Clinical follow-up in the rat experimental model of African trypanosomiasis.

Animal models of Human African Trypanosomiasis (HAT) have been developed to understand the pathogenic mechanisms leading to the passage into the neurological phase, most of them referring to histological aspects but not clinical or behavioral data. Our study aimed at defining simple clinical and/or behavioral markers of the passage between the hemolymphatic phase and the meningo-encephalitic stage of the disease. Sprague-Dawley rats (n=24) were infected with Trypanosoma brucei brucei AnTat 1.1E. Food intake and body weight were measured daily from the day of infection until death. Hematocrit w

[Rapid tolerance to ethanol and high voluntary alcohol consumption in mice selected for brain weight].

Mice of two strains selected for small and large brain weight (SB and LB, respectively) had free access to 10% alcohol and water within three months. At the end of this period, they consumed alcohol in daily dose of 6.9 +/- 0.9 and 7.5 +/- 0.8 g/kg, respectively. After a period of imposed three-day abstinence, the alcohol consumption by the mice of these strains increased by 68.6 and 49.3%, respectively. Exploratory behavior of independent groups of mice from these strains was studied in the closed cross-maze. The animals were injected with ethanol (2.4 g/kg, i.p.) or vehicle twice with a week

Behavioral arousal blocks light-induced phase advances in locomotor rhythmicity but not light-induced Per1 and Fos expression in the hamster suprachiasmatic nucleus.

Both photic and nonphotic stimuli entrain circadian rhythms. Although the adaptive significance of nonphotic clock resetting is unknown, one possibility is that nonphotic cues modulate circadian responses to light. Results of studies on the interaction between photic and nonphotic stimuli support this idea. During the day, light blocks the effects of nonphotic stimuli on the phase of locomotor rhythms and on expression of clock genes in suprachiasmatic nucleus (SCN) neurons. At night, novelty-induced activity prior to and during exposure to light attenuates the phase-shifting response to that

Facilitation of ejaculation after methamphetamine administration in paradoxical sleep deprived rats.

This study investigated the effects of methamphetamine (MA) on genital reflexes in paradoxical sleep deprived (PSD) rats. Different doses of MA (0, 4, 16 and 64 mg/kg) were acutely given after PSD or the equivalent time to control animals. We observed enhancement of spontaneous ejaculation in PSD rats with larger doses of MA, the highest of which induced ejaculation in 100% of the PSD rats. This was significantly higher than the 30% in the control. Although testosterone exerts motivational effects on male sexual behavior, our data shows that testosterone levels were lower after the PSD period

Stereotyped behavior: effects of d-amphetamine and methylphenidate in the young rat.

The proclivity of d-amphetamine and methylphenidate to induce perseverative motoric and vocal side effects detracts from the clinical efficacy of these stimulants in the treatment of Attention-Deficit Hyperactivity Disorder (ADHD). In an attempt to develop a model for these deleterious treatment effects, this study explored the behavioral influences exerted by d-amphetamine and methylphenidate in the young laboratory rat. This experiment revealed that doses of these stimulants that typically induce stereotypy provoke diverging behavioral profiles: while animals given 5 mg/kg d-amphetamine exhi

Intracerebroventricular administration of cholecystokinin reduces stereotypy in dopamine-supersensitive rats.

Cholecystokinin (CCK-8) coexists with dopamine in some neurons and modulates dopaminergic neurotransmission. In the present study we determined the effect of CCK-8 on stereotyped behavior in supersensitive dopaminergic system. Adult male Wistar rats, weighing 200-250 g, were used. Dopaminergic supersensitivity was induced by long-term haloperidol (HAL) treatment (30 days: 1.0 mg/kg twice a day). Seventy-two hours after HAL withdrawal animals received CCK-8 (14.5 nmol/5 microliters) or saline intracerebroventricularly (icv) before being tested for apomorphine (APO, 0.6 mg/kg, sc)-induced stereo

Rapid cessation of focally induced generalized seizures in rats through microinfusion of lidocaine hydrochloride into the focus.

An experimental animal model of complex partial seizures which become secondarily generalized is produced by microinfusion of the GABA antagonist bicuculline (BIC) into the deep prepiriform cortex (DPC) of rats. In the present study, we investigated the effects of microinfusion of the local anesthetic, lidocaine hydrochloride, directly into the BIC focus in the DPC and demonstrated that direct inactivation of the focus arrested a focal seizure that was in progress. A measure of the integrated amplitude of the electrocorticogram (ECoG) and behavioral seizure scores from unanesthetized and freel

The effect of luteinizing hormone releasing hormone on the copulatory behavior of hyperprolactinemic male rats.

The present study was undertaken to test the hypothesis that the deficits in copulatory behavior observed in hyperprolactinemic male rats may be related to a reduction in hypothalamic release of luteinizing hormone releasing hormone (LHRH). Adult male Fischer 344 rats were made hyperprolactinemic by ectopic pituitary grafts or were sham operated and 30 min prior to being tested for copulatory performance received a single subcutaneous injection of 500 ng LHRH, 100 ng LHRH, or saline. On different occasions, testosterone (T) levels were measured in plasma collected 30 min following identical tr

The intestinal microbiota and anorexia nervosa: cause or consequence of nutrient deprivation.

The intestinal microbiota is a diverse microbial community that colonizes the gastrointestinal tract of animals. Abnormal changes in intestinal microbiota has been associated with multiple diseases including inflammatory bowel diseases and obesity; however, emerging evidence suggests a role for the gut microbiota in anxiety and depression via the gut-brain axis. As this microbial community is associated with weight dysregulation and host behavior it is not surprising that the intestinal microbiota may have a role to play in anorexia nervosa (AN). In this review we examine recent studies linkin

Generational Effects of Opioid Exposure.

The inheritance of substance abuse, including opioid abuse, may be influenced by genetic and non-genetic factors related to the environment, such as stress and socioeconomic status. These non-genetic influences on the heritability of a trait can be attributed to epigenetics. Epigenetic inheritance can result from modifications passed down from the mother, father, or both, resulting in either maternal, paternal, or parental epigenetic inheritance, respectively. These epigenetic modifications can be passed to the offspring to result in multigenerational, intergenerational, or transgenerational i

Sigma (σ) receptors as potential therapeutic targets to mitigate psychostimulant effects.

Many psychostimulants, including cocaine and methamphetamine, interact with sigma (σ) receptors at physiologically relevant concentrations. The potential therapeutic relevance of this interaction is underscored by the ability to selectively target σ receptors to mitigate many behavioral and physiological effects of psychostimulants in animal and cell-based model systems. This chapter begins with an overview of these enigmatic proteins. Provocative preclinical data showing that σ ligands modulate an array of cocaine and methamphetamine effects are summarized, along with emerging areas of resear

Can ophthalmologists repair the brain in infantile esotropia? Early surgery, stereopsis, monofixation syndrome, and the legacy of Marshall Parks.

Can ophthalmologists repair defects of visual cortex circuitry in infants who have esotropia? The answer to this question encompasses both sensory and motor behaviors because the clinical hallmarks of the disorder are stereoblindness and absence of motor fusion, which manifests as convergently deviated eyes. Functional recovery of sensory and motor fusion in infantile esotropia was a consuming interest, if not career-defining passion, of Marshall Parks. The purpose of this work is to pay tribute to Parks' legacy by showing how human and animal studies, conducted largely during the last 25 year

MRI visualization of focal induced neocortical malformations of the rat.

In an attempt to determine whether small focal malformations of the neocortex can be visualized in vivo, focal microgyria were induced in the neocortex of otherwise normal rats by freezing injury to the developing cortical plate, and in adulthood the malformation was visualized using MRI. Induced microgyria of varying size were successfully visualized with MRI, and the location and extent of the malformation was confirmed on subsequent histology. This work has potential implications for the field of experimental neuropathology by enhancing the ability to study the behavioral and connectional c

Psychophysiology of stress in dermatology. The psychobiologic pattern of psychosomatics.

Stress is an abnormal or extreme adjustment in the physiology of an animal to cope with adverse effects of its environment and management. The adverse effect is designated the stressor. In this article, the authors try to focus on the main types of host factors that can influence stress (genetics, perception) and on the numerous types of stressors (environmental, behavioral, psychological). Moreover, the authors outline the relevance of psychosomatic medicine, proposing the examination of psyche and soma as a unit "in sickness and in health." They focus their attention on the new aspects of bi

Effect of aging on striatal dopamine receptor subtype recovery following N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline blockade and relation to motor function in Wistar rats.

Recovery of D1- and D2-dopamine receptors in Wistar rat corpora striata were assessed following N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ blockade). Absolute recovery declines during senescence approximately 25 and 40% for the D1- and D2-subtypes, respectively. Net biosynthetic reductions are comparable to the overall age-related decreases in receptor concentrations for this rat strain. EEDQ administration also induces catalepsy behavior and impairs ability of animals to remain on an inclined screen. Recovery of inclined screen performance is also reduced with age, but is not strict

[Effect of newborn rat testes on the male sexual behavior of the testosterone-treated adult].

Neonatal male rats were castrated either at 0, 6 or 24 hrs. after birth. As adults, testosterone was delivered by subcutaneous implantation of a Silastic capsule containing this hormone. The probability to display mounting behavior in presence of an estrous female was lower when the animals were castrated at 0 hr. than at 6 or 24 hrs. or when they received a subcutaneous injection of 1 microgram of testosterone propionate, at the time of castration at 0 hr. These results suggest that in the rat, during the 6 hrs. following birth, neonatal testes influence the sensitivity of the adult central n

Enclosure design and space utilization by Indian leopards (Panthera pardus) in four zoos in Southern India.

Enclosure design and the use of enclosure space influence the activity budget of cap-tive leopards. The study laid out in grids all enclosures on the base map and segregated these grids into 4 zones. Every 5 min, the study recorded the proportion of time spent in these zones with the leopards' behavior. Captive leopards most frequently used the "edge" zone. Almost all leopards used the edge zone for stereotypic pacing, the "back" zone for resting, and the "other" zone for activity. The study positively corre-lated the proportion of time spent in the "enriched" zone with activity levels exhibit

Effect of color in rearing and testing environment on the behavior of young domestic chicks.

Young domestic chicks were reared individually in compartments of four different colors and then tested either in a color-preference test with stationary objects (N equals 128) or with a moving stimulus (N equals 128). For each type of test we used four background colors in the test arena. The dependent variables included color preferences, following behavior and emotional responses of the young chicks. The results showed no significant effects of the color of the arena-background in either test situation. The color of the rearing compartment was a significant factor affecting color preference

Acute psilocybin or ethocybin leads to a sustained increase in Pavlovian aversion memory in male mice.

There is considerable interest in the concept that psychedelic compounds might facilitate reorganization of emotional processing with therapeutic benefit; in particular, amelioration of excessive focus on past, current and potential-future aversive events, a dominant emotional-cognitive state in stress-related affective disorders. Translational animal models can advance understanding of specific aspects of this concept, namely, those relating to subconscious emotional memories and their neurobiology. In male mice, chronic social stress (CSS) leads to excessive tone/context-foot shock Pavlovian
