Growth pattern and sister chromatid exchanges of bone marrow cells in acute lymphoblastic leukaemia.

An in vitro BrDU chromosome labelling technique has been used for analysing the growth pattern and incidence of sister chromatid exchanges (SCE) in the bone marrow cells of twelve patients with acute lymphoblastic leukaemia (ALL): six with only diploid cells and six with aneuploid cells, containing various numerical and structural chromosome abnormalities, with or without concurrent diploid cells. There was no clear-cut karyotypic relevance to the growth pattern or SCE incidence in such cells. An increased SCE incidence was observed during or immediately after treatment with certain chemothera

The pathogenesis of autoimmunity in New Zealand mice.

In the past few years there has been an explosion of information in the area of NZ mouse research. This has reflected a general increase in knowledge and technology in cellular immunology and lymphocyte biology. Continuing research in many specific areas promises to expand this increasing data base. The numerous abnormalities of CMI in NZ mice chronicled in the past are currently being analyzed in terms of cell synergism rather than as simple effector mechanisms. Such analysis is indicating more selective rather than general impairment of CMI in aging NZ mice. Similarly, the allogeneic H-2 ide

Untransformed xeroderma pigmentosum cells are not hypersensitive to sister-chromatid exchange production by ethyl methanesulphonate--implications for the use of transformed cell lines and for the mechanism by which SCE arise.

A transformed cell line, XP12RO(SV40) has previously been found to be hypersensitive to several chemical mutagens including ethyl methanesulphonate, as judged by sister-chromatid exchange (SCE) formation. The hypersensitivity of this line has been confirmed for SCE formation and extended to cell survival. Measurements of SCE formation and survival show, however, that the hypersensitivity of the XP12RO(SV40) cell line is not typical of the primary strain (XP12RO) from which the transformed line was derived, nor it is typical of other primary strains also belonging to complementation group A (XP

Increased sister chromatid exchange in bone marrow and blood cells from Bloom's syndrome.

Bone-marrow cells from a patient with Bloom's syndrome cultured for 48 h in the presence of BudR exhibited a striking increase in the number of sister chromatid exchanges (SCEs) in comparison to that in the marrow cells of a patient with treated polycythemia vera (PV). Thus, it appears that an increased incidence of SCE in Bloom's syndrome occurs in various differentiated types of cells, not just blood lymphocytes, and constitutes the syndrome's most characteristic cytogenetic feature. In contrast, the incidence of SCE was not increased in marrow cells and lymphocytes of the particular PV pati

Histocompatibility-linked genetic control of disease susceptibility. Murine lymphocytic choriomeningitis virus infection.

Acute necrotizing inflammatory disease after intracerebral injection of LCM virus is largely dependent on the host immune response to the virus and is controlled, in part, by a dominant gene which is closely linked to the H-2 locus. The F(1) hybrid (H-2(q/k)) from mating a susceptible SWR/J mouse (H-2(q/q)) to a resistant C3H/HeJ mouse (H-2(k/k)) is susceptible to LCM virus disease. When such hybrids (H-2(q/k)) are backcrossed to susceptible parents (H-2(q/q)), all F(2) offspring (H-2(q/q), H-2(q/k)) are highly susceptible. In contrast, hybrid (H-2(q/k)) backcross to resistant parents (H-2(k/k

Antigenic variation and protective immunity in Plasmodium knowlesi malaria.

Rhesus monkeys were immunized with defined strains and variants of Plasmodium knowlesi and their immunity on challenge was correlated with serum levels of schizont agglutinins and specific inhibitory antibody assayed by in vitro parasite culture. The results indicate that the inhibitory antibody provides a consistent index of immune status and probably represents the protective antibody which initiates specific immune reactions in vivo. The relationship between inhibitory and schizont agglutinating antibodies is discussed. Inhibitory antibody is predominantly specific for those variants which

A Batch-Aware, Subject-Grouped Pipeline to Prioritize HIIT-Associated DNA Methylation Candidates with Cross-Cohort Evaluation in Skeletal Muscle.

Skeletal muscle DNA methylation is closely associated with molecular adaptation to exercise training, but longitudinal studies in this area are constrained by the difficulty of repeated muscle biopsy sampling and extended follow-up. The resulting datasets are typically small, with repeated measurements within individuals and complex sub-cohort or batch structure. To address these limitations, we developed a methylation candidate-discovery pipeline that integrates batch-aware statistical screening, subject-grouped nested cross-validation, and a binary/multi-class/temporal three-axis consensus s

Seasonal influenza vaccination is the strongest correlate of cross-reactive antibody responses in migratory bird handlers.

UNLABELLED: Avian species are reservoirs of influenza A viruses and could harbor viruses with significant pandemic potential. We examined the antibody and cellular immune responses to influenza A viruses in field or laboratory workers with a spectrum of occupational exposure to avian species for evidence of zoonotic infections. We measured the seroprevalence and T cell responses among 95 individuals with various types and degrees of prior field or laboratory occupational exposure to wild North American avian species using whole blood samples collected in 2010. Plasma samples were tested using

Comparative Proteomic Quantification Reveals Novel Longevity Regulators.

Proteomics is a powerful tool for profiling the protein landscape underlying cellular and physiological processes. Advances in mass spectrometry-based methods, such as tandem mass tags (TMT) isobaric labeling-based proteomics at both MS2 and MS3, label-free quantification (LFQ) with data-dependent acquisition (DDA) or data-independent acquisition (DIA)-based proteomics, and stable isotope labeling with amino acid (SILAC) spike-in-assisted proteomics, enable quantitative proteome comparisons across biological conditions. These five quantitative proteomic methods offer distinct advantages in cov

STR typing from punched FFPE tissues: resolving suspected contamination and fetal-maternal mixture in two cases.

The analysis of mixed or contaminated biological materials represents a persistent challenge in both forensic genetics and diagnostic pathology. This study presents two casework applications demonstrating the analytical value of a targeted FFPE punching strategy coupled with STR genotyping for resolving complex scenarios. In the first case, an unexpected focus of high-grade urothelial carcinoma within a prostatic surgical specimen raised concerns regarding cross-sample contamination. STR profiling of microdissected punches-respectively containing the carcinoma and benign prostatic tissue-yield

Evolving skin cancer therapeutics: molecular and genetic drivers, immunotherapy challenges, and advances in nanogel-based nanomedicine.

Skin cancer is among the most common malignancies worldwide, with a rising global incidence. Cancer develops through complex molecular and genetic changes. Current treatments, such as surgery, chemotherapy, targeted therapy, radiation, and immunotherapy, offer hope. However, challenges like immune evasion, drug resistance, and systemic toxicities prevent long-term success. Nanotechnology-based delivery systems may offer a potential solution to these challenges. Cross-linked polymeric nanogel networks are unique nanocarriers with high drug-loading capacity, biocompatibility, stimuli-responsive

GenoME: a MoE-based generative model for individualized, multimodal prediction and perturbation of genomic profiles.

The non-coding genome operates through a complex, multiscale regulatory system where regulated gene expressions are closely associated with cell-type-specific histone modifications, transcription factor binding, and 3D conformation. Developing computational models that can integrate these patterns to predict and interpret the regulatory system remains challenging. Here, we present GenoME, a Mixture of Experts (MoE)-based generative model that uses DNA sequence and cell-type-specific ATAC-seq signals to predict a unified genomic profiles encompassing epigenomics, transcriptomics, and chromatin

Metagenomic analysis of antimicrobial resistance, virulence, and mobile genetic elements in the gut microbiota of Caprinae species.

The livestock gut microbiota serves as a reservoir for antimicrobial resistance (AMR), yet Caprinae species remain understudied. Here, we present a large-scale metagenomic analysis of 779 gut samples from Caprinae animals, primarily originating from China (95.38%), including Capra hircus (79.85%) and Ovis aries (17.33%). We reconstruct 17,023 metagenome-assembled genomes (MAGs), and identify 2,440 antimicrobial resistance genes (ARGs) and 5,401 virulence factor genes (VFGs). Escherichia coli represents a major host for both. Correlation analyses between ARGs, VFGs, and mobile genetic elements

Artificial selection for resistance to copper and off-target physiological and behavioral effects in Drosophila melanogaster.

Pollution resulting from mining, industry, and agriculture has played an important role in shaping the evolution of diverse organisms. Heavy metal contamination is of particular concern due to the health effects for humans and cross-tolerance effects that may influence pesticide resistance. We used a replicated artificial selection approach to examine the response to copper selection in Drosophila melanogaster collected from a retired mine and active fruit orchard. We tracked shifts in resistance to the target trait, adult copper resistance, as well as off-target effects on cadmium and lead re

Co-Cultivation of Cross-Kingdom Microorganisms Effectively Triggers the Production of Tryptophol and Its Heterologous Expression in E. coli.

Genome sequencing has revealed that microorganisms have the potential to produce many more natural products than previously thought; the challenge is to establish efficient ways to "wake up" those "sleeping" biosynthetic pathways or genes, which are undoubtedly expressed in nature under specific conditions that are not normally reproduced in the laboratory. To activate these cryptic natural products, co-cultivation of cross-kingdom microorganisms between Candida albicans and Streptomyces longshengensis was performed in this study. A novel peak generated through co-culture was isolated and anal

MethyAnno: An Interpretable Automated Annotation Method Leveraging Multi-Scale Information and Metric Learning Framework for scDNAm Data.

Single-cell DNA methylation (scDNAm) sequencing provides unique insights into epigenetic heterogeneity and cell-specific regulatory landscapes. However, accurate cell type annotation for scDNAm data remains challenging, as the distinct data distribution of scDNAm hinders the adaptation of annotation methods from other omics, and specialized annotation tools for scDNAm are currently lacking. Here, MethyAnno is proposed as an interpretable deep metric learning framework that leverages multi-scale information for accurate cell type annotation of scDNAm data. Additionally, MethyAnno enables genera

Functional genes with their expression and horizontal gene transfer drive microbial interactions in anammox systems: Critical review and potential applications.

The anaerobic ammonium oxidation (anammox) process, a low-carbon and energy-efficient biological nitrogen removal technology, is crucial for sustainable wastewater treatment and energy self-sufficiency. However, its performance stability is influenced by complex microbial interactions, and the gene-level mechanisms, particularly horizontal gene transfer (HGT), remain underexplored. This review comprehensively examines the interactions between anammox bacteria and their syntrophic partners, focusing on the functional genes involved in substrate degradation, electron transfer, cofactor biosynthe

A domestication-selected CIPK-MYB-HAK module improves salt tolerance in modern maize.

Domestication has profoundly reshaped the maize genome, yet its impact on abiotic stress resilience remains largely unresolved. Here we show that modern maize exhibits markedly greater salt tolerance than its wild progenitor, teosinte. Using a maize-teosinte (W22-T8759) recombinant inbred line population, we identify the transcriptional repressor MYB28 as a major genetic determinant underlying this divergence. MYB28 negatively regulates salt tolerance by impairing shoot Na+ exclusion. In modern maize (W22), however, salt stress activates CIPK20, which phosphorylates MYB28 to relieve its repres
