Differential expression of the p75 nerve growth factor receptor in glia and neurons of the rat dorsal root ganglia after peripheral nerve transection.

Sympathetic nerve terminals on blood vessels within the dorsal root ganglia sprout after sciatic nerve lesions in the rat. The mechanism underlying this phenomenon is not clear, but might be predicted to involve nerve growth factor or its homologs because these factors are known to trigger collateral sprouting of undamaged sympathetic noradrenergic terminals. We have found that sciatic nerve lesions lead to a decreased expression of neuronal p75, the low-affinity receptor for the neurotrophins, but an increased expression of glial p75 in ipsilateral dorsal root ganglia. Intriguingly, the incre

On the site of transcranial magnetic stimulation of the facial nerve: electrophysiological observations in two patients after transection of the facial nerve during neuroma removal.

The site of stimulation of the facial nerve after transcranial temporo-occipital magnetic stimulation is being controversially discussed, particularly whether the nerve is stimulated in the root exit zone in the cerebellopontine angle or whether stimulation originates within the bony canal of the facial nerve. In two case reports, the neurophysiological findings after the surgical transection of the facial nerve during the extirpation of a large acoustic and a facial nerve neuroma are presented. In both cases, transcranial magnetic stimulation of the facial nerve produced compound muscle actio

TrkB-like immunoreactivity in trigeminal sensory nerve fibers of the rat molar tooth pulp: development and response to nerve injury.

The localization of TrkB, a signal transducing receptor for brain-derived neurotrophic factor and neurotrophin-4, was studied in the rat mandibular molar pulp during development and following nerve injury. Sections were incubated with rabbit polyclonal antiserum against the catalytic part of the TrkB receptor, thus only binding to full-length TrkB receptors, and examined by immunofluorescence microscopy and EM immunocytochemistry. At embryonic day 17, strongly TrkB-positive fibers were located in mandibular nerve trunks, in nerve fibers of deeper mesenchyme in close spatial relation to sites o

In vivo release of glutamate in nucleus tractus solitarii of the rat during hypoxia.

1. An attempt has been made to test the hypothesis that, in the caudal part of nucleus tractus solitarii (NTS) where carotid sinus nerve (CSN) afferents project, L-glutamate (Glut) modulates the hypoxic ventilatory response. 2. Unanaesthetized, peripherally chemodenervated (carotid body denervated; CBD) and sham-operated, freely moving rats were used. During peripheral chemoreceptor stimulation by hypoxia (10% O2 for 30 min) or doxapram (Dox) infusion (2 mg kg-1 (30 min)-1), ventilation was recorded and successively, under the same conditions, the extracellular Glut concentration ([Glut]o) in

Transganglionic neuropeptide Y response to sciatic nerve injury in young and aged rats.

Gracile neuroaxonal dystrophy (NAD) is a hallmark of the aging human and rodent sensory nervous systems which may represent an abnormal transganglionic response to peripheral axonal injury. To examine the structural plasticity of central dorsal root ganglia (DRG)-derived axons in the gracile nucleus, we evaluated the response of the lumbar DRG and their central projections to sciatic nerve injury in young and old rats. In uninjured rats neither the DRG nor its central projections contained histochemical immunoreactivity for neuropeptide Y (NPY). However, within 1 week of sciatic nerve crush or

Inhibition of a cutaneous nociceptive reflex by a noxious visceral stimulus is mediated by spinal cholinergic and descending serotonergic systems in the rat.

The present study examined the spinal pathway and receptors that mediate nocigenic inhibition of the tail-flick (TF) reflex produced by conditioning colorectal distension (CRD). Conditioning CRD (80 mmHg; 30 s) inhibited the TF reflex in all rats studied (n = 29). In 19 rats where intensity-dependent effects of CRD were studied, conditioning CRD in 7 rats facilitated the TF reflex at lesser, non-noxious intensities (mean 7.9 +/- 2.1 mmHg) and inhibited the TF reflex at greater, noxious intensities (40-100 mmHg); conditioning CRD at all intensities tested only inhibited the TF reflex in the oth

Response of facial and rubrospinal neurons to axotomy: changes in mRNA expression for cytoskeletal proteins and GAP-43.

Neurons confined within the mammalian CNS usually do not regenerate after axonal injury, while axonal regeneration is the rule in the PNS. It has been hypothesized that this may be related to differences in the microenvironment of the PNS versus CNS and to differences in the neuronal response to injury. In order to test the latter hypothesis, we compared changes in gene expression after axotomy in two populations of neurons: rat facial motoneurons and rat rubrospinal neurons. In situ hybridization with cDNA probes for the medium and light neurofilament protein revealed a reduced mRNA content i

Heritability of symptoms in an experimental model of neuropathic pain.

Male and female rats underwent transection and ligation of the sciatic and saphenous nerves, and the development of autonomy was monitored. The deafferented animals were then interbred, always selecting males and females that expressed relatively high and, alternatively, relatively low levels of autotomy. Offspring were similarly operated and interbred. By the sixth generation of selective breeding, lines were achieved in which autotomy was consistently high (HA) or consistently low (LA). There was no indication of sex linkage. Thermal and mechanical nocifensive responsiveness co-selected with

Repair of severed peripheral nerves: comparison of the "de Medinaceli" and standard microsuture methods.

The return of function following transection of the rat sciatic nerve has been assessed after repair by either standard microsurgical techniques (i.e., the use of microsutures to coapt the severed ends) or the new repair technique introduced by de Medinaceli and coworkers. The regeneration after transection was compared with that following sciatic nerve crush, i.e., a lesion in which the return of function is near optimal. Return of function was monitored serially using walking track analysis (i.e., the sciatic functional index, which indicates overall functional performance), the ability to s

Safflower yellow alleviates sciatic nerve crush injury in rats: associated with TRPV1-pERK signaling pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Peripheral nerve injury (PNI) clinically resembles the traditional Chinese medical "Bi" and "Blood-Stasis" syndromes. Carthamus tinctorius L. (safflower) is a classic blood-activating and stasis-resolving herb commonly prescribed for PNI, whereas the protective effect of its major active component safflower yellow (SY) against PNI remains unclear. AIM OF THE STUDY: To explore the efficacy and potential mechanisms of SY in ameliorating PNI using a rat sciatic nerve crush (SNC) model. MATERIALS AND METHODS: Rats were randomly assigned to four groups (n=10): Sham

Electromyography After Total Hip Arthroplasty: A Systematic Review of Neuromuscular Alterations and Functional Movement Patterns.

Background: Electromyography (EMG) is increasingly used to characterize neuromuscular alterations after total hip arthroplasty (THA), yet available evidence remains fragmented and inconsistent. This systematic review synthesizes postoperative EMG findings during gait, functional tasks, and static assessments, highlighting clinical implications and future research needs. Methods: Peer-reviewed studies employing surface, needle, or high-density EMG after THA were systematically examined. Extracted variables included activation amplitude, timing (onset, offset, burst duration), co-activation patt

Liver transplantation-related osteoporosis and fragility fractures: pathogenic pathways, risk stratification and novel targeted therapy advances.

Liver transplantation (LT) serves as the definitive treatment for end-stage liver disease. Yet, osteoporosis (OP) and fragility fractures remain frequently overlooked skeletal complications that may impair the long-term prognosis of recipients. This review systematically summarizes recent evidence regarding epidemiological characteristics, multifactorial pathogenic pathways, risk stratification approaches, and advances in targeted interventions for LT-related bone disorders. Skeletal damage distributes across pre-transplant waiting, early post-transplant acute and long-term chronic phases; rap

Motor Neuron Disease with Guillain-Barré Syndrome? Motor Band Sign with Anti-GQ1b Antibodies.

A 79-year-old former marathoner, with memory impairment since age 78, developed increasing stumbling and progressively worsening waddling gait. Three months after gait disturbance onset, she noted mild dysphagia. With declining walking distance and endurance, she presented to our hospital six months after onset, exhibiting frontal signs, Parkinsonism with marked trunk rigidity, and hyperreflexia of the jaw and limbs. L-dopa challenge tests showed no improvement. At seven months post-onset, she had difficulty rising. By nine months, she relied on a walker, and speech disturbance appeared. At 10

Fentanyl-Induced Diaphragmatic Discoordination during Overdose.

UNLABELLED: Synthetic opioids like fentanyl are a leading cause of overdose mortality. Although the hallmark of fentanyl overdose is ventilatory depression, fentanyl also induces tonic activation of skeletal musculature, including the diaphragm, which may advance progression of overdose towards death. While tonicity may further restrict diaphragmatic contractility, phase-specific dysregulation may also reflect a larger state of discoordination in respiratory control. Using urethane-anesthetized mice exposed to fentanyl, we test the hypothesis that fentanyl-induced diaphragm tonicity results fr
