Platelet rich plasma (PRP) induces chondroprotection via increasing autophagy, anti-inflammatory markers, and decreasing apoptosis in human osteoarthritic cartilage.

OBJECTIVES: Autophagy constitutes a defense mechanism to overcome aging and apoptosis in osteoarthritic cartilage. Several cytokines and transcription factors are linked to autophagy and play an important role in the degradative cascade in osteoarthritis (OA). Cell therapy such as platelet rich plasma (PRP) has recently emerged as a promising therapeutic tool for many diseases including OA. However, its mechanism of action on improving cartilage repair remains to be determined. The purpose of this study is to investigate the effect of PRP on osteoarthritic chondrocytes and to elucidate the mec

Pyrroloquinoline quinone alleviates age-related osteoarthritis via nuclear factor erythroid 2-related factor 2-mediated stress response and insulin-like growth factor 1 receptor upregulation.

BACKGROUND: Age-related knee osteoarthritis (OA) arises from cumulative oxidative damage, chondrocyte senescence and extracellular matrix loss; yet safe and effective disease‑modifying interventions for aging‑associated OA are lacking. Pyrroloquinoline quinone (PQQ; molecular formula C14H6N2O8) is a naturally bioactive compound that has been reported to activate nuclear factor erythroid 2-related factor 2 (Nrf2), a transcription factor that regulates antioxidant and cytoprotective gene expression. However, its effects on age-related OA and the underlying mechanisms remain unclear. METHODS: Tw

Loss of mechanical stress induces synovitis, fibrosis and articular cartilage degeneration via distinct synovial cell subsets.

Joint function is impaired by disuse, as well as overuse. However, the underlying mechanisms remain unclear. Here, we elucidate the mechanisms of synovial and cartilage changes using a minimized mechanical stress (MMS) mouse model by combining knee joint immobilization and unloading. In this model, synovitis appeared by day 3, followed by subsequent fibrosis leading to joint contracture within two weeks. In contrast, articular cartilage degeneration developed gradually after the synovial alterations. Notably, synovial changes were attenuated by discontinuation of joint immobilization, while ca

CCR1+ monocytes facilitating bronchopulmonary dysplasia through regulation of S100A8 and MMP8.

Bronchopulmonary dysplasia (BPD) is the most common chronic lung disease in preterm infants and is characterized by aberrant late-stage lung development driven by multiple factors. Although previous studies have demonstrated that BPD is associated with prenatal and postnatal inflammatory stimuli, the inflammatory mechanisms involved immune cell populations have not been fully elucidated. Here, we systematically investigated the roles of myeloid cells in the inflammatory processes underlying BPD. Using single-cell transcriptomic data of normal and BPD mouse lung, we found that the proportions o

Identification of Differential Fibrocartilage Degradation Between Sexes in a Burn-Synovectomy Temporomandibular Osteoarthritis Mouse Model.

BACKGROUND: Temporomandibular joint osteoarthritis is a degenerative disease with a higher prevalence in women, yet its sex-specific mechanisms remain unclear. This study investigates fibrocartilage degradation in a burn-synovectomy TMJ OA mouse model, hypothesizing that female mice exhibit greater cartilage degradation due to differential inflammatory responses. METHODS: PRG4-CreERT2; Ai9 mice (12-week-old males and females) underwent burn-synovectomy injuries and were euthanized at 1, 3, and 9 weeks for histological and immunofluorescence analyses. PRG4 and MMP-13 expression were assessed t

Nano-Drug Delivery Systems Targeting MMPs: A Promising Treatment for Gliomas.

Gliomas are the most prevalent Central Nervous System (CNS) tumors. Among them, glioblastoma (grade IV) is the most challenging brain cancer because of its highly aggressive nature, treatment resistance and poor prognosis. Matrix metalloproteinase (MMP) is a family of zinc-dependent protein hydrolases. In recent years, MMPs have become a research focus owing to their central role in tumor microenvironment remodeling, angiogenesis, invasion, metastasis. Clinical studies have shown that the expression levels of MMPs in glioma tissues exhibit a significant positive correlation with the degree of
