important for translational investigation of potential molecular mimics. This w

Beyond CAR-T Cells: exploring CAR-NK, CAR-M, and CAR-γδ T strategies in solid tumor immunotherapy.

Adoptive cell therapy (ACT) employing chimeric antigen receptor (CAR) engineering represents a transformative advancement in cancer immunotherapy. CAR-T cell therapies have demonstrated significant clinical success in hematological malignancies, yet their application to solid tumors faces persistent challenges. Key limitations include the paucity of tumor-specific antigens, poor intratumoral infiltration, immunosuppressive tumor microenvironment (TME), and treatment-related toxicities such as cytokine release syndrome (CRS) and neurotoxicity. In contrast, CAR natural killer (CAR-NK) cells show

HLA-G 14 bp polymorphism: Implications and its associated risk to malaria severity among iron-fortified Ghanaian infants and young children.

BACKGROUND OF THE STUDY: Malaria is a paramount health concern mostly among infants and young children. The World Health Organization recommends iron fortification for children with iron deficiency anaemia living in malaria-endemic regions like Ghana. However, the intricate interplay between genetic polymorphisms and nutritional interventions in malaria susceptibility and severity remains unclear. The Human Leukocyte Antigen-G (HLA-G) locus within the Major Histocompatibility Complex (MHC) genes has surfaced as a critical player in regulating immune responses and influencing disease outcomes.
