'Tis better to receive than to give: the relative failure of the African American community to provide organs for transplantation.

There is a serious, continuous and increasing shortfall of organs, especially kidneys, for the purpose of transplantation. This shortfall is especially remarkable in African American populations. Because the incidence of hypertension (HTN) and associated end-stage renal diseases (ESRD) is 17 times greater in African Americans, this minority group, which comprises only 12% of the U.S. population, represents 34% of the dialysis population and 30% of the national kidney waiting list. Furthermore, while black individuals comprise 22-24% of kidney recipients, they comprise only 8-11% of donors. Bec

The immune system is a key factor in the etiology of psychosocial disease.

The immune system is proposed as the key to understanding the etiology and treatment of psychosocial disease. There is a dense communication network between the immune system and the central nervous system (CNS). Immune cell cytokines, via direct action on the CNS, induce fever, alter sleep, pain perception and pituitary hormone release and reduce appetite and activity in animals. Interleukin-2 and interferon given to humans result in global behavioral and cognitive pathology. Activation of the immune system by pathogens produces global cognitive and behavioral pathology also. Recently, contro

Three-dimensional observation of the rat endocrine pancreas by a scanning electron microscope.

A three-dimensional observation was performed on chemically digested rat pancreata and vascular corrosion casts of it with a scanning electron microscope. The Langerhans islets were sporadically situated in the exocrine tissue and were surrounded by the fibrous capsule, which was so tough that it could not be chemically digested easily. This capsule was considered to play different roles in the collection of the endocrine cells in a functional unit, in the prevention of secreted hormones spreading into the adjacent exocrine and stromal tissue, in the support of the microvascular structure, and

Identification of osteoarthritis-associated chondrocyte subpopulations and key gene-regulating drugs based on multi-omics analysis.

The mechanism by which chondrocytes respond to mechanical stress in joints significantly affects the balance and function of cartilage. This study aims to characterize osteoarthritis-associated chondrocyte subpopulations and key gene targets for regulatory drugs. To begin, single-cell and transcriptome datasets were obtained from the Gene Expression Omnibus (GEO) database. Cell communication and pseudo-temporal analysis, as well as High-dimensional Weighted Gene Co-expression Network Analysis (hdWGCNA), were conducted on the single-cell data to identify key chondrocyte subtypes and module gene

Disrupting the balance: how acne duration impacts skin microbiota assembly processes.

Growing interest in the impact of microbial balance on health has driven studies on the ecological processes shaping the skin microbiota. Skin diseases, which alter the skin's local environment, can disrupt the microbial structure and interact with the disease itself. However, research on microbial assembly in diseased skin remains limited. In this study, we applied ecological models to characterize the processes shaping the skin microbiota in acne patients, considering the impact of disease duration on both skin pores and surfaces using bacterial amplicon sequencing. Our results revealed a si

Interspecies metabolite transfer fuels the methionine metabolism of Fusobacterium nucleatum to stimulate volatile methyl mercaptan production.

The major oral odor compound methyl mercaptan (CH3SH) is strongly associated with halitosis and periodontitis. CH3SH production stems from the metabolism of polymicrobial communities in periodontal pockets and on the tongue dorsum. However, understanding of CH3SH-producing oral bacteria and their interactions is limited. This study aimed to investigate CH3SH production by major oral bacteria and the impact of interspecies interactions on its generation. Using a newly constructed large-volume anaerobic noncontact coculture system, Fusobacterium nucleatum was found to be a potent producer of CH3

Barriers to Buprenorphine Dispensing by Medicaid-Participating Community Retail Pharmacies.

IMPORTANCE: Controlled substances have regulatory requirements under the US Federal Controlled Substance Act that must be met before pharmacies can stock and dispense them. However, emerging evidence suggests there are pharmacy-level barriers in access to buprenorphine for treatment for opioid use disorder even among pharmacies that dispense other opioids. OBJECTIVE: To estimate the proportion of Medicaid-participating community retail pharmacies that dispense buprenorphine, out of Medicaid-participating community retail pharmacies that dispense other opioids and assess if the proportion disp

What motivated men to start PrEP? A cross-section of men starting PrEP in Buffalo city municipality, South Africa.

BACKGROUND: Compared to women, South African men are less likely to know their HIV status (78% vs. 89%), have suppressed viral loads (82% vs. 90%), or access HIV prevention services. To achieve epidemic control where heterosexual sexual behavior drives transmission, interventions to improve the uptake of HIV testing services (HTS) and prevention services must also target cis-gendered, heterosexual men. There is limited understanding of these men's needs and wants with regards to accessing pre-exposure prophylaxis (PrEP). METHODS: Adult men (≥ 18 years) from a peri-urban community in Buffalo C

Investigation and analysis of mental health status of the older adult in western rural areas.

BACKGROUND: The mental health of the older adult in western rural areas is an area of growing concern. Mental health, as defined by the World Health Organization, encompasses emotional, psychological, and social well-being, affecting how individuals think, feel, and act. In this study, we operationalize mental health through four validated dimensions: depression (emotional well-being), anxiety (psychological stability), loneliness (social connectedness), and life satisfaction (overall subjective well-being), which together provide a comprehensive assessment of mental health status. Engineering acid-sensing ion channel 1a with disrupted desensitization: A computer-guided approach.

Human Acid-Sensing Ion Channel 1a (hASIC1a) is a complex, multi-state membrane macromolecule whose rapid desensitization limits its application as a natural biosensor. To manipulate these structural dynamics, we designed and applied an integrative computational approach, combining protein residue network analysis and a machine learning-based functional site model, to identify functionally important positions governing hASIC1a desensitization. Focusing on structural communication regions, we pinpointed novel regulatory positions modulating the channel's conformational dynamics involving the des

The Brain-Gut-Microbiome Axis: Exploring the Link Between Depression and Irritable Bowel Syndrome.

The brain-gut-microbiome system is an integrated communication network linking the central nervous system, enteric nervous system, immune pathways, and intestinal microbiota. Increasing evidence indicates that dysregulation of this bidirectional signaling system contributes to both gastrointestinal and psychiatric disorders. Irritable bowel syndrome (IBS), a disorder of gut-brain interaction, and major depressive disorder (MDD) frequently coexist and share multiple biological and environmental risk factors. Advances in microbiome science, neuroimaging, and multi-omics technologies have reveale

Plasmid-Host Networks Reveal Persistent Mobile Elements Across Wastewater Treatment.

Plasmids drive horizontal gene transfer and antimicrobial resistance spread in water resource recovery facilities, yet their in situ dynamics in complex microbial communities remain poorly characterized. We apply Hi-C metagenomics to resolve plasmid-host associations across influent, activated sludge, and effluent at three facilities. We identify 944 plasmid clusters with facility- and stage-specific distributions. Plasmid host range narrows from influent to effluent, indicating that treatment selectively constrains plasmid-host associations. Persistent plasmid clusters with detectable mobilit
