SHANK3 regulates nuclear hnRNP K and oxidative stress-associated DNA damage signaling in neurons.

SHANK3 is a major postsynaptic scaffold protein that plays essential roles in synaptic organization and excitatory neurotransmission. Beyond its established synaptic functions, SHANK3 has been shown to translocate from excitatory synapses to the nucleus following synaptic stimulation. Here, we demonstrate that nuclear SHANK3 localization changes during the differentiation of human induced pluripotent stem cell-derived neurons, with transient enrichment at the neural rosette stage. To identify potential nuclear interaction partners, we analyzed SHANK3-associated protein complexes in HEK293T cel

Effects of testosterone upon the survival of new neurons in the dentate gyrus of adult male rats.

Prolonged testosterone exposure promotes adult hippocampal neurogenesis, but it remains unclear whether exposure during specific time periods of neural development is necessary for testosterone to enhance neuron survival. Our experiment tested the effects of testosterone on the later stages of neural development, when functional synapses are forming. Adult male rats (n = 6-8/group) were bilaterally castrated and given a single injection of bromodeoxyuridine (BrdU; 200 mg/kg). Daily injections of testosterone (0.500 mg/rat) or sesame oil (0.1 ml/rat) were given 1-30, 1-15, or 16-30 days after B

Alcohol drinking sex-dependently regulates interleukin-1 pro-inflammatory signaling in the prefrontal cortex of mice and rhesus macaques.

Alcohol use disorder (AUD) causes executive dysfunction, leading to reduced treatment adherence, worse clinical outcomes, and relapse. Current medications do not address these symptoms. Targeting the neuroimmune interleukin-1 (IL-1) system has potential, but its mechanistic role in prefrontal cortex (PFC) dysfunction is unclear. Here we investigated how chronic alcohol consumption shifts IL-1 regulation of the rodent and non-human primate PFC. We found that alcohol drinking decreased reference memory and shifted the search strategy in mice of both sexes, and this deficit correlated with altere

Two populations of heteromeric acetylcholine receptors are present at Caenorhabditis elegans neuromuscular junctions.

Ligand-gated acetylcholine receptors (AChRs) are pentameric transmembrane proteins that display extensive molecular diversity through combinatorial assembly of subunits. In the nematode Caenorhabditis elegans, this diversity is exceptionally expanded, with nearly 60 AChR subunit genes, yet the composition and function of many receptor subtypes remain poorly defined. At neuromuscular junctions (NMJs), L-AChRs are heteromeric AChRs sensitive to levamisole, a nematode-specific L-AChR agonist, that play a crucial role in locomotion. Forward genetic screens for mutants resistant to levamisole have

Temporal transcriptomic changes during neurodevelopment in a mouse model of Smith-Lemli-Opitz syndrome.

Smith-Lemli-Opitz syndrome (SLOS) is a cholesterol biosynthesis disorder caused by mutations in the DHCR7 gene, leading to reduced cholesterol production and accumulation of its precursor, 7-dehydrocholesterol. SLOS displays a wide range of neurodevelopmental defects, intellectual disability, and behavioral problems. However, an in-depth study of the temporal changes in gene expression in developing brains has not been conducted before. In this work, we carried out the transcriptomic analysis of whole brains from WT and Dhcr7-KO mice at embryonic day 12.5 (E12.5), E14.5, E16.5, and postnatal d

An R83W mutation in Rab3A causes autosomal-dominant cerebellar ataxia.

Spinocerebellar ataxias (SCAs) are a group of progressive neurodegenerative disorders caused by pathogenic variants in more than 40 genes with diverse cellular functions. In this study, we identified the c.247C > T p.(Arg83Trp) variant in RAB3A, encoding a small GTPase involved in membrane-associated regulated exocytosis, in two families with cerebellar ataxia. Affected individuals presented with adult-onset, gradually progressive cerebellar symptoms, often accompanied by mild gait spasticity and tremors. Variable features of neurodevelopmental disorders were also observed. Brain MRI consisten

Cognitive loss after brain trauma results from sex-specific activation of synaptic pruning processes.

Cognitive losses resulting from severe brain trauma have long been associated with the focal region of tissue damage, leading to devastating functional impairment. For decades, researchers have focused on the sequelae of cellular alterations that exist within the perilesional tissues; however, few pharmacological therapies are available to patients. To examine whether expansive global synaptic damage underlies cognitive losses associated with brain injury, we evaluated the influence of D-serine on synaptic damage in male and female wild-type mice as well as mice deficient in microglial serine

Loss of vesicular monoamine transporter 2 in striatum of long COVID and relationship to neuropsychiatric symptoms.

BACKGROUND: Dopaminergic neurons are vulnerable to injury from gliosis and have high density of ACE2 receptors, but the integrity of dopaminergic neurons has not been investigated in long COVID. This study examined whether vesicular monoamine transporter 2 (VMAT2) binding, index of dopamine-releasing neuron density, is reduced in the striatum in long COVID and associated with neuropsychiatric symptoms. METHODS: This case-control study (Aug 2022-Apr 2025, Toronto, Canada) included 24 adults with long COVID and 24 age-matched healthy controls, and the healthy control sample was extended to 43 f

Phosphoproteomic profiling reveals reversal of dysregulated kinase signaling by nitric oxide inhibition in the Shank3 mouse model of autism.

Autism spectrum disorder (ASD) is a wide-ranging neurodevelopmental disorder characterized by behavioral deficits, including difficulties in social communication and interaction, restricted or repetitive behaviors, and limited interests. The SHANK3 gene encodes a scaffolding protein in the postsynaptic density of glutamatergic excitatory synapses and is one of the highest-confidence ASD risk genes. Our previous studies showed abnormally elevated levels of nitric oxide (NO) in Shank3 models of ASD, and inhibition of the neuronal NO synthase (nNOS) reversed ASD-like behavioral and synaptic pheno

Digging Deep into Alzheimer Disease: How Electron Microscopy Helps Elucidating Its Pathogenesis.

Alzheimer disease (AD), first described by Alzheimer and Perusini in the early twentieth century, is a devastating neurodegenerative disorder without a definitive cure. Unraveling the subcellular alterations underlying AD is essential to elucidate disease mechanisms, track progression, link cellular abnormalities to functional deficits, and develop therapeutic strategies aimed at preventing, slowing or reverting the disease course. Electron microscopy (EM) has been pivotal in this field since the 1960s, when Terry and Kidd characterized the ultrastructure of amyloid-beta (Aβ) deposits and pair

Evolving Roles of Primary Cilia in CNS Development and Neural Circuit Function: From Human Disease to Molecular Underpinnings.

Primary cilia are small organelles acting as cellular antennae that sense and transduce diverse signals, including developmental signaling pathways in the developing central nervous system (CNS). This signaling is essential for normal CNS development, as evidenced by the prevalence of neurodevelopmental phenotypes in conditions arising from primary cilia dysfunction (ciliopathies). Though significant research has focused on the roles of primary cilia during CNS development, the functions of primary cilia in the mature CNS have only recently become a focus of investigation. Primary cilia are pr

CRISPR inhibition of activity-dependent Arc expression in the adult mouse brain has limited effects on plasticity in visual cortex and nucleus accumbens.

Neuronal activity drives long-lasting change in circuit function by inducing the expression of gene products that modulate the function of synapses. Arc is one of the most robustly activity-regulated neuronal genes, being induced broadly in neurons by a wide range of physiologically-relevant stimuli. The ability of Arc to promote internalization of AMPA-type glutamate receptors plays an important role in activity-dependent refinement of synaptic connectivity during development. However, the consequences of Arc induction for circuit plasticity in the adult brain are less well understood. We rea

Recovery of Retinal Terminal Fields after Traumatic Brain Injury: Evidence of Collateral Sprouting and Sexual Dimorphism.

The central nervous system is characterized by its limited regenerative potential, yet striking examples of functional recovery after injury in animal models and humans highlight its capacity for repair. Little is known about repair of pathways/circuits after traumatic brain injury (TBI), which results in disruption of connectivity. Here we utilize a mouse model of diffuse traumatic axonal injury (impact-acceleration TBI) in order to explore, for the first time, the evolution of structural and functional changes in the terminal fields of the injured visual system. Retinal ganglion cell (RGC) a

Climbing fibres recruit disinhibition to enhance Purkinje cell calcium signals.

Climbing fibre (CF) inputs to Purkinje cells (PCs) instruct plasticity and learning in the cerebellum1-3. Paradoxically, CFs also excite molecular layer interneurons (MLIs)4,5, a cell type that inhibits PCs and can restrict plasticity and learning6,7. However, two types of MLI with opposing influences have recently been identified: MLI1s inhibit PCs, reduce dendritic calcium signals and suppress plasticity of granule cell to PC synapses2,6-9, whereas MLI2s inhibit MLI1s and disinhibit PCs8. To determine how CFs can activate MLIs without also suppressing the PC calcium signals necessary for pla

EA1-linked Kv1.1 dysfunction enhances susceptibility to cerebellar spreading depression and a transient cerebellar refractory state.

Episodic ataxia type 1 (EA1) is a neurological channelopathy caused by loss-of-function mutations in the KCNA1 gene, which encodes the Kv1.1 α-subunit of voltage-gated potassium channels. Clinically, EA1 is characterized by interictal myokymia and transient episodes of generalized ataxia. While the molecular basis of the disease is well established, the mechanisms underlying the paroxysmal nature of attacks remains unclear. Drawing parallels with migraine with aura, an episodic disorder in which cortical spreading depression (CSD), a propagating wave of neuronal and glial depolarization follow

Persisting Region-Specific Microglia Activation and Synaptic Imbalance During Remission in a DSS-Induced Colitis Model.

Ulcerative colitis (UC) patients experience cycles of active gut inflammation and remission, with neuropsychiatric comorbidities persisting even during clinical remission. While the dextran sulfate sodium (DSS) mouse model was previously applied to study gut-brain interactions, those studies focused on acute protocols missing the chronic, relapsing-remitting nature of human UC, when patients continue to experience central nervous system symptoms. Thus, we employed a chronic DSS treatment regimen comprising three cycles of active intestinal inflammation followed by remission phases to investiga

Disease modeling of myocilin mutation-dependent normal tension glaucoma: human retinal ganglion cell susceptibility to unfolded protein response and mTOR signaling.

Glaucoma represents a group of diseases where the unifying theme is the progressive degeneration of retinal ganglion cells (RGCs), causing irreversible vision loss. Mutations in the myocilin (MYOC) gene represent one of the most common genetic factors associated with primary open-angle glaucoma (POAG). However, the mechanism underlying MYOC mutation-associated POAG is poorly understood. Here, using human disease modeling of MYOC mutation (A445V)-dependent POAG, which is usually without ocular hypertension, we have tested a hypothesis that human RGCs (hRGCs) are the target of the mutant protein

Therapeutic potential of Cola nitida against dexamethasone- and scopolamine-induced cognitive and cardiac dysfunction.

BACKGROUND: Cognitive dysfunction and cardiometabolic disturbances frequently coexist in neurological and metabolic disorders. Chronic systemic inflammation in cardiometabolic syndrome may promote neuroinflammation, damaging neurons and synapses and impairing memory pathways. Cola nitida is a medicinal plant widely recognized for its stimulant and therapeutic properties, yet its neuroprotective and cardiometabolic effects remain underexplored. Objective: This study investigated the effects of methanol extract of Cola nitida (CN) in a mouse model of memory and metabolic dysfunction induced by s

Impaired long-term depression mediated by group I metabotropic glutamate receptors, but not muscarinic acetylcholine receptors in juvenile-onset diabetes rats.

Type 1 diabetes mellitus is a chronic disease where the destruction of insulin-producing pancreatic β cells causes hyperglycemia and its onset typically occurs during childhood or adolescence. We previously reported that long-term depression (LTD) in the CA1 region of the hippocampus, induced by a low-frequency stimulation to the Schaffer collaterals, was attenuated in juvenile-onset diabetes (JDM) rats. We herein investigated whether LTD was triggered by the activation of G-protein coupled group I metabotropic glutamate receptors (mGluR-LTD) or M1 muscarinic acetylcholine receptors (mAChR-LTD

Multiple spinal muscular atrophy disease-modifying effects of a Hspa8G470R synaptic chaperone variant.

Spinal muscular atrophy (SMA) is an oft-fatal infantile-onset neuromuscular disease caused by homozygous loss of the Survival of Motor Neuron 1 (SMN1) gene and, consequently, low SMN protein. Administration of SMN-inducing agents to SMA newborns prevents early mortality, but therapeutic outcomes vary considerably, and disease mechanisms remain poorly understood. Genetic modifiers can provide clues to disease mechanisms and serve as targets for novel treatments. Here, we describe how one such modifier, an Hspa8G470R synaptic chaperone variant we identified, suppresses SMA in model mice. Our res
