Sequential delivery of cardioactive drugs via microcapped microneedle patches for improved heart function in post myocardial infarction rats.

After myocardial infarction, the heart undergoes adverse remodeling characterized by a series of pathological changes, including inflammation, apoptosis, fibrosis, and hypertrophy. In addition to cardiac catheter-based re-establishment of blood flow, patients typically receive multiple medications that aim to address these different mechanisms underlying left ventricular remodeling. The current study aims to establish a versatile multi-drug delivery platform for the controlled and sequential delivery of multiple therapeutic agents in a single treatment. Toward this goal, we generated a microca

Expression of CD31 in Mycosis Fungoides.

BACKGROUND/AIM: Mycosis fungoides (MF) is the most common primary cutaneous T-cell lymphoma (CTCL) characterized by malignant proliferation of mature T lymphocytes and primary skin involvement. Recent reports suggest that angiogenesis may play a role in the growth and spread of this malignancy. Cluster of differentiation 31 (CD31) is a protein classified into the Ig-superfamily of cell adhesion molecules, expressed on endothelial cells and majority of hematopoietic non-erythroid cells. The aim of our study was to explore the role of angiogenesis in MF. MATERIALS AND METHODS: We evaluated the

Combination of stromal-derived factor-1alpha and vascular endothelial growth factor gene-modified endothelial progenitor cells is more effective for ischemic neovascularization.

BACKGROUND: Recruitment and entrapment of bone marrow-derived endothelial progenitor cells (EPCs) is important in vascular endothelial growth factor (VEGF)-induced angiogenesis. EPC mobilization and differentiation are modulated by stromal-derived factor-1alpha (SDF-1alpha/CXCL12), another important chemokine. In this study, we investigated the hypothesis that SDF-1alpha and VEGF might act synergistically on EPC-mediated vasculogenesis. METHODS: EPCs were isolated and cultured from human peripheral blood, then transduced with retroviral vectors pBabe containing human VEGF(165) complimentary D

Topical timolol in dermatology: infantile haemangiomas and beyond.

Timolol, a nonselective β-adrenergic receptor blocker, is well-tolerated and is becoming increasingly popular in dermatology, especially in the management of infantile haemangioma (IH). Its effects are mainly due to vasoconstriction, inhibition of angiogenesis and keratinocyte migration promotion for re-epithelialization and wound healing. We review the evidence behind the use of timolol in several dermatological conditions including IH, pyogenic granulomas, Kaposi sarcoma, chronic wound healing, postsurgical wounds, acne vulgaris, rosacea, eczema and red scrotum syndrome.

Vascular endothelial growth factors (VEGFs) and stroke.

Vascular endothelial growth factors (VEGFs) have been shown to participate in atherosclerosis, arteriogenesis, cerebral edema, neuroprotection, neurogenesis, angiogenesis, postischemic brain and vessel repair, and the effects of transplanted stem cells in experimental stroke. Most of these actions involve VEGF-A and the VEGFR-2 receptor, but VEGF-B, placental growth factor, and VEGFR-1 have been implicated in some cases as well. VEGF signaling pathways represent important potential targets for the acute and chronic treatment of stroke.

Studies in mice reveal a role for anthrax toxin receptors in matrix metalloproteinase function and extracellular matrix homeostasis.

The genes encoding Anthrax Toxin Receptors (ANTXRs) were originally identified based on expression in endothelial cells suggesting a role in angiogenesis. The focus of this review is to discuss what has been learned about the physiological roles of these receptors through evaluation of the Antxr knockout mouse phenotypes. Mice mutant in Antxr genes have defects in extracellular matrix homeostasis. We discuss how knowledge of physiological ANTXR function relates to what is already known about anthrax intoxication.

Interdependent biological systems, multi-functional molecules: the evolving role of tissue factor pathway inhibitor beyond anti-coagulation.

Coagulation, innate immunity, angiogenesis, and lipid metabolism represent fundamental and interdependent biological systems. While tissue factor pathway inhibitor (TFPI) is the major physiological inhibitor of TF, its unique structure and endothelial expression allow multi-modal interactions with constituent molecules in each of these systems. We review emerging data describing roles for TFPI beyond simply opposing the action of TF, particularly with regard to the highly basic c-terminus of TFPI, and highlight potentially exciting new areas for future research.

Umbilical cord blood cells and brain stroke injury: bringing in fresh blood to address an old problem.

Degeneration of brain tissue following stroke leads to functional impairment with limited brain self-repair. New evidence suggests that delivery of circulating CD34(+) human umbilical cord blood cells can produce functional recovery in an animal stroke model with concurrent angiogenesis and neurogenesis leading to some restoration of cortical tissue. While some alternative interpretations of this data are offered herein, the study provides encouraging evidence of functional recovery from stroke in an animal model using stem cell therapy.

Revascularization-driven nanozyme Therapy: Disrupting the vicious cycle of ROS and insufficient vascularization for chronic non-healing wound treatment.

The pathophysiology of chronic non-healing wound is characterized by a deleterious interplay between impaired angiogenesis and excessive generation of reactive oxygen species (ROS), which perpetuates a vicious cycle exacerbated by persistent inflammation. Current therapeutic strategies lack efficacy in addressing this multifaceted pathophysiology. To dismantle this vicious cycle, we design a composite nanozyme (TCC) by encapsulating ultrasmall ceria nanoclusters (CeNC) and introducing tryptophan as competitive ligands within a biocompatible cobalt-based zeolitic imidazolate framework (Co-ZIF).

Evaluation of the anticancer effects of ellagic acid and cisplatin in cisplatin-sensitive and -resistant MDA-MB-231 breast cancer cells.

Ellagic acid (EA) is a natural polyphenol noted for its antiproliferative and pro-apoptotic effects. This study investigates the impact of EA, alone or combined with cisplatin (CIS), on the expression of angiogenesis, apoptosis, metastasis, and chemoresistance-related genes and proteins in cisplatin-sensitive and -resistant MDA-MB-231 breast cancer cells. Cell viability was evaluated by cytotoxicity assay, while gene and protein expression levels were analyzed via qPCR and immunocytochemistry. Molecular docking was used to assess EA's binding affinity to target proteins. The IC₅₀ values of EA

Angiogenic switching in cerebral cavernous malformations driven by MAP3K3-PIK3CA synergy.

Cerebral cavernous malformations are common vascular anomalies in the CNS that predispose individuals to seizures and haemorrhagic stroke. Familial forms are linked to germline loss-of-function mutations in CCM1-3, and sporadic lesions frequently harbour somatic gain-of-function mutations in MAP3K3 and PIK3CA. However, the mechanisms by which these somatic mutations drive lesion development remain incompletely understood, and no medical therapies are currently available. Here, we investigated the cooperative effects of MAP3K3I441M and PIK3CAH1047R mutations using transgenic neonatal and adult

Functional noncoding variants within the TBX1 enhancer contribute to tetralogy of Fallot.

The TBX1 deletion is frequently observed in patients with tetralogy of Fallot (TOF), the most prevalent cyanotic congenital heart defect (CHD); however, the role of noncoding variants in its regulatory region remains unclear. We performed whole-genome sequencing (WGS) on a cohort of 428 patients diagnosed with TOF, which identified the presence of 22q11.2 deletion in 31 cases, including the TBX1 genomic region, as well as 7 functional noncoding variants in its enhancer (EnhTBX1). These findings accounted for a total of 9.81% (42/428) of TOF cases. We then knocked out EnhTBX1 (EnhTBX1-/-) in ES

MicroRNAs in endometriosis: bioinformatics resources, machine learning strategies, and multi-omics perspectives.

BACKGROUND: Endometriosis is a heterogeneous gynecological disorder characterized by chronic pain, infertility, and substantial impairment of quality of life. Increasing evidence indicates that microRNAs (miRNAs) are key regulators of endometriosis pathogenesis through their effects on inflammation, angiogenesis, cell proliferation, fibrosis, and hormone-responsive pathways. METHODS: In this review, we summarize the biological roles of miRNAs in endometriosis and discuss their emerging value as diagnostic biomarkers and therapeutic targets. We further examine major bioinformatics resources an
