Human Cytomegalovirus pUL11, a CD45 Ligand, Disrupts CD4 T Cell Control of Viral Spread in Epithelial Cells.

Human cytomegalovirus (HCMV) encodes numerous immunomodulatory genes that facilitate its persistence. Previously described mechanisms by which HCMV avoids T cell control typically involve evasion of detection by infected cells. Here, we show that the virus also inhibits T cells directly via an interaction between the pUL11 glycoprotein on infected cells and the CD45 phosphatase on T cells. The antiviral functions of CD4 T cells are impaired as a result of this interaction, largely via induced interleukin 10 (IL-10) secretion in the CD4 T cell central memory compartment, resulting in enhanced v

MicroRNAs as Potential Orchestrators of Alzheimer's Disease-Related Pathologies: Insights on Current Status and Future Possibilities.

Alzheimer's disease (AD) is a progressive and deleterious neurodegenerative disease, strongly affecting the cognitive functions and memory of seniors worldwide. Around 58% of the affected patients live in low and middle-income countries, with estimates of increasing deaths caused by AD in the coming decade. AD is a multifactor pathology. Mitochondrial function declines in AD brain and is currently emerging as a hallmark of this disease. It has been considered as one of the intracellular processes severely compromised in AD. Many mitochondrial parameters decline already during aging; mitochondr

Detection of Children/Youth With Fetal Alcohol Spectrum Disorder Through Eye Movement, Psychometric, and Neuroimaging Data.

Background: Fetal alcohol spectrum disorders (FASD) is one of the most common causes of developmental disabilities and neurobehavioral deficits. Despite the high-prevalence of FASD, the current diagnostic process is challenging and time- and money- consuming, with underreported profiles of the neurocognitive and neurobehavioral impairments because of limited clinical capacity. We assessed children/youth with FASD from a multimodal perspective and developed a high-performing, low-cost screening protocol using a machine learning framework. Methods and Findings: Participants with FASD and age-mat

Causal role of immune cells in major depressive disorder and bipolar disorder: Mendelian randomization (MR) study.

BACKGROUND: Major depressive disorder (MDD) and bipolar disorder (BD) are prevalent psychiatric conditions linked to inflammatory processes. However, it is unclear whether associations of immune cells with these disorders are likely to be causal. METHODS: We used two-sample Mendelian randomization (MR) approach to investigate the relationship between 731 immune cells and the risk of MDD and BD. Rigorous sensitivity analyses are conducted to assess the reliability, heterogeneity, and horizontal pleiotropy of the findings. RESULTS: Genetically-predicted CD27 on IgD+ CD38- unswitched memory B c

Impact of Repetitive Mild Traumatic Brain Injury on Behavioral and Hippocampal Deficits in a Mouse Model of Chronic Stress.

Clinical studies examining the interaction between traumatic brain injury (TBI) and stress-related disorders (e.g., post-traumatic stress disorder) are often complicated by methodological constraints, such as heterogeneity in injury type and severity, time post-trauma, and predisposing risk factors. Developing relevant animal models whereby many variables can be efficiently controlled is thus essential to understanding this elusive relationship. Here, we use our repeated unpredictable stress (RUS) paradigm, in combination with our established mouse model of repetitive mild TBI (r-mTBI), to ass

Delusional disorder and its differentiation from schizophrenia: A narrative review.

The nosological status and psychopathology of delusional disorder have been a subject of debate since Kraepelin distinguished it from schizophrenia and affective psychoses. Contemporary diagnostic manuals define delusional disorder primarily by the presence of delusions, offering limited guidance for its differentiation from schizophrenia. Notably, DSM-5 introduced a major, seemingly unexplained change by allowing bizarre delusions in delusional disorder, contrary to ICD-11, prior DSM editions, and classical descriptions. This narrative review revisits the seminal works of six classical author

The transcriptomics profiling of blood CD4 and CD8 T-cells in narcolepsy type I.

BACKGROUND: Narcolepsy Type I (NT1) is a rare, life-long sleep disorder arising as a consequence of the extensive destruction of orexin-producing hypothalamic neurons. The mechanisms involved in the destruction of orexin neurons are not yet elucidated but the association of narcolepsy with environmental triggers and genetic susceptibility (strong association with the HLA, TCRs and other immunologically-relevant loci) implicates an immuno-pathological process. Several studies in animal models and on human samples have suggested that T-cells are the main pathogenic culprits. METHODS: RNA sequen

Anomalous prefrontal-subcortical activation in familial pediatric bipolar disorder: a functional magnetic resonance imaging investigation.

BACKGROUND: The neurobiological features of pediatric bipolar disorder (BD) are largely unknown. Children and adolescents with BD may be important to study with functional neuroimaging techniques because of their unique status of early-onset BD and high familial loading for the disorder. Neuroimaging studies of adults with BD have implicated the dorsolateral prefrontal cortex (DLPFC) and anterior cingulate cortex (ACC) in the development of this disorder. OBJECTIVES: To study children and adolescents with BD via functional magnetic resonance imaging using cognitive and affective tasks and to

Disintegrating ipseity: a gerontological exegesis of Alzheimer's disease and relational identity in Alice Munro's fiction.

BACKGROUND: Alzheimer's is a progressive disorder of the brain that gradually affects memory, personality, behaviour, identity, and interpersonal relationships, particularly among older adults. Alice Munro's novella "The Bear Came Over the Mountain" offers a compelling literary framework for exploring these impacts through narrative gerontology, a lens that emphasizes storytelling's role in shaping identity and relational bonds during ageing and illness. METHOD: This study applies narrative gerontology to analyze Munro's depiction of Alzheimer's, focusing on the experiences of Fiona and Grant

Decrement in sustained attention and working memory in young, treatment-naïve women with polycystic ovary syndrome: Preliminary findings.

Objective: Polycystic ovary syndrome (PCOS) is a complex endocrine disorder associated with hyperandrogenism and insulin resistance. Although its reproductive and metabolic consequences are well established, research on the impact of PCOS on cognitive function remains inconsistent. This study aimed to evaluate the cognitive profiles across multiple domains in young, treatment-naïve patients with PCOS. Method: This cross-sectional study included 35 patients with PCOS (aged 18-35 years) and 30 age-matched healthy controls from Northern India. Participants underwent standardized computer-based ps

Differential effects of Lactobacillus casei in probiotic and paraprobiotic forms on behavioral performance, oxidative stress, neuroinflammation, and cholinergic dysfunction in a streptozotocin-induced model of sporadic Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline, oxidative stress, neuroinflammation, and cholinergic dysfunction. Increasing evidence suggests that gut microbiota disturbances contribute to AD progression, encouraging the investigation of microbiota-modulating approaches such as probiotics and paraprobiotics. This study evaluated the effects of Lactobacillus casei probiotic and its thermally inactivated paraprobiotic on cognitive, behavioral, oxidative, cholinergic, and inflammatory alterations in a sporadic AD model induced by intracere

Effects of quetiapine on cognitive functioning in schizophrenia: evidence for the remyelination hypothesis?

Postmortem findings, neuroimaging data, and in-vitro models suggest a decrease in number and density of oligodendrocytes is driving cognitive deficits in schizophrenia (SCZ). Second-generation antipsychotics are discussed to improve oligodendrocyte dysfunction with most conclusive evidence available for quetiapine (QET). We postulate that sustained QET treatment leads to cognitive improvement in SCZ, particularly, in tests with high demands for working memory function. We further hypothesize that these effects are moderated by polygenic factors associated with hippocampus-related brain volumes

Cannabidiol attenuates seizure progression and recognition memory deficit induced by hippocampal HCN1 knockdown in the kindling model of epilepsy in male rats.

Epilepsy is a neurological disorder characterized by excessive neuronal firing, frequently originating in the hippocampus. Hyperpolarization-activated cyclic nucleotide-gated channel-1 (HCN1) regulates neuronal excitability and resting membrane potential, yet its role in seizure progression remains unclear. Cannabidiol (CBD), an effective anticonvulsant, may exert part of its effects through HCN1. This study investigated the contribution of HCN1 to seizure progression, synaptic plasticity, and CBD-mediated neuroprotection. Rats were implanted with stimulation electrodes in the perforant path (

Peripheral metabolic dysfunction is associated with cognitive frailty in a mouse model of type 2 diabetes.

Type 2 diabetes mellitus (T2DM) is a chronic metabolic disorder increasingly associated with cognitive impairment and neurodegeneration. Peripheral metabolic dysfunction, particularly insulin resistance and glycation stress, represents a critical link between diabetes and brain pathology. This study investigated the relationship between metabolic alterations and cognitive frailty using a murine T2DM model induced by a high-fat diet (HFD) combined with low-dose streptozotocin (STZ). Male C57BL/6J mice were longitudinally followed from adulthood to senescence and subjected to HFD alone or HFD pl

Targeted high-definition transcranial direct current stimulation over the left dorsolateral prefrontal cortex to modify cognitive biases in young adults with social anxiety: a randomized controlled trial.

Cognitive model suggests that social anxiety disorder (SAD) is maintained by persistent biases in attention, interpretation, and memory. Neuroscientific perspectives further implicate that hyperactivation of amygdala and default mode network, alongside prefrontal cortex dysregulation may underlie the neural mechanisms of SAD. This study is the first to investigate the effects of offline high-definition transcranial direct current stimulation (HD-tDCS) on multiple cognitive biases in young adults with social anxiety. Seventy-four university students with elevated social anxiety were randomly as

Pre-treatment subjective sleep quality as a predictive biomarker of tDCS effects in preclinical Alzheimer's disease patients: Secondary analysis of a randomised clinical trial.

BACKGROUND: Despite transcranial direct current stimulation (tDCS) has demonstrated encouraging potential for modulating the circadian rhythm, little is known about how well and sustainably tDCS might improve the subjective sleep quality in older adults. This study sought to determine how tDCS affected sleep quality and cognition, as well as how well pre-treatment sleep quality predicted tDCS effects on domain-specific cognitive functions in patients with mild neurocognitive disorder due to Alzheimer's disease (NCD-AD). METHODS: This clinical trial aimed to compare the effectiveness of tDCS a
