Mesenchymal Stromal Cell-Based Cell-Drug Conjugates for the Treatment of Acute Liver Failure.

Acute liver failure (ALF) is a life-threatening syndrome characterized by severe inflammatory injury and high mortality. Mesenchymal stromal cells (MSCs) hold therapeutic promise for ALF because of their immunomodulatory and tissue-repair capacities. However, in the ALF inflammatory microenvironment, characterized by abundant activated M1 macrophages and elevated pro-inflammatory cytokines, MSCs are susceptible to apoptosis, resulting in low survival and limited therapeutic efficacy. Here, we combined the anti-inflammatory small-molecule rosiglitazone with MSCs to construct a cell-drug conjuga

The Role of Plastin in Bone, Cartilage, and Related Diseases.

The plastin family (PLS1, PLS2, PLS3) comprises highly conserved actin-binding proteins (ABPs) that regulate cytoskeletal dynamics by cross-linking actin microfilaments and play key roles in bone and cartilage development, homeostasis, and disease. PLS1 primarily stabilizes tight actin bundles in epithelial brush-border microvilli and has also been reported to support osteoblast differentiation via regulating intracellular Ca2+ homeostasis. In bone, PLS3 mutations lead to X-linked osteoporosis through mechanisms involving the inhibition of the WNT pathway, activation of TH17 cell differentiati

Flow-mediated endothelial remodeling and inflammation drive developmental vascular susceptibility in ldlr loss of function.

Atherosclerosis, the leading cause of cardiovascular disease, is associated with aberrant lipid metabolism, endothelial dysfunction, and chronic inflammation, yet its early manifestations and mechanisms remain incompletely understood. As low-density lipoprotein receptor loss of function is the most common monogenic cause of atherosclerosis, we employed low-density lipoprotein receptor knockout (ldlr-/-) zebrafish to investigate the developmental origins of atherosclerotic cardiovascular disease. Single-cell RNA-sequencing under differential flow conditions in embryonic ldlr-/- zebrafish identi

Serum Proteomic Profiling Uncovers Dysregulated Keratinisation and Immune-Related Pathways in Hidradenitis Suppurativa.

Hidradenitis suppurativa (HS) is an autoinflammatory keratinisation disease affecting the pilosebaceous unit and hallmarked by a complex and multifactorial pathogenesis. Although genomic and transcriptomic investigations have substantially advanced our understanding of key mechanisms underlying HS pathogenesis, proteomic studies remain limited, despite the significant potential of serum proteomics to identify molecular signatures reflecting both cutaneous and systemic inflammatory activity. This exploratory study presents a serum proteomic analysis of patients with moderate-to-severe HS, ident

Basophil Extracellular Traps in Immunity and Disease.

Basophil extracellular traps (BETs) represent a recently recognized extracellular effector mechanism whose biology remains far less defined than that of neutrophil extracellular traps. This review critically examines the terminology, molecular composition, signaling requirements, and disease relevance of BET formation. Current evidence supports a rapid, predominantly non-lytic process in which mitochondrial reactive oxygen species promote the externalization of mitochondrial DNA, particularly after IL-3 priming followed by FcεRI or C5a receptor activation. The resulting networks contain mtDNA

CD4+ T cell-mediated immunity protects from VSV-SUD lethal challenge in a mouse model of Sudan virus infection.

Recurring outbreaks of filoviruses, including Ebola virus and Sudan virus, threaten to cause large epidemics. No treatment or vaccine is available for Sudan virus and how protective immunity is achieved remains unknown. To unravel mechanisms contributing to protection, we used a surrogate mouse model of Sudan virus infection amenable to deep functional analysis. Mice vaccinated with a live viral vector based on live-attenuated YF17D expressing Sudan virus glycoprotein were protected against lethal challenge with a surrogate virus, a chimeric recombinant vesicular stomatitis virus expressing Su

Dynamic tethering of M protein drives pathological inflammation during group A Streptococcus infections.

The M protein of Group A Streptococcus (GAS) is an essential virulence factor that promotes both superficial and invasive infections, as well as the development of immune sequela, including acute rheumatic fever and rheumatic heart disease. M protein is a prototypical sortase-anchored surface protein, yet has also been observed free of the GAS surface during human infection. This suggests a mechanism for its release, and this change in localization could influence innate immune detection, effector functions, and antigenic potential. Here, we show the protease SpeB cleaves M protein from the mi

The spleen tyrosine kinase inhibitor entospletinib resolves inflammation to promote repair following acute kidney injury.

Nonresolving inflammation and maladaptive renal repair contribute to the pathogenesis of acute kidney injury (AKI) transition to chronic kidney disease (CKD). Few therapies have been identified that can modulate these injurious pathways following AKI. Spleen tyrosine kinase (SYK) is an immune regulator expressed in the kidney and a potential therapeutic target for AKI. The effect of the selective SYK inhibitor entospletinib was studied in AKI-to-CKD transition. Entospletinib was administered to mice undergoing unilateral renal ischemia-reperfusion injury (IRI), with kidneys analyzed over 14 da

ZEB1 promotes chemoimmunotherapy resistance in pancreatic cancer models by downregulating chromatin acetylation of CXCL16.

Pancreatic cancer (PC) is notoriously resistant to both chemotherapy and immunotherapy, presenting a major therapeutic challenge. Epigenetic modifications play a critical role in PC progression, yet their contribution to chemoimmunotherapy resistance remains poorly understood. Here, we identified the transcription factor ZEB1 as a critical driver of chemoimmunotherapy resistance in PC. ZEB1 knockdown synergized with gemcitabine and anti-PD-1 therapy, markedly suppressed PC growth, and prolonged survival in vivo. Single-cell and spatial transcriptomics revealed that ZEB1 ablation promoted tumor

Engraftment syndrome after PD-1 blockade in classical Hodgkin lymphoma treated by autologous stem cell transplantation.

Immune checkpoint inhibitors (CPIs) targeting PD-1, such as nivolumab and pembrolizumab, have become integral to the treatment of relapsed or refractory classical Hodgkin lymphoma (cHL), frequently serving as a bridge to autologous stem cell transplantation (ASCT). However, recent reports suggest that CPI exposure prior to ASCT may increase the risk of engraftment syndrome (ES), an inflammatory complication that emerges during neutrophil recovery. We present three cases of cHL treated by ASCT was performed following anti PD-1-based salvage therapy. Two of the three patients developed ES, with

Transcription factor ELF-1 protects against colitis by maintaining intestinal epithelium homeostasis.

Inflammatory bowel disease (IBD) is a chronic, relapsing, and remitting disease characterized by chronic inflammation in the gastrointestinal tract. The exact etiology and pathogenesis of IBD remain elusive. Although ELF-1 has been known to be highly expressed in epithelial cells for past twenty years, little is known about its function in epithelial cells and epithelial-related IBD. Here, we demonstrated that ELF-1 deficiency in mouse lead to exacerbated DSS-induced colitis, marked by inflammation dominated by neutrophil infiltration and activation of IL-17 signaling pathways in various immun

Articular involvement in Henoch-Schönlein Purpura: A review of literature.

BACKGROUND: Henoch-Schönlein purpura (HSP), the most typical kind of pediatric vasculitis, can also affect adults. Over the last 10 years, research has been increasing on improvements in HSP diagnosis, physiopathology, symptoms, and therapy. Joint involvement is highly frequent in this condition; however, it typically undergoes spontaneous resolution and does not lead to long-term complications. OBJECTIVE: To provide a deeper understanding of the constituting pathogenic mechanisms and clinical presentation of articular involvement, focusing on the effect of neutrophil activation on systemic s

Lachnospiraceae bacterium alleviates alcohol-associated liver disease by enhancing N-acetyl-glutamic acid levels and inhibiting ferroptosis through the KEAP1-NRF2 pathway.

Alcohol-associated liver disease (ALD) is a prevalent global health issue primarily caused by excessive alcohol consumption. Recent studies have highlighted the gut-liver axis's protective role against ALD, mainly through gut microbiota. However, the precise mechanism remains ill-defined. Our results showed a significant reduction in Lachnospiraceae bacterium in the gut microbiota of ALD patients and ethanol (EtOH)-fed mice, as revealed by 16S rDNA sequencing. Supplementation with Lachnospiraceae bacterium strains in mice significantly reduced inflammation, hepatic neutrophil infiltration, oxi

Temporal trends of a cellular host response test for sepsis and a comparison with selected biomarkers of inflammation and infection.

Sepsis, a condition characterized by immune dysregulation, is the leading cause of in-hospital mortality and requires rapid treatment. Assessing immune dysregulation has been challenging. The IntelliSep Index (ISI), a novel biomarker which leverages microfluidic deformability cytometry to assess immune activation, has been evaluated as a test for Emergency Department use but it is unclear how the signal evolves as patient condition evolves. In a 47-patient cohort with hospital stays ≤ 14 days, ISI provided a good indicator of disease progression by blinded physician review. ISI trends correlat

Emerging molecular targets in deep vein thrombosis: from inflammation to coagulation.

Deep vein thrombosis (DVT), a prevalent vascular disorder driven by venous stasis, endothelial injury, and hypercoagulability, imposes a significant global health burden due to life-threatening complications like pulmonary embolism. Recent advances highlight inflammation as a pivotal contributor to DVT pathogenesis, intricately linked with coagulation through immunothrombosis. This review synthesizes emerging molecular targets bridging these pathways, focusing on neutrophil extracellular traps (NETs), peptidylarginine deiminase 4 (PAD4), P-selectin, high-mobility group box 1 (HMGB1), tissue fa

The immunopathogenesis of a cytokine storm: The key mechanisms underlying severe COVID-19.

A cytokine storm is marked by excessive pro-inflammatory cytokine release, and has emerged as a key factor in severe COVID-19 cases - making it a critical therapeutic target. However, its pathophysiology was poorly understood, which hindered effective treatment. SARS-CoV-2 initially disrupts angiotensin signalling, promoting inflammation through ACE-2 downregulation. Some patients' immune systems then fail to shift from innate to adaptive immunity, suppressing interferon responses and leading to excessive pyroptosis and neutrophil activation. This amplifies tissue damage and inflammation, crea
