Congenital heart disease (CHD) is the most prevalent human birth defect and remains a leading cause of mortality in childhood. Although advancements in surgical and medical interventions have significantly reduced mortality rates among infants with critical CHDs, many survivors experience substantial cardiac and extracardiac comorbidities that affect their quality of life. The etiology of CHD is multifactorial, involving both genetic and environmental factors, yet a definitive cause remains unidentified in many cases. Recent advancements in genetic testing technologies have improved our abilit

Efficient management for electrochemical reactions within Pt/C electrodes, specifically the oxygen reduction reaction (ORR) and methanol oxidation reactions (MOR), is critical to the performance and long-life stability of direct methanol fuel cells (DMFCs). Optimizing the hierarchical macro/mesoscale structures of Pt/C electrodes plays a decisive role in regulating the mass transport pathways and electrochemical reactions. In this work, bead-like Pt/C-ionomer hybrid porous nanofibrous networks are constructed via electrospinning. Ascribing to the hierarchical architecture consisting of continu

Non-obstructive azoospermia (NOA) is a severe form of male infertility, yet its underlying molecular mechanisms remain poorly understood. This study aimed to identify key regulatory non-coding RNAs (ncRNAs) and hub genes associated with NOA through an integrative bioinformatics approach. Using microarray analysis, we examined 4956 ncRNAs and identified 29 differentially expressed ncRNAs (14 upregulated, 15 downregulated) in NOA compared to healthy individuals. Co-expression analysis revealed significant interactions between lncRNAs, miRNAs, and mRNAs, predicting 31 target mRNAs within the regu

Cell surface receptors play a crucial role in mediating cellular communication and precisely regulating cellular functions. Dysregulation of these receptors is closely associated with various diseases. To enable precise and dynamic receptor control, we developed a gas-responsive aptameric binder based on a chemically masked configuration, enabling stimulus-triggered reactivation. Specifically, the receptor-targeting activity of the aptamer is sterically blocked via azide modifications at critical residues, with selective restoration of function upon exposure to hydrogen sulfide (H2S), enabling

Myelin, along with the oligodendrocytes (OLs) that produce it, is essential for proper central nervous system (CNS) function in vertebrates. Although the accurate targeting of myelin to axons and its maintenance are critical for CNS performance, the molecular pathways that regulate these processes remain poorly understood. Through a combination of zebrafish genetics, mouse models, and primary OL cultures, we find that FBXW7, a recognition subunit of an E3 ubiquitin ligase complex, is a regulator of adult myelination in the CNS. Loss of Fbxw7 in myelinating OLs results in increased myelin sheat

Atopic dermatitis (AD) and allergic asthma (AA) are interconnected allergic diseases that frequently co-occur in early childhood, representing a critical sequence in the "atopic march". The rising global prevalence of these conditions, driven by environmental factors and epigenetic changes, underscores an urgent clinical need to understand their shared pathogenesis and disrupt disease progression. Evidence implicates multifaceted mechanisms in AD-to-AA progression, including skin barrier dysfunction, type 2 inflammation, microbiome dysbiosis, and systemic immune priming. These pathways not onl

N-methyl-d-aspartate (NMDA) receptor-mediated autoimmune encephalitis (NMDAR-AE) is the most common cause of autoimmune encephalitis, especially in children and young adults. The disorder is caused by antibodies directed against the GluN1 protein, an obligatory constituent of NMDA receptors, which are key signaling molecules in brain development, learning and memory, and executive function. The manuscript by Zhou et al. offers key insights into aberrant development of cortical pathways that may underly persistent sensorimotor deficits associated with this encephalitis in a newly generated mous

Chemosensory dysfunction, defined as an altered or lost taste and smell, is a prevalent side effect of cancer treatment, with 93% of patients complaining of taste and 60% complaining of changes in smell. Despite their impact, it is an underrecognized symptom, impairing nutritional intake, quality of life, and treatment outcomes. Surprisingly, taste and smell changes are rarely assessed or addressed in oncology care. This commentary highlights the educational gap faced by clinicians and reports the results of a pilot educational intervention consisting of a 15-min podcast. The results indicate

The mammalian inner ear exhibits a striking dichotomy in regenerative capacity: while vestibular hair cells retain limited ability to regenerate, cochlear hair cells permanently lose this potential after birth. Exploiting this disparity, we employed single-cell comparative transcriptomics combined with lineage tracing to identify c-Fos as a critical early responder selectively activated in the vestibular epithelium following ototoxic injury. Transcriptomic and chromatin profiling revealed that c-Fos initiates regenerative reprogramming by transcriptionally priming Atoh1, the master regulator o

Neutrophil-fibroblast crosstalk drives inflammatory pathology across organ systems through both shared and tissue-specific mechanisms. This review synthesizes evidence from skin, lung, gut, cardiovascular, joint, sinus, and oral diseases, revealing conserved molecular pathways where fibroblasts secrete chemokines (CXCL1/8/12) to recruit neutrophils, which, in turn, release neutrophil extracellular traps (NETs), elastase, and cytokines to modulate fibroblast function. Additionally, we identify critical tissue-specific differences, including the predominance of IL-36 signaling in COPD, IL-17-car

The central nervous system (CNS) governs critical physiological processes, and its dysregulation drives severe pathologies, particularly glioma, a life-threatening malignancy with limited therapeutic options. β-elemene (ELE), the bioactive compound derived from Curcuma wenyujin, exhibits potent anti-glioma activity as both a monotherapy and in synergy with chemo- or radiotherapy. Beyond glioma, ELE demonstrates therapeutic versatility across CNS disorders, including traumatic brain injury, ischemic stroke, spinal cord injury, neuropathic pain, experimental autoimmune encephalomyelitis, and obe

Electronic Health Records (EHRs) are a leading source of epidemiological data, but often lack information on a patient's disability status. This gap hampers our ability to analyse the full scope of health inequities faced by people with disabilities. Current approaches to identify disability within EHRs have limitations because of inadequate proxies for disability or issues linking data sources. Machine learning (ML) offer unprecedented opportunities to create disability markers within EHRs, such as through unsupervised learning to classify disability groups and Natural Language Processing to

The neural control of breathing is both dynamic and essential, ensuring life-sustaining gas exchange while protecting the respiratory system from harm. Peripheral neurons innervating the respiratory tract exhibit remarkable diversity, continuously relaying sensory feedback to the brain to regulate breathing, trigger protective reflexes such as coughing and sickness behaviors, and even influence emotional states. Understanding this airway-brain axis is especially critical given the increasing global burden of respiratory diseases, as it holds implications for both human health and broader brain

Chlamydia trachomatis is an obligate intracellular pathogen and the leading cause of bacterial sexually transmitted infections and infectious blindness worldwide. All Chlamydia species share a unique biphasic developmental cycle, alternating between infectious elementary bodies (EBs) and replicative reticulate bodies (RBs). The pathogenesis of C. trachomatis is driven by a sophisticated arsenal of adhesins, conventional type III secretion system effector proteins, and inclusion membrane proteins that subvert host cellular processes to establish infection and promote survival. In this review, w

Persistent psychological stress can affect immune homeostasis and is a key factor in the development of depression. Many efforts are focused on the identifcation of pathways that link the immune system and mood disorders. Here, we found that psychological stress caused an increase in the frequency of brain-associated neutrophils and the level of neutrophil-specific antigen CD177 on peripheral neutrophils in male mice. Upregulated levels of blood CD177 are associated with depression in humans. Neutrophil depletion or Cd177 deficiency protected mice from stress-induced behavioral deficits. Impor

Podocytes are highly specialized epithelial cells that surround the capillaries of the glomeruli in the kidney. Together with the glomerular endothelial cells, these postmitotic cells are responsible for regulating filtrate from the circulating blood with their organized network of interdigitating foot processes that wrap around the glomerular basement membrane. Although podocyte injury and subsequent loss is the hallmark of many glomerular diseases, recent evidence suggests that the cell-cell communication between podocytes and other glomerular and nonglomerular cells is critical for the deve
