Role of LIGHT in the Inflammatory Mechanisms of Psoriasis via Upregulation of Proliferation and Cytokine Production of Keratinocytes.

BACKGROUND: The pathogenesis of psoriasis is associated with abnormalities in immune pathways. HVEM is known as a receptor of LIGHT (homologous to lymphotoxins, inducible, and competes with HSV glycoprotein D), which is a newly identified member of the TNF superfamily. The expression of HVEM and LTBR (another LIGHT receptor) has been found to be increased in the skin of psoriasis patients. This indicates the potential role of LIGHT and its receptors in the pathogenesis of psoriasis. Therefore, the objective of this study was to examine the effect of LIGHT on keratinocyte proliferation and its

Diagnostic Usefulness of Immunohistochemical Staining for EMA in Psoriasis and Atopic Dermatitis Showing Psoriasiform Hyperplasia.

BACKGROUND: Atopic dermatitis (AD) and psoriasis (PSO) can share clinical and histopathologic features, complicating differentiation. Epithelial membrane antigen (EMA), a marker of epithelial differentiation, is used primarily in neoplastic diseases but is rarely assessed in inflammatory dermatoses. EMA expression in AD remains poorly defined. OBJECTIVE: To evaluate whether EMA immunohistochemistry (IHC) helps distinguish PSO from AD with psoriasiform hyperplasia. METHODS: We retrospectively reviewed 15 AD and 15 PSO patients showing psoriasiform hyperplasia. Clinical, histopathologic featur

Associations of plasma proteomic and polygenic profiling with incident psoriasis risk: a prospective cohort study.

BACKGROUND: Psoriasis is a common chronic immune-mediated disease with substantial systemic and public health burden, yet early molecular signatures associated with its onset remain insufficiently characterised. Although both circulating proteins and genetic susceptibility may contribute to psoriasis development, their prospective and joint associations with incident psoriasis remain unclear. We aimed to evaluate the individual and combined effects of proteomic and polygenic profiling on psoriasis risk. METHODS: We identified psoriasis-related protein signature using plasma proteomic data (29

NLRP1 inflammasome activation in skin equivalents reveals mechanistic insights into the roles of keratinocytes in psoriasis.

The pro-inflammatory cytokines interleukin(IL)-1β and IL-36γ are key drivers of psoriasis, an inflammatory skin disease for which a causal therapy is not available. However, the mechanisms underlying regulation of these cytokines in psoriasis remain poorly understood. Generation of IL-1β activity is regulated by inflammasomes. We activated the NLRP1 inflammasome in human keratinocytes cultivated in three-dimensional skin equivalents. NLRP1 activation induced histological and molecular features highly reminiscent of psoriasis. Mechanistically, the phenotype was dependent on IL-1, which triggere

Targeting cutaneous inflammation: A review on advanced topical delivery systems and precision medicine in psoriasis.

Psoriasis is a chronic autoimmune-mediated inflammatory skin disorder that is characterized by increased keratinocyte proliferation, abnormalities in the immune system, and abnormalities in the epidermal barrier. The conventional topical treatments for the management of psoriasis include corticosteroids, vitamin D analogues, and keratolytic. The long-term efficacy of these topical treatments for the management of psoriasis is marred by issues such as skin penetration difficulties, side effects, tachyphylaxis, and poor patient compliance. In recent times, nanotechnology-based drug delivery syst

The multifaceted role of NF-κB signaling in psoriasis: from inflammatory amplification to epidermal remodeling.

Psoriasis is a chronic immune-mediated inflammatory skin disease characterized by persistent epidermal hyperplasia and relapsing inflammation. Although extensive progress has been made in elucidating individual pathogenic pathways, a unifying framework that integrates innate immune activation, adaptive immune maintenance, and keratinocyte-driven amplification remains incomplete. Accumulating evidence indicates that nuclear factor-κB (NF-κB) functions as a central convergence hub that coordinates these diverse inflammatory inputs in psoriasis. In this review, we synthesize recent advances demon
