The expression and function of E3 ligase SIAH2 in acute T lymphoblastic leukemia.

INTRODUCTION: The seven in absentia homolog 2 (SIAH2) protein plays a significant role in human cancer by regulating hypoxia-inducible factor-a (HIF-1α); however, its role in T-cell acute lymphoblastic leukemia (T-ALL) is less clear. METHODS: Immunofluorescence evaluation of SIAH2 protein expression and location were conducted in Jurkat cell (a T-ALL cell line) as well as in bone marrow mononuclear cells (BMMNCs) from T-ALL and idiopathic thrombocytopenic purpura (ITP) patients. The expression of SIAH2 mRNA was also examined by quantitative real-time PCR (qRT-PCR) in these cells. Lentivirus-p

Bacillus anthracis' PA63 Delivers the Tumor Metastasis Suppressor Protein NDPK-A/NME1 into Breast Cancer Cells.

Some highly metastatic types of breast cancer show decreased intracellular levels of the tumor suppressor protein NME1, also known as nm23-H1 or nucleoside diphosphate kinase A (NDPK-A), which decreases cancer cell motility and metastasis. Since its activity is directly correlated with the overall outcome in patients, increasing the cytosolic levels of NDPK-A/NME1 in such cancer cells should represent an attractive starting point for novel therapeutic approaches to reduce tumor cell motility and decrease metastasis. Here, we established the Bacillus anthracis protein toxins' transport componen

P-body-mediated RNA decay controls neural crest EMT.

Neural crest cells, a multipotent vertebrate stem cell population, undergo a tightly regulated epithelial-mesenchymal transition (EMT) to delaminate from the neural tube and initiate migration. Neural crest EMT is controlled by a transient pulse of Draxin, which acts as a molecular rheostat of canonical Wnt signaling, and is then rapidly downregulated by unknown mechanisms. To address this, here we use in vivo reporter assays and live RNA imaging to show that Draxin mRNA is rapidly degraded posttranscriptionally via its 3' untranslated region. We found that Draxin transcripts are targeted to c

tRF-Glu-CTC-005 Promotes Pathological Retinal Neovascularization via ACSL4 in Microvascular Endothelial Cells.

Pathological retinal neovascularization (RNV), a leading contributor to blindness, remains poorly understood, especially regarding the involvement of transfer RNA-derived small RNAs (tsRNAs). In this study, the mouse model of oxygen-induced retinopathy (OIR) was induced in vivo, and human retinal endothelial cells (HRECs) were applied in vitro. CCK-8, cell migration, and tube formation assays were conducted to evaluate alterations in HREC functions. tRF-Glu-CTC-005 was significantly upregulated in OIR retinas compared with controls. Overexpression of tRF-Glu-CTC-005 accelerated retinal angioge

Role of the H19/miR-423-5p/DTX3L Axis in Enhancing the Malignant Phenotype of Nasopharyngeal Carcinoma Cells.

Deltex E3 ubiquitin ligase 3L (DTX3L) is a well-established ubiquitin ligase implicated in various cancers, but its role in nasopharyngeal carcinoma (NPC) progression remains elusive. In this study, we confirmed for the first time that DTX3L was highly expressed in C666-1 and NPC/HK1 NPC cells. DTX3L overexpression promoted NPC cell proliferation, invasion, and migration. Conversely, DTX3L knockdown suppressed these malignant phenotypes. Notably, DTX3L activated the β-catenin pathway, as evidenced by increased β-catenin nuclear translocation, increased transcriptional activity, and elevated ex

Inhibition of replication factor C4-induced DNA damage repair reverses the chemoresistance of glioblastoma to temozolomide and reveals the synthetic lethal effect of combined targeting of checkpoint kinase 1.

Glioblastoma (GBM) is a highly aggressive malignant tumor of the central nervous system, and resistance to temozolomide (TMZ) remains a major cause of treatment failure and tumor recurrence. DNA damage repair (DDR) serves as a critical mechanism underlying chemoresistance, with replication factor C4 (RFC4) emerging as a key mediator. We aimed to investigate the mechanism by which RFC4 induces TMZ resistance in GBM through DDR activation. Analysis of clinical specimens revealed that RFC4 is overexpressed in glioma tissues and correlates with poor prognosis in patients with GBM. Functional assay

CircUBAP2 promotes intrahepatic cholangiocarcinoma progression via PRMT1-p65 methylation and the miR-642b-5p/IL-1β axis.

Intrahepatic cholangiocarcinoma (ICC) frequently exhibits inflammatory signaling and poor outcomes, yet upstream regulators that maintain NF-κB activity remain incompletely defined. Here, we identified circUBAP2 (hsa_circ_0001851) by circRNA microarray and validated it in 76 paired ICC tissues and cell lines. Higher circUBAP2 levels were associated with advanced stage and poorer survival. Gain- and loss-of-function assays showed that circUBAP2 enhanced proliferation and migration in vitro and promoted tumor growth and liver metastasis in vivo, accompanied by increased NF-κB activity. HP1γ promotes the progression of colorectal cancer through interaction with RBPJ of the Notch signaling pathway.

Epigenetic dysregulation plays a pivotal role in cancer progression. Unlike HP1α and HP1β, which localize primarily to heterochromatin, HP1γ predominantly occupies euchromatin, suggesting a direct role in transcriptional regulation. Here, we identify HP1γ as a high-risk oncogene in colorectal cancer (CRC). Genome-wide analysis and disease-free survival (DFS) correlation studies revealed that high expression of HP1γ is associated with poor DFS in CRC patients, and we confirmed HP1γ upregulation in colon adenocarcinoma (COAD), the predominant CRC subtype. Functional assays demonstrated that HP1γ

Discovery of dimeric costunolide-triazole conjugates as antihepatoma agents via integrating dimerization and molecular hybridization.

Costunolide, a natural germacranolide sesquiterpenoid, exhibits only moderate anti-HCC activity. To enhance its efficacy and tumor selectivity, a series of 37 dimeric costunolide-1,2,3-triazole conjugates was designed and synthesized by integrating dimerization and molecular hybridization strategies. Evaluation of their antiproliferative effects on HepG2, Huh-7, and SK-Hep-1 cells suggested that 25 compounds were more potent than either costunolide or sorafenib. The most active dimer 19 exhibited significant activity with IC50 values of 1.6, 1.3, and 0.7 μmol·L-1, which were 13.1, 14.2, and 34

5-Methoxyflavone alleviates high-fat diet-induced atherosclerosis in ApoE-/- mice through modulation of ACADM.

Atherosclerosis (AS) is a chronic vascular pathology and a leading cause of global cardiovascular morbidity and mortality, with dysregulated lipid metabolism acting as a primary driver of its progression. 5-Methoxyflavone (5-MF), a natural flavonoid, exhibits known lipid-modulating and antioxidant properties; however, its specific atheroprotective potential remains elusive. This study investigated the therapeutic efficacy and underlying metabolic mechanisms of 5-MF against AS. We employed a high-fat diet (HFD)-fed ApoE-/- mouse model alongside a cholesterol-induced vascular smooth muscle cell

SAT1 promotes prostate cancer progression and decreases enzalutamide sensitivity by enhancing spermine catabolism.

Prostate cancer is a major contributor to cancer-related mortality in men, and enzalutamide is a primary treatment option for advanced prostate cancer. Polyamines are ubiquitous amine molecules involved in tumorigenesis, cancer development, and drug resistance. This study aims to explore how spermidine/ spermine N1-acetyltransferase 1 (SAT1)-mediated polyamine metabolism influences prostate cancer progression and enzalutamide sensitivity. Through comprehensive bioinformatics screening, SAT1 was identified as a key regulator negatively associated with spermine abundance in prostate cancer. Anal

CRISPR/Cas9-mediated claudin-2 knockout in HCT116 cells reveals its key role in colorectal cancer progression.

Colorectal cancer (CRC) progression involves complex mechanisms of invasion and metastasis. Claudin-2 (CLDN2), a tight junction protein, has emerged as a key regulator paracellular permeability and its dysregulation is implicated in chronic inflammatory diseases and cancer. The present study aimed to determine the mechanisms by which CLDN2 deletion affects genes associated with motility and invasion of colon cancer cells. CRISPR/Cas9 was used to knock out CLDN2 in HCT116 cells. Subsequently, gene expression was analyzed using reverse transcription-quantitative PCR and migratory capacity was as

RHOBTB2 enhances cell proliferation of acute myeloid leukemia by modulating Hippo-YAP1 signaling and dependent of KLHL13.

Acute myeloid leukemia (AML) is a heterogeneous hematologic malignancy associated with poor clinical outcomes. RHOBTB2, an atypical member of the Rho-GTPase family, contains a conserved GTPase domain at its N-terminus. While previous studies have implicated RHOBTB2 in AML progression, its underlying mechanisms remain inadequately defined. In this study, bioinformatics analyses revealed that RHOBTB2 is markedly upregulated in AML and correlates with unfavorable prognosis. To elucidate its functional role, we overexpressed or silenced RHOBTB2 in human AML cell lines KG-1 and MOLM-13. Functional

Crosstalk of NPY and TGFβ orchestrates the signaling to facilitate perineural invasion of oral squamous cell carcinoma.

BACKGROUND: Perineural invasion (PNI) frequently occurs in oral squamous cell carcinoma (OSCC) and predicts poor prognosis. Although PNI is increasingly recognised as a process driven by tumour-nerve crosstalk, the underlying molecular mechanisms remain unclear. We investigated the role of sympathetic nerve-derived neuropeptide Y (NPY) and its receptor NPY1R in OSCC PNI. METHODS: NPY/NPY1R expression was assessed in human OSCC tissues by immunostaining, qPCR, and TCGA data analysis. Functional studies using Cal27 and SCC9 cells included migration, invasion, and sphere assays. The causal role

Apatinib triggers ferroptosis in gastric cancer via HDAC1/HIF1α/CA9 signaling axis.

Gastric cancer (GC) poses significant therapeutic challenges due to frequent late-stage diagnosis and limited treatment efficacy. Although Apatinib demonstrates clinical benefits in GC, acquired resistance remains problematic. Ferroptosis induction represents a promising strategy to overcome such resistance. Integrated computational target prediction (SwissTargetPrediction) and ferroptosis suppressor gene screening (FerrDb) identified carbonic anhydrase IX (CA9) as Apatinib's putative target. Functional validation employed EdU, Transwell, sphere-formation, and flow cytometry assays. Ferroptosi

CD36 Inhibits Triple-Negative Breast Cancer Progression by Transcriptionally Upregulating Caveolin-1 and Promoting Lipid-Reactive Oxygen Species-Related Ferroptosis.

Triple-negative breast cancer (TNBC) is an aggressive subtype with limited therapeutic options and poor prognosis. Cluster of differentiation 36 (CD36), a fatty acid transporter, plays controversial roles in tumor progression. Here, we report a tumor-suppressive function of CD36 in TNBC. Analysis of The Cancer Genome Atlas and Gene Expression Omnibus databases, along with validation in clinical samples, revealed that CD36 expression was significantly downregulated in TNBC tissues, and its low expression correlated with advanced disease stage and poorer patient prognosis. TPI1 promotes p53 ubiquitination in bladder cancer by recruiting AKT to enhance MDM2 phosphorylation.

Bladder cancer (BCa) is an aggressive malignancy with limited effective treatment options, and its poor outcomes largely result from delayed detection and therapeutic resistance. Triosephosphate isomerase 1 (TPI1) has been associated with tumor progression in various cancers, but its specific function in BCa remains poorly characterized. This study evaluated cancer-related markers and identified glycolysis as a key factor negatively impacting survival in BCa. Additionally, TPI1 was recognized as a potential prognostic marker, with its expression significantly elevated in BCa tissues compared t
