Alleviation of aspirin-induced fetotoxicity by calcium was investigated in rats. ASA (500 mg/kg s.c.) decreased the plasma Ca level in pregnant rats and that in the feto-placenta units. 0.05 M CaCl2 given to dams as tap water on days 8 through 20 of gestation inhibited ASA-induced hypocalcemic effect in maternal plasma. Tap water of 0.002-0.05 M CaCl2, Ca(OH)2 and Ca-lactate on days 8 through 20 of gestation alleviated malformations elicited by the administration of ASA (500 mg/kg s.c.) on the 11th day of gestation resulting in a decrease in the fetotoxicity in rats.

Four unrelated patients who had the clinical appearance of Miller-Dieker syndrome, also called lissencephaly syndrome, were studied. All four had a typical clinical course with failure to thrive, severe psychomotor retardation, opisthotonos, seizures, and death early in life. Aging mice treated with radio-detoxified endotoxin and subsequently inoculated intracerebrally with lymphocytic choriomeningitis (LCM) virus died earlier and in a higher rate than the virus infected and otherwise untreated controls. Thus, in mice with insufficient T lymphocyte function due to aging, the radio-detoxified endotoxin pretreatment contributed to the outcome of LCM virus infection in the form of lethal meningitis. That indicates a stimulatory effect on the reduced cellular immunological reaction to the virus infection.

The time-course of changes in baroreceptor reflex sensitivity during the reversal of Goldblatt two-kidney one-clip hypertension was studied in male Wistar rats. Groups of animals were studied before and 1, 3, 7, 14 and 25 days after removal of a left renal artery clip of 0.18 mm internal diameter. Normotensive rats of comparable ages were included as controls. Acute toxicity of ammonium metavanadate solutions in normal saline (pH 6.7) or 0.1 M Tris-HCl-NaCl buffers (pH 7.2 or pH 7.8) was studied in BALB/c mice at 20 mg V/kg. Murine trisomy (Ts) 16 occurs in the fetal and neonatal progeny of males doubly heterozygous for the Robertsonian metacentric chromosomes Rb(16.17)7Bnr/Rb(9.16)9Rma and "all acrocentric" females. No hepatotoxicity was demonstrated after 90 days in rats treated with two dose levels of disulfiram. A reversible inhibition of microsomal p-nitroanisole demethylase activity was found. In phenobarbital-treated rats disulfiram 100 mg/kg did not alter the induction response as indicated by the cytochrome P-450 content, but inhibited the p-nitroanisole activity to control levels. As no sign of histological liver damage was found, we conclude that the rare cases of disulfiram hepatotoxicity in man may be due to an allergic reaction.

We have developed an in vitro technique for producing myocardial rupture in lamb hearts, which relates tensile strength to a variety of conditions which can prevail in normal and infarcted human hearts. Oral barbital treatments consisting of average daily doses of 200 mg/kg with durations of 15-50 weeks have been used to induce functional tolerance and physical dependence. Tolerance is usually studied with a hexobarbital threshold while increased excitation has been studied by various means, such as induction of convulsions with pilocarpine or choline. Oral administration of delta 9-tetrahydrocannabinol or cannabinol to female mice late in pregnancy and during early lactation alters body weight regulation and pituitary-gonadal function and suppresses adult copulatory activity in their male offspring. These findings suggest that both psychoactive and nonpsychoactive constituents of marihuana can affect the development of male reproductive functions in mice.
