"From inflammation to fibrosis: a proposed role of mucosal-associated invariant T cells in lichen sclerosus - a conceptual framework".

Lichen sclerosus (LS) is a chronic inflammatory and fibrosing dermatosis with a well-documented autoimmune background and a dominant Th1/Th17-driven immune response. Despite growing insight into its immunopathogenesis, the mechanisms linking persistent inflammation to progressive fibrosis remain unclear. Mucosal-associated invariant T cells (MAIT cells) are innate-like lymphocytes enriched in epithelial tissues, characterized by rapid activation and robust production of proinflammatory cytokines. Although alterations in MAIT cells frequency and function have been described in autoimmune and fi

Bacterial components-driven intrahepatic CXCR5hi B cells are important population for MASH progression through inducing inflammation.

Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by severe liver inflammation and fibrosis due to an imbalanced immune response caused by enhanced bacterial components. The progression of MASH is closely linked to increased permeability of intestinal mucosal barrier facilitating enter of bacterial components into hepatic portal venous system. B cells are important immune cells for adaptive responses and enhance hepatic inflammation through cytokine production and T cell activation. B cells are influenced by gut microbiota, but the specific B cell populations in MASH and

Brevibacillus laterosporus BL1 improves intestinal barrier function and immune homeostasis and modulates gut microbiota in high-fat diet-fed mice.

BACKGROUND: High-fat diet (HFD)-induced intestinal damage contributes to metabolic dysfunction and systemic inflammation. Brevibacillus laterosporus, a broad-spectrum antimicrobial, has been approved as a microbial feed additive in the breeding industry and may improve intestinal health. This study investigated whether B. laterosporus BL1 enhances intestinal health in HFD-fed mice. C57BL/6 mice were fed an HFD with or without B. laterosporus BL1 supplementation for 8 weeks. RESULTS: The findings showed that B. laterosporus BL1 reduced intestinal morphological damage, enhanced epithelial barri

Bifidobacteria and Celiac Disease: Mechanisms of Probiotic Action in Reducing Gluten-Induced Cytotoxicity and Inflammation.

Celiac disease (CD) is an immune-mediated systemic disorder triggered by gluten peptides present in the diets of genetically susceptible individuals, leading to a range of intestinal and extra-intestinal manifestations. Although managed by a gluten-free diet (GFD), symptoms persist in 30%-50% of treated individuals despite apparent dietary compliance. Accordingly, the present review explores how bifidobacteria may mediate cytotoxic and proinflammatory responses induced by gluten-derived peptides, contributing to the modulation of CD symptoms. Experimental in vitro studies, primarily using Caco

Efficient Tonsillar T Follicular Helper Cell Processing and Functional Analysis through High-dimensional Flow Cytometry.

T follicular helper cells (Tfh) are a subset of CD4+ T helper cells that aid in B cell isotype switching, germinal center (GC) formation, somatic hypermutation, and affinity maturation, thereby helping to orchestrate adaptive humoral immune responses in secondary lymphoid tissues during infection, autoimmunity, and vaccination. Understanding the development and function of Tfh cells is crucial for designing effective vaccines and developing targeted treatment strategies for diseases involving this population. Human tonsil cells are accessible mucosal lymphoid organs. They offer a unique opport

Epithelial OPA1 links mitochondrial fusion to inflammatory bowel disease.

Dysregulation at the intestinal epithelial barrier is a driver of inflammatory bowel disease (IBD). However, the molecular mechanisms of barrier failure are not well understood. Here, we demonstrate dysregulated mitochondrial fusion in intestinal epithelial cells (IECs) of patients with IBD and show that impaired fusion is sufficient to drive chronic intestinal inflammation. We found reduced expression of mitochondrial fusion-related genes, such as the dynamin-related guanosine triphosphatase (GTPase) optic atrophy 1 (OPA1), and fragmented mitochondrial networks in crypt IECs of patients with

Recombinant IL-34 alleviates bacterial enteritis in Megalobrama amblycephala by strengthening the intestinal barrier.

The potential role of interleukin-34 (IL-34), an important cytokine that regulates intestinal function in mammals, is currently being explored in the context of fish intestinal diseases. In this study, we assessed the preventive potential of recombinant Megalobrama amblycephala IL-34 (rMaIL-34) protein in bacterial enteritis. MaIL-34 exhibited conserved structural and evolutionary features with teleost IL-34 and was found to be involved in the intestinal immune response to Aeromonas hydrophila infection. Furthermore, treatment with rMaIL-34 significantly improved the survival rate of M. amblyc

IgA facilitates the persistence of the mucosal pathogen Helicobacter pylori.

IgA antibodies have an important role in clearing mucosal pathogens. In this study, we have examined the contribution of IgA to the immune control of the gastrointestinal bacterial pathogens Helicobacter pylori and Citrobacter rodentium. Both bacteria trigger a strong local IgA response that results in bacterial IgA coating in mice and in gastritis patients. Class switching to IgA depends on Peyer's patches, T-cells, eosinophils, and eosinophil-derived TGF-β in both models. In the case of H. pylori, IgA secretion and bacterial coating also depend on a functional bacterial type IV secretion sys

Contemporary Perspectives on the Role of Vitamin D in Enhancing Gut Health and Its Implications for Preventing and Managing Intestinal Diseases.

Vitamin D, a crucial fat-soluble vitamin, is primarily synthesized in the skin upon exposure to ultraviolet radiation and is widely recognized as a bone-associated hormone. However, recent scientific advancements have unveiled its intricate association with gut health. The intestinal barrier serves as a vital component, safeguarding the intestinal milieu and maintaining overall homeostasis. Deficiencies in vitamin D have been implicated in altering the gut microbiome composition, compromising the integrity of the intestinal mucosal barrier, and predisposing individuals to various intestinal pa

Oral administration of egg ovalbumin allergen induces dysregulation of tryptophan metabolism in sensitized BALB/c mice.

Food allergy (FA), triggered by specific dietary allergens, has emerged as a substantial global concern for food safety and public health. While studies have elucidated changes in immune cells and cytokines associated with allergen exposure, a comprehensive analysis of the host's metabolic features and the interaction between metabolites and the gut microbiota has not been conducted. In this study, egg allergen ovalbumin (OVA) was administered by the oral route to sensitized BALB/c mice to faithfully replicate key aspects of human FA, including severe allergic diarrhea, mast cell infiltration,

Optical imaging of the small intestine immune compartment across scales.

The limitations of 2D microscopy constrain our ability to observe and understand tissue-wide networks that are, by nature, 3-dimensional. Optical projection tomography (OPT) enables the acquisition of large volumes (ranging from micrometres to centimetres) in various tissues. We present a multi-modal workflow for the characterization of both structural and quantitative parameters of the mouse small intestine. As proof of principle, we evidence its applicability for imaging the mouse intestinal immune compartment and surrounding mucosal structures. We quantify the volumetric size and spatial di

Development of a microfluidic device to observe dynamic flow around the villi generated by deformation of small intestinal tissue.

The mucosal immune system in the small intestine is crucial for human health. For the immune response, mutual contact between gut bacteria and intestinal epithelial cells is important because there are unique epithelial cells specialized in gut bacteria sampling on the villi. The travel of gut bacteria to the villi is led by a complex dynamic flow in the small intestine. However, the complex dynamic flow around the villi has not yet been explored at a micro scale. In this study, we proposed a microfluidic device to observe the flow around the villi generated by the dynamic deformation of small

Polyamine metabolism impacts T cell dysfunction in the oral mucosa of people living with HIV.

Metabolic changes in immune cells contribute to both physiological and pathophysiological outcomes of immune reactions. Here, by comparing protein expression, transcriptome, and salivary metabolome profiles of uninfected and HIV+ individuals, we found perturbations of polyamine metabolism in the oral mucosa of HIV+ patients. Mechanistic studies using an in vitro human tonsil organoid infection model revealed that HIV infection of T cells also resulted in increased polyamine synthesis, which was dependent on the activities of caspase-1, IL-1β, and ornithine decarboxylase-1. HIV-1 also led to a
