A possible relationship between KCl symport and basolateral K(+)-conductance in Necturus gallbladder epithelial cells.

1. Apical membrane potential (Va), transepithelial potential (VT), fractional apical voltage ratio (FVa = delta Va/delta VT), tissue resistance (RT), and intracellular Cl- (aiCl) and K+ (aiK) activities were measured in isolated gallbladders maintained between oxygenated bicarbonate-free physiological media (23 degrees C, pH 7.2 or 8.2) in a divided chamber. The basolateral membrane potential (Vb) was calculated from the measured values of Va and VT. 2. Cl- removal from the serosal medium (which should accelerate coupled basolateral KCl exit) significantly depolarized Vb, decreased aiCl, decre

Erythropoietic protoporphyria: lipid peroxidation and red cell membrane damage associated with photohemolysis.

The mechanism by which long wavelength ultraviolet light hemolyzes red cells obtained from patients with erythropoietic protoporphyria (EPP) was investigated. Previous studies had suggested that irradiation of these red cells with wavelengths of light capable of eliciting dermatological manifestations led to oxygen-dependent colloid osmotic hemolysis through the formation of peroxides. In the present report, lipid peroxidation during in vitro irradiation of EPP red cells with long ultraviolet light was demonstrated by: (a) the formation of 2-thiobarbituric acid reactants; (b) the presence of c

Lipid Peroxidation in Cancer Therapy: Molecular Mechanisms Involving Oxidative Stress, Cell Death, and Therapeutic Response.

Lipid peroxidation (LPO) is a process where polyunsaturated fatty acids (PUFA) in cellular membranes are oxidized. This process is mediated by reactive oxygen species (ROS) and leads to the formation of reactive products, including 4-hydroxynonenal (4-HNE), malondialdehyde (MDA), and oxidized phospholipids. At low concentrations these products act as second messengers in adaptive redox signalling and metabolic homeostasis, whereas at higher concentrations they compromise membrane integrity and promote cell death. Lipid peroxidation plays a crucial role in anticancer therapies. Here we focus on

Recent Advances in ZIF-8 Performance for Electrochemical Applications: A Comprehensive Review.

Zeolitic imidazolate framework-8 (ZIF-8) has emerged as a material for electrochemical energy conversion, serving dual primary roles in fuel cell technologies: (i) as an electrocatalyst precursor for oxygen reduction reaction (ORR) and methanol oxidation reaction (MOR) through pyrolysis-derived N-doped porous carbons and metal-nitrogen-carbon (M-N-C) structures, and (ii) as a membrane component that enhances proton conductivity via imidazole-mediated Grotthuss hopping while suppressing fuel crossover through molecular sieving. This comprehensive review systematically evaluates ZIF-8 performanc

Applications of hollow-fiber systems in bone, liver, and pancreatic tissue engineering.

BACKGROUND: Hollow-fiber bioreactors (HFBs) have emerged as promising platforms for tissue engineering because their capillary-like architecture can improve perfusion, molecular exchange, and three-dimensional cell organization. Their high surface-area-to-volume ratio and compartmentalized design help reduce the diffusion limitations commonly observed in static cultures, particularly in large or metabolically active tissue constructs. METHODS: This review discusses recent advances in hollow-fiber-based systems for bone, liver, and pancreatic tissue engineering from experimental, computational

Synthesis of cationic N-acylated thiazolidine for selective activity against Gram-positive bacteria and evaluation of N-acylation's role in membrane-disrupting activity.

The evolution of antimicrobial-resistant strains jeopardizes the existing clinical drugs and demands new therapeutic interventions. Herein, we report the synthesis of cationic thiazolidine bearing a quaternary pyridinium group, in which thiazolidine was N-acylated with fatty acid to establish a hydrophilic-lipophilic balance that disrupts bacterial membranes. The bacterial growth inhibition assays and hemolytic activity against human red blood cells indicate that the N-acylated cationic thiazolidine (QPyNATh) inhibits Gram-positive bacteria at lower minimum inhibitory concentrations (MIC) and

RIPK1 inhibition contributes to lysosomal membrane stabilization in ischemic astrocytes via a lysosomal Hsp70.1B-dependent mechanism.

Receptor-interacting protein kinase 1 (RIPK1) contributes to necroptosis. Our previous study showed that pharmacological or genetic inhibition of RIPK1 protects against ischemic stroke-induced astrocyte injury. In this study, we investigated the molecular mechanisms underlying RIPK1-mediated astrocyte injury in vitro and in vivo. Primary cultured astrocytes were transfected with lentiviruses and then subjected to oxygen and glucose deprivation (OGD). In a rat model of permanent middle cerebral artery occlusion (pMCAO), lentiviruses carrying shRNA targeting RIPK1 or shRNA targeting heat shock p

Addition of seminal plasma to thawed stallion spermatozoa did not repair cryoinjuries.

Freezing and thawing processes induce structural and functional damage to sperm plasma membranes and internal organelles. Adding seminal plasma (SP) has been found to minimize or repair the cryoinjuries in some species. The objective of this study was to investigate whether adding SP from stallions of known freezability after thawing could repair cryoinjuries. Semen was collected from warmblood stallions (n = 8, three ejaculates/stallion) and processed by Single Layer Centrifugation (SLC) to remove SP prior to freezing. Pooled SP (5%) from bad freezer (BF) or good freezer (GF) stallions was ad

Cardiac mitochondrial damage and inflammation responses in sepsis.

BACKGROUND AND PURPOSE: Studies in sepsis suggest that mitochondria mediate multiple organ dysfunction, including cardiac failure; however, the underlying molecular mechanisms remain elusive. This study examined changes in mitochondrial membrane integrity, antioxidant activities, and oxidative stress in the heart after infectious challenge (intratracheal Streptococcus pneumoniae, 4 x 10(6) colony-forming units). Inflammation responses also were examined. METHODS: Cardiac tissues were harvested from Sprague-Dawley rats 4, 8, 12, and 24 h after bacterial challenge (or intratracheal vehicle for

Proteinuria in passive Heymann nephritis is associated with lipid peroxidation and formation of adducts on type IV collagen.

Passive Heymann nephritis (PHN) is a model of human membranous nephropathy that is characterized by formation of granular subepithelial immune deposits in the glomerular capillary wall which results in complement activation. This is causally related to damage of the filtration barrier and subsequent proteinuria. The local accumulation of injurious reactive oxygen species (ROS) is a major effector mechanism in PHN. ROS may induce tissue damage by initiating lipid peroxidation (LPO). In turn, this leads to adduct formation between breakdown products of LPO with structural proteins, such as forma

Chiaroscuro of P(V) porphyrins cytotoxicity: Balancing light and dark effects.

In order to search for new more effective photosensitizers for photodynamic therapy, three phosphorus(V) porphyrins with donor meso-substituents (p-methoxyphenyl, p-methylphenyl, p-tert-butylphenyl) and axial ethoxy groups were synthesized. All complexes were sufficiently photostable and capable of effectively generating singlet oxygen with quantum yields of 0.68-0.74 in ethanol. The study revealed that complexes with meso-p-methylphenyl and meso-p-methoxyphenyl fragments demonstrated pico- and nanomolar photocytotoxicity both upon 450 nm (IC50 light = 330 pM - 150 nM) and 660 nm (IC50 light =

AKAP1 orchestrates mitochondrial dynamics and metabolic reprogramming to promote hypoxia tolerance in yak cardiac fibroblasts.

High-altitude hypoxia poses a sustained energetic and oxidative challenge to the heart, often leading to mitochondrial dysfunction and metabolic inflexibility in nonadapted mammals. Yaks (Bos grunniens) exhibit remarkable cardiac tolerance to chronic hypoxia; however, the underlying intracellular mechanisms remain poorly defined. Herein, we identify A-kinase anchoring protein 1 (AKAP1) as an important regulator of hypoxia tolerance in yak cardiac fibroblasts. Through the genetic manipulation of AKAP1 combined with live-cell mitochondrial imaging, functional assays, and untargeted metabolomics,

Lapatinib suppresses cysteine deprivation-induced ferroptotic cell death by modulating mitochondrial function.

Ferroptosis, an iron-dependent regulated cell death driven by lipid peroxidation, has emerged as a promising therapeutic target for cancer. Although lapatinib has been reported to induce ferroptosis in several cancer types, we found that it exerts a ferroptosis-suppressive effect under cysteine deprivation. Lapatinib alone induced a mild ferroptotic phenotype (< 10% cell death), accompanied by increased intracellular reactive oxygen species (ROS) generation, lipid peroxidation, and intracellular labile iron (Fe²⁺) accumulation. In contrast, under cysteine deprivation, lapatinib markedly attenu

Alpha- and Gamma-Synucleins Regulate Energy Metabolism and Xanthine Oxidase Activity in Brain Cells.

The process of signal transmission and transformation in the central nervous system requires active energy metabolism with high consumption of glucose and oxygen. Reactive oxygen species (ROS) produced as a result of these processes participate in intracellular signaling, but their overproduction leads to oxidative stress. Oxidative stress and α-synuclein aggregation are recognized as activators of neuronal death in Parkinson's disease. However, much less is known about the physiological role of monomeric synucleins. Using acute brain slices and primary co-cultures of cortical neurons and glia

Molecular pathways in cardiovascular disease under hypoxia: Mechanisms, biomarkers, and therapeutic targets.

Chronic hypoxia is a key factor in the pathogenesis of cardiovascular diseases, including ischemia, heart failure, and hypertension. Under hypoxia, oxygen deficiency disrupts oxidative phosphorylation in mitochondria, impairing ATP production and generating reactive oxygen species (ROS). These reactive species induce mitochondrial dysfunction, leading to oxidative stress, calcium imbalance, and activation of apoptosis pathways. The mitochondrial ATP-sensitive potassium channel (mitoK ATP) and mitochondrial permeability transition pore (mPTP) channels are particularly affected, contributing to

Ampere-level co-electrosynthesis of formate from CO2 reduction paired with formaldehyde dehydrogenation reactions.

Current catalysts face challenges with low formate selectivity at high current densities during the CO2 electroreduction. Here, we showcase a versatile strategy to enhance the formate production on p-block metal-based catalysts by incorporating noble metal atoms on their surface, refining oxygen affinity, and tuning adsorption of the critical oxygen-bound *OCHO intermediate. The formate yield is observed to afford a volcano-like dependence on the *OCHO binding strength across a series of modified catalysts. The rhodium-dispersed indium oxide (Rh/In2O3) catalyst exhibits impressive performances

Retinal Pigment Epithelium Pigment Granules: Norms, Age Relations and Pathology.

The retinal pigment epithelium (RPE), which ensures the normal functioning of the neural retina, is a pigmented single-cell layer that separates the retina from the Bruch's membrane and the choroid. There are three main types of pigment granules in the RPE cells of the human eye: lipofuscin granules (LG) containing the fluorescent "age pigment" lipofuscin, melanoprotein granules (melanosomes, melanolysosomes) containing the screening pigment melanin and complex melanolipofuscin granules (MLG) containing both types of pigments simultaneously-melanin and lipofuscin. This review examines the func

PEG300 Protects Mitochondrial Function By Upregulating PGC-1α to Delay Central Nervous System Oxygen Toxicity in Mice.

Central nervous system oxygen toxicity (CNS-OT) is a complication of hyperbaric oxygen (HBO) treatment, with limited prevention and treatment options available. In this study, we aimed to explore the effect of polyethylene glycol 300 (PEG300) on CNS-OT and underlying mechanisms. Motor and cognitive functions of mice in normobaric conditions were evaluated by Morris water maze, passive active avoidance, and rotarod tests. HBO was applied at 6 atmospheres absolute (ATA) for 30 min after drug administration. The latency period of convulsion in mice was recorded, and hippocampal tissues were extra
