Macrophagic "Crown-like Structures" Are Associated with an Increased Risk of Breast Cancer in Benign Breast Disease.

In breast adipose tissue, macrophages that encircle damaged adipocytes form "crown-like structures of breast" (CLS-B). Although CLS-B have been associated with breast cancer, their role in benign breast disease (BBD) and early carcinogenesis is not understood. We evaluated breast biopsies from three age-matched groups (n = 86 each, mean age 55 years), including normal tissue donors of the Susan G. Komen for the Cure Tissue Bank (KTB), and subjects in the Mayo Clinic Benign Breast Disease Cohort who developed cancer (BBD cases) or did not develop cancer (BBD controls, median follow-up 14 years)

Ciliary Tissue Transplantation in the Rabbit Eye: Does the Localization of the Graft Affect Survival?

PURPOSE: To examine whether ciliary body transplantation is applicable, the graft is viable and the localization of the graft material affects graft survival. METHODS: Fifteen female New Zealand white rabbits were used. Three of them were used as donors, and a ciliary body graft was prepared from their enucleated eyes. There are two groups in the study according to the localization of the ciliary body graft in the anterior chamber. The graft was placed on the iris surface close to the pupil margin in group 1 and adjacent to the anterior chamber angle in group 2. Immunosuppressive treatment wi

Current strategies on kidney regeneration using tissue engineering approaches: a systematic review.

INTRODUCTION: Over the past two decades, there has been a notable rise in the number of individuals afflicted with End-Stage Renal Disease, resulting in an increased demand for renal replacement therapies. While periodic dialysis is beneficial, it can negatively impact a patient's quality of life and does not fully replicate the secretory functions of the kidneys. Additionally, the scarcity of organ donors and complications associated with organ transplants have underscored the importance of tissue engineering. Regenerative medicine is revolutionized by developing decellularized organs and tis

The median nerve´s system of connective tissue distal to the pronator teres to the carpal tunnel.

BACKGROUND: Connective tissue serves a role beyond mere spatial filling. Furthermore, there is increasing evidence that connective tissue plays an important role in the pathogenesis of conditions such as carpal tunnel syndrome (CTS). According to our hypothesis, the median nerve (MN) is surrounded by a system of connective tissue distal to the pronator teres and extending up to, and including, the carpal tunnel. METHODS: To visualize the connective tissue surrounding the median nerve, we dissected the forearms of 15 body donors from pronator teres to the carpal tunnel, created plastination sl

Human Islet miR-199a-5p and miR-214-3p Associate With Donor Age, BMI, and Sex and Mediate Cellular Aging in Islet-Derived Cells.

OBJECTIVES: Human islets are widely researched to understand pathophysiological mechanisms leading to diabetes. Sex, age, and body mass index (BMI) are key donor traits influencing islet function, which is also regulated by an intricate network of microRNAs. METHODS: Here, we profiled 754 microRNAs and 58 191 gene transcripts (19 919 protein-coding) in up to 131 different human islet donor preparations (without diabetes) and assessed their association with donor traits. Additionally, the effect of the age-associated key microRNAs on relative telomere length in human islet-derived cells was ev

Heart recipient outcomes following transplantation of donor hearts with impaired versus normal function: a study protocol for IMPROVED Heart, a prospective multicentre observational study.

BACKGROUND: Cardiac transplantation remains the most effective treatment for patients with end-stage heart failure, but its use is limited by donor organ shortage. Expanding donor acceptance criteria may increase the availability of transplantable hearts. Left ventricular dysfunction due to neurogenic stunned myocardium is common among organ donors and is characterised by rapid functional and structural recovery. The IMPROVED Heart (IMPact of donor heart function on Recipient Outcomes: a prospectiVE observational study to increase the utilisation of Donor Hearts) study aims to increase the num

DHCR24 in cholesterol metabolism and diseases of the nervous system.

DHCR24 (3β-hydroxysterol 24-reductase) is a critical enzyme that plays an essential role in the cholesterol biosynthesis pathway, which catalyzes the reduction of the sterol intermediate C-24 double bond. DHCR24 is also involved in cell growth, senescence, and cellular responses to carcinogenesis and oxidative stress. DHCR24 is extensively studied, particularly in neurodegenerative diseases like Alzheimer’s disease. It is thought to have neuroprotective effects such as resistance to amyloid β-peptide (Aβ) toxicity and oxidative stress. Nevertheless, there is a notable absence of systematic rev

Achieving zero waitlist mortality for children on the pediatric liver transplant waitlist.

Children with end-stage liver disease are at high risk of waitlist morbidity and mortality; children <1 year have the highest mortality of all age groups in the United States (US) (18.6 deaths per 100 patient-years). Pediatric candidates receive fewer offers of organs than adults, and excessive wait times contribute to delays in development that can have a lasting impact. Policy changes and outreach efforts have been insufficient to increase organ availability for children. Living donor liver transplantation (LDLT) is underutilized in the US, accounting for 14.6% of all pediatric transplants i

Clinical and immunological predictors of receiving deceased donor kidney transplantation and waitlist dropout: a single-center study in Japan.

BACKGROUND: In Japan, where the annual number of deceased donor kidney transplantation (DDKT) is limited, candidates typically wait about 15 years for transplantation, and many leave the waitlist before a graft becomes available. We investigated baseline factors that influence the chance of DDKT and the risk of dropout to guide individualized care. METHODS: This retrospective single-center study included 1,050 candidates who were actively listed for DDKT between 1997 and 2024. Kaplan-Meier curves described time to DDKT and dropout. Independent predictors were identified with multivariable Cox

Isolation and functional characterization of primary human proximal tubular epithelial cells from brain-dead organ donors.

Kidney proximal tubules (PT) utilize endocytosis to maintain body homeostasis. Effective PT repair mechanisms after acute kidney injury (AKI) are crucial to avoid the transition to chronic kidney disease (CKD). Cultured PT cells are often utilized to study these processes, but studies demonstrate loss of functional transporter expression in nearly all available cell lines. Here, we describe acquisition and culture of human PT cells isolated from the kidneys of brain-dead donors (bddPTC) not suitable for transplantation and designated to research. The kidney cortex was digested, and the cells f

Evaluation of dnDSA risk stratification using the updated PIRCHE-T2 model in two kidney transplant cohorts.

BACKGROUND: Development of de novo donor-specific antibodies (dnDSA) remains a key risk factor for antibody-mediated rejection and graft loss in kidney transplantation. The PIRCHE-T2 model estimates immunogenicity by predicting donor-derived HLA peptides presented via the recipient's HLA class II molecules. A recent update to the model integrates a new neural network-based peptide-binding predictor "Frost", which requires further testing of clinical performance. METHODS: We compared the predictive performance of the previous and updated PIRCHE-T2 models in two independent kidney transplant co

Donor Risk Factors Affecting Graft Survival in Pediatric Kidney Transplants: Protocol for a Systematic Review and Meta-Analysis.

BACKGROUND: Pediatric patients with end-stage kidney disease require kidney transplants (KTs) throughout their lifetime. Long-term graft survival is dependent on multiple factors, which are broadly categorized as donor- and recipient-related factors. Advances in transplant care and changes in donor population demographics necessitate an updated analysis on donor risk factors to guide clinical decision-making. OBJECTIVE: In this systematic review and meta-analysis, we will focus on the impact of donor factors on graft survival in pediatric KT, excluding transplants from donation after circulat

When Fewer Complications Cost More: A Value-Based Reanalysis of Donor-Site Reconstruction.

BACKGROUND: Reconstructive strategies are commonly evaluated according to complication rates and technical success at the individual patient level. However, interventions that reduce complications may simultaneously increase cumulative procedural exposure, operative utilization, and healthcare resource consumption across patient populations. This distinction reflects a broader epidemiologic trade-off between universal prophylactic intervention and selective escalation following clinically significant failure. METHODS: We performed a value-based reinterpretation of the donor-site reconstructio

A Flow Cytometry Crossmatch With Combined Cytotoxicity and IgG Detection: Ten Years of Use in Deceased Donor Kidney Allocation.

Virtual crossmatch (VXM) is increasingly used in deceased-donor kidney allocation, raising questions about the added value of a physical crossmatch (PXM) when donor-specific antibody (DSA) are absent. We applied our previously described flow cytometry-based assay (FCtox), combining complement-mediated cytotoxicity and IgG binding, across 3236 routine crossmatches performed between 2018 and 2024. All donor offers underwent VXM followed by prospective PXM. Among 704 patients, 95% of FCtox assays were performed in VXM- cases. FCtox positivity was uncommon in VXM- cases (9%), and overall concordan

Different States of Lung Allograft Injury Assessed by Plasma Donor-Derived and Total Cell-Free DNA.

Lung allografts are susceptible to myriad injury types, including acute rejection (AR), infectious disease (ID), baseline lung allograft dysfunction (BLAD), and chronic lung allograft dysfunction (CLAD), that affect outcomes. Donor-derived cell-free DNA (dd-cfDNA) is validated for detecting AR after lung transplantation (LT). However, data are limited regarding the ability of dd-cfDNA or total cell-free DNA (TcfDNA) to detect or differentiate other clinical conditions. This study stratified patients into Stable, AR, CLAD, and ID cohorts. Stable double LT recipients were further stratified into

Responders vs. non-responders to mesenchymal stromal cells in knee osteoarthritis: Mechanistic correlates of donor cell and patient features.

Mesenchymal stromal cell (MSC) injection has afforded heterogeneous outcomes in knee osteoarthritis (KOA). Herein, a framework that dually correlates KOA patient responsiveness with baseline autologous bone marrow-derived MSC(M) donor batch attributes and baseline clinical and biomarker features is provided. Using clinical trial data (NCT02351011), we demonstrated that MSC(M) with increased immunomodulatory potency are more efficacious. Multivariable MSC(M) genes correlated strongly with responder status and 12- and 24-month improvements in Knee Injury and Osteoarthritis Outcome Scores. Respon
