A rat model of bone cancer pain.

This study describes the first known model of bone cancer pain in the rat. Sprague-Dawley rats receiving intra-tibial injections of syngeneic MRMT-1 rat mammary gland carcinoma cells developed behavioural signs indicative of pain, including: mechanical allodynia, difference of weight bearing between hind paws and mechanical hyperalgesia. The development of the bone tumour and structural damage to the bone was monitored by radiological analysis, quantitative measurement of mineral content and histology. Intra-tibial injections of 3 x 10(3) or 3 x 10(4) syngeneic MRMT-1 cells produced a rapidly

Bone marrow failure and extramedullary hematopoiesis in McCune-Albright syndrome.

In fibrous dysplasia/McCune-Albright syndrome (FD/MAS), bone and bone marrow are, to varying degrees, replaced by fibro-osseous tissue typically devoid of hematopoietic marrow. Despite the extensive marrow replacement in severely affected patients, bone marrow failure is not commonly associated with FD/MAS. We present a 14-year-old girl with FD/MAS, who developed pancytopenia and extramedullary hematopoiesis (EMH) with no identified cause, in the setting of iatrogenic thyrotoxicosis and hyperparathyroidism. Pancytopenia, requiring monthly blood transfusions, persisted despite multiple strategi

Mechanisms, impact and prevention of pathological bone regeneration in spondyloarthritis.

PURPOSE OF REVIEW: To discuss different aspects of new bone formation in patients with spondyloarthritis based on emerging data from clinical trials, prospective cohort studies and translational laboratory investigations. RECENT FINDINGS: New bone formation potentially leading to ankylosis of the spine and sacroiliac joints remains an important concern for patients with axial spondyloarthritis. New therapeutic strategies, in particular targeting of interleukin-17, have emerged in addition to the antitumor necrosis factor drugs, but we still fail to fully understand the mechanisms of structura

Application of automatic image segmentation to tibiae and vertebrae from ovariectomized rats.

Automatic contextual segmentation algorithms were developed to objectively identify bone compartments in pQCT images of tibiae, femora, and vertebrae. Principal advantages of this approach over existing techniques such as histomorphometry are as follows: (a) the algorithms can be implemented in a fast, uniform, nonsubjective manner across many images, allowing unbiased comparisons of therapeutic efficacy; (b) much larger volumes in the region of interest can be analyzed to derive true volumetric parameters for trabecular and cortical bone compartments; and (c) pQCT can be used to quantitate bo

Viable Gata2 deficient mice exhibit T cell development independent of adult definitive hematopoiesis.

Gata2 is indispensable for hematopoiesis, with knockout mice typically exhibiting embryonic lethality attributed to severe anemia and defective hematopoietic stem cell (HSC) development. Intriguingly, we obtained viable adult Gata2-/- mice harboring a 17-nucleotide deletion in exon 2, which displayed intact T-cell development despite a significantly reduced bone marrow HSC pool. While mutant HSCs exhibited attenuated lymphoid potential, they retained partial myeloid reconstitution capacity. Single-cell transcriptomic analysis revealed compensatory upregulation of key hematopoietic regulators,

Functions of Matricellular Proteins in Dental Tissues and Their Emerging Roles in Orofacial Tissue Development, Maintenance, and Disease.

Matricellular proteins (MCPs) are defined as extracellular matrix (ECM) associated proteins that are important regulators and integrators of microenvironmental signals, contributing to the dynamic nature of ECM signalling. There is a growing understanding of the role of matricellular proteins in cellular processes governing tissue development as well as in disease pathogenesis. In this review, the expression and functions of different MP family members (periostin, CCNs, TSPs, SIBLINGs and others) are presented, specifically in relation to craniofacial development and the maintenance of orofaci

Bone Organoids: Bridging Natural Bone with Advanced Organoid Technologies.

Bone tissue engineering has long been a focal point of research, aiming to address critical large segmental bone defects resulting from severe trauma, tumors, and other bone-related diseases. Despite significant advancements in conventional bone tissue engineering, the simulation of the intricate microenvironment characteristic of natural bone tissue remains inadequate. Natural bone is characterized by intricate macroscopic and microscopic architectures, along with a dynamic microenvironment that facilitates processes such as bone formation, remodeling, and repair. Bone organoids-three-dimensi

Hypocalcaemia in patients with prostate cancer treated with a bisphosphonate or denosumab: prevention supports treatment completion.

BackgroundMost patients with advanced prostate cancer develop bone metastases, which often result in painful and debilitating skeletal-related events. Inhibitors of bone resorption, such as bisphosphonates and denosumab, can each reduce the incidence of skeletal-related events and delay the progression of bone pain. However, these agents are associated with an increased risk of hypocalcaemia, which, although often mild and transient, can be serious and life-threatening. Here we provide practical advice on managing the risk of hypocalcaemia in patients with advanced prostate cancer who are rece

Effects of condensation and compressive strain on implant primary stability: A longitudinal, in vivo, multiscale study in mice.

AIMS: Surgeons and most engineers believe that bone compaction improves implant primary stability without causing undue damage to the bone itself. In this study, we developed a murine distal femoral implant model and tested this dogma. METHODS: Each mouse received two femoral implants, one placed into a site prepared by drilling and the other into the contralateral site prepared by drilling followed by stepwise condensation. RESULTS: Condensation significantly increased peri-implant bone density but it also produced higher strains at the interface between the bone and implant, which led to s

Hypophosphatemia, Severe Bone Pain, Gait Disturbance, and Fatigue Fractures After Iron Substitution in Inflammatory Bowel Disease: A Case Report.

Intravenous infusions of different iron formulations are recognized as a cause of hypophosphatemia. Chronic hypophosphatemia can alter bone metabolism and bone material structure. As a consequence, osteomalacia may develop and lead to bone fragility. Herein, we report a patient with Crohn's disease presenting with persistent hypophosphatemia and insufficiency fractures while receiving regular iron infusions due to chronic gastrointestinal bleeding. Previously, the patient regularly received vitamin D and also zoledronic acid. The patient underwent bone biopsy of the iliac crest that showed typ

Navigating the Planning and Initiation of a First Prospective Clinical Trial Through a Regional Pediatric Oncology Consortium Within the Middle East, Africa, and Asia-A Roadmap.

BACKGROUND: Prospective pediatric oncology clinical trials remain scarce across low- and middle-income countries (LMICs). We developed the first multicountry prospective clinical trial within the Pediatric Oncology East and Mediterranean (POEM) group. Localized osteosarcoma was selected due to heterogeneous real-world management, including alternative regimens in settings where methotrexate administration is not feasible. This descriptive report details the feasibility of a regionally coordinated, resource-adapted clinical trial across diverse LMIC settings. METHODS: We describe the planning,

Bone age advancement in non-obese children with premature adrenarche: relationship to growth acceleration and predicted adult height outcomes.

OBJECTIVES: Premature adrenarche (PA) is commonly associated with bone age (BA) acceleration, but it is difficult to assess the independent contribution of adrenal androgens to skeletal maturation in studies that include obese children. This study examined BA advancement in non-obese children with PA and evaluated predicted adult height (PAH) estimates in this population. METHODS: The medical records of children diagnosed with PA between April 2022 and April 2023 were reviewed retrospectively. Children who were obese, had congenital adrenal hyperplasia, central precocious puberty, genetic or

Association Between Bone Maturation, Menstrual Cycle, Hypermobility, Scoliosis, and Low Back Pain in Pre-Professional Female Adolescent Dancers.

BACKGROUND: Many adult dancers suffer from low back pain (LBP), which interferes with their movement strategies. A group that merits further attention are adolescent dancers as high levels of physical activity are associated with a higher risk to develop LBP. OBJECTIVE: The aims of this cross-sectional study were (i) to investigate if delayed bone maturation and aberrant menstrual cycle were present in pre-professional female adolescent dancers and (ii) whether these ailments or other physical impairments could be related to LBP. OUTCOME MEASURES: Bone maturation, menstrual cycle, hypermobil

Management of bilateral osteochondritis dissecans of the trochlea in a skeletally immature patient.

Osteochondritis dissecans (OCD) lesions are injuries that occur more commonly in the skeletally immature population. In most cases, the aetiology is not well understood, but fortunately, many OCD lesions may heal on their own over time, particularly in skeletally immature patients with open physes. Conversely, if the lesion is considered unstable, surgical intervention may be required. This case demonstrates an especially rare presentation of bilateral OCD lesions within the lateral femoral trochlear facet. The lesions became symptomatic approximately 1 year apart without a specific injury. No

Limb specific Acvr1-knockout during embryogenesis in mice exhibits great toe malformation as seen in Fibrodysplasia Ossificans Progressiva (FOP).

PURPOSE: This study analyzes Prx1-specific conditional knockout of Acvr1 aiming to elucidate the endogenous role of Acvr1 during limb formation in early embryonic development. ACVR1 can exhibit activating and inhibiting function in BMP signaling. ACVR1 gain-of-function mutations can cause the rare disease fibrodysplasia ossificans progressiva (FOP), where patients develop ectopic bone replacing soft tissue, tendons and ligaments. METHODS: Whole-mount in situ hybridization and skeletal preparations revealed that following limb-specific conditional knockout of Acvr1, metacarpals and proximal ph

Bi-allelic Loss-of-Function Mutations in the NPR-C Receptor Result in Enhanced Growth and Connective Tissue Abnormalities.

The natriuretic peptide signaling pathway has been implicated in many cellular processes, including endochondral ossification and bone growth. More precisely, different mutations in the NPR-B receptor and the CNP ligand have been identified in individuals with either short or tall stature. In this study we show that the NPR-C receptor (encoded by NPR3) is also important for the regulation of linear bone growth. We report four individuals, originating from three different families, with a phenotype characterized by tall stature, long digits, and extra epiphyses in the hands and feet. In additio
