Haem homeostasis is regulated by the conserved and concerted functions of HRG-1 proteins.

Haems are metalloporphyrins that serve as prosthetic groups for various biological processes including respiration, gas sensing, xenobiotic detoxification, cell differentiation, circadian clock control, metabolic reprogramming and microRNA processing. With a few exceptions, haem is synthesized by a multistep biosynthetic pathway comprising defined intermediates that are highly conserved throughout evolution. Despite our extensive knowledge of haem biosynthesis and degradation, the cellular pathways and molecules that mediate intracellular haem trafficking are unknown. The experimental setback

Bidirectional Hypoxic Extracellular Vesicle Signaling Between Müller Glia and Retinal Pigment Epithelium Regulates Retinal Metabolism and Barrier Function.

The retina is highly sensitive to oxygen and blood supply, and hypoxia plays a key role in retinal diseases such as diabetic retinopathy (DR) and age-related macular degeneration (AMD). Müller glial cells, which are essential for retinal homeostasis, respond to injury and hypoxia with reactive gliosis, characterized by the upregulation of the glial fibrillary acidic protein (GFAP) and vimentin, cellular hypertrophy, and extracellular matrix changes, which can impair retinal function and repair. The retinal pigment epithelium (RPE) supports photoreceptors, forms part of the blood-retinal barrie

BACKGROUND: Pelvic inflammatory disease (PID) is a prevalent gynecological infection, that poses significant therapeutic challenges. Sargentodoxa cuneata and Patrinia villosa (S&P) demonstrate empirical efficacy in PID treatment within traditional Chinese medicine practice. However, their active components and precise anti-inflammatory mechanisms should be systematically elucidated. PURPOSE: This study aimed to investigate the pharmacologically active components of S&P and decipher their anti-inflammatory mechanism through integrated multi-omics approaches. MATERIALS AND METHODS: CCK-8 and N

Epstein-Barr Virus Latent Membrane Protein 1 Subverts IMPDH pathways to drive B-cell oncometabolism.

UNLABELLED: Epstein-Barr virus (EBV) is associated with multiple types of cancers, many of which express the key viral oncoprotein Latent Membrane Protein 1 (LMP1). LMP1 is the only EBV-encoded protein whose expression is sufficient to transform both epithelial and B-cells. Although metabolism reprogramming is a cancer hallmark, much remains to be learned about how LMP1 alters lymphocyte oncometabolism. To gain insights into key B-cell metabolic pathways subverted by LMP1, we performed systematic metabolomic analyses on B cells with conditional LMP1 expression. This approach highlighted that L

Targeting PFKFB3 to enhance CDK4/6 inhibitor response in ER+ breast cancer.

BACKGROUND: Cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitors are widely used in the treatment of estrogen receptor-positive (ER⁺) breast cancer; however, the metabolic adaptations induced by CDK4/6 inhibition remain incompletely defined. In ER⁺ breast cancer, estrogen signaling plays a central role in coordinating cell cycle progression and metabolic programs that support tumor growth. Glycolytic flux is regulated at the level of phosphofructokinase-1 (PFK1) through the inducible enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3), which is transcriptionally regulated by

Endogenous CD28 Drives the Persistent Activity of CAR T Cells in Myeloma and Lymphoma Models.

UNLABELLED: Chimeric antigen receptor (CAR) T-cell therapy has reshaped the therapeutic landscape for multiple myeloma, yet most patients treated with BCMA-targeted CAR T cells experience disease relapse. Consequently, we sought to determine if inhibition of CD28 survival signaling could increase multiple myeloma sensitivity to CAR T-cell therapy. Contrary to expectations, blockade of CD28 interaction with CD80/86 accelerated tumor regrowth in preclinical multiple myeloma and lymphoma CAR T-cell therapy models. Knockout studies revealed that endogenous CD28 on 4-1BB costimulated CAR T cells pr

HOXA9 drives lymphatic metastasis by activating the c-MYC-glycolysis-lactate axis in gastric cancer.

BACKGROUND: The molecular mechanisms underlying lymph node metastasis (LNM) in gastric cancer (GC) remain poorly understood. This study investigated HOXA9’s role in driving LNM via metabolic reprogramming. METHODS: Integrated analysis of gastric cancer RNA sequencing data and clinical specimens was performed. Functional validation involved HOXA9 overexpression and knockdown in AGS and HGC-27 cell lines, c-MYC silencing by siRNA, and glycolytic inhibition using 2-deoxyglucose (2-DG, 2.5 mM). In vitro assays evaluated proliferation (CCK-8), apoptosis (Annexin V/PI), migration/invasion (Transwell

GSC State Plasticity Drives Therapeutic Resistance and Recurrence in Glioblastoma.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults, and treatment failure is closely linked to the presence of GBM stem cells (GSCs). Rather than representing a fixed marker-defined state, GSC identity is better understood as a dynamic and reversible stem-like state. GBM cells exhibit transcriptional plasticity, most clearly reflected by the proneural-to-mesenchymal transition, although direct interconversion among neural progenitor cell-like, oligodendrocyte precursor cell-like, astrocyte-like, and mesenchymal-like states remains unproven. GBM cells can also transition bi

Regulating the iron-ROS cycle via electronic structure-engineered nanoregulators for Parkinson's disease therapy.

Iron-driven reactive oxygen species (ROS) amplification forms a self-sustaining pathological cycle in Parkinson's disease (PD), yet remains difficult to therapeutically interrupt due to the lack of strategies that coordinate metal homeostasis and redox regulation. Here, we report an electronic-structure-engineered nanoregulatory strategy that regulates the iron-ROS cycle rather than passively scavenging ROS. Nitrogen-doped, herbal-derived carbon dots (CD) were rationally constructed to establish a conjugated electronic framework, in which heteroatom-induced electron delocalization defined redo

Targeting tumor-associated bacteria in digestive system cancers: carcinogenic mechanisms and nano-regulate platform design.

Tumor-associated bacteria (TAB) and their products can reprogram host signaling and metabolism, disrupt tissue homeostasis, and remodel antitumor immunity, collectively creating a cooperative protumor ecosystem. Accumulating evidence indicates that TAB promote the initiation and progression of digestive system malignancies and can also influence therapeutic outcomes. Although conventional bacterial modulation approaches can alter the composition and functions of TAB, their clinical application is hindered by major limitations, including poor in vivo stability, limited targeting specificity, di

Entorhinal cortex astrocytes in early Alzheimer disease: Human evidence, cross-species circuit rationale, and translational boundaries.

Alzheimer disease (AD) pathology affects the transentorhinal/entorhinal region early but whether entorhinal cortex (EC) astrocytes are causal drivers or therapeutic targets remains unresolved. This narrative review evaluates EC astrocytes through an explicit evidence hierarchy. We distinguish: (1) human EC-specific neuropathology, imaging, and transcriptomic findings, (2) human AD astrocyte data lacking EC-cell resolution, (3) mechanistic evidence from animal and cellular models, and (4) translational hypotheses. Human data support early EC vulnerability and region- and stage-dependent astrocy

Integrin α7 defines a profibrotic adipose stromal population targeted for nanoparticle PAI-1 gene silencing in obesity.

Obesity-associated metabolic dysfunction is driven not only by adipocyte expansion but also by pathological remodeling of adipose tissue mediated by distinct stromal cell subpopulations. However, the lack of strategies to selectively interrogate and therapeutically modulate these pathogenic cells has limited clinical progress. Here, we identify integrin α7 (ITGA7)-expressing adipose stromal cells (ASCs) as a proinflammatory and profibrotic population that contributes to adipose tissue dysfunction in obesity and introduce a targeted nanotherapy to selectively reprogram their inflammatory signal

CMGC Kinases in Viral Infection and Human Disease.

Cellular processes rely heavily on protein phosphorylation, a mechanism essential for organismal physiology and pathology. The CMGC family comprises a large group of serine/threonine kinases defined by a conserved catalytic core and closely related kinase domains. While several CMGC members have been extensively studied, others, including the RCK and CDKL subfamilies, remain less studied. Here, we synthesize current knowledge of CMGC kinases, emphasizing their structural organization, mechanisms of activation, and roles in infection and disease. CMGC kinases such as CDKs and DYRKs are activate

Cyclins and Cyclin-Dependent Kinases: Structure, Biological Functions, and Innovative Targeting Strategies in Cancer.

Cyclins and cyclin-dependent kinases (CDKs) are frequently dysregulated in human cancers and represent compelling therapeutic targets. Beyond their well-recognized roles in cell cycle control, CDK/cyclin complexes orchestrate diverse oncogenic processes, including transcription, genome maintenance, epigenetics, metabolism, and immune regulation. Deciphering the multifaceted biology of CDK/cyclin will provide valuable insights and rationales for the development of CDK/cyclin-targeting strategies and modalities. While the clinical success of CDK4/6 inhibitors has validated CDKs as druggable targ

Isolate-Specific Modulation of Growth, Carbon Allocation, and Transcriptomic Responses in Spirodela polyrhiza by Duckweed-Associated Bacteria.

Duckweed-associated plant growth-promoting bacteria have attracted attention for their potential to enhance duckweed biomass production. However, the mechanisms underlying isolate-specific growth promotion are unclear. This study compared the effects of two duckweed-associated bacterial isolates, Terrimicrobium sp. PS02 and Aeromicrobium sp. PS05, on the growth, biomass composition, colonization behavior, and transcriptomic responses of Spirodela polyrhiza. Biomass composition and transcriptome analyses were performed to characterize the host responses. Both isolates enhanced duckweed biomass;

Probiotics-Armed Cationic Photosensitizer for Combating Chronic Wound Infections with Multidrug-Resistant Bacteria Through Dismantling Biofilms and Reprogramming Hostile Wound Milieu.

Chronic wound infections caused by multidrug-resistant (MDR) bacteria present a serious global health challenge. Clinical treatment outcomes are significantly impeded by two critical pathological barriers: stubborn bacterial biofilms that block antimicrobial penetration, and a highly oxidative, inflammatory microenvironment that hinders tissue regeneration. To overcome these challenges, we developed a novel integrated therapeutic platform to concurrently dismantle biofilms and reprogram the hostile wound milieu, thereby enabling synergistic healing of chronic wound. This platform, LGG@TBTCP-BO

Diving into AA9-mediated transglutaminase 2 inhibition reveals ferroptosis as driver of anticancer effects.

Ferroptosis recently gained considerable attention in breast cancer research, particularly in triple-negative subtypes. In this context, emerging evidence links transglutaminase 2 (TG2) to aggressive tumour phenotypes, highlighting pharmacological TG2 inhibition as a promising therapeutic approach in aggressive breast cancer settings. Comparative analysis of independent RNA-sequencing datasets from distinct cellular models treated with irreversible TG2 inhibitors revealed ferroptosis as the only significantly enriched and commonly deregulated process. In triple-negative breast cancer cells, tr

Synthetic microbial community-assisted regulation of arsenic transport and metabolism reduces grain arsenic accumulation in rice across developmental stages.

Arsenic contamination in paddy soils threatens crop productivity and results in the accumulation of toxic arsenic in rice grains. Here, we demonstrate that a defined synthetic microbial community (SynCom) of Priestia flexa and Pseudomonas putida mitigates arsenic toxicity and restricts arsenic accumulation in rice (Oryza sativa var. Sarju-52). Under arsenic stress [As(III), 18 mg/kg; As(V), 50 mg/kg], plants exhibited impaired growth, reduced photosynthetic performance, and increased oxidative stress. Inoculation with SynCom restored physiological function and metabolic balance, as evidenced b

Epigenetic regulation of nephrogenesis: from Waddington's landscape to adult disease.

The establishment of lifelong nephron endowment depends on the tightly coordinated regulation of nephron progenitor cell (NPC) self-renewal, lineage specification, and differentiation during kidney development. Conrad H. Waddington's developmental landscape provides a powerful conceptual framework for understanding how NPCs navigate sequential fate decisions toward mature renal epithelial identities. Within this paradigm, the topology of the landscape is actively shaped by epigenetic mechanisms, including DNA methylation, histone posttranslational modifications, noncoding RNA-mediated gene reg
