Transglutaminase-mediated cross-linking of a peptic fraction of omega-5 gliadin enhances IgE reactivity in wheat-dependent, exercise-induced anaphylaxis.

BACKGROUND: Patients with wheat-dependent, exercise-induced anaphylaxis (WDEIA) experience recurrent anaphylactic reactions when exercising after ingestion of wheat products. We have identified omega-5 gliadin (Tri a 19) as a major allergen in WDEIA, but the role of exercise in eliciting the symptoms remains obscure. OBJECTIVE: The aim was to examine whether tissue transglutaminase (tTG)-mediated cross-linking could be involved in modulating the IgE-binding ability and in vivo reactivity of digested omega-5 gliadin peptides in WDEIA. METHODS: Purified omega-5 gliadin was digested with pepsin

Targeting Aromatase and HDACs in Hormone Receptor-Positive Breast Cancer: Molecular Interplay, Endocrine Resistance and Therapeutic Opportunities.

Breast cancer is the most frequently diagnosed malignancy in women, and hormone receptor-positive disease represents its predominant subtype. Aromatase inhibitors (AIs) are a cornerstone of endocrine therapy, particularly in postmenopausal patients, but their long-term efficacy is frequently limited by intrinsic or acquired resistance. Increasing evidence indicates that epigenetic dysregulation, especially aberrant histone deacetylase (HDAC) activity, contributes to altered estrogen receptor signaling, transcriptional plasticity, activation of compensatory survival pathways, and endocrine trea

The alleviation of oxidative damage in celiac disease by black soybean peptides and their mechanism.

BACKGROUND: Celiac disease (CD) is a chronic autoimmune condition that occurs in genetically predisposed individuals when they consume gluten. The α-gliadin peptide exacerbates the CD pathological process by inducing oxidative stress, intestinal cell damage, and apoptosis. Addressing oxidative stress in CD may offer a valuable strategy for supplementary treatment. RESULTS: Black soybean peptides (BSPs) have been shown to inhibit oxidative stress and inflammation, although the specific mechanisms are not fully understood. This study investigated the effects of BSPs on Caco-2 cytotoxicity induc

YTHDF2 drives malignant progression in cutaneous melanoma through m⁶A-dependent HMOX1-mediated ferroptosis.

N6-methyladenosine (m6A) modification plays a critical role in eukaryotic mRNA and is significantly associated with cutaneous melanoma. This study investigated the role of the m6A-binding protein YTHDF2 in the pathogenesis of cutaneous melanoma and its potential therapeutic applications. Analyses of public databases indicated that YTHDF2 exhibited elevated expression in various cancers, including cutaneous melanoma, where its levels positively correlated with PD-L1 and negatively correlated with immune and stromal scores. Functionally, the downregulation of YTHDF2 in melanoma cells suppressed

Injectable hydrogel-mediated spermidine delivery triggers tumor pyroptosis via oxidative metabolism for enhanced cancer therapy.

Intracellular polyamine metabolic homeostasis is closely linked to therapeutic outcomes in cancer. However, their intrinsic antitumor mechanisms have not been fully clarified. Herein, we demonstrate that the catabolism of spermine (Spm) and spermidine (Spd) mediated by polyamine oxidase directly generates two potent cytotoxic metabolites, namely acrolein and hydrogen peroxide. These metabolites act synergistically to trigger a marked elevation in intracellular reactive oxygen species (ROS), which in turn activate the caspase-1/GSDMD and caspase-3/GSDME signaling pathways, ultimately driving tu

Starvation of leukemic cells enhances DNA damage-induced apoptosis in vitro via ROS/p38 MAPK and prevents leukemia progression in fasting xenograft mice.

Most children with acute lymphoblastic leukemia (ALL) achieve long-term survival due to intensive multimodal chemotherapy. However, the use of cytotoxic DNA-damaging agents is frequently associated with severe long-term side effects, prompting continued efforts to improve treatment strategies. This study explores the potential of starving the leukemic cells to enhance the efficacy of DNA-damaging therapy in ALL. Previous work demonstrated that cAMP signaling attenuates DNA damage-induced apoptosis in ALL cells, both in vitro and in a xenograft model. The current findings show that glucose and

Mechanism of the Non-Host Flavonoid Nobiletin in Inhibiting the Phytopathogen Sclerotinia sclerotiorum.

Sclerotinia sclerotiorum is a devastating phytopathogenic fungus with a broad host range, which is partly attributed to its ability to detoxify a wide array of host-derived secondary metabolites. Among these, flavonoids are one of the most widespread classes of plant secondary metabolites and are ubiquitously present in both host and non-host plants of S. sclerotiorum. Understanding the mechanisms by which non-host flavonoids inhibit this pathogen is therefore crucial, not only for elucidating the biochemical basis of non-host resistance but also for discovering novel plant-derived fungicides.

A Perturb-seq map of a differentiation hub reveals synergistic vulnerabilities in KMT2A-rearranged acute myeloid leukemia.

KMT2A-rearranged acute myeloid leukemia is driven by epigenetic dependencies yet remains clinically resistant to therapies targeting individual regulators, indicating that resistance reflects compensatory regulation across an epigenetic network. A systematic understanding of this compensatory network has been lacking. To address this gap, we utilized Perturb-seq screening to systematically map the functional architecture of this network. We uncovered a compensatory epigenetic circuit, where a synergistic hub including KAT6A, Menin, and DOT1L converges to silence a core differentiation program

Quercetin alleviates LPS-induced inflammation and immunosuppression in broiler spleen via regulating the mtDNA/cGAS/STING axis.

Quercetin has significant biological activities in anti-oxidation, anti-inflammation, immunomodulation and anti-cancer. Lipopolysaccharide, a key factor in inducing sepsis, is widely used to simulate the pathological process of endotoxemia. The study sought to elucidate the ameliorative role of quercetin and its underlying mechanisms mitigate lipopolysaccharide-induced splenic damage in broiler. The results showed that lipopolysaccharide stimulation caused splenic tissues edema and inflammatory infiltration, increased reactive oxygen species levels, suppressed antioxidant enzyme activities, an

Rosuvastatin calcium and pravastatin sodium regulate the antioxidant system in zebrafish liver through the PI3K/Nrf2/ARE signaling pathway.

Rosuvastatin calcium (ROV) and pravastatin sodium (PRA) are common drugs used for the treatment and prevention of cardiovascular diseases. They have been frequently detected in the global aquatic environment, but their toxic effects and molecular mechanisms on the antioxidant system of fish remain unclear. In this study, zebrafish were exposed to ROV or PRA for 48 h to investigate their effects on the hepatic antioxidant response system of zebrafish and the possible underlying molecular mechanisms. The results showed that after exposure to the two drugs, the levels of reactive oxygen species (

Hijacking tumor metabolism via sonocatalysis to orchestrate cuproptosis-pyroptosis coupling for potent antitumor immunity.

In response to the high metabolic resilience and immunosuppression characteristic of triple-negative breast cancer, this study established a CuGaO2 (CGO) nano sonocatalytic platform to reshape the therapeutic landscape through a metabolic hijacking dual death cascade strategy. The ultrasound activates CGO to generate reactive oxygen species (ROS), and facilitates the release of Cu+ and Ga3+ in tumor. The Ga3+ precisely disrupts iron metabolism and suppresses the expression of Fe-S cluster proteins via the metal mimicry effect, thereby achieving metabolic disarmament at a fundamental level. The

Lycorine protects against inflammatory hemorrhagic skin injury via suppression of neutrophil-driven NADPH oxidase activation.

Lycorine (LYC) , an alkaloid derived from Amaryllidaceae plants, exhibits broad biological activities, including anti-inflammatory, antiviral, and antitumor effects. However, its role in inflammatory hemorrhagic skin diseases and whether LYC modulate Nicotinamide Adenine Dinucleotide Phosphate(NADPH)oxidase activation remain unclear. In this study, we demonstrate that LYC dose-dependently alleviates lipopolysaccharide (LPS) and tumor necrosis factor-α (TNF-α) co-induced hemorrhagic inflammation in a murine Local Shwartzman Reaction (LSR) model. LYC treatment markedly reduced cutaneous hemorrha

PLL-g-HPA Hydrogel Microneedles Loaded With Rhoifolin Target Macrophages to Alleviate Fibroblast Senescence for Diabetic Wound Healing.

In this study, we demonstrate that during diabetic wound healing, interleukin-11 (IL-11) promotes the M1 polarization of macrophages, which subsequently triggers fibroblast senescence through the enhanced release of pro-inflammatory cytokines IL-1β and IL-6. Notably, Rhoifolin (Rho) effectively inhibits IL-11-induced M1 polarization, alleviates the secretion of IL-1β and IL-6, and concurrently promotes the release of choline, thereby mitigating fibroblast senescence. Based on these mechanistic findings, we constructed a targeted delivery system by loading Rho into mesoporous silica nanoparticl

Optimization, characterization and biological activity of siderophore produced by marine Streptomyces coelicolor.

Two Streptomyces spp. were isolated from open ocean waters of the Gulf of Mannar. Of the two isolates, one was identified as Streptomyces coelicolor through conventional and molecular approaches, and it was found to produce a siderophore. Characterization revealed the siderophore to be of trihydroxamate type with hexadentate iron-binding capacity. FTIR analysis indicated the presence of aromatic rings with C-O and C = C stretching, while 1H and 13C NMR together with mass spectrometry confirmed the hydroxamate nature and identified the siderophore as ferrioxamine. A narrow shift in λ max indica

Nanomedicine design strategy for preferential elimination of cancer stem cells by targeted HIF-Warburg-immune evasion axis: Systematic review.

Cancer stem cells (CSCs) drive tumor recurrence, metastasis, and therapeutic resistance.Rather than exhibiting a uniform glycolytic phenotype, CSCs occupy tumor- and state-dependent metabolic configurations ranging from glycolysisdominant to oxidative phosphorylation(OXPHOS)-dependent and hybrid states. In hypoxic and hypoxia-inducible factor-1α(HIF-1α)-active CSC subpopulations, aerobic glycolysis can support stemness, treatment resistance, and immune evasion by increasing glucose consumption, lactate production, and microenvironmental acidification. Accordingly, the HIF-Warburg axis represen

Herbal extracts as antibiotic adjuvants against ESKAPE pathogens: mechanisms and therapeutic potential.

The escalating crisis of antimicrobial resistance, particularly driven by ESKAPE pathogens, poses a serious threat to global public health. With the stagnant pipeline of novel antibiotic discovery, repurposing herbal extracts as antibiotic adjuvants to revive existing therapies is a promising strategy. This review systematically elaborates the multi-faceted mechanisms by which herbal extracts and their bioactive constituents counteract multi-drug resistance in ESKAPE pathogens. These mechanisms include the inhibition of antibiotic-degrading enzymes, alteration of bacterial membrane permeabilit
