Clinical trial landscape and translational prospects of natural killer cell-based immunotherapy: an analysis based on the INFORMA database.

Natural killer (NK) cell-based immunotherapy is an increasingly important cancer treatment strategy because of its innate cytotoxicity, allogeneic potential, and compatibility with off-the-shelf manufacturing. We analyzed 287 clinical trials identified in the INFORMA database to characterize the global development of NK-cell therapies across phases, regions, indications, combinations, cell sources, product origins, and engineering platforms. Most trials were planned or early-phase, and combination regimens were more frequent than monotherapy. The USA, China, and South Korea led clinical activi

Closing the Translational Gap: Closed-Loop AI Discovery Frameworks for Experimental Validation and Clinical Implementation in Cancer Therapeutics.

The development of novel cancer therapeutics is a protracted, costly endeavor with high attrition rates, largely attributed to tumor heterogeneity and acquired resistance. Artificial intelligence (AI) is emerging as a powerful technology to enhance the drug discovery pipeline, employing multimodal datasets to identify therapeutic targets, design de novo candidates, and discover biomarkers. However, a significant validation gap persists. AI models frequently hallucinate chemically implausible molecules, overfit to biased training datasets (particularly immortalized cell lines that poorly repres

Biomarkers of Hypercoagulability and Thromboinflammation in Cervical Cancer-Associated Thrombosis: A Systematic Review with Translational Insights from Breast Cancer.

Cancer-associated thrombosis (CAT) is a major cause of morbidity and mortality in patients with malignancy. Biomarkers of hypercoagulability and thromboinflammation may improve risk stratification and support personalised thromboprophylaxis, but evidence remains heterogeneous, particularly in cervical cancer. A systematic review was conducted according to PRISMA 2020. PubMed/MEDLINE, Scopus, Embase, and Web of Science were searched for studies published between January 2009 and March 2025 evaluating biological, molecular, genetic, and imaging biomarkers associated with hypercoagulability and t

Protein-protein interactions as therapeutic targets in breast cancer: integrated computational and experimental advances, mechanisms and clinical translation.

Breast cancer is a highly heterogeneous malignant tumour, whose initiation and progression are closely associated with aberrant rewiring of protein-protein interactions (PPIs). This review systematically summarizes the core regulatory mechanisms of PPIs in breast cancer: focusing on neomorphic interactions (neoPPIs) and hypomorphic interactions (hypoPPIs) induced by driver mutations, and dissecting their regulatory effects on key pathways such as apoptotic pathways, DNA damage repair, and hormone receptor signalling. It also summarizes the application progress of critical technologies for PPI

Global development of bispecific antibodies in oncology: a bibliometric analysis of geographic disparities, collaborative models, and emerging translational strategies.

BACKGROUND: Bispecific antibodies (bsAbs) have rapidly emerged as one of the most promising therapeutic strategies in oncology because of their ability to simultaneously engage multiple molecular targets and enhance antitumor immunity. Although the number of publications has increased substantially, a comprehensive bibliometric characterization of the global research landscape and its developmental trajectory remain lacking. METHODS: A bibliometric analysis was conducted using publications retrieved from the Web of Science Core Collection and Scopus from January 2014 to July 2026. After data

Spatially guided translation from histology images to transcriptomic profiles using foundation model-driven contrastive learning.

Spatial transcriptomics (ST) enhances single-cell RNA sequencing by revealing transcript distribution, offering critical insights into heterogeneous diseases such as breast cancer. However, the high cost and lengthy processes of generating high-quality ST data limit clinical application. Recent deep learning methods predict ST from histology images, but often fail to capture both morphological features and spatial context. We introduce FOCST, a foundation model-driven framework for ST imputation that leverages spatial guided contrastive learning. FOCST begins with UNI, a large histopathology f

Context-dependent roles of cGAS-STING signaling in glioblastoma: Therapeutic resistance, drug delivery, and heterogeneity-informed translation.

Glioblastoma (GBM) is an aggressive primary brain tumor with poor prognosis, profound spatial and cellular heterogeneity, and a highly immunosuppressive microenvironment. The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central cytosolic DNA-sensing axis that activates innate immune responses, largely through the induction of type I interferons and inflammatory signaling. Increasing evidence indicates that the role of cGAS-STING signaling in GBM is highly context dependent. Acute activation, particularly in immune cells, may enhance antitumor immunity by pro

Quantifying VMAT2 target occupancy at effective valbenazine doses and comparing to a novel VMAT2 inhibitor: a translational PET study.

Positron emission tomography (PET) is frequently used to obtain target occupancy (%TO) of central nervous system (CNS) drug candidates during clinical development. Obtaining %TO with PET can be particularly powerful when the %TO associated with efficacy is known for a protein target. Using the radiotracer [18F]AV-133, the relationship between plasma concentration (PK) and %TO of NBI-750142, an experimental inhibitor of the vesicular monoamine transporter type 2 (VMAT2) was obtained in both nonhuman primate (NHP) and human. This work established [18F]AV-133 PET as capable of providing a VMAT2 i

FBXW7 Gene Mutation and Expression in Colorectal Cancer (CRC): A Systematic Review from Molecular Mechanisms to Clinical Translation.

In the context of precision oncology, understanding the molecular drivers of colorectal cancer (CRC) is critical for improving prognosis and guiding targeted therapy. FBXW7 is a tumor suppressor that plays a pivotal role in CRC by regulating the degradation of key oncogenic proteins, influencing tumor initiation, growth, therapeutic response, and metastatic behavior. Mutations in FBXW7 occur in 6-10% of CRC. Despite its biological relevance, the prognostic and predictive role of FBXW7 in CRC remains unclear, with inconsistent findings across studies. This systematic review collects and analyze

Hypoxia-Induced circPRELID2 Promotes Gastric Cancer Metastasis by Facilitating ZEB2 Translation via PCBP1 O-GlcNAcylation.

Circular RNAs (circRNAs) are involved in the occurrence and development of various carcinomas. However, the biogenesis, function and underlying mechanism of hypoxia-induced circRNA in gastric cancer (GC) are poorly understood. Here, a novel circRNA, circPRELID2, which is upregulated by HIF1A under hypoxic conditions, is identified. circPRELID2 is highly expressed in GC tissues, and positively correlates with lymph node invasion, vascular invasion in GC patients. Elevated circPRELID2 promotes the epithelial-mesenchymal transition (EMT) and metastasis of GC cells both in vitro and in vivo under

TOPK Inhibition Promotes Anti-Tumor Immunity Via eIF4F Complex Mediated STAT1 Translation in Gastric Cancer.

Immune checkpoint blockade-directed immunotherapy emerges as a revolutionary therapy in gastric cancer (GC). However, the proportion of patients who can benefit from it and its overall efficacy remain limited. Here the aim is to identify key dual-function targets that both inhibit proliferation and suppress immune evasion. Using whole genome-wide CRISPR-Cas9-based screening, the serine/threonine kinase T-lymphokine-activated killer cell-originated protein kinase (TOPK) is identified as a key regulator of PD-L1 in gastric cancer upon IFN-γ stimulation. Mechanical study is performed to explore t

Characterisation of post-translational and transcriptional reprogramming of the immune response to ISAV and IPNV infections in salmon head kidney cells.

Viral diseases remain a major barrier to the sustainable production of farmed fish, primarily attributable to the absence of effective prevention and treatment options. Understanding host-pathogen interactions can guide the development of vaccines, antiviral therapies, or gene editing strategies. Ubiquitination is a post-translational modification capable of regulating protein activation, structure, and degradation. As such, it is known to regulate many aspects of immune functions in model species, but is currently understudied in fish. This study leverages ubiquitin-enriched mass spectroscopy

Mechanisms and clinical translation of ICOS/ICOSL signaling pathway blockade in delaying vascular complications of type 2 diabetes.

BACKGROUND: Type 2 diabetes mellitus (T2DM) is often complicated by vascular conditions such as atherosclerosis, which contribute significantly to morbidity and mortality. The ICOS/ICOSL signaling pathway has emerged as a promising target for mitigating these complications. This study aims to investigate the effects of ICOS/ICOSL pathway blockade on vascular inflammation and endothelial dysfunction in T2DM and atherosclerosis, and to assess its potential for clinical translation. METHODS: Peripheral blood mononuclear cells (PBMCs) were collected from T2DM patients, with and without atheroscle

Weight trajectories in aging humanized APOE mice with translational validity to human Alzheimer's risk population: A retrospective analysis.

Translational validity of mouse models of Alzheimer's disease (AD) is variable. Because change in weight is a well-documented precursor of AD, we investigated whether diversity of human AD risk weight phenotypes was evident in a longitudinally characterized cohort of 1,196 female and male humanized APOE (hAPOE) mice, monitored up to 28 months of age which is equivalent to 81 human years. Autoregressive Hidden Markov Model (AHMM) incorporating age, sex, and APOE genotype was employed to identify emergent weight trajectories and phenotypes. In the hAPOE-AD mouse cohort, five distinct weight traj

Critical analysis of translational potential of rodent models of white matter pathology across a wide spectrum of human diseases.

Rodents are the most commonly used laboratory animals in medical research. However, significant evolutionary divergences between humans and rodents, particularly in the complexity of white matter connectome, which are fundamentally shaped by myelin as their major structural component, pose critical challenges in modeling the human neurological diseases. Given the divergences and central roles of myelin in pathology, a thorough reevaluation of the rodent models used in contemporary research is critical, alongside the careful selection, optimization, or de novo development of models that faithfu

Therapeutic Agents Targeting the Nrf2 Signaling Pathway to Combat Oxidative Stress and Intestinal Inflammation in Veterinary and Translational Medicine.

This review synthesizes research on nuclear factor erythroid 2-related factor 2 (Nrf2) in intestinal health across human, livestock, and mouse models. The Nrf2 signaling pathway serves as a master regulator of cellular antioxidant defenses and a key therapeutic target for intestinal inflammatory disorders, including ulcerative colitis and Crohn's disease. The interplay between oxidative stress, Nrf2 signaling, and NF-κB inflammatory cascades represents a critical axis in the pathogenesis and resolution of intestinal inflammation. Under normal physiological conditions, Nrf2 remains sequestered

Vesicular Rps6 Released by Astrocytes in an Experimental Model of AD Regulates Local Translation and Enhances Synaptic Integrity in Neurones.

In neurones, like in any other cell, their function often relies on the fine-tuning of their protein levels, which is achieved by the balance between protein synthesis and turnover. Defects in protein homeostasis frequently lead to neuronal dysfunction and neurological disorders. Given their extreme morphological complexity and high compartmentalization, neurones highly depend on the asymmetrical distribution of their proteome. The common belief is that proteins that sustain axonal, dendritic and synaptic functions are synthesized in the soma and then transported to distal neuronal compartment
