Rhabdomyosarcoma (RMS) is the most common soft tissue malignancy in children and adolescents and is characterized by expression of myogenic regulatory factors without terminal differentiation. Treatment of RMS remains challenging, with nearly one-third of patients experiencing relapse due to chemotherapy resistance. However, the molecular mechanisms underlying RMS growth and chemoresistance remain poorly understood. Fibroblast growth factor-inducible 14 (FN14), a receptor for the TWEAK cytokine, has been implicated in tumor progression in several cancers, but its role in RMS is unknown. Here,

Cholangiocarcinoma (CCA) is a highly aggressive malignancy with limited treatment options and frequent chemoresistance. Peanut hairy root culture crude extract (PCE) has previously demonstrated selective cytotoxicity toward CCA cells. Here, we investigated the combinatorial effects of PCE with standard chemotherapeutic agents on CCA cell lines. In KKU-213 A cells, PCE exhibited antagonistic and additive interactions with gemcitabine (Gem) and 5-fluorouracil (5-FU), respectively. In contrast, both combinations showed synergistic effects in KKU-100 cells, with the strongest synergy observed for

Metastasis remains a major therapeutic challenge in hepatocellular carcinoma (HCC), yet its underlying molecular mechanisms are not fully understood. This study reveals that ZCCHC4 acts as a key promoter of HCC metastasis. We demonstrate that ZCCHC4 enhances the metastatic capacity of HCC cells both in vitro and in vivo. Mechanistically, ZCCHC4 upregulates key lipid biosynthesis enzymes (HMGCR, SQLE, FASN, and SCD), leading to intracellular accumulation of cholesterol and fatty acids which drive metastasis. Concurrently, ZCCHC4 transcriptionally activates TMEM97, thereby augmenting Wnt signali

Gastric cancer (GC) is a very aggressive cancer with a high potential for metastasis. A naturally occurring flavonoid, luteolin (Lut) has metabolic regulation and anti-cancer properties. This work investigates whether Lut modulates glucose metabolism and targets AKR1B1 to prevent the epithelial-mesenchymal transition (EMT) in GC. Lut was used to treat human GC cell lines that were grown in both normal and high-glucose environments. Western blotting and qPCR were used to assess the expression levels of EMT markers and AKR1B1. The CCK-8 assay, the scratch assay, and the Transwell assay were used

KRAS G12S mutations in non-small cell lung cancer (NSCLC) remain refractory to current targeted therapies, with few clinical options and frequent resistance. While CRISPR/Cas9 enables mutation-specific gene disruption, its pulmonary application is limited by systemic clearance, hepatic tropism, and airway mucus barriers. Here, we present lipid nanoparticles (LNPs) specifically engineered for pulmonary delivery of Cas9 mRNA and KRAS G12S-targeting sgRNA, optimized through mRNA surrogate screening and orthogonal mixture design to guide lipid composition and Cas9:sgRNA weight-to-weight ratios. BACKGROUND: Glioblastoma (GBM) remains highly lethal, and resistance to temozolomide (TMZ) is a major barrier to durable therapeutic response. Although MGMT-mediated DNA repair is a well-established mechanism of TMZ resistance, how GBM cells maintain proteostasis under sustained chemotherapy stress remains less defined.
METHODS: We analyzed RUVBL1 expression and clinical relevance in glioma datasets from TCGA and validation cohorts. Functional studies were performed using RUVBL1 knockdown, TMZ-resistant GBM cells, proteasome activity assays, protein aggregation analysis, ER stress assessment,

Liposomes represent versatile drug delivery shuttles in clinics for cancer therapy. Nevertheless, traditional PEG-modified liposomes encounter difficulties, including (1) poor blood-brain barrier (BBB) transcytosis and tumor targeting without ligand-decoration; (2) accelerated blood clearance (ABC) resulting from anti-PEG antibodies and complement proteins. To overcome these challenges, we employed a ligand-free, BBB-permeable, and glioblastoma (GBM)-targeting zwitterionic polyphosphorylcholine (PMPC)-modified liposomal formulation for siRNA delivery (PMPC-Lipo@siRNA). PMPC-modified formulatio

BACKGROUND: Acquired resistance to osimertinib, a third-generation EGFR tyrosine kinase inhibitor, remains a major clinical challenge in the treatment of non-small cell lung cancer (NSCLC). Although circular RNAs (circRNAs) have been increasingly implicated in drug resistance, most studies have focused on their canonical role as microRNA sponges, while their capacity to encode functional micropeptides remains largely unexplored. This study aimed to identify novel circRNAs involved in osimertinib resistance and to characterize their regulatory functions at the protein level.
METHODS: Osimertin

Nicotinamide adenine dinucleotide (NAD+) plays an important role in tumor progression, but its role in non-small cell lung cancer with brain metastasis (NSCLC BM) remains unclear. Herein, we investigated NAD+ biosynthesis targeting as a new therapeutic strategy for NSCLC BM. Therapeutic activity of nicotinamide phosphoribosyl transferase (NAMPT) inhibitors was evaluated in mouse models of NSCLC BM and using various assays such as NAD+ quantitation, cell viability, and apoptosis assays. To explore impact on downstream signaling, RNA sequencing was used in NAMPT inhibitor-treated and control cel

KRASG12D mutation is a prevalent gain-of-function mutation that drives pancreatic cancer tumorigenesis, but the underlying mechanisms that promote KRAS-induced cell proliferation and tumor formation remain elusive. To uncover the molecular pathways that facilitate KRASG12D-driven malignant transformation, we measured the transcriptomic alterations at various time points after induction of KRASG12D expression in human pancreatic normal epithelial cells. KEGG pathway enrichment of the differentially expressed genes (DEGs) showed that the major DEGs were located in pathways that regulate nicotina

Pancreatic cancer (PC) is a highly malignant and lethal tumor in gastrointestinal tract. Lactate accumulation is a classical feature of metabolic reprogramming in cancers. Lactate-derived lysine lactylation (Kla) is identified as a new type of post-translational modifications (PTMs), which is confirmed to be involved in multiple biological processes. However, the cancer-specific regulation of protein Kla in PC requires further elucidation. Here, we report a range of dysregulated Kla sites specifically related to RNA splicing in human pancreatic cancer, leading to the observation that the Kla a

Cholangiocarcinoma (CCA), a highly aggressive form of cancer, is known for its high mortality rate. A Disintegrin and Metalloprotease Domain-like Protein Decysin-1 (ADAMDEC1) can promote the development and metastasis in various tumors by degrading the extracellular matrix. However, its regulatory mechanism in CCA remains unclear. Public databases and clinical tissue samples were used to evaluate whether ADAMDEC1 expression was correlated with the prognosis of CCA. We investigated the expression of ADAMDEC1-related regulatory genes and proteins in CCA and assessed the biological behaviors of C

The resistance of oestrogen receptor-positive (ER+) breast cancer to tamoxifen (TAM) therapy represents a significant challenge in the clinical management of ER + breast cancer. It has been demonstrated that tamoxifen-resistant breast cancer is sensitive to ferroptosis. Consequently, the targeted intervention of Solute Carrier Family 7 Member 11 (SLC7A11) to promote ferroptosis represents a promising means of treating this form of cancer. Cryptotanshinone (CTS), a fat-soluble diterpene derivative extracted from Salvia miltiorrhiza, has been demonstrated to possess favorable anti-breast cancer

Hepatocellular carcinoma (HCC), the most common primary liver cancer, faces treatment challenges due to drug resistance and poor bioavailability, with sorafenib, a key therapy, characterized by rapid clearance and significant side effects. This paper describes the development of amphiphilic graft copolymers for efficient loading and delivery of sorafenib through controlled Atom Transfer Radical Polymerization (ATRP). The amphiphilic graft copolymer PHEA-g-IB-(pButMA)-g-PEG-Bt was synthesized to enhance tumor specificity via biotin-mediated targeting. The synthesis involved a three-step process

Cisplatin (CDDP) resistance remains a major clinical challenge in the treatment of head and neck squamous cell carcinoma (HNSC). Our group identified ATPase copper transporting beta (ATP7B) as a mediator of CDDP resistance through its role in drug efflux and small extracellular vesicle (sEV) secretion. Herein, we uncovered a novel mechanism by which ATP7B regulates sEV dynamics and the intercellular transmission of CDDP resistance. Using transcriptomic analyses of HNSC datasets, we demonstrate that ATP7B expression correlates with endocytosis- and epithelial-mesenchymal transition (EMT)-relate

In recent years, there has been a rapid increase in the incidence of thyroid carcinoma (TC). Our study focuses on the regulatory effect of circular RNAs on metabolism of TC, aiming to provide new insights into the mechanisms of progression and a potential therapeutic target for TC. In this study, we identified high expression levels of circPSD3 in TC tissues through RNA sequencing. Papillary thyroid cancer tissue cohorts verified the circPSD3 expression level was positively correlated with larger tumor size. circPSD3 promoted the proliferation of TC cells and reduced apoptosis both in vitro an

Cuproptosis, a recently identified regulated cell death pathway, is emerging as a promising therapeutic target for cancer. However, nonsmall cell lung cancer (NSCLC) exhibits inherent resistance to copper-induced toxicity, which is primarily due to enhanced glycolytic activity. Herein, a coordination polymer (Cu-Ox@HA) is designed by the chelation of copper ions with the glycolysis inhibitor oxamate (Ox), using hyaluronic acid (HA) as a biocompatible template. In the tumor microenvironment, Cu-Ox@HA disassembles in response to glutathione (GSH), enabling the synchronous release of copper ions
