Anthracycline resistance in murine leukemic P388 cells. Role of drug efflux and glutathione related enzymes.

Energy-dependent drug efflux is a major factor in cellular resistance of P388/R84 mouse leukemic cells to anthracyclines such as doxorubicin (DOX), and blocking of efflux increases sensitivity. However, efflux does not play a significant role in resistance to N-trifluoroacetyladriamycin-14-valerate (AD 32), a DOX analog. Since drug efflux alone cannot account for resistance to anthracyclines, we have, in the present study, measured cellular glutathione (GSH) content and activity of GSH cycle related enzymes to determine their role in resistance. Cellular GSH content was similar in DOX-sensitiv

Epidemic Venezuelan equine encephalitis in North America in 1971: vertebrate field studies.

Epidemic Venezuelan equine encephalitis in North America in 1971: vertebrate field studies. Am J Epidemiol 101:36-50, 1975.-In June 1971, epidemic Venezuelan equine encephalitis (VEE) invaded the lower Rio Grande Valley in south Texas. The Boca Chica area of Cameron County was selected as a study site to investigate vertebrate involvement in the natural cycle of epidemic VEE on the basis of considerable evidence of VEE virus activity there in equines, humans, and mosquito vectors. Only one VEE virus isolation was made from 4739 wild and domestic non-equine vertebrates, although numerous equine

Umbralisib, a novel PI3Kδ and casein kinase-1ε inhibitor, in relapsed or refractory chronic lymphocytic leukaemia and lymphoma: an open-label, phase 1, dose-escalation, first-in-human study.

BACKGROUND: Umbralisib (TGR-1202) is a novel next-generation inhibitor of phosphatidylinositol 3-kinase (PI3K) isoform p110δ (PI3Kδ), which is structurally distinct from other PI3Kδ inhibitors and shows improved isoform selectivity. Umbralisib also uniquely inhibits casein kinase-1ε, a major regulator of protein translation. The aim of this first-in-human phase 1 study was to establish the safety and preliminary activity profile of umbralisib in patients with haematological malignancies. METHODS: We did an open-label, phase 1, dose-escalation study at seven clinics in the USA. We recruited pa

Fumarate hydratase deficiency and cancer: activation of hypoxia signaling?

Molecular genetic analysis of hereditary leiomyomatosis and renal cell cancer (HLRCC) unexpectedly revealed germline defects in the gene encoding the Krebs cycle enzyme fumarate hydratase (FH), stimulating great interest in the underlying mechanism of oncogenesis. It has been proposed that the associated accumulation of fumarate competitively inhibits the 2-oxoglutarate-dependent dioxygenases that regulate hypoxia-inducible factor (HIF), thus activating oncogenic hypoxia pathways. In this issue of Cancer Cell, Pollard and colleagues describe a genetic mouse model of FH deficiency that recapitu

The reproducibility of depressive and hypomanic symptoms across repeated episodes in patients with rapid-cycling bipolar disorder.

The purpose of this study is to determine the stability of symptoms of hypomania and depression across repeated affective episodes in patients with rapid-cycling bipolar disorder. Nine patients had a total of 30 depressive episodes and 31 hypomanic episodes during the period of observation. Standardized observer ratings indicated that the three symptoms most consistently reported during depressive episodes were fatiguability, decreased work activities and hypersomnia. These results as well as those from the standardized observer ratings of hypomania indicate that depression in this population

Biology of African trypanosomes in the tsetse fly.

African trypanosomes present several features of interest to cell biologists. These include: a repressible single mitochondrion with a large mass of mitochondrial DNA, the kinetoplast; a special organelle, the glycosome, which houses the enzymes of the glycolytic chain; a surface coat of variable glycoprotein which enables the parasite to evade the mammalian host's immune response; and a unique flagellum-to-host attachment mechanism associated with novel cytoskeletal elements. Trypanosome development during the life cycle involves cyclical activation and repression of genes controlling these a

Mating in the rhesus monkey (Macaca mulatta) after conception and its relationship to oestradiol and progesterone levels throughout pregnancy.

Behavioural and hormonal data were collected throughout pregnancy in captive rhesus monkeys. Heterosexual pairs observed in daily time-limited mating tests showed two distinct periods of increased sexual interaction, as measured by the incidence of ejaculation. One period was coincident with the preovulatory oestradiol peak, while the other occurred between the 6th and 10th weeks of pregnancy. The positive relationship found during the menstrual cycle between the ratio of circulating oestradiol: progesterone and degree of sexual activity continued for the first 8 weeks of pregnancy. However, s

Application of Panax notoginseng Saponins in Skin Wound Repair: Mechanisms and Research Progress.

Skin wound repair involves four stages: haemostasis, inflammation, proliferation and remodelling. Refractory wounds feature prolonged healing cycles, severe scar hyperplasia and limited clinical therapeutic strategies. Panax notoginseng saponins (PNS), primarily the ginsenosides Rg1 and Rb1 monomers, display high safety and multitarget regulatory properties across the entire wound repair cycle. PNS modulates the inflammatory microenvironment and induces M2-type polarization of macrophages by inhibiting NF-κB, MAPK and other signalling pathways; activates the PI3K/AKT pathway to promote the pro

Circadian Disruption Through Light-Dark Cycle Alteration Induced Alzheimer's Disease-like Pathology in Mice.

Circadian disruption (CD) has emerged as a critical factor compromising human health in contemporary society. Increasing evidence suggests that disturbances in circadian rhythms are involved in the pathogenesis of neurodegenerative diseases, such as Alzheimer's disease (AD). The hyperphosphorylation of tau and the deposition of amyloid-β (Aβ) are recognized as major pathological hallmarks of AD. In this study, we aimed to explore the impact of long-term CD on AD-like pathological changes and to explore the underlying molecular mechanisms using a mouse model. To mimic the CD experienced by shif

Bee venom and thymoquinone combination inhibits cancer cells by inducing cell cycle arrest and apoptosis.

Natural products have gained significant interest in cancer therapy. Thymoquinone (TQ), a bioactive compound mainly derived from Nigella sativa seeds, and bee venom (BV), a complex mixture of bioactive components secreted by honeybees (Apis mellifera), are notable examples. This study aimed to evaluate the synergistic anticancer effects of TQ-BV combination on selected cancer cell lines—HeLa, MCF-7, HCT—and normal human skin fibroblasts (HSF). BV was collected using a novel remote-controlled extraction device designed to maintain venom purity by isolating it from environmental contaminants and

Redox-coupled coordination strategy for xanthate detoxification via self-catalysis over a broad pH range.

This study developed an effective strategy to construct a Cu(II)/Cu(I) redox cycle catalytic system by anchoring Cu(II)/Cu(I) onto multi-walled carbon nanotubes (MWCNTs) via wet ball milling for the efficient degradation of xanthate compounds. The catalyst exhibited dual functionality: Cu(II) coordinated with electron-rich S-CS moiety in xanthate molecules, facilitating strong adsorption of xanthate and generation of stable Cu(I), which subsequently activated H2O2 to regenerate Cu(II) and generate reactive oxygen species (•OH, •O2⁻, and 1O2). DFT calculations verified the enhanced adsorption o

The cellular protein phosphatase 2A is a crucial host factor for Marburg virus transcription.

UNLABELLED: Little is known regarding the molecular mechanisms that highly pathogenic Marburg virus (MARV) utilizes to transcribe and replicate its genome. Previous studies assumed that dephosphorylation of the filoviral transcription factor VP30 supports transcription, while phosphorylated VP30 reduces transcription. Here, we focused on the role of the host protein phosphatase 2A (PP2A) for VP30 dephosphorylation and promotion of viral transcription. We could show that MARV NP interacts with the subunit B56 of PP2A, as previously shown for the Ebola virus, and that this interaction is importa

Extracorporeal photopheresis induces NETosis in neutrophils derived from patients with chronic graft-vs-host disease.

INTRODUCTION: Extracorporeal photopheresis (ECP) is considered an effective treatment for patients with chronic graft vs host disease (cGVHD) and demonstrates efficacy in ameliorating GVHD. The mechanism by which ECP acts against cGVHD is not fully understood. Preliminary observations have hinted at the potential involvement of neutrophil extracellular traps (NETs) formation in the pathogenesis of cGVHD. We aimed to assess the influence of ECP on the formation of NETs in patients with cGVHD as a potential mechanism in this setting. METHODS: Patients treated with ECP for cGVHD at the Rabin Med

A Proactive Approach to Prevent Hematopoietic Exhaustion During Cancer Chemotherapy in Older Patients: Temporary Cell-Cycle Arrest.

Age is associated with the decline of multiple organ systems. In older patients, hematological toxicities associated with chemotherapy are often dose limiting, impairing dose intensity and treatment efficacy. Contrary to the classical path using growth factors to activate tissue regeneration, a novel strategy is emerging to prevent chemotherapy toxicity that involves temporary cell-cycle arrest of normal cells, such as hematopoietic or epithelial precursors. This proactive approach may allow the sparing of the stem cell reserve of these tissues. Two molecules are included in this new category,

Boron improves cardiac contractility and fibrotic remodeling following myocardial infarction injury.

Myocardial fibrosis is a major determinant of clinical outcomes in heart failure (HF) patients. It is characterized by the emergence of myofibroblasts and early activation of pro-fibrotic signaling pathways before adverse ventricular remodeling and progression of HF. Boron has been reported in recent years to augment the innate immune system and cell proliferation, which play an important role in the repair and regeneration of the injured tissue. Currently, the effect of boron on cardiac contractility and remodeling is unknown. In this study, we investigated, for the first time, the effect of
