Integration of metabolic and evolutionary processes to construct efficient xylose-utilizing strain of Zymomonas mobilis for lignocellulosic ethanol production.

Xylose is a major component of lignocellulose, and it is crucial to enable microorganisms the capability of efficient xylose utilization for economical lignocellulosic biochemical production. Zymomonas mobilis is a natural ethanologenic bacterium with xylose-utilizing recombinant strains engineered for commercial lignocellulosic ethanol production. However, low efficiency of xylose utilization is still the major hurdle for lignocellulosic biochemical production, especially in the lignocellulosic hydrolysate containing inhibitory compounds and mixed sugars of glucose and xylose. In this study,

Phenotypic Physiological and Metabolomic Analyses Reveal Crucial Metabolic Pathways in Quinoa (Chenopodium quinoa Willd.) in Response to PEG-6000 Induced Drought Stress.

Drought stress seriously threatens human food security, and enhancing crops' drought tolerance is an urgent problem to be solved in breeding. Quinoa is known for its high nutritional value and strong drought tolerance, but its molecular mechanism in response to drought stress is still unclear. In this study, we used drought-tolerant (D2) and drought-sensitive (ZK1) quinoa varieties, and PEG-6000 was used to simulate drought stress in quinoa seedlings. Phenotypic and physiological biochemical indicators were measured during the seedling stage, and LC-MS was used for a metabolite analysis of dro

Dynamic Characterization of Antioxidant-Related, Non-Volatile, and Volatile Metabolite Profiles of Cherry Tomato During Ripening.

Cherry tomato is a notable dietary source of metabolites associated with antioxidant functions. However, how ripening reshapes primary, specialized, and volatile metabolites remains incompletely resolved. Green-ripe and red-ripe fruits were comparatively analyzed using targeted HPLC assays for quality indices and vitamins, UPLC-MS/MS for non-volatile metabolites, and HS-SPME-GC-MS for volatiles. Ripening was accompanied by a pronounced accumulation of lycopene and an increase in soluble solids, reflecting a shift of sugars toward glucose and fructose while sucrose remained low. Organic acids d

Nutritional Status of Puerto Rican Families in 1966 - What can be learned from Retrospective Surveys?

Previous nutritional surveys have been conducted with Puerto Rican families, but extensive analyses of dietary results have been lacking due to databases with insufficient nutrient composition information. Herein, we determine the nutritional status of Puerto Rican families from the results of a survey conducted in 1966 using the Minnesota's Nutritional Data System for Research 2020 from which we report macronutrients, vitamins, minerals, and other nutrients, with the results being evaluated for compliance with current dietary recommendations. These data establish eating patterns that could ha

Enhanced methane production from bloom algal biomass using hydrothermal and hydrothermal-alkaline pretreatment with anaerobic digestion.

Coalbeds have the potential as geobioreactors for producing renewable natural gas from biomass derived from photosynthesis. This brings about a number of benefits, including support for sustainable energy and the sequestration of carbon dioxide in coal. In this study, freshwater bloom algae were employed as the substrate to examine the influence of hydrothermal and hydrothermal-alkaline pretreatment on methane production using an inoculum from an anaerobic digester. The morphology and chemical structures of the biomass, as well as the volatile fatty acids (VFAs) in the liquid fraction of the p

Diet in Knee Osteoarthritis-Myths and Facts.

Knee osteoarthritis (OA) is a common degenerative joint disease affecting global health. Its increasing prevalence, particularly among aging populations, remains a leading cause of disability. Besides conventional pharmacological and surgical treatments, dietary interventions are promising strategies to alleviate OA symptoms and progression. Unfortunately, scientific evidence does not support many commonly used, misleading ideas about nutrition in knee OA. Recent data highlight the detrimental effects of high-carbohydrate and high-fat diets, particularly those rich in refined sugars and satura

Two new members of the family Aerococcaceae: Tuanshanicoccus yangjingiae gen. nov., sp. nov. and Tuanshanicoccus lijuaniae sp. nov., isolated from the trachea of Marmota himalayana.

Nine strains of two Gram-positive, facultative anaerobic, catalase-, oxidase- and urease-negative coccoid species, designated as zg-252T, zg-1578, zg-B36, zg-ZUI334, and zg-1292T, zg-A91, zg-BR9, zg-BR22 and zg-BR33, were isolated from the trachea of Marmota himalayana. The nine strains grew optimally at 37 °C, pH 7.0 and 0.5% NaCl. Comparative analysis of the 16S rRNA gene revealed that the two novel type strains were most closely related to each other, with low similarity to known species in the family Aerococcaceae. Phylogenetic analyses based on the 16S rRNA gene and genome suggested that

Na⁺/Mg²⁺ ratio: a new physiological trait for salt resistance in faba bean (Vicia faba L.).

Magnesium plays a vital role in enhancing plant resilience under salt stress. However, its specific function in maintaining ion homeostasis, particularly in regulating sodium uptake, remains unclear. Recognizing that magnesium deficiency leads to increased potassium uptake and accumulation, and given that sodium and potassium possess the same charge, we hypothesize that salt stress disrupts ion homeostasis to a greater extent in magnesium-deficient plants compared to those deficient in potassium. To test this hypothesis, Vicia faba plants were cultivated hydroponically and subjected to moderat

Middle-aged dogs with low and high Aβ CSF concentrations show differences in energy and stress related metabolic profiles in CSF.

BACKGROUND: Amyloid beta (Aβ) accumulation in the brain is one of the earliest findings in Alzheimer's disease (AD). The dog is a natural animal model for amyloid processing and early brain amyloid pathology. The goal of this study is to examine which differences in metabolomic profiles in cerebrospinal fluid (CSF) could be detected in dogs with a difference in CSF Aβ concentrations before amyloid accumulation occurs. METHOD: Metabolic profiling was performed on CSF from 4 to 8 year old dogs with different CSF Aβ concentrations. RESULTS: Metabolomic profiling of CSF showed differences in bra

Influence of dietary and lifestyle factors on levels of inflammatory markers (IL-6, IFN-γ and TNF-α) in obese subjects.

INTRODUCTION: The low-grade inflammation occurring in obese individuals leads to many diseases, including cardiovascular disease (CVD). Dietary patterns, food groups or nutrients in a well-balanced diet may reduce the level of pro-inflammatory markers and the risk of obesity-related morbidities. Our study aims to describe three cytokines in obese patients in relation to dietary habits, lifestyle and body composition. MATERIAL AND METHODS: Serum samples were collected from 84 obese adult volunteer subjects [body mass index (BMI) ≥ 30 kg/m2] to analyze the concentrations of interleukin 6 (IL-6)

Athletes' medical preventive behaviors: the case of oral health and ultraendurance trail runners.

Ultra-endurance runners are particularly exposed to caries risk factors due to the continuous intake of sugars in liquid or sticky forms during long-distance exercise. The risk of caries increases due to a failure to perform oral hygiene during races. Ultra-endurance runners had good oral health status despite these particularities. Our hypothesis is that high compliance with preventive oral health recommendations (toothbrushing and preventive or regular dental visits) counterbalance the risks associated with their exposure to caries. We aimed to gain a better understanding of preventive denta

Recent Advancements in Harnessing Lactic Acid Bacterial Metabolites for Fruits and Vegetables Preservation.

Postharvest losses in fruits and vegetables exert substantial economic and environmental repercussions. Chemical interventions are being widely utilized for the past six decades which may lead to significant health complications. Bioprotection of fruits and vegetables is the need of the hour in which use of lactic acid bacteria (LAB) with GRAS status predominantly stands out. Incorporation of LAB in postharvest fruits and vegetables suppresses the growth of spoilage organisms by synthesizing various antimicrobial compounds such as bacteriocins, organic acids, hydrogen peroxide (H2O2), exopolys

Toward the apoplast metabolome: Establishing a leaf apoplast collection approach suitable for metabolomics analysis.

The leaf apoplast contains several compounds that play important roles in the regulation of different physiological processes in plants. However, this compartment has been neglected in several experimental and modelling studies, which is mostly associated to the difficulty to collect apoplast washing fluid (AWF) in sufficient amount for metabolomics analysis and as free as possible from symplastic contamination. Here, we established an approach based in an infiltration-centrifugation technique that use little leaf material but allows sufficient AWF collection for gas chromatography mass spectr
