YBX1 Modulated Corneal Neovascularization Induced by Alkali Burn via m5C-Dependent Regulation of the STAT3/HIF-1α/VEGFA Axis.

PURPOSE: Effective management of corneal neovascularization (CoNV) remains challenging, and the role of epitranscriptomic regulation, particularly N5-methylcytosine (m5C) modification, in this process remain incompletely defined. This study investigated the function and mechanism of the RNA-binding protein YBX1 in CoNV following alkali burn (AB). METHODS: An AB-induced CoNV model was generated using C57BL/6 mice. In vitro, human umbilical vein endothelial cells (HUVECs) underwent hypoxia/reoxygenation (H/R). Multi-omics approaches including transcriptome sequencing, RNA immunoprecipitation se

Promotive effects of 830-nm Photobiomodulation on corneal epithelial wound healing and rho-gtpase expression in a rat alkali burn model.

INTRODUCTION: Corneal injuries can lead to permanent damage that disrupts the integrity of the cornea and impairs vision. Photobiomodulation (PBM) is a non-invasive therapy that uses specific wavelengths of light to stimulate biological processes, leading to a range of therapeutic benefits. In this study, the effect of PBM on the alkali burn model was investigated, with a focus on its potential to promote cell proliferation and migration and reduce inflammation using 830 nm of light. A wavelength of 830 nm was used for PBM treatment. After Alkali burn induction, rats were treated with 830 nm l

Protective efficacy of a PcrV-based vaccine formulated with chitosan nanoparticles against Pseudomonas aeruginosa infection in burn and sepsis mouse models.

Pseudomonas aeruginosa is an opportunistic Gram-negative pathogen and a major clinical concern due to its multidrug resistance and extensive virulence repertoire. In this study, recombinant PcrV protein was formulated with Alum and chitosan nanoparticles (CS-NPs) as co-adjuvants to enhance immunogenicity. CS-NPs were prepared via ionic gelation and characterized for morphology and charge. BALB/c mice were immunized with formulations containing r-PcrV + Alum + Nanochitosan, r-PcrV + Alum, r-PcrV + Nanochitosan, or Nanochitosan + Alum or PBS (control). This was followed by burn induction, after

GM-CSF and IL-1α secreted by cryopreserved porcine skin promote angiogenesis in burn wounds by activating the JAK2/STAT3 pathway.

OBJECTIVE: Cryopreserved porcine skin (CPS) is widely applied in skin grafting for burns. In this study, we aimed to investigate the effects and underlying mechanisms of CPS on burn wound healing. METHODS: Cryopreserved porcine skins were acquired. Guinea pig burn models were established and grafted with thawed-CPS. Human umbilical vein endothelial cells (HUVECs) were cultured in CPS-conditioned medium. Histological structure was assessed by hematoxylin-eosin (HE) staining. Protein levels were detected using enzyme-linked immunosorbent assay (ELISA), western blot, and immunohistochemistry. HU

SPARC-overexpressing mesenchymal stem cell-hydrogel system accelerates burn wound healing and improves tissue repair in diabetic mice.

Burn injuries, especially in diabetic patients, are difficult to treat because of persistent inflammation, impaired angiogenesis, and defective remodeling of the extracellular matrix (ECM). A hydrogel based on hyaluronic acid, chitosan, and alginate was developed to deliver adipose-derived mesenchymal stem cells (MSC) overexpressing SPARC (secreted protein acidic and rich in cysteine). SPARC is an ECM glycoprotein that promotes angiogenesis, collagen deposition, and matrix remodeling and whose activity is reduced in diabetic wounds - was overexpressed to restore these regenerative functions, a

Personalized Clinical Workflow for Compression Therapy in Post-Burn Hypertrophic Scars Using 3D Imaging and Computational Modeling: A Feasibility Study.

Background: Hypertrophic scarring remains a common and clinically challenging consequence of burn injury. Although compression therapy is widely used, its effectiveness is limited by variability in pressure delivery, lack of personalization and inconsistent treatment protocols. Methods: This prospective single-centre feasibility study included adult patients with post-burn scars following deep partial-thickness or full-thickness injuries. The study was designed as a two-phase workflow. In Phase I, scars were documented using standardized photography and 3D surface scanning and were evaluated u

Investigating the prevalence of class 1, 2, and 3 integrons in carbapenem-resistant Acinetobacter baumannii isolated from burn wound infections.

Acinetobacter baumannii is a significant antibiotic-resistant pathogen with high morbidity and mortality in hospitalized patients, especially in burn units. Acquiring mobile genetic elements, such as integrons, is significant in developing multidrug-resistant (MDR) hospital isolates. Therefore, this study aimed to determine the prevalence of class 1, 2, and 3 integrons in A. baumannii. The clinical isolates were collected from burned patients with wound infections. The isolates were identified using standard biochemical and microbiological tests and were confirmed by detecting the blaoxa-51 ge

Burned out and thinking of quitting: A moderated-mediation model of frontline health care workers and occupational stigma during COVID-19.

BACKGROUND: Work burnout and job turnover intentions pose challenges for health care workers. An understudied area of impact is COVID-19 occupational stigma consciousness or the awareness that one's occupation has been stigmatized because of proximity and exposure to COVID-19. PURPOSES: Using job demands-resources theory, we examine the impact of COVID-19 occupational stigma consciousness, a novel job demand, on work burnout and job turnover intentions and the effectiveness of common job resources to buffer these effects among nurses in the United States. METHODOLOGY/APPROACH: We surveyed fr

Exploring the Role of Meaning in Life in Relation to Burn-Out Symptoms Among Early and Mid-Career Nurses: A Cross-Sectional Study.

BACKGROUND: Burnout negatively affects nurses' well-being and patient care. This study examines whether meaning in life, conceptualized as presence (sense that life is meaningful) and search (active pursuit of meaning), is associated with burnout symptoms among early- and mid-career nurses. Additionally, we explore whether job resources (influence at work, supervisor and colleague support, and professional development) moderate these associations. METHODS: A cross-sectional analysis was conducted using baseline data from a longitudinal panel study among 375 nurses (≤ 10 years of experience) a

Developing and Consumer Testing Fire and Burn and Cold Injury Prevention Education With People Who Are Unhoused: Equity in Injury Prevention Toolkit.

Fifteen percent of people receiving care for major burn injuries in urban burn centers across North America were homeless pre-injury. The number and risk of such injuries are increasing due to greater numbers of people experiencing homelessness (PEH) and the frequency of extreme climate events. Prevention education, along with passive and active fire and cold weather protections, is critical for preventing these injuries. To increase acceptability, understandability, and actionability, prevention education needs to be in plain language, contextualized, and consumer-tested. We aimed to test new

Influence of Surgical Procedures on C-Reactive Protein Levels in Severely Burned Patients: Preliminary Analysis on Implications for Early Sepsis Diagnosis.

The local treatment of deep burn wounds involves the excision of the necrosis and covering the wounds with skin grafts. Surgical procedures are thought to have an impact on the inflammatory response, especially in severe burn patients requiring treatment in an intensive care unit. Currently, there are no available data in the literature regarding the correlation of the type of surgical procedure and the levels of the inflammatory markers. This study investigates the importance of monitoring c-reactive protein (CRP) around the time of surgical burn procedures and how it can aid in assessing the

A multi-omics approach exploring the gut-liver axis following combined radiation exposure and burn injury in a Sinclair minipig model.

While radiation and burn injury have distinct local and systemic effects, they both negatively impact intestinal permeability/function. In a nuclear attack, combined thermal burns and radiation exposure (e.g., combined injury (CI)) would be a dominant injury pattern. Despite this, how burns affect gastrointestinal acute radiation syndrome, and vice versa, has been largely unstudied. Next-generation sequencing has revealed a strong bidirectional link between the liver and gut. Here, we used a porcine model to evaluate the impact of burn, radiation, and CI on the gut microbiota and liver transcr

Early extubations in children intubated prior to arrival in Paediatric Burn ICU: A single center retrospective study over 1520 admissions.

BACKGROUND: In adult burns intensive care units, more than 30% of patients arriving intubated, are extubated within 2 days (potentially unnecessary intubation, PUNI). Such data are lacking in paediatric populations. Exploring this paediatric PUNI rate was the primary aim of the study. METHODS: Data from all the admissions to our paediatric burn intensive care unit were retrospectively analyzed over an 8-years period. Extubations within the first two days among patients arriving intubated were assessed as the primary outcome (PUNI rate). Using a univariate logistic regression and a multivariat

Analgesic effect of diluted nitrous oxide in rehabilitation training of patients with burn scar contracture: Study protocol for a randomized controlled trial.

BACKGROUND: Burn patients experience severe pain when undergoing rehabilitation after skin grafting, which negatively affects their recovery. Traditional analgesic methods (such as opioids) have the risk of addictiveness and side effects. Nitrous oxide, which has rapid analgesic and sedative effects, is commonly used for conscious analgesia. The purpose of this study was to determine whether diluted nitrous oxide reduces pain compared to Placebo(oxygen) during rehabilitation after burn surgery. METHOD: This single-center, randomized, double-blind, and controlled trial will enroll 80 patients.
