Infrapatellar fat pad exosomes promote osteoarthritis by reprogramming macrophage polarization.

The infrapatellar fat pad (IPFP) is a crucial mediator in knee osteoarthritis (OA), but the mechanisms of its pathogenic signaling remain elusive. This study investigates the role of IPFP-derived exosomes in OA progression. We found that exosomes from end-stage OA patient IPFP (OA-EXOs) were secreted abundantly. In vitro, OA-EXOs were internalized by macrophages, reprogramming them to a pro-inflammatory M1 phenotype while inhibiting anti-inflammatory M2 polarization, thereby creating a catabolic microenvironment that disrupted chondrocyte extracellular matrix homeostasis. Furthermore, OA-EXOs

Limited Added Effect of Proline-Rich P6 Peptide to Cross-Linked Hyaluronic Acid After Partial Lateral Meniscectomy in Sheep.

This study evaluates whether a proline-rich P6 peptide provides added benefit to a cross-linked hyaluronic acid (HA) hydrogel after partial lateral meniscectomy-associated osteoarthritis-like change in sheep. The corrected four-group framework includes Intact no-surgery/no-injection reference knees (n = 7) and 16 surgically injured knees from eight sheep assigned to Mnx (meniscectomy only, n = 4), Mnx + HA (n = 6), or Mnx + HA + P6 (n = 6). Mnx + HA and Mnx + HA + P6 knees received intra-articular injections at Weeks 0, 1, 2, and 4. Osteochondral tissues were evaluated at Week 12 by microCT, H

The ITGB2-COPS3-SOX2 Axis and SOX2 Liquid-Liquid Phase Separation: Dual Mechanisms Governing Osteosarcoma Stemness.

Osteosarcoma is a highly malignant bone tumor prevalent in adolescents, whose therapeutic challenges stem largely from cancer stem cell (CSC)-mediated chemoresistance and pulmonary metastasis. High-frequency amplification of the 17p11.2 chromosomal region in osteosarcoma represents a key molecular event driving malignant progression. This study focuses on the key gene COPS3 in this region, delving into its molecular mechanisms regulating cancer stemness. The findings show that elevated COPS3 expression strongly correlates with stemness features. Mechanistically, COPS3 binds directly to transcr

Elucidating the mechanism of polycyclic aromatic hydrocarbons-induced osteoporosis through network toxicology and molecular docking approaches: BaP as a model compound.

This work showcases a network‑toxicology strategy to uncover the osteotoxic effects and molecular pathways of polycyclic aromatic hydrocarbons, using BaP‑induced osteoporosis as a model. Potential targets of BaP were extracted from the Comparative Toxicogenomics Database (CTD) and ChEMBL, while osteoporosis-related targets were obtained from GeneCards. A total of 93 overlapping genes between osteoporosis-related targets and BaP-perturbed genes were identified. Subsequent protein-protein interaction (PPI) network analysis was constructed using STRING and visualized with Cytoscape. Seven core ge

Immuno-Mechanical Signaling Network Integration in Temporomandibular Joint Pathology: A TMID Conceptual Framework.

Temporomandibular disorders (TMDs) are multifactorial conditions traditionally attributed to excessive mechanical loading on the temporomandibular joint, leading to clinical manifestations ranging from joint sounds to structural deformation. Contributing factors include trauma, occlusal abnormalities, psychological stress, and bruxism. However, immune and molecular alterations associated with early disease activity are not systematically integrated into structure-centered TMD frameworks. Emerging evidence indicates that temporomandibular joint osteoarthritis (TMJOA) involves activation of inna

Mammalian Osteoderm Ultrastructure in the Armored Acomys Spiny Mouse Tail.

The skin of mammals acts as a barrier against pathogens, ultraviolet radiation, and mechanical trauma. Embedded within the dermal tail skin of spiny mice (genus Acomys) lie osteoderms-finely structured, symmetrically patterned, bone-like armor plates akin to those in armadillos and non-avian reptiles. Using 2D and 3D microscopic characterization techniques, we provide details on Acomys osteoderm patterning, cell size and distribution, and an ultrastructural overview of collagen-mineral relationships in the extracellular matrix. Osteoderms are organized into a periodic array with A-B-A-B-A-B pa

CD31+ Cell Enrichment Enhances Therapeutic Effects of Stromal Vascular Fraction in Experimental Primary Osteoarthritis: A Preclinical Study in the Dunkin Hartley Guinea Pig Model.

Despite the growing burden of osteoarthritis (OA), effective disease-modifying treatments remain limited. Hyaluronic acid, platelet-rich plasma, and hydrogels are in clinical use, but their efficacy beyond placebo is debated. Cell-based therapies are continuously investigated for their disease-modifying potential. Stromal vascular fraction (SVF), a minimally processed and clinically accessible cell product, has shown therapeutic potential, but remains poorly characterized in respect to its pro-regenerative cell types and molecular signaling. To determine whether CD31+ cell enrichment enhances

STK40 inhibits profibrotic Arg1+ macrophage differentiation and renal fibrosis by regulating poly-ubiquitination of STAT3.

Monocyte-derived macrophages are central drivers of chronic renal inflammation and fibrosis, yet the regulatory mechanisms that restrain their profibrotic differentiation remain poorly defined. Here, we identified serine/threonine kinase 40 (STK40) as a suppressor of profibrotic macrophage differentiation and renal fibrosis progression. Myeloid-specific Stk40 deletion exacerbated renal fibrosis in multiple mouse models. Single-cell RNA sequencing revealed expansion of Arg1⁺ macrophages in STK40-deficient kidneys. In vitro, STK40 restrained the differentiation of profibrotic Arg1⁺ macrophages i

TNAP and PHOSPHO1 Function Synergistically to Afford Critical Control Over the Mineralization of the Postnatal Murine Skeleton.

Biomineralization is essential for skeletal integrity, yet the synergistic roles of tissue non-specific alkaline phosphatase (TNAP) and PHOSPHO1 in postnatal bone remain unclear. We generated a novel murine model with Prx1-driven deletion of Alpl (AlplPrx1/Prx1) on a global Phospho1-/- background, circumventing the perinatal lethality associated with dual global deletion and premature death of Alpl global knockouts. Multi-modal spatial phenotyping of the limbs revealed that mice lacking both TNAP and PHOSPHO1 exhibit distinct mineralization defects and altered anatomical structures at postnata

Separable microneedles based on thermosensitive hydroxypropyl chitin with hyaluronic acid separation layer via green process for prolonged piroxicam release in osteoarthritis treatment.

Osteoarthritis is a chronic degenerative joint disease with local inflammation that needs sustained, localized anti-inflammatory therapy. In this study, a separable drug-loaded microneedle system based on thermosensitive hydroxypropyl chitin (HPCH) was developed via a green fabrication process without using organic solvents or chemical crosslinkers for prolonged transdermal piroxicam delivery. Each PrxC@HPCH microneedle contained a tip loaded with size-controlled piroxicam microcrystals (PrxC), prepared via an alkaline dissolution-acid neutralization antisolvent strategy, and a rapidly separat

Phosphate homeostasis and endocrine regulators.

Phosphate homeostasis is essential for skeletal integrity, cellular metabolism, and endocrine regulation. In humans, circulating phosphate is typically maintained between 2.5 and 4.5 mg/dL by coordinated processes that balance gastrointestinal uptake from the diet, storage in cells and bone matrix, and renal reclamation that regulates excretion. Dietary phosphate enters primarily through the small intestine via a low‑affinity paracellular diffusion and a high‑affinity, sodium‑coupled transcellular route mediated by transporters such as NPT2b, PIT1, and PIT2. After absorption, phosphate is inco

Single-cell analysis reveals multi-faceted features of B cell development, together with age-associated B cell subpopulations.

The development and maturation of B lymphocytes are critical for adaptive immunity, relying on tightly regulated gene programs within specialized microenvironments shaped by extracellular matrix and neighboring cells. However, high-dimensional, integrated analyses of B cell heterogeneity, gene regulation, and external factors during development remain limited. Here, we analyze single-cell transcriptomic and B cell receptor (BCR) sequencing data from B cells and surrounding cells in bone marrow, tonsil, and peripheral blood. We reveal the dynamics of gene regulation, the heterogeneity of conven

Pycnodysostosis: Report of Two Novel CTSK Variants in a Child.

Background: Pycnodysostosis is a rare autosomal recessive skeletal disorder caused by biallelic pathogenic variants in CTSK, which encodes cathepsin K, a lysosomal cysteine protease required for osteoclast-mediated degradation of bone matrix. Case Report: We describe a girl with short stature, skeletal deformities, osteosclerosis, craniofacial features, clavicular dysplasia, and radiological evidence of fractures. Clinical exome sequencing identified two heterozygous CTSK variants, c.85T > C (p.Trp29Arg) and c.679A>T (p.Ile227Phe), both currently classified as variants of uncertain significanc

Mechanisms and therapeutic targets in distant metastasis of lung adenocarcinoma.

Distant metastasis, characterized by organotropism, is a major cause of mortality in lung adenocarcinoma (LUAD). In this study, digital spatial profiling (DSP), multiplex immunofluorescence (mIF), and clinical data from 52 LUAD patients were integrated to develop organ-specific metastasis risk models, and the molecular mechanisms underlying metastatic organotropism were investigated. Random forest models based on primary tumor spatial transcriptomics accurately predicted metastasis to the brain (AUC = 0.974), liver (AUC = 0.975), adrenal gland (AUC = 0.929), and bone (AUC = 0.907). SPP1hi macrophages in fibrin niches promote hyperplastic tissue remodeling in rheumatoid arthritis synovium.

In chronic inflammatory diseases, maladaptive tissue remodeling is driven by a complex interplay of resident cells, immune infiltrates, and the extracellular matrix. In the autoimmune disorder rheumatoid arthritis (RA), synovial tissue undergoes massive expansion to form an invasive pannus that drives the erosion of cartilage and bone. The mechanisms mediating this aggressive growth are incompletely defined. Using spatial transcriptomic profiling of patient tissue, we detected an abundance of proliferating fibroblasts near the synovial tissue lining surface and adjacent to macrophages expressi

Berberine Chloride Suppresses Growth of Canine Osteosarcoma Cells via Regulating MMP2/AKT/β-Catenin Pathway.

Canine osteosarcoma is an aggressive primary bone malignancy associated with a poor prognosis and a high propensity for lung metastasis. Due to the limitations and side effects of conventional chemotherapy, there remains a pressing need for alternative therapeutic strategies and adjuvants. In this study, we examined the effects of berberine chloride (BBRC), a bioactive isoquinoline alkaloid derived from the dried roots of Coptidis Rhizoma, on canine osteosarcoma D-17 and DSN cell lines. BBRC reduced the viability and migratory capacity of D-17 and DSN cell lines in three-dimensional (3D) spher

DNA methylation subtypes dictate metastatic heterogeneity of osteosarcoma via distinct tumor-stromal interactions: Multi-omics profiling and decitabine validation.

Osteosarcoma (OS), the most prevalent primary bone malignancy in adolescents, is characterized by aggressive progression and early metastasis. However, the epigenetic drivers of its metastatic heterogeneity remain poorly understood. Herein, we integrated bulk DNA methylation profiling and single-cell RNA sequencing (scRNA-seq) to elucidate the epigenetic mechanisms driving OS metastatic heterogeneity. Consensus clustering identified two methylation subtypes (K = 2) with distinct survival outcomes, where hypermethylated (MSO-high) tumors exhibited poor prognosis. Weighted gene co-expression net
