Application of Causal Inference to Establish Assay Effect in the Absence of a Bridging Study: A Case Study of MenACWY-CRM Conjugate Vaccine Data.

During a vaccine development program, if the assay used to measure immunological endpoints is changed, ideally, a bridging study is performed to establish the relationship between results obtained with the new and previous assay. However, this is not always feasible, and when bridging study data are absent, this can limit the ability to use historical study information to strengthen evidence generated in the clinical program. We present a case study on GSK's quadrivalent meningococcal vaccine (MenACWY-CRM), where the immunogenicity assay was changed over time. A large amount of study data was

Endothelial leak and knowledge gaps: yellow fever virus non-structural protein 1 and the antibodies that bind it.

Yellow fever virus, family Orthoflaviviridae, is the causative agent of yellow fever - a lethal mosquito-borne disease endemic to South America and Africa. Recent advances in understanding the mechanism of orthoflavivirus pathogenesis, particularly for dengue (DENV), demonstrate a role for the secreted viral non-structural protein 1 (NS1) in both animal and human models. The NS1 protein's most well described pathogenic mechanism is its ability to induce vascular endothelial permeability in a tissue organ specific manner, contributing to orthoflavivirus disease severity. Surprisingly, there is

Bacterially expressed full length Hemagglutinin of Avian Influenza Virus H5N1 forms oligomers and exhibits hemagglutination.

Avian influenza poses a significant global health threat, with the potential for widespread pandemics and devastating consequences. Hemagglutinin (HA), a critical surface glycoprotein of influenza viruses, plays a pivotal role in viral entry and serves as a primary target for subunit vaccine development. In this study, we successfully cloned, expressed, and purified hemagglutinin from the circulating strain of H5N1 influenza virus using a robust molecular biology approach. The cloning process involved insertion of the synthetic HA gene into the pET21b vector, confirmed through double digestion

Intranasal bovine β-defensin-5 enhances antituberculosis immunity in a mouse model by a novel protein-based respiratory mucosal vaccine.

Respiratory mucosal immunization is an effective immunization strategy against tuberculosis (TB), and effective mucosal vaccines require adjuvants that can promote protective immunity without deleterious inflammation. Mucosal BCG (Bacille Calmette-Guerin) is effective, but it causes a severe inflammatory response in the lung. A novel less cytotoxic mucosal vaccine AH-PB containing Mycobacterium tuberculosis (Mtb) cell surface antigens Ag85A and HspX (AH), as well as polyinosinic-polycytidylic acid (Poly IC) and bovine neutrophil β-defensin-5 (B5) adjuvants were prepared, with the overarching g

The Cryptosporidium parvum gp60 glycoprotein expressed in the ciliate Tetrahymena thermophila is immunoreactive with sera of calves infected with Cryptosporidium oocysts.

Cryptosporidium parvum is a protozoan parasite of the phylum Apicomplexa responsible for cryptosporidiosis in calves, a disease that causes significant diarrhea and impairs gain of body weight, generating important production losses. As to now, no effective drugs or vaccines are available for the treatment or prevention of bovine cryptosporidiosis. Several reports suggest that development of a vaccine to prevent cryptosporidiosis is feasible, but relatively few vaccine candidates have been characterized and tested. The most prominent C. parvum antigen is gp60, an O-glycosylated mucin-like prot

Immunisation of chickens with live Salmonella vaccines - Role of booster vaccination.

It is accepted that booster vaccinations of chickens with live Salmonella vaccines are essential part of vaccinations schemes to induce an effective adaptive immune response. As manufacturer of registered live Salmonella vaccines recommend different times of booster the question raises whether the duration between the first and second immunisation might influence the protective effect against Salmonella exposure. Chickens were immunised with a live Salmonella Enteritidis vaccine on day 1 of age followed by a booster vaccination at different intervals (day 28, 35 or 42 of age) to study the effe

Identification of vaccine candidate antigens of Staphylococcus pseudintermedius by whole proteome characterization and serological proteomic analyses.

The recent emergence of methicillin-resistant Staphylococcus pseudintermedius (MRSP) has complicated considerably the treatment of infections caused by these bacteria. Therefore new treatment strategies are urgently needed, namely through the development of vaccines towards the control of bacterial infections. Our study describes an extensive characterization of the proteome of S. pseudintermedius through a 2-DE MALDI-TOF/TOF approach, followed by SERological Proteome Analysis (SERPA) to identify potential vaccine candidate antigens. We were able to identify 361 unique proteins, of which 39 ar

Fatty acid metabolism in the Plasmodium apicoplast: Drugs, doubts and knockouts.

The malaria parasite Plasmodium possesses a relict, non-photosynthetic plastid known as the apicoplast. The apicoplast is essential for parasite survival, and harbors several plant-like metabolic pathways including a type II fatty acid synthesis (FASII) pathway. The FASII pathway was discovered in 1998, and much of the early research in the field pursued it as a therapeutic drug target. These studies identified a range of compounds with activity against bloodstage parasites and led to the localization and characterization of most enzymes in the pathway. However, when genetic studies revealed F

Hepatitis C virus in the new era: perspectives in epidemiology, prevention, diagnostics and predictors of response to therapy.

Despite the great successes achieved in the fields of virology and diagnostics, several difficulties affect improvements in hepatitis C virus (HCV) infection control and eradication in the new era. New HCV infections still occur, especially in some of the poorest regions of the world, where HCV is endemic and long-term sequelae have a growing economic and health burden. An HCV vaccine is still no available, despite years of researches and discoveries about the natural history of infection and host-virus interactions: several HCV vaccine candidates have been developed in the last years, targeti

Hepatitis E virus: neutralizing sites, diagnosis, and protective immunity.

There have been increased attentions on HEV and its associated diseases in recent years as a result of an increased number of reports on autochthonous patients from many developed countries. Vaccine development and better disease management are expected from protective immunity with increased knowledge on the pathogenesis and virology of HEV. This review summarizes the current understanding of the HEV virology, the key neutralization sites (epitopes) on the surface of the viral capsid, the host humoral immune responses for HEV infection, and the protective immunity conferred by natural infecti

Antigenic analysis of divergent genotypes human Enterovirus 71 viruses by a panel of neutralizing monoclonal antibodies: current genotyping of EV71 does not reflect their antigenicity.

In recent year, Enterovirus 71 (EV71)-associated hand, foot and mouth disease (HFMD) has become an important public health issue in China. EV71 has been classified into genotypes A, B1-B5 and C1-C5. With such genetic diversity, whether the convalescent or recovery antibody responses can cross-protect infections from other genotypes remains a question. Understanding of the antigenicity of such diverse genetic EV71 isolates is crucial for the EV71 vaccine development. Here, a total of 186 clones anti-EV71 MAbs was generated and characterized with Western blot and cell-based neutralization assay.

ACAM2000: the new smallpox vaccine for United States Strategic National Stockpile.

Smallpox was eradicated more than 30 years ago, but heightened concerns over bioterrorism have brought smallpox and smallpox vaccination back to the forefront. The previously licensed smallpox vaccine in the United States, Dryvax (Wyeth Laboratories, Inc.), was highly effective, but the supply was insufficient to vaccinate the entire current US population. Additionally, Dryvax had a questionable safety profile since it consisted of a pool of vaccinia virus strains with varying degrees of virulence, and was grown on the skin of calves, an outdated technique that poses an unnecessary risk of con

Bacterial ghosts as a delivery system for zona pellucida-2 fertility control vaccines for brushtail possums (Trichosurus vulpecula).

The introduced brushtail possum is a serious pest in New Zealand and there is much interest in the development of an immunocontraceptive vaccine for population control. Immunisation of female possums against recombinant possum zona pellucida protein-2 (ZP2) is known to reduce embryo production by 72-75% but successful development of fertility control will depend on a delivery system that is effective for field use. Bacterial ghost vaccine technology is a promising system to formulate a non-living vaccine for bait or aerosol delivery. The N-terminal (amino acid residues 41-316, ZP2N) and C-term

Bactericidal activity of a monoclonal antibody against a recombinant 40-kDa outer membrane protein of Porphyromonas gingivalis.

BACKGROUND: We have cloned the gene for a 40-kDa outer membrane protein (40-kDa OMP) from Porphyromonas gingivalis 381. The recombinant (r)40-kDa OMP has become the subject of considerable interest because of its potential role in the development of a vaccine useful for passive immunization. To develop such a vaccine, it is essential to fully understand the functions of anti-r40-kDa OMP antibody in the host defense against P. gingivalis. To that end, we developed a panel of monoclonal antibodies by immunizing mice with purified r40-kDa OMP. The objective of this study was to determine the bact

COVID-19: vaccine's progress.

Under the pressure of the COVID-19 pandemic, vaccines were developed and rolled out into mass vaccination campaigns at incredible speed. What normally takes a decade was worked out within a year. Vaccines were produced along many different platforms ranging from inactivated whole virus vaccines over adenovirus-vectored vaccines, recombinant protein vaccines and nanoparticles to mRNA vaccines. Several vaccines went through preclinical testing and completed successful phase 1 to phase 3 clinical trials. The first evaluations of national vaccination campaigns document astonishing high levels of p

A comparative study of four live measles vaccines in Israel.

While an earlier live measles vaccine induced a high degree of immunity it also caused clinical reactions of variable severity in some vaccinated children. A number of attenuated measles vaccine strains have been developed to avoid these problems, and this paper reports the results of small-scale trials with 3 attenuated measles vaccines and a new further-attenuated vaccine on children from 9 months to over 4 years old.All 4 vaccines produced a satisfactory serological response. The new further-attenuated vaccine (Moraten) produced less fever and fewer rashes in the vaccinated children than th

Acute viscerotropic disease following vaccination against yellow fever.

Acute viscerotropic disease following yellow fever vaccination (YEL-AVD) is a rare but serious complication of vaccination with 17D yellow fever vaccine. This paper reviews the existing literature regarding YEL-AVD and discusses possible etiologic mechanisms. A greater understanding of this condition is essential to assuring safe and effective prevention of yellow fever and vaccination against other arboviral diseases for which 17D-based vaccines are being developed.

Polyarthropathy, orbital myositis and posterior scleritis: an unusual adverse reaction to influenza vaccine.

We report a patient who developed an acute symmetrical polyarthropathy, orbital myositis and posterior scleritis following inoculation with the 1993 Fluvirin (inactivated influenza vaccine: surface antigen) vaccine. We believe that this represents a hypersensitivity reaction to the vaccine as extensive investigations revealed no underlying infective, inflammatory or systemic cause. We review the current indications for vaccination against the influenza virus and the known adverse reactions to the vaccine.

Structure-based design and immunogenicity of an H5 hemagglutinin stem nanoparticle vaccine candidate.

Highly pathogenic avian influenza viruses (HPAIVs), particularly the H5N6 subtype, pose a serious global public health threat. Conventional influenza vaccines primarily target the highly variable globular head of hemagglutinin (HA), resulting in limited breadth of protection. Here, we employed a structure-guided vaccine design strategy to develop a self-assembling nanoparticle immunogen targeting conserved epitopes within the HA stem region. The HAstem antigen was rationally engineered to incorporate major neutralizing epitopes and further stabilized through the introduction of a GCN4 trimeriz
