Treatment of phantom shocks: A case report.

Implantable cardioverter-defibrillators have become standard preventive treatment for patients with ventricular arrhythmias and other life-threatening cardiac conditions. The advantages and efficiency of the device are supported by multiple clinical trials and outcome studies, leading to its popularity among cardiologists. Implantation of the device is not without adverse outcomes. Implantable cardioverter-defibrillator placement has been found to lead to negative psychological and psychosocial sequelae such as apprehension to engage in physical activity, chronic anxiety, decreased physical an

Effects of major depression on remission and relapse of substance dependence.

BACKGROUND: The effects of major depressive disorder (MDD) on the course of substance dependence may differ depending on the temporal relationship of depression to dependence. We investigated the effects of MDD on the outcome of substance dependence under 3 circumstances: (1) lifetime onset of MDD prior to lifetime onset of dependence onset, (2) current MDD occurring during a period of abstinence, and (3) current MDD during substance use that exceeded the expected effects of intoxication or withdrawal. METHODS: A sample of 250 inpatients with DSM-IV cocaine, heroin, and/or alcohol dependence

Astrocytes as a target of transcranial direct current stimulation (tDCS) to treat depression.

Transcranial direct current stimulation (tDCS) has been reported to be effective in treating mood disorders such as major depressive disorder, however, its detailed mechanism of action is not fully understood. Human and animal experiments have demonstrated that tDCS promotes brain plasticity and have suggested that this consequence may underlie its therapeutic benefits. Nonetheless, the specific neurobiological underpinnings of tDCS-induced brain plasticity have only recently begun to be investigated. While brain plasticity occurs in synapses formed by neurons, astrocytes, a major glial cell t

Socio-demographic and clinical risk factors of treatment-resistant depression: A Danish population-based cohort study.

BACKGROUND: Knowledge of risk factors of treatment-resistant depression (TRD) contributes to understand the underlying mechanisms and to identify patients at risk. However, there is still a lack of studies on how different risk factors associate with TRD. The objective of this study was to determine the independent association of several socio-demographic and clinical risk factors with TRD. METHODS: 194.074 patients with a first-time hospital contact for depression identified in the Danish National Patient Registry (DNPR) from 1996 through 2014 were followed for TRD for 12 months after diagno

MiR-129-5p alleviates depression and anxiety by increasing astrocyte ATP production partly through targeting deubiquitinase Mysm1.

Major depressive disorder (MDD) is a major global mental concern that severely affects quality of life, yet current pharmacological treatments remain limited in their effectiveness. Long-term chronic stress has been shown to increase the incidence of depression and anxiety. Micro RNAs (miRNAs) have been revealed to participate in the pathological process of depression and represent promising therapeutic targets. In this study, we found that microRNA-129-5p (miR-129-5p) was significantly decreased in the brains of depressive mice. Overexpression of miR-129-5p in the hippocampus effectively alle

Neural vulnerability to stress in adolescents: a longitudinal study using polyconnectomic scoring of depression risk.

BACKGROUND: Adolescence is a sensitive period of brain development when perceived stress can shape emotional and cognitive outcomes. However, it remains unclear how neural vulnerability modulates these effects. METHODS: In a longitudinal cohort of 407 adolescents, we assessed perceived stress, emotional symptoms, and cognitive function at baseline and follow-up. Neural vulnerability was indexed using polyconnectomic scoring for major depressive disorder (PCS-MDD), derived from large-scale functional connectivity patterns. An independent cohort of 80 adolescents with clinically diagnosed depre

Development of a novel HDAC6 PET imaging agent uncovers associations between HDAC6 overexpression and neuroinflammation in depression.

Histone deacetylase 6 (HDAC6) represents a compelling target in major depressive disorder (MDD) pathophysiology, yet in vivo investigation has been constrained by inadequate imaging capabilities. Here, we report the development and validation of [18F]PB200, a novel positron emission tomography (PET) radiotracer specifically targeting brain HDAC6. PB200 was engineered with nanomolar affinity, high HDAC6 selectivity, and excellent blood-brain barrier permeability. [18F]PB200 was successfully synthesized in a radiochemical yield of 13 ± 4 % and validated through in vitro autoradiography and in vi

The multilevel exploration test, a novel paradigm to measure exploratory behavior in depression animal models and the involvement of the PL-ZI circuit.

Diminished drive is one of the core symptoms of major depressive disorder (MDD) diagnosis, yet its underlying neural mechanisms remain elusive, primarily due to a lack of appropriate animal models. We developed a novel Multilevel Exploration Test (MET) apparatus to evaluate exploratory behavior, which is captured as a dynamic, stage-dependent process involving "search", "attend/investigate", and "approach" phases. We employed fiber photometry to measure real-time dopamine dynamics in the nucleus accumbens. We further combined cFos staining and neural circuit tracing to identify relevant brain

Beyond total depressive symptoms: Differential associations between depressive domains, HIV status and disease characteristics.

BACKGROUND: People with HIV (PWH) are at two times greater risk for major depressive disorder (MDD) than people without HIV (PWoH), which manifests in symptoms across cognitive, somatic, affective, apathy, and anhedonia domains that may differentially impact clinical outcomes. However, few studies have examined whether HIV and its characteristics relate to depressive symptom domains. METHODS: This secondary, cross-sectional analysis included 3456 participants enrolled in studies at the UCSD HIV Neurobehavioral Research Program and CHARTER sites between 2000 and 2023 (79 % PWH, 78 % male, Age:

Considering Comorbidities and Individual Differences in Testing a Gaming Behavioral Activation App for Perinatal Depression and Anxiety: Open Trial Pilot Intervention Study.

BACKGROUND: There is increasing interest in the development of scalable digital mental health interventions for perinatal populations to increase accessibility. Mobile behavioral activation (BA) is efficacious for the treatment of perinatal depression; however, the effect of comorbid anxiety and depression (CAD) on symptom trajectories remains underexplored. This is important given that at least 10% of women in the perinatal period experience CAD. OBJECTIVE: We assessed whether there were differences in symptom trajectories in pregnant participants with CAD as compared to those with depressio

Large responses to antidepressants or methodological artifacts? A secondary analysis of STAR∗D, a single-arm, open-label, nonindustry antidepressant trial.

OBJECTIVES: To replicate Stone et al's (2022) finding that the distribution of response in clinical antidepressant trials is trimodal with large, medium-effect, and small subgroups. METHODS: To apply finite mixture modeling to pre-post Hamilton Depression Rating Scale (HDRS) differences (n = 2184) of STAR∗D study's level 1, a single-arm, open-label study. For a successful replication, the best fitting model had to be trimodal, with comparable components as in Stone et al. Secondary/sensitivity analyses repeated the analysis for different baseline levels of depression severity, imputed values,

Explanatory and modifying factors of the association between sex and depression onset during adolescence: An exploratory study.

BACKGROUND: The prevalence of Major Depressive Disorder (MDD) is twice as high in women as in men and this difference already emerges during adolescence. Because the mechanisms underlying this sex-difference remain poorly understood, we took a bottom-up approach to identify factors explaining the sex-MDD relationship. METHODS: Data came from the TRacking Adolescents' Individual Lives Survey (TRAILS), a population study investigating youths' development from age 11 into adulthood. We assessed multiple baseline covariates (e.g., demographic, social and psychological) at ages 11-13 years and MDD

Intranasal delivery of paroxetine: A preclinical study on pharmacokinetics, depressive-like behaviour, and neurochemical sex differences.

Treatment of major depressive disorder remains a major unmet clinical need. Given the advantages of intranasal administration for targeted brain delivery, the present study aimed at investigating the pharmacokinetics of paroxetine, after its intranasal instillation and assessing its potential therapeutic effect on female and male mice subjected to unpredictable chronic mild stress (UCMS) protocol. IN administration revealed direct nose-to-brain paroxetine delivery but dose- and sex-dependent differences. Pharmacokinetics was nonlinear and paroxetine concentrations were consistently higher in p
