Conformational cycling of the Wntless transporter drives trafficking and secretion of Wnt morphogens.

Wnt proteins are lipid-modified morphogens fundamental in development and disease. During Wnt biogenesis, the G-protein-coupled receptor (GPCR)-like transporter Wntless (WLS) escorts lipidated Wnts from the endoplasmic reticulum to the plasma membrane, then transfers them to extracellular carriers, forming active and soluble morphogen-carrier complexes. To dissect the mechanisms involved, we solve cryo-EM structures of Wnt-bound WLS and unliganded WLS, and perform structure-guided functional experiments. Wnts engage WLS via three conserved hairpins, which are all required for Wnt trafficking t

Photodynamic activity of a mitochondria-localized G-quadruplex-responsive fluorescent ligand: Cellular effects and photo-enhanced cytotoxicity.

Mitochondrial localization concentrates photosensitizing molecules near redox-sensitive organelles. In this study, we investigated SPN, a mitochondria-targeted G-quadruplex (G4)-responsive ligand. Photochemical characterization confirmed that SPN generates robust light-triggered ROS and exhibits photooxidative activity in vitro. In cancer cell models under dark conditions, SPN treatment was associated with cell-cycle perturbation, loss of mitochondrial membrane potential, altered mitochondrial morphology, elevated mitochondrial superoxide-sensitive fluorescence, and accumulation of MDC-positiv

Lactoferrin-Derived Peptides in Cancer Therapy: Structural Features, Mechanistic Insights and Clinical Translation Prospects.

Lactoferrin (LF)-derived peptides (LDPs) are short cationic and amphipathic fragments generated primarily from the N-terminal lobe of LF through pepsin-mediated proteolytic processes. The best-characterized LDPs include lactoferricin (LFcin), lactoferrampin (LFampin), and LF1-11. In addition to these native peptides, a growing range of engineered LDPs has been developed by modifying the LFcin-derived RRWQWR motif through the incorporation of non-natural amino acids, cyclization, multimerization, and conjugation with chemotherapeutic agents. LDPs have garnered significant interest as potential

Redox-Regulated Mechanisms of Cytotoxic Effect of Iron Nitrosyl Complex - Activator of Transcription Factor Nrf2 in Glioblastoma Cells In Vitro.

We performed a comprehensive in vitro study of a potential antiglioma compound from the class of NO donors, a nitrosyl iron complex with N-ethylthiourea (ETM), as an activator of redox-regulated mechanisms of cellular response. The effect of ETM on glioblastoma cells is accompanied by enhanced ROS generation, accumulation of intracellular NO, a decrease in mitochondrial membrane potential, and induction of caspase-3-dependent apoptosis. The studied complex acts as an activator of Nrf2 transcription factor, and the Nrf2/HO-1-dependent pathway plays a major role in regulating redox processes tri

Design, synthesis, and activity evaluation of novel arylpyrrolidinones NMT antifungal inhibitors.

N-Myristoyltransferase (NMT) is essential for fungal pathogenicity by regulating the membrane localization and biological functions of key signaling proteins. Based on this target, three series of novel arylpyrrolidinone-based NMT inhibitors were designed using a scaffold-growth strategy, synthesized, and biologically evaluated. Among them, compounds 8b, 8c, and 9b showed potent NMT inhibitory activity, with IC50 values ranging from 93.20 to 152.3 nM, and exhibited broad-spectrum antifungal activity against both wild-type and multidrug-resistant fungal isolates. Mechanistic studies indicated t

Methyltransferase CPmtA drives dormancy-associated synthesis of coproporphyrin III tetramethyl ether via a "carousel" mechanism in Mycobacterium smegmatis.

It was shown previously that mycobacteria (Mycobacterium smegmatis and Mycobacterium tuberculosis) accumulate tetramethyl ether of coproporphyrin III (TMC) during transition to dormant state. However, the enzymes involved in TMC synthesis remained unknown. Here we have identified a new M. smegmatis methyltransferase MSMEG_0614 (CPmtA) that catalyzes the S-adenosyl-L-methionine-dependent conversion of coproporphyrin (CP) into TMC through sequential methylation of its carboxyl groups. Overexpression of MSMEG_0614 in both M. smegmatis and M. tuberculosis leads to increase in intracellular TMC lev

Analysis of Intracellular Trafficking of Cholera Toxin B Subunits Depending on Ganglioside GM1 in Tissue-Resident Macrophages.

Cholera toxin (CT) is a bacterial exotoxin secreted by Vibrio cholerae. CT is comprised of one toxic A subunit (CTA) and five nontoxic B subunits (CTB). CTB is required for internalization of CT through binding with a membrane glycolipid ganglioside GM1. Previously, we clarified that CTB could induce production of proinflammatory cytokine interleukin-1β (IL-1β) from murine resident peritoneal macrophages (RPMs) in synergy with lipopolysaccharide (LPS) through a ganglioside GM1-dependent manner. Then, CTB was incorporated into endoplasmic reticulum (ER) depending on GM1 and activate ER stress s

Piperazine and Morpholine Protoporphyrin IX Derivatives Intensify Biomolecules Oxidation and Phototoxicity in Triple-Negative Breast Cancer.

Photodynamic therapy (PDT) is a clinically established approach that relies on the localized generation of reactive oxygen species (ROS) mediated by a photosensitizer (PS), light, and molecular oxygen. Protoporphyrin IX (PpIX) is an approved PS; however, its intracellular retention is limited, factor that can alter its overall photodynamic efficiency. In this study, we report how the design of PpIX derivatives bearing piperazine (PpIX-Pip) or morpholine (PpIX-Morp) substituents may modulate lipophilicity, cellular uptake, oxidative stress, and phototoxicity. The photodynamic activity of these

The E3 ubiquitin ligase Trip12 semi-selectively attenuates Wnt signaling.

Wnt signaling is critical for the development and maintenance of many cell lineages, including hematopoietic stem cells (HSCs). The tight regulation of Wnt signals is essential, as overactivation can drive tumorigenesis. Numerous Wnt ligands and Frizzled (Fzd) receptors exist, and the negative regulation of particular Fzd signals is the focus of this study. We previously identified the requirement of Wnt9a-Fzd9b pairing for early HSC proliferation. However, the mechanisms controlling activation and signal termination are unclear. Here, we show that the E3 ubiquitin ligase Trip12 (thyroid hormo

Lipidomic Signatures in Pediatric Metabolic Disorders.

Lipids are essential biomolecules involved in membrane structure, energy storage, and intracellular signaling. Dysregulation of lipid metabolism (dyslipidemia) plays a central role in a wide spectrum of pediatric metabolic disorders, including both inherited and acquired conditions. Recent and rapid advances in mass spectrometry-based lipidomics have enabled high-resolution profiling of more than one-thousand lipid species, facilitating the discovery of disease-specific lipid signatures that were previously undetectable with conventional biochemical assays. In parallel, the rising prevalence o

ZnO colludes with C. acnes in healing delay and Scar hyperplasia by barrier destruction.

As an important component of sunscreen products for sensitive skin, the potential damage mechanism of ZnO nanoparticles on skin surface with barrier structure or function defect caused by Cutibacterium acnes (C. acnes) has not been elucidated, which poses a serious challenge for reasonable selection of sunscreen products for acne-infected skin. In this work, we demonstrated for the first time that C. acnes induced significant changes in the membrane permeability and intracellular pH of fibroblasts through lipase up-regulation and lipid peroxidation, promoting endocytosis and ionization of ZnO

Nitrosoamine-3-Hydroxyflavothione Hybrids Enable Oxygen-Tolerant Photoredox Catalysis for Red-Light-Stimulated CO/NO Release for Combating Bacteria.

Single gas donors have proved to have vast therapeutic potential; however, the investigation of multigas releasing molecules is limited. Besides, porphyrins and their derivatives are essential energy and electron transfer agents for activating gaseous signaling molecule (GSM) donors, despite, the catalysis of the latter is greatly limited in oxygen-rich environments. We, therefore, report photosensitive small-molecule hybrids composed of oxo-/thio-flavonols as the singlet oxygen (1O2) scavengers for catalytic stability and N-nitrosoamine moieties for the competent co-release of carbon monoxide

Circulating FABP5 released by dying macrophages function as a DAMP to exacerbate septic inflammation.

Damage-associated molecular patterns (DAMPs) often cause an exaggerated immune response. Fatty acid-binding protein 5 (FABP5) is traditionally considered a cytosolic protein responsible for the transport of fatty acids. However, little is known about the role of FABP5 in sepsis. Herein, we found that circulating FABP5 levels were higher in patients with sepsis and were associated with adverse outcomes. The circulating FABP5 originated mainly from macrophage pyroptosis in the later stages of sepsis despite a decrease in cytoplasm-resident FABP5 expression. Functionally, circulating FABP5 penetr

Identification of α-Azacyclic Acetamide-Based Inhibitors of P. falciparum Na+ Pump (PfATP4) with Fast-Killing Asexual Blood-Stage Antimalarial Activity by Phenotypic Screening.

Malaria treatments are compromised by drug resistance, creating an urgent need to discover new drugs. We used a phenotypic high-throughput screening (HTS) platform to identify new antimalarials, uncovering three related pyrrole-, indole-, and indoline-based series with a shared α-azacyclic acetamide core. These compounds showed fast-killing activity on asexual blood-stage Plasmodium falciparum parasites, were not cytotoxic, and disrupted parasite intracellular pH and Na+ regulation similarly to cipargamin (KAE609), a clinically advanced inhibitor of the P. falciparum Na+ pump (PfATP4). Surface expression of antitoxin on engineered bacteria neutralizes genotoxic colibactin in the gut.

Colibactin, a metabolite produced by gut bacteria carrying the polyketide synthase (pks) island, is associated with host genotoxicity and tumorigenesis. However, no Food and Drug Administration-approved therapeutics directly target colibactin. Here we show that expression of the intracellular colibactin self-resistance protein (ClbS) on the surface of engineered bacteria shields the host from genotoxic effects across multiple pks+ isolates. The surface display, due to the fusion of ClbS with outer membrane protein A (ClbS-OmpA) in Escherichia coli, effectively reduced colibactin-induced DNA da
