Interleukin-33 enhances ASIC currents in mouse dorsal root ganglion neurons.

Cytokine interleukin-33 (IL-33) signaling in primary sensory neurons plays a crucial role in pain. However, the underlying molecular mechanisms remain poorly understood. Therefore, we investigated whether IL-33 signaling affects ion channels in nociceptive dorsal root ganglion (DRG) neurons. Herein, we reported that the application of IL-33 enhanced the electrophysiological activity of acid-sensing ion channels (ASICs). IL-33 dose-dependently increased acid-evoked ASIC currents in mouse DRG neurons. IL-33 enhanced the maximum responses of ASICs, whereas the sensitivity to acidic stimuli remain

DIRAS2 modulates MAPK pathway-mediated ferroptosis to regulate excitation/inhibition balance and seizure susceptibility.

Epilepsy is a common neurological disorder that is widely believed to be associated with an imbalance between neuronal excitation and inhibition (E/I). DIRAS2, a Ras-related GTPase, has not been well understood regarding its role and function within the nervous system. In this study, we found that DIRAS2 is downregulated in the hippocampus during the epileptogenesis phase in a kainic acid-induced epilepsy model, while it is upregulated during the chronic phase in this epilepsy model and in patients with temporal lobe epilepsy. Overexpression of DIRAS2 alleviates epileptic seizure susceptibilit

Ocular Toxicities of Anticancer Therapies in the Era of Precision Oncology: A Practical, Clinically Oriented Narrative Review.

The introduction of modern anticancer therapies, including targeted therapies (TTs), immune checkpoint inhibitors (ICIs), and antibody-drug conjugates (ADCs), has significantly improved survival across a wide range of malignancies. At the same time, these agents have expanded the spectrum of treatment-related adverse events, with ocular toxicities emerging as a clinically relevant and increasingly recognized complication. Ocular adverse events may affect multiple anatomical structures, including the ocular surface, cornea, anterior and posterior segments, and optic nerve, often reflecting drug

Metabotropic glutamate receptors in the testis: An integrative bioinformatic review of neuroendocrine, reproductive, and Neuropsychopharmacological aspects.

Metabotropic glutamate receptors (mGluRs) are best known for modulating synaptic transmission, yet accumulating evidence shows that they are also widely expressed in peripheral tissues, including the testis. Methodologically, this work integrates a narrative synthesis of the literature with an in-silico bioinformatic analysis of bulk (GTEx) and single-cell (HPA, CellxGene) RNA-seq datasets, aiming to clarify how mGluRs contribute to testicular physiology. After outlining the striking structural and metabolic parallels between brain and testis-tight barrier systems, selenium-dependent redox con

Carbon Monoxide: A Context-Dependent Regulator of the Stress Axis.

Carbon monoxide (CO) is a gasotransmitter generated by heme oxygenase (HO) isoforms during heme catabolism. The inducible HO-1 produces CO under conditions of redox imbalance, such as oxidative stress and inflammation. On the other hand, HO-2 constitutively generates CO, primarily during the physiological turnover of heme. Extensive evidence indicates that CO exerts autocrine effects by targeting hemoproteins, including soluble guanylyl cyclase, cyclooxygenase, and cytochromes. Furthermore, CO regulates many biological processes within the brain, including mitochondrial biogenesis, potassium c

A reduction in A-to-I RNA editing by ADAR disrupts Pumilio-mediated inhibition of MAPK signaling to drive intestinal stem cell aging.

Aging impairs intestinal stem cell (ISC) function, disrupting epithelial homeostasis and regenerative repair. Loss of ISC quiescence promotes intestinal dysfunction and contributes to organismal aging. Here, we report an epitranscriptomic mechanism through which a decrease in adenosine-to-inosine (A-to-I) RNA editing by the adenosine deaminase ADAR in ISCs during aging disrupts a conserved signaling axis that maintains ISC quiescence. In Drosophila melanogaster, ISC-specific loss of dADAR triggered hyperproliferation and tissue aging, whereas dADAR overexpression reduced age-related gut dysfun

c-FLIP as a master regulator of immune homeostasis and disease mechanisms.

Cellular FLICE (FADD-like IL-1β-converting enzyme)-inhibitory protein (c-FLIP) was discovered more than three decades ago and has since emerged as a multifunctional regulator of cell fate. Initially identified through its homology with viral FLIP (v-FLIP) proteins and its ability to inhibit death receptor-induced apoptosis, c-FLIP is now recognized as a pivotal molecule at the crossroads of apoptosis, necroptosis, autophagy, and inflammation. Beyond its classical anti-apoptotic role, c-FLIP modulates key signaling pathways, including nuclear factor-κB (NF-κB), mitogen-activated protein kinase

Biological Roles of Melanin and Natural Product-Derived Approaches for Its Modulation.

Melanin produced in melanocytes contributes to photoprotection, oxidative stress reduction, immune regulation, and epidermal homeostasis, while its dysregulation underlies diverse pigmentary disorders. Natural products modulate melanogenesis by regulating tyrosinase activity, intracellular signaling pathways such as extracellular signal-regulated kinase/mitogen-activated protein kinase (ERK/MAPK) and cyclicAMP/protein kinase A/cAMP response element-binding protein (cAMP/PKA/CREB), and cellular redox balance. Anti-melanogenic effects have been reported for various fruit-derived phytochemicals,

ZAK kinase at the crossroads of cellular stress: functional plasticity, human pathobiology, and precision therapeutics.

Mitogen-activated protein kinase kinase 20 (MAP3K20/ZAK) has emerged as a critical biophysical sensor, uniquely poised at the intersection of environmental stress and cellular fate. Its versatile signaling is primarily orchestrated by two structurally distinct splice variants: ZAKα, which drives the ribotoxic stress response (RSR) upon detecting ribosome collisions, and ZAKβ, which mediates homeostatic mechanotransduction. However, emerging evidence indicates that the biological roles of ZAK cannot be strictly categorized as either pathogenic or protective. It exhibits profound functional plas

Progressive non-immune hydrops fetalis associated with RASA1 mutation: prenatal imaging and genomic insights.

Hydrops fetalis (HF) is the pathological accumulation of fluid in two or more fetal compartments. While immune-mediated HF was historically predominant, non-immune hydrops fetalis (NIHF) is now increasingly common. Advances in genetic testing have revealed monogenic causes, including RASopathies (a group of genetic syndromes caused by dysregulation of the RAS-mitogen-activated protein kinase signalling pathway). We report a young primigravida, referred at 20+3 weeks for unilateral fetal pleural effusion. Serial ultrasound scans showed rapid progression to bilateral effusion, hepatomegaly, poly

GLP-1 Receptor Agonists in Periodontology: Mechanisms, Clinical Evidence, and Implications for Care.

GLP-1 receptor agonists (GLP-1RAs) are widely used in the treatment of type 2 diabetes and obesity and are increasingly relevant in periodontal and implant practice. This review covers mechanisms, preclinical and early human evidence, and practical periodontal considerations; the structured database search is conducted in accordance with the Scale for the Assessment of Narrative Review Articles (SANRA) and the International Committee of Medical Journal Editors (ICMJE) principles. Two pathways explain GLP-1RAs' relevance: indirect effects from better glycemic control, weight loss, and reduced i

Epidermal METTL1-Mediated m7G Modification Drives Psoriatic Inflammation by Stabilizing Bdkrb1 and Orchestrating Neutrophil Recruitment.

The functional significance of RNA modifications, specifically N7-methylguanosine (m7G), in inflammatory conditions such as psoriasis remains not fully elucidated. This study demonstrates that methyltransferase-like 1 (METTL1), an m7G methyltransferase, is significantly upregulated in epidermal keratinocytes of human psoriatic lesions and imiquimod (IMQ)-induced murine models. Utilizing mice with an inducible keratinocyte-specific Mettl1 deletion (Mettl1fl/flKrt14-CreERT2), the research reveals significantly attenuated psoriasiform inflammation and decreased neutrophil infiltration relative to

Network Pharmacology and Experimental Validation Elucidate the Anti-Angiogenic Mechanism of Silibinin.

Retinopathy of prematurity (ROP) is a sight-threatening vascular disorder driven by pathological neovascularization. Silibinin (SIL), a major flavonolignan from milk thistle, possesses antioxidant and anti-inflammatory properties. Mechanistically, its therapeutic effect arises from attenuated oxidative stress, which suppresses mitogen-activated protein kinase (MAPK) signaling upstream. This study employed an integrated network pharmacology and experimental strategy to elucidate SIL's antiangiogenic mechanism. Network analysis identified the MAPK pathway as a key target, and topological analysi

GRN rs5848 variant associates with TDP-43 pathology and cancer in opposite directions.

Epidemiologic studies have reported that cancer survivors have a relatively low risk of developing dementia, but the mechanisms underlying that inverse relationship are mostly unknown. The Granulin (GRN) gene single nucleotide variant rs5848 T allele is associated with increased risk of limbic-predominant age-related TDP-43 encephalopathy neuropathologic change (LATE-NC) and hippocampal sclerosis of aging (HS-Aging). The T allele is also associated with lower expression of the cognate protein progranulin (PGRN), which is a mitogen implicated in neoplasia. We examined whether the rs5848 variant

Conserved and rewired MAPK networks in human fungal pathogens.

Mitogen-activated protein kinase (MAPK) cascades are among the most ancient and evolutionarily conserved signalling modules in eukaryotes, translating extracellular cues into adaptive transcriptional and metabolic outputs through a hierarchical three-tier kinase architecture. In yeasts, these pathways control osmotic and oxidative stress adaptation, mating, morphogenesis and cell wall remodelling, while in fungal pathogens they have been adapted to support virulence-factor production, host adaptation and antifungal tolerance. Here, we summarise the current understanding of MAPK signalling in t

ATAC sequencing and transcriptomics reveal OsCHR732 function in regulating chromatin accessibility and gene expression in pollen and ovule development in rice.

BACKGROUND: The fertility of pollen and ovules directly influences seed set and overall yield in rice, however, the molecular regulatory mechanisms governing rice pollen and ovule development remain poorly understood. RESULTS: In this study, we discovered that functional knockout mutants of the rice chromatin remodeling factor OsCHR732 exhibited significantly reduced pollen viability, accompanied by abnormal ovule development. Quantitative reverse transcription PCR (qRT-PCR) and β-glucuronidase (GUS) staining revealed that OsCHR732 was highly expressed in young panicles, ovaries, and anthers

Epichloë bromicola Enhances Barley Disease Resistance Through Temporally Coordinated Defense Responses.

Epichloë endophytes enhance plant defense and biotic stress resistance through mutualistic interactions in natural hosts. However, whether these protective effects are retained in annual non-native hosts such as barley (Hordeum vulgare L.) remains unclear. An integrated multi-omics approach was used to characterize defense responses in Epichloë bromicola-infected barley (LD1) and endophyte-free barley (CK) following pathogen challenge. The results show that LD1 had 20.0% lower disease incidence and 24.1% lower disease index than CK. LD1 also exhibited 21.1% lower malondialdehyde (MDA) content

Distinct Host and Bacterial Profiles in Murine Single Versus Co-Infection with Foal-Isolated Streptococcus equi and Streptococcus dysgalactiae.

This study investigated the interactive pathogenic characteristics of co-infection with Streptococcus equi (S. equi) and Streptococcus dysgalactiae (S. dysgalactiae) isolated from diseased foals. Integrating genomic analysis, in vitro phenotypic assays, and a murine intranasal infection model, we comparatively analyzed the biological traits and pathogenic differences between single and dual infections. Comparative genomic analysis revealed divergent virulence profiles between the two isolates, and both species harbored open pan-genomes. In vitro, the biofilm biomass of strain F1 was higher tha

Identification of CD55 as a downstream factor of EP4 receptor signaling in colorectal cancer cells.

Prostaglandin E2 (PGE2) signaling through the E-type prostanoid 4 (EP4) receptor has been implicated in the pathophysiology of colorectal cancer (CRC). We herein identified decay-accelerating factor, also known as CD55, as a novel CRC-associated downstream factor of the EP4 receptor. The integration of transcriptomic profiling of PGE2-stimulated HCA-7 human colon cancer cells with analyses of cancer genomic databases predicted CD55 as a potential EP4 receptor-regulated target. Inhibitor-based experiments showed the induction of CD55 after a PGE2 stimulation required the EP4 receptor and Gi pro
