HECTD1 Directs HIF-1α to Facilitate Intestinal Inflammation and Carcinogenesis via Promoting Th17 Differentiation and Glycolysis.

Targeting glucose metabolism in Th17 cells may be beneficial for treating inflammatory bowel disease (IBD) and colitis-associated cancer (CAC), however, the molecular mechanisms orchestrating Th17 cell metabolism remain unclear. Here, we revealed that HECTD1 regulates IGF2BP3-dependent HIF-1α translation to promote Th17 cell glycolysis and differentiation and that HECTD1 expression correlates with IBD and CAC pathogenesis. T-cell-specific deletion of HECTD1 mitigated the severity of colitis and CAC in murine models. RNA-seq revealed enrichment of Th17 differentiation, glycolysis/gluconeogenesi

Recent discoveries of CRBN-binding molecular glue degraders: Focus on protein kinases.

Cereblon (CRBN)-based molecular glue degraders (MGDs) have emerged as one of the most rapidly evolving therapeutic modalities in targeted protein degradation. Upon binding of MGDs such as immunomodulatory drugs, CRBN undergoes an allosteric transition into a degradation-competent closed conformation, enabling the recruitment of structurally compatible neosubstrates for subsequent ubiquitination and proteasomal degradation. Recently, the repertoire of CRBN neosubstrates has rapidly expanded beyond the classical IMiD neosubstrates such as IKZF1/3 and CK1α. In particular, many therapeutically rel

Aptamer-Based Targeted Protein Degradation: Mechanistic Principles, Delivery Strategies, and Translational Opportunities.

Targeted elimination of pathogenic proteins remains a fundamental challenge in therapeutic intervention, particularly for proteins considered "undruggable" by conventional inhibition-based approaches. Aptamer-based targeted protein degradation (TPD) has emerged as a versatile strategy that leverages the high specificity, structural programmability, and chemical tunability of nucleic acid aptamers to redirect disease-relevant proteins toward endogenous degradation pathways. Intracellular platforms, exemplified by aptamer-conjugated proteolysis-targeting chimeras (PROTACs), induce ubiquitination

Research progress and prospect of β-carotene hydroxylase gene mediating plant carotenoid metabolism in response to abiotic stress.

Carotenoid metabolism plays a significant role in plants response to abiotic stress. The β-carotene hydroxylase gene (BCH) is an important gene located downstream of the carotenoid metabolic pathway. Its main function is to hydroxylate β-carotene to form zeaxanthin, which is the direct valve regulating the carotenoid xanthophyll cycle and also an important controller in the upstream of ABA synthesis. Most carotene components contain a molecular structure with conjugated double bonds, which determines that they have a powerful antioxidant function. When the BCH gene is overexpressed, Reactive o

PANoptosis In Microglia and Neurons Following Traumatic CNS Injury: A Systematic Review of Pathological Features and Therapeutic Implications.

BACKGROUND: Traumatic brain injury (TBI) and spinal cord injury (SCI) induce PANoptosis, a pro-inflammatory programmed cell death modality orchestrated by the PANoptosome. This modality integrates pyroptosis, apoptosis, and necroptosis molecular machinery and contrasts with canonical single-pathway cell death. METHODS: This systematic review (PROSPERO CRD420261383435) followed PRISMA and searched PubMed, Web of Science, and Cochrane Library. The in vivo studies confirming PANoptosis at the protein level were included; quality was assessed using CRIME-Q, and shared genes/protein interactions w

The Mechanism of the Parkin-CMA-NLRP3-PANoptosis Signaling Axis in Parkinson's Disease: From Molecular Basis to Therapeutic Strategies.

Parkinson's Disease (PD) is a common neurodegenerative disorder. Existing clinical interventions merely relieve symptoms and cannot reverse progressive neuronal injury. Neuroinflammation and dysregulated programmed cell death are central pathological drivers of PD. PANoptosis is an integrated pan-programmed cell death, can simultaneously trigger apoptosis, pyroptosis and necroptosis, with the NOD‑like receptor family pyrin domain‑containing 3 (NLRP3) inflammasome serving as a core molecular hub for PANoptosis initiation. Apart from its well‑characterized role in mitophagy, Parkin directly inte

Pyroptosis in premature ovarian insufficiency: pathogenic mechanisms, pathway crosstalk, and therapeutic strategies.

Premature ovarian insufficiency (POI) is characterised by pathological loss of ovarian function in women younger than 40 years, resulting in premature exhaustion of the ovarian follicle pool and impaired fertility. Although apoptosis has long been considered the dominant mechanism driving granulosa cell (GC) death and follicular atresia, it does not fully explain the severe inflammatory microenvironment and tissue fibrosis observed during POI progression. Recently, pyroptosis, a proinflammatory form of programmed cell death mediated by the gasdermin family, has emerged as a critical driver of

Tangeretin Targets Macrophage M1 Polarization to Alleviate Liver Injury in a Murine Model of Hepatic Sinusoidal Obstruction Syndrome.

Hepatic sinusoidal obstruction syndrome (HSOS) is a life-threatening liver microvascular disorder characterized by hepatic sinusoidal endothelial cells (HSECs) injury, for which safe and effective pharmacological treatments are currently lacking. Tangeretin (TAN), a natural polymethoxylated flavone, exhibits anti-inflammatory, antioxidant, and hepatoprotective properties. However, its therapeutic potential and mechanisms in HSOS remain unclear. This study investigated the protective role and molecular mechanisms of TAN against monocrotaline (MCT)-induced HSOS. In the murine HSOS model, TAN int

Rational design, molecular characterization and dissociation mechanisms of PROTACs targeting hFAAH.

Fatty acid amide hydrolase (FAAH) is a key enzyme in the endocannabinoid system and a promising therapeutic target for pain and depression. However, traditional FAAH inhibitors have faced clinical setbacks due to their limited efficacy and off-target effects. Here, we present the rational design and molecular characterization of C13p, a novel PROTAC molecule targeting hFAAH, composed of a 13-carbon linker connecting the FAAH ligand Carb4d and the CRBN ligand pomalidomide. Our results demonstrate that C13p significantly enhances the protein-protein interactions between hFAAH and CRBN, with a bi

Penfluridol directly targets PDPK1 to suppress AKT1 phosphorylation and stabilize CTR1, inducing cuproptosis in colorectal cancer.

Colorectal cancer (CRC) is a major cause of cancer mortality worldwide. Here, we identified the antipsychotic drug penfluridol as a potent anticancer agent that induces cuproptosis in CRC through a newly defined signaling mechanism. Using real-world clinical datasets, we demonstrated that PDPK1 is significantly upregulated in CRC and further increases in its expression level are correlated with a worse outcome, whereas CTR1 is downregulated in CRC and further decreases in its expression level are correlated with favourable outcomes, directly establishing the clinical relevance of these two pro

A new candidate tumor suppressor tRF-Ser inhibits gastric cancer progression by regulating the CNBP/HSPA8 axis.

Gastric cancer (GC) is a highly aggressive malignancy with a poor prognosis. Transfer RNA-derived small RNAs (tsRNAs) are implicated in tumorigenesis, but their precise mechanistic roles in GC progression remain incompletely understood. We performed high-throughput sequencing in four paired GC/normal tissues to profile tsRNAs. The functional and mechanistic role of a candidate tsRNA was systematically investigated, alongside a suite of techniques including fluorescence in situ hybridization, RNA immunoprecipitation, RNA pull-down, chromatin immunoprecipitation, and luciferase reporter assays.

Targeting ANXA1/TRKA axis enhances immunotherapy sensitivity in neural invasion-positive gastric cancer.

Gastric cancer (GC) remains a leading cause of cancer-related morbidity and mortality worldwide. Neural invasion (NI) is a common pathological behavior that worsens the prognosis in GC. However, the immune microenvironment of neural invasion-positive GC (NI+GC) and its potential therapeutic implications remain poorly defined. Here, we performed single-cell RNA sequencing (scRNA-seq) on tumor specimens from patients with NI+GC and neural invasion-negative GC (NI-GC) to comprehensively delineate the immune landscape. Our analysis revealed a significant enrichment of exhausted ANXA1+CD8+T cells w

Post-translational modifications in alzheimer's disease: proteome dynamics and emerging therapeutic strategies.

Alzheimer’s disease (AD) is a progressive neurodegenerative condition marked by the accumulation of amyloid-β (Aβ), tau hyperphosphorylation, synaptic dysfunction, and ongoing neuroinflammation. Recent findings emphasize the role of post-translational modifications (PTMs) such as phosphorylation, ubiquitination, SUMOylation, methylation, acetylation, palmitoylation, prenylation, and O-GlcNAcylation as crucial molecular switches that influence protein stability, localization, aggregation, and signaling. Disrupted PTMs interfere with APP processing, increase Aβ production, encourage tau misfoldi

Serial comparative transcriptomics reveals dynamic and coordinated immune-metabolic responses in the liver-spleen axis of Oplegnathus punctatus to Edwardsiella piscicida infection.

Oplegnathus punctatus is a high-value marine fish in aquaculture, yet its production is severely threatened by pathogen infections such as Edwardsiella piscicida (the old name called as Edwardsiellatarda). Understanding the systemic host response is essential for developing effective control strategies. This study investigated the dynamic and coordinated immune-metabolic reactions in the liver and spleen of O. punctatus following E. piscicida infection. Serial transcriptomic analyses of liver and spleen were conducted at 0, 12, 24, 48, 72 h post-infection (hpi). Results revealed that the splee

Protein Modifications and Quality Control System: Target for Alzheimer's Disease Therapy.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by memory loss and cognitive decline. Its main pathological features are extracellular plaques composed of aggregated amyloid-β (Aβ) peptides and intracellular neurofibrillary tangles formed by hyperphosphorylated tau. The Aβ hypothesis proposes that Aβ accumulation is a key driver of AD, influencing tau pathology, neuroinflammation, and neurodegeneration. However, therapies that reduce Aβ have shown limited clinical benefits. This suggests that the mechanisms underlying peptide-mediated modulation of AD patholo

TNFAIP1 as a novel therapeutic target in ulcerative colitis and salidroside as its potential inhibitor.

BACKGROUND: The absence of known inhibitors for TNFAIP1, a TNF-α-induced protein, has limited its exploration as a therapeutic target for ulcerative colitis (UC). This study aimed to validate TNFAIP1's pathogenic role in UC and to identify the first direct TNFAIP1-targeting compound from natural compounds. METHODS: We integrated bioinformatics analysis, DSS-induced colitis models in Tnfaip1-knockout mice, and virtual screening to identify Salidroside as a potential TNFAIP1-targeting compound. Cellular thermal shift and ubiquitination assays confirmed its direct interaction with TNFAIP1 and it

BRCC3 promotes progression and immune evasion in non-small cell lung cancer through regulation of PD-L1 and B7-H3.

While elevated levels of the deubiquitinating enzyme BRCC3 have been documented in a range of cancers, its capacity to influence immune checkpoint expression or enable tumor immune evasion in non-small cell lung cancer (NSCLC) remains uncharacterized. BRCC3 expression and clinical relevance were assessed using TIMER2, UALCAN, and Kaplan-Meier Plotter databases, and validated in NSCLC tissues and cell lines. Functional impacts were evaluated through proliferation, migration, invasion, and syngeneic tumor models. CD8+ T cell cytotoxicity and cytokine secretion were measured in co-culture systems

The splicing factor hnRNPA1 promotes osimertinib resistance in lung adenocarcinoma by regulating NEDD4L alternative splicing.

Resistance to osimertinib in lung adenocarcinoma presents a significant hurdle in contemporary lung cancer therapy, with splicing dysregulation being instrumental in tumorigenesis and progression. The mechanism via which alternate splicing facilitates osimertinib resistance in lung cancer is still ambiguous. We aimed to examine the pivotal function of the splicing factor hnRNPA1 in osimertinib resistance in lung cancer. The impact of hnRNPA1 on osimertinib resistance in lung cancer was confirmed by small interfering RNA and CDX models. RNA-seq, RIP, CLIP-qPCR, and COIP methodologies were utili

TEAD upregulation promotes osimertinib resistance in NSCLC by recruiting YAP into the nucleus.

Osimertinib, a third-generation tyrosine kinase inhibitor (TKI), is the first-line standard therapy for epidermal growth factor receptor (EGFR)-mutated non-small cell lung cancer (NSCLC). However, acquired resistance is inevitable, with underlying mechanisms incompletely elucidated, leading to limited subsequent treatments and posing major clinical challenges. This study established osimertinib-resistant NSCLC models in vitro and in vivo, revealing a novel resistance mechanism mediated by TEAD upregulation but independent of the classical Hippo pathway. Osimertinib treatment enhances TEAD prot
