Intestinal alkaline phosphatase (AP) detoxifies lipopolysaccharides, which has been exploited in treating infection of Gram-negative bacterial pathogens such as Escherichia coli. Orally administered AP has the potential to modulate the gut microbiota, which is in part ascribed to its ability to degrade ATP, a well-known inhibitor of gut commensal bacteria. In addition, AP can fortify the gut barrier. Therefore, we hypothesized that the enzyme might also be used to control intestinal Gram-positive pathogens such as Clostridium perfringens. Herein, broiler chickens were challenged with 3 × 108 c

α-synuclein (αS) is an abundant, neuronal protein that assembles into fibrillar pathological inclusions in a spectrum of neurodegenerative diseases that include Lewy body diseases (LBD) and Multiple System Atrophy (MSA). The cellular and regional distributions of pathological inclusions vary widely between different synucleinopathies contributing to the spectrum of clinical presentations. Extensive cleavage within the carboxy (C)-terminal region of αS is associated with inclusion formation, although the events leading to these modifications and the implications for pathobiology are of ongoing

Botulinum neurotoxins (BoNTs) are produced as protein complexes by bacteria of the genus Clostridium that are Gram-positive, anaerobic and spore forming (Clostridium botulinum, C. butyricum, C. baratii and C. argentinense spp.). BoNTs show a high immunological and genetic diversity. Therefore, fast, precise, and more reliable detection methods are still required to monitor outbreaks and ensure surveillance of botulism. The botulinum toxin field also comprises therapeutic uses, basic research studies and biodefense issues. This review presents currently available detection methods, and new meth

BACKGROUND: Aromatic L-amino acid decarboxylase (AADC) deficiency is a rare genetic disorder with heterogeneous phenotypic spectrum resulting from disease-causing variants in the dopa decarboxylase (DDC) gene. Consensus guidelines recommend dopamine agonists, monoamine oxidase inhibitors, and other symptomatic treatments, but most patients have an unrelenting disease course with no response to these therapies.
CASE PRESENTATION: We describe 2 African American siblings with AADC deficiency and identify 2 DDC gene variants not previously associated with the disorder. The patients were evaluated

OBJECTIVE: Vascular endothelial growth factor (VEGF) is elevated in joint fluids from patients diagnosed with osteoarthritis (OA). VEGF is known to contribute to vascular tidemark invasion and osteophyte formation, which are classic features of advanced OA. Among the factors that may drive VEGF accumulation in diseased joints, stromal cell-derived factor-1α (SDF-1α) is a likely culprit, as it is enriched in synovial fluids from osteoarthritic joints and is a potent inducer of VEGF expression. Chondrogenic progenitor cells (CPCs) that overexpress SDF-1α are abundant in osteoarthritic cartilage,

The ability of certain species of Chlamydia to inhibit the biogenesis of phagolysosomes permits their survival and replication within macrophages. The survival of macrophage-adapted chlamydiae correlates with the multiplicity of infection (MOI), and optimal chlamydial growth occurs in macrophages infected at an MOI of ≤1. In this study, we examined the replicative capacity of Chlamydia muridarum in the RAW 264.7 murine macrophage cell line at different MOIs. C. muridarum productively infected these macrophages at low MOIs but yielded few viable elementary bodies (EBs) when macrophages were inf

We have previously shown that type I procollagen pro-alpha1(I) chains from an osteogenesis imperfecta patient (OI26) with a frameshift mutation resulting in a truncated C-propeptide, have impaired assembly, and are degraded by an endoplasmic reticulum-associated pathway (Lamandé, S. R., Chessler, S. D., Golub, S. B., Byers, P. H., Chan, D., Cole, W. G., Sillence, D. O. and Bateman, J. F. (1995) J. Biol. Chem. 270, 8642-8649). To further explore the degradation of procollagen chains with mutant C-propeptides, mouse Mov13 cells, which produce no endogenous pro-alpha1(I), were stably transfected

We have studied the effect of tri-phenyl tin benzimadazolethiolcopper chloride (TPT-CuCl(2)), a novel bimetallic compound, on the regulation of apoptosis in HeLa cells, MCF-7 cells, and in vivo Wistar rat model. TPT-CuCl(2) induces significant apoptosis in HeLa cell line characterized by DNA fragmentation and chromosome condensation. Comet assay revealed that TPT-CuCl(2) targets and causes severe damage to the DNA. Treatment of HeLa cells with TPT-CuCl(2) rescues the accumulation of p53 from the suppression of human papilloma virus E6, resulting in a dramatic up-regulation of Bax and Bak and d

Insulin like growth factor-1 (IGF-1) is established as an anabolic factor that can induce skeletal muscle growth by activating the phosphoinositide 3-kinase/Akt/mammalian target of rapamycin (mTOR) pathway. Although this signaling pathway has been the subject of much study, the molecular mechanisms linking IGF-1 binding to mTOR activation remain poorly defined in muscle. The present study aimed to test the hypothesis that IGF-1 activation of mTOR in C2C12 myotubes requires a phosphorylation-dependent, altered distribution of the tuberous sclerosis complex (TSC)1/TSC2 complex from the membrane

Parkinson's disease is associated with amyloid fibrillation of alpha-synuclein (α-syn) that involves the aggregation of protein into insoluble fibrils. In this study, we investigate the effect of luteolin (LUT) on α-syn aggregation in vitro, in silico, and in Caenorhabditis elegans. The decrements in light scattering and ThT fluorescence reveal that there is less formation of α-syn aggregates with an increase in the concentration of LUT. Also, a decrease in the ANS fluorescence depicts less exposure of hydrophobic patches in α-syn in the presence of LUT. Far UV CD data shows that LUT resists c

Spinal cord injury (SCI) triggers robust neuroinflammation, in which microglial activation and dysregulated programmed cell death exacerbate secondary damage. PANoptosis, a recently defined inflammatory cell death modality integrating pyroptosis, apoptosis, and necroptosis, has been implicated in central nervous system (CNS) injury, yet its cellular specificity and regulation remain unclear. In this study, integrative bioinformatic analyses identified microglia as the predominant PANoptotic cell population after SCI, with PANoptosis inversely correlated with adenosine monophosphate (AMP)-activ

X-linked inhibitor of apoptosis protein (XIAP) deficiency is a monogenic immunodeficiency disorder with a broad clinical spectrum that includes inflammatory bowel disease (IBD). The inflammatory mechanisms underlying XIAP-associated IBD remain incompletely understood, and resistance to anti-tumor necrosis factor (TNF) therapy represents a significant therapeutic challenge. We report a case of a pediatric patient with XIAP-associated IBD caused by a novel pathogenic frameshift variant in the XIAP gene (c.498dupA, p.Leu167Ilefs*30). The clinical course was characterized by refractoriness to anti

Pyroptosis is involved in the progression of chronic obstructive pulmonary disease (COPD). Andrographolide (AG) possesses anti-inflammatory activity; however, its role in alveolar macrophage (AM) pyroptosis in COPD is still unclear. This study is to explore AG’s effects on AM pyroptosis and its underlying mechanism. A murine COPD model was generated by cigarette smoke (CS) exposure, and lung pathological changes were evaluated. A cigarette smoke extract (CSE)-induced MH-S cell model was established in vitro, with cells pretreated with AG or the c-jun N-terminal kinase (JNK) inhibitor SP600125.

This study retrospectively analyzed the clinical data of the first reported case in China of myeloproliferative neoplasm (MPN) driven by a homozygous germline SH2B3 mutation, aiming to deepen the understanding of this rare MPN subtype. The patient was a 59-year-old female admitted due to intermittent fatigue and fever for 3 years, which had worsened 4 days prior to admission. Next-generation sequencing revealed a homozygous germline SH2B3 gene mutation (c.1481C>A, p.S494*) with a variant allele frequency (VAF) of 100%. Family analysis confirmed its germline origin, and her children were all he

Hepatocellular carcinoma (HCC) remains a lethal malignancy with limited therapeutic targets. P53 and DNA damage-regulated gene 1 (PDRG1) has emerged as an oncogene in multiple cancers, yet its role and regulatory mechanism in HCC remain unclear. Here, we demonstrated that PDRG1 expression was significantly upregulated in HCC tissues compared to normal liver, correlating with advanced tumor stage, higher grade, and poor patient survival. Functionally, PDRG1 knockdown suppressed HCC cell proliferation, migration, and invasion in vitro and inhibited tumor growth and lung metastasis in vivo, where

Pathological ectopic calcification of soft tissues can arise from reduced or absent levels of inorganic pyrophosphate (PPi), a key inhibitor of calcium hydroxyapatite deposition in soft connective tissues. The role of PPi in regulating mineralization has been recognized for decades, thanks to the pivotal work of Herbert Fleisch and colleagues; and its clinical relevance has been underscored by the identification of hereditary metabolic disorders, collectively termed PPi deficiency syndromes. These are caused by pathogenic variants in the essential genes for maintaining PPi homeostasis: ATP-bin

Parkinson's disease (PD) is a prevalent neurodegenerative disorder lacking effective disease-modifying therapies, with neuronal death critically linked to endoplasmic reticulum stress (ERS) and the activation of the activating transcription factor 4/activating transcription factor 3/C/EBP homologous protein (ATF4/ATF3/CHOP) pro-apoptotic pathway. This study investigated whether the alkaloid tetramethylpyrazine (TMP) confers neuroprotection by modulating this pathway. Our approach combined bioinformatics, which suggested ATF4 as a potential regulatory node, with mechanistic experiments in cellu
