Structural and Functional Analysis of the Recombinant Aureobasidium pullulans Cutinase Variants for Efficient Polycaprolactone Degradation.

Polycaprolactone (PCL) is a biodegradable polyester widely used in industry, but it degrades slowly in the environment. Enzymatic hydrolysis catalyzed by cutinases represents a promising approach for PCL waste utilization. Through screening, we selected the Aureobasidium pullulans VKM 1116 strain capable of degrading PCL. We amplified the ApCUT1 cutinase gene from the A. pullulans VKM 1116 genomic DNA. Heterologous expression in Komagataella phaffii yielded ~60 mg/L of the target protein. The recombinant enzyme exhibited maximum activity at pH 6.0-6.5 and 30-40°C. Using site-directed mutagenes

Carbon Shell-Mediated Electronic Modulation of NiFe Alloy Electrocatalysts for Efficient CO2 Electroreduction.

Developing highly efficient and stable catalysts for electrochemical carbon dioxide reduction reaction (CO2RR) remains a significant challenge, particularly for transition metal-based systems that often suffer from excessive hydrogen evolution and catalyst degradation. In this work, we report a carbon-coated NiFe alloy (NiFe@NC) synthesized via a substrate-anchored pyrolysis strategy, in which the carbon shell serves as an electronic modulator and protective layer. DFT calculations and in situ spectroscopic analysis reveal that the carbon layer induces notable electronic reconstruction at the

Efficient photocatalytic propane oxidative dehydrogenation with ultralow loading of PdAg intermetallic nanoparticle catalyst.

Propane oxidative dehydrogenation (ODH) is a promising route for propylene production; however, the conventional thermal catalysts suffer from carbon deposit induced by harsh reaction conditions at high temperature. Developing a highly efficient light-driven photocatalytic system is a benign environmentally-friendly alternative. Herein, an ultra-low loading PdAg intermetallic compound (0.1 wt% Pd and 0.05 wt% Ag) supported on TiO2 catalyst was employed via a facile photodeposition (PD) method. The introduction of Ag significantly improved the activation of oxygen driven by the unique interface

Bimetallic PtRu alloy nanowires confined on MXene nanosheets for highly efficient methanol electrooxidation.

Herein, we present the oriented synthesis of 1D ultrafine bimetallic PtRu alloy nanowires in situ confined on 2D Ti3C2Tx MXene nanosheets (PtRu NW/MXene) via a convenient and robust solvothermal assembly strategy. Both experimental and theoretical investigations demonstrate that this distinct 1D/2D heterointerface design not only effectively stabilizes the integral hybrid structure through a "line-to-face" contact model, but also significantly ameliorates the electronic structure and catalytic environment of the PtRu component. Accordingly, the newly developed Pt70Ru30 NW/MXene catalyst exhibi

Exploration of highly stable and efficient pseudohalide double perovskite doping schemes for solar cells.

Double perovskites offer a chemically versatile platform for photovoltaic applications, yet the systematic design of pseudohalide doping strategies, including the selection of pseudohalide species, host compositions, and substitution levels, remains insufficiently explored. In this work, a machine-learning-assisted screening workflow combined with first-principles calculations was developed to explore pseudohalide doping schemes for double perovskites. By systematically screening combinations of pseudohalide species, host compositions, and substitution coefficients within a large compositional

EventDiff: A Unified and Efficient Diffusion Model Framework for Event-based Video Frame Interpolation.

Video Frame Interpolation (VFI) is a fundamental yet challenging task in computer vision, particularly under conditions involving large motion, occlusion, and lighting variation. Recent advancements in event cameras have opened up new opportunities for addressing these challenges. While existing event-based VFI methods have succeeded in recovering large and complex motions by leveraging handcrafted intermediate representations such as optical flow, these designs often compromise high-fidelity image reconstruction under subtle motion scenarios due to their reliance on explicit motion modeling.

Efficient Binding Affinity Estimation for Fragment-Based Compounds Using a Separated Topologies Approach.

Fragment-based drug discovery (FBDD) is a widely used strategy in early-stage drug development, but accurately predicting the binding affinities of fragments and their elaborated analogs poses unique computational challenges. These difficulties arise from weak binding affinities, diverse chemical scaffolds, and limited structural overlap between fragments and their optimized derivatives. While several free-energy methods exist, few are tailored to the specific requirements of FBDD. In this study, we evaluate the Separated Topologies (SepTop) approach for modeling fragment-based transformations

Guanosine 10 methylation by TRMT11-TRMT112 optimizes human mitochondrial tRNAs for efficient translation.

Modified nucleotides are essential determinants of RNA function, and identifying the enzymes that install them is fundamental to understanding their cellular roles. Here, we show that the human RNA methyltransferase TRMT11 and its cofactor TRMT112 are imported into mitochondria via N-terminal targeting signals. Using a recently developed N2-methylguanosine (m2G)-sensitive DNAzyme, we demonstrate that TRMT11 catalytic activity and interaction with TRMT112 are required for installation of m2G at position 10 in 13 mitochondrial (mt-)tRNAs. The crosslinking profile of TRMT11 on mt-tRNAs experiment

Transferring strongly reducing electrons to bidentate-adsorbed CO2 for efficient and selective formate photosynthesis.

The photocatalytic conversion of carbon dioxide into formate offers substantial economic value; however, it suffers from the inherent efficiency-selectivity trade-offs. Here, we introduce phthalocyanine polymer dots coexposed with defined phthalocyanine nitrogen (Npc) and phthalocyanine nitrogen-hydrogen (Npc─H) sites as promising photocatalysts, achieving synergistic enhancement in efficiency and selectivity. Npc sites act as electron nodes, directing photogenerated electrons from photosensitizers toward carbon dioxide-adsorbed phthalocyanine, boosting the reactive charge density by 6.5-fold.

Surface Halogen Passivation Enables Ultra-Stable Metal Sulfide for Efficient Methanol Photoactivation.

Transition metal sulfides exhibit promising photocatalytic activity for high-value-added chemical synthesis; however, they suffer from significant photocorrosion and rapid deactivation during reactions. In this study, we propose a surface halogen (Cl, Br, and I) passivation strategy to improve the stability of photocatalysts, which would provide a protective barrier against chemical attack, preventing oxidative and reductive surface degradation. By taking the typical metal sulfide catalysts of CdS as a representative model, we show that the introduced surface halogen can effectively suppress s

Synergistic pulsed electrocatalysis enables green and efficient synthesis of nylon monomer adipic acid.

Adipic acid (AA), a pivotal monomer, is produced via high-temperature nitric acid oxidation of cyclohexanone-cyclohexanol mixture (KA oil); a NO2-intensive process with a large carbon footprint. Electricity-powered electro-oxidative upgrading of KA is appealing, but impractical due to poor O2 utilization, oxygen evolution, and rapid catalyst passivation under potentials, causing an activity-stability-selectivity trade-off. We present an integrated electrocatalytic platform combining an α-FeOOH/NiFe layered double hydroxide (NiFe-LDH) heterojunction catalyst with pulsed electrolysis (PE) for gr

Layer-by-Layer Assembled Polyelectrolytes on Ta3N5 Pyramid Arrays for Efficient Photoelectrochemical Water Splitting.

Ta3N5 is a promising material for overall water splitting due to its suitable band positions and high theoretical photocurrent (12.9 mA cm-2). However, pristine Ta3N5 suffers from significant optical, recombination, and resistive losses, resulting in photocurrents and efficiencies far below theoretical expectations. To address these limitations, this work focuses on enhancing photocurrent and overall performance through structural and morphological engineering and surface functionalization. We investigated the potential of polyelectrolytes, poly(sodium-p-styrenesulfonate) (PSS) with anionic gr

Efficient bioimaging using rare-earth-doped carbon quantum dots: A doping strategy.

This study addressed the limitations of carbon quantum dots (CQDs) in bioimaging, specifically low photoluminescence quantum yield (PLQY) and complex purification processes, by developing lanthanide-doped CQDs (L-CQDs). Using a microwave-assisted solvothermal method in a glycerol-water system with precursor mass ratios of citric acid monohydrate: urea: NdCl3·6H2O:CeCl3 at 30:3:10:2 and a glycerol/water volume ratio of 2:1, we achieved one-pot synthesis with cerium and neodymium doping. This approach significantly enhanced the PLQY from 0.43% to 69%. The resulting L-CQDs exhibited a graphene-li

Design of a functionalized lacunary polyoxometalate catalyst for efficient epoxide ring-opening and biodiesel production.

Developing robust and efficient heterogeneous catalysts remains a key challenge in catalytic organic transformations. In this work, a lacunary polyoxometalate-based catalyst (LPOM) was synthesized and subsequently functionalized with 3-(aminopropyl)trimethoxysilane (APTMS) and then 1,3-propanesultone (1,3-PS) to achieve LPOM/APTMS/1,3-PS. Comprehensive characterization using various physicochemical techniques confirmed the successful modification of the lacunary POM framework with desired functional groups. The catalytic behavior of the prepared material was assessed through two representative

Metabolic and transport engineering enable efficient de novo biosynthesis of patchoulol in Saccharomyces cerevisiae.

Patchoulol is a valuable sesquiterpene for fragrance and cosmetics, but plant extraction is slow and resource-intensive. Here, we combined enzyme, pathway and transporter engineering to build a yeast chassis for high-titer patchoulol production. Semi-rational engineering of patchoulol synthase (PTS) increased catalytic efficiency; together with multi-copy PTS expression, the shake-flask titer rose from 0.40 ± 0.05 mg/L (PT01) to 258.72 ± 27.35 mg/L. Strengthening glycolysis and the mevalonate pathway and down-regulating ERG9 redirected flux to FPP. Guided by docking and transcript analysis, we
