B7-CD28 co-stimulation modulates central tolerance via thymic clonal deletion and Treg generation through distinct mechanisms.

The molecular and cellular mechanisms mediating thymic central tolerance and prevention of autoimmunity are not fully understood. Here we show that B7-CD28 co-stimulation and B7 expression by specific antigen-presenting cell (APC) types are required for clonal deletion and for regulatory T (Treg) cell generation from endogenous tissue-restricted antigen (TRA)-specific thymocytes. While B7-CD28 interaction is required for both clonal deletion and Treg induction, these two processes differ in their CD28 signaling requirements and in their dependence on B7-expressing dendritic cells, B cells, and

Mass cytometry reveals cellular fingerprint associated with IgE+ peanut tolerance and allergy in early life.

IgE-mediated peanut allergic is common, often serious, and usually lifelong. Not all individuals who produce peanut-specific IgE will react upon consumption of peanut and can eat the food without adverse reactions, known as sensitized tolerance. Here, we employ high-dimensional mass cytometry to define the circulating immune cell signatures associated with sensitized tolerance and clinical allergy to peanut in the first year of life. Key features of clinical peanut allergic are increased frequency of activated B cells (CD19hiHLADRhi), overproduction of TNFα and increased frequency of peanut-sp

Pathogenesis of thyroid autoimmune disease: the role of cellular mechanisms.

Hashimoto's thyroiditis (HT) and Graves' disease (GD) are two very common organ-specific autoimmune diseases which are characterized by circulating antibodies and lymphocyte infiltration. Although humoral and cellular mechanisms have been classically considered separately in the pathogenesis of autoimmune thyroid diseases (AITD), recent research suggests a close reciprocal relationship between these two immune pathways. Several B- and T-cell activation pathways through antigen-presenting cells (APCs) and cytokine production lead to specific differentiation of T helper (Th) and T regulatory (Tr

Unilateral Pigmented Extramammary Paget's Disease of the Axilla Associated with a Benign Mole: A Case Study and a Review of Literature.

Pigmented extramammary Paget's disease (PEMPD) is an uncommon intraepithelial adenocarcinoma and a rare variant of Paget's disease affecting skin that is rich in apocrine sweat glands such as the axilla, perianal region and vulva. It most commonly occurs in postmenopausal women and presents as a superficial pigmented scaly macule, mimicking a melanocytic lesion. The histological presentation is adenocarcinoma in situ with an increased number of melanocytes scattered between the Paget's cells. Therefore, PEMPD may be misdiagnosed as a melanocytic tumour both clinically and histologically. The t

HIV target cells in Schistosoma haematobium-infected female genital mucosa.

The parasite Schistosoma haematobium frequently causes genital lesions in women and could increase the risk of human immunodeficiency virus (HIV) transmission. This study quantifies the HIV target cells in schistosome-infected female genital mucosa. Cervicovaginal biopsies with and without schistosomiasis were immunostained for quantification of CD4(+) T lymphocytes (CD3, CD8), macrophages (CD68), and dendritic Langerhans cells (S100 protein). We found significantly higher densities of genital mucosal CD4(+) T lymphocytes and macrophages surrounding schistosome ova compared with cervicovaginal

Potential factors induced by filoviruses that lead to immune supression.

The filoviruses, Ebola (EBOV) and Marburg (MARV), are among the deadliest of human pathogens, causing acute diseases typified by rapidly fatal hemorrhagic fevers. Upon filoviral infection, innate immune cells become paralyzed and lose the capacity to properly co-stimulate and activate filovirus-specific, T-cell responses. Deleterious inflammation and upregulation of co-inhibitory molecules expressed by monocytic lineage cells (e.g., dendritic cells) and their co-inhibitory receptors on T- and B-cells may lead to incomplete humoral and T-cell immunity, anergy, exhaustion, apoptosis, and subsequ

[Pigmented Paget's disease of the male nipple].

BACKGROUND: Mammary Paget's disease unfrequently occurs in males, and may be pigmented in rare instances. Differential diagnosis with malignant melanoma relies on immunohistochemical studies. CASE REPORT: A case of Paget's disease of the nipple in a 76 year-old male is reported, clinically mimicking a malignant melanoma because of massive pigmentation. Histologically, large Paget's clear cells were intermingled with numerous melanin-rich dendritic melanocytes. An underlying ductal carcinoma was found. After differential immunohistochemical staining, diagnosis of Paget's disease could be unequ

Germinal center B cell apoptosis requires both caspase and cathepsin activity.

Follicular dendritic cells (FDCs) select B cells during germinal center (GC) reactions. The B cells that are able to bind to the FDCs receive a signal that leads to the termination of endonuclease activity in the nuclei of those B cells. This signal must be in addition to the signals transferred through the cross-linkage of the B cell receptors and signals resulting from the interactions of the adhesion molecules lymphocyte function-associated Ag-1 and very late Ag-4 with ICAM-1 and VCAM-1, respectively. In this report, we present evidence that the FDCs silence all apoptotic processes in GC B

Triple-Coupling Regulation Enables Dendrite-Free Subzero Aqueous Zn-Ion Batteries.

Aqueous Zn-ion batteries (AZIBs) suffer severe degradation at subzero temperatures due to electrolyte freezing, sluggish Zn2+ kinetics, and dendrite-induced interfacial instability. Herein, a triple-coupling regulation strategy integrating solvation structure, interfacial chemistry, and crystallographic orientation through a formamide-ethanol deep eutectic electrolyte (ZEFW2) was developed. Molecular dynamics simulations and spectroscopic analyses reveal that ZEFW2 redistributes the primary Zn2+ solvation environment, reducing H2O and acetate coordination while introducing formamide coordinati

Synchronously Tuning Crystal Facets and Constructing a Multifunctional Interface toward a High-Performance Zn Anode.

Aqueous zinc-ion batteries (AZIBs) face challenges such as dendrite growth and byproduct accumulation in practical applications. Herein, an innovative strategy of alkali corrosion is utilized to tune crystal facets of a Zn electrode, selectively weakening the active (101) plane and highlighting the (002) plane. Furthermore, a protective interface composed of coordination polymers and inorganic zinc compounds has been synchronously fabricated, exhibiting remarkable mechanical robustness. The engineered multifunctional protective layer (MPL) also possesses abundant zincophilic sites that can sig

Dendrite-Free Zinc Ion Hybrid Supercapacitors with ZnCl2-Modified Lignocellulose Hydrogel Electrolyte for Modulating Zinc Ion Solvation and Directed Deposition.

Chargeable-dischargeable zinc-ion hybrid supercapacitors (ZHSCs) face significant challenges regarding low coulombic efficiency and short cycle life, primarily due to water-induced side reactions and uncontrolled zinc anode dendrite growth. Herein, a Zn-friendly hydrogel electrolyte is developed using ZnCl2-modified lignocellulose (LC), which effectively facilitates the coordination of Zn2+ with -OH by disrupting the strong hydrogen bonding between the LC chains, exposing more -OH binding sites, and thus modulating the deposition behavior and interfacial chemistry of Zn on the Zn electrode. Loss of X-linked Protocadherin-19 differentially affects the behavior of heterozygous female and hemizygous male mice.

Mutations in the X-linked gene Protocadherin-19 (Pcdh19) cause female-limited epilepsy and mental retardation in humans. Although Pcdh19 is known to be a homophilic cell-cell adhesion molecule, how its mutations bring about female-specific disorders remains elusive. Here, we report the effects of Pcdh19 knockout in mice on their development and behavior. Pcdh19 was expressed in various brain regions including the cerebral cortex and hippocampus. Although Pcdh19-positive cells were evenly distributed in layer V of wild-type cortices, their distribution became a mosaic in Pcdh19 heterozygous fem

Expression of C-type lectin receptors and Toll-like receptors in decidua of patients with unexplained recurrent spontaneous abortion.

In the present study, the mechanisms underlying the pathogenesis of unexplained recurrent spontaneous abortion (URSA) were explored. The protein and mRNA expression of two C-type lectin-like receptors (CLRs), namely dendritic cell-specific intercellular adhesion molecule-3-grabbing non-integrin (DC-SIGN) and mannose receptor (MR), and two Toll-like receptors (TLRs), namely TLR2 and TLR4, in the decidua and dendritic cells (DCs) was compared between URSA patients and normal controls. URSA patients had significantly lower protein and mRNA expression of DC-SIGN and significantly higher expression

Microwave therapy for cutaneous human papilloma virus infection.

BACKGROUND: Human papilloma virus (HPV) infects keratinocytes of the skin and mucous membranes, and is associated with the induction of cutaneous warts and malignancy. Warts can induce significant morbidity and disability but most therapies, including cryotherapy, laser, and radiofrequency devices show low efficacy and induce discomfort through tissue destruction. Microwaves are readily capable of passing through highly keratinised skin to deliver energy and induce heating of the tissue in a highly controllable, uniform manner. OBJECTIVES: To determine the effects of microwave on cutaneous HP

Transient cerebral ischemia increases CA1 pyramidal neuron excitability.

In human and experimental animals, the hippocampal CA1 region is one of the most vulnerable areas of the brain to ischemia. Pyramidal neurons in this region die 2-3 days after transient cerebral ischemia whereas other neurons in the same region remain intact. The mechanisms underlying the selective and delayed neuronal death are unclear. We tested the hypothesis that there is an increase in post-synaptic intrinsic excitability of CA1 pyramidal neurons after ischemia that exacerbates glutamatergic excitotoxicity. We performed whole-cell patch-clamp recordings in brain slices obtained 24 h after

The immunology of Leishmania infection and the implications for vaccine development.

Leishmania parasites are vector-borne protozoal pathogens found in tropical and subtropical regions of both the Old and New World. These parasites can cause visceral or cutaneous disease, and the pathology of the infection is determined by both host immune factors and species/strain differences of the parasite. Dogs are an important reservoir for maintaining the population of Leishmania parasites that can lead to visceral leishmaniasis in humans, and a vaccination approach may be an effective method for reducing the numbers of infected dogs. Resistance to leishmaniasis has been consistently as

Dedifferentiated liposarcoma: a report of nine cases with a peculiar neurallike whorling pattern associated with metaplastic bone formation.

Nine cases of dedifferentiated liposarcoma with both a peculiar neurallike or meningeallike whorling pattern and metaplastic bone formation are reported. The tumors predominated in the retroperitoneum of elderly adults. All nine tumors were resected, and five of seven that were followed-up recurred locally, but none metastasized after a follow-up of 2 to 7 years. Grossly, most of the tumors were huge masses, ranging from 2.5 cm to 60 cm. Histologically, the tumors revealed, in addition to areas of well-differentiated liposarcoma, discrete nodules consisting of hypercellular, spindled to ovoid

Age-related hearing loss begins with gain, not silence: physiological compensation in dorsal cochlear nucleus fusiform cells during auditory aging.

Age-related hearing loss (ARHL) is the most common cause of sensorineural hearing loss. The cochlear nucleus, the first central auditory structure to receive input from the cochlea, has been shown to be disrupted by ARHL. Fusiform cells (FCs), the principal output cell of the dorsal cochlear nucleus (DCN), mature physiologically during hearing onset. Specifically, FCs increase in the rate of action potential (AP) rise and decay, stabilizing by postnatal day 14 (P14) in mice. However, whether FC intrinsic electrophysiological properties or morphological characteristics continue to change throug

Signal Transduction and Transcription Activator 1 in Pan-Cancer: Prognostic and Immunological Value.

STAT1, a key mediator of cellular responses to interferons and cytokines, has not been comprehensively analyzed in pan-cancer studies regarding its role in tumor biology. This study investigated STAT1 expression, its prognostic value, and its correlation with immune status in various types of cancer. We analyzed STAT1 expression, methylation, genomic alterations, survival, immune cell infiltration, and functional pathways using the TCGA, GTEx, GEO, TIMER, CCLE, and The Human Protein Atlas (HPA) databases. STAT1 expression was significantly higher in tumor tissues than in normal tissues and cor

[Role of myeloid cell abnormalities in the pathogenesis of aplastic anemia:a review of research advances].

Aplastic anemia is a disorder characterized by bone marrow failure and peripheral pancytopenia. T cell-mediated injury to hematopoietic stem and progenitor cells (HSPCs) is recognized as a key immunological mechanism underlying its pathogenesis. Recent evidence indicates that myeloid cell abnormalities also contribute to the bone marrow immunopathology of aplastic anemia. Myeloid populations-including dendritic cells, mononuclear phagocytes, myeloid-derived suppressor cells, and mast cells-may disrupt the local bone marrow immune and hematopoietic microenvironment through their involvement in
