The dispositions of radiolabeled diethylstilbestrol (DES) and estradiol (E2) in the fetal rat were compared to determine whether kinetic differences accounted for their differences in teratogenic potency. 14C (from DES) was concentrated in fetal tissues relative to plasma, whereas 3H (from E2) was largely retained protein-bound in fetal plasma. Spironolactone and digitoxin have previously been shown to interact with cytosol androgen and estrogen receptors, respectively, in the rat. Hormone replacement therapy (HRT) is given to relieve the climacteric symptoms of menopause. All current hormonal contraceptives have side effects and contraindications related to estrogens and progestins. Users are often dissatisfied with this. There is a great need for a new contraceptive drug without these hormones with the associated side effects and contraindications; with also a favorable bleeding profile; that can be used orally and not daily; that can serve as a contraceptive and after-care and can also be used on demand. Mifepristone 50 mg seems to provide the answer to all these wishes but is not (yet) registered in the Netherlands.

Gynecomastia is a common finding in males, with an incidence that varies widely globally. In 10-25% of cases, it is caused by drugs. Its pathophysiologic mechanism includes exposure to exogenous estrogens and medications that cause hypogonadism, antiandrogenic effects and hyperprolactinemia. Gynecomastia is caused by drugs in 10 - 25% of all cases. The pathophysiologic mechanism for some drugs includes exogenous estrogens exposure, medications that cause hypogonadism, anti-androgenic effects and hyperprolactinemia. Pretreatment plasma concentrations of total testosterone, prolactin, and total estradiol-17 beta (E2) were measured in 123 prostatic cancer patients who were categorized into groups according to the UICC classification.
