Lymphedema are characterized by interstitial edema leading to swelling of extremities. They can be divided into primary and secondary lymphedema. Developmental abnormalities of the lymphatic system are responsible for the primary form of lymphedema. The secondary form of lymphedema is caused by damage of the lymphatic system due to external factors. Lymphedema can rarely be observed in patients with tuberous sclerosis complex (TSC), which is a neurocutaneous syndrome caused by pathogenic variants in the genes TSC1 or TSC2. Patients with TSC usually present with neurological manifestations and

Caudal regression syndrome (CRS) represents a spectrum of clinical phenotypes with varying degrees of malformation of the lower body with involvement of structures deriving from all 3 layers of the trilaminar embryo. We review areas of active investigation in the diagnosis, etiology, epidemiology, and treatment of the disease with a focus on underlying genetics. CRS pathobiology is complex and multifactorial with a significant contribution from environmental factors as evidenced in twin studies. Contemporary genomic and genetic investigations in both human primary tissue and murine in vitro an

BACKGROUND: Chromosome deletions of the long arm of chromosome 4 in 4q syndrome are characterized by mild facial and digital dysmorphism, developmental delay, growth retardation, and skeletal and cardiac anomalies, which is regarded as an autism spectrum disorder. Moreover, some scarce reports indicate that patients with 4q interstitial deletion and 7p duplication may present symptoms associated with hearing loss.

Down syndrome (DS) is the most common genetic disorder leading to developmental disability. The phenotypes associated with DS are complex and vary between affected individuals. Placental abnormalities in DS include differences in cytotrophoblast fusion that affect subsequent conversion to syncytiotrophoblast, atypical oxidative stress/antioxidant balance, and increased expression of genes that are also upregulated in the brains of individuals with Alzheimer's disease. Placentas in DS are prematurely senescent, showing atypical evidence of mineralization. Fetuses with DS are especially suscepti

Methylmercury (MeHg) is a ubiquitous environmental neurotoxicant, with human exposures predominantly resulting from fish consumption. Developmental exposure of zebrafish to MeHg is known to alter their neurobehavior. The current study investigated the direct exposure and transgenerational effects of MeHg, at tissue doses similar to those detected in exposed human populations, on sperm epimutations (i.e., differential DNA methylation regions [DMRs]) and neurobehavior (i.e., visual startle and spontaneous locomotion) in zebrafish, an established human health model. F0 generation embryos were exp

RAS signaling plays a key role in controlling appropriate cell responses to extracellular stimuli and participates in early and late developmental processes. Although enhanced flow through this pathway has been established as a major contributor to oncogenesis, recent discoveries have revealed that aberrant RAS activation causes a group of clinically related developmental disorders characterized by facial dysmorphism, a wide spectrum of cardiac disease, reduced growth, variable cognitive deficits, ectodermal and musculoskeletal anomalies, and increased risk for certain malignancies. Here, we r

Notch signaling is critical for cell fate decisions during development. Caenorhabditis elegans and vertebrate Notch ligands are more diverse than classical Drosophila Notch ligands, suggesting possible functional complexities. Here, we describe a developmental role in Notch signaling for OSM-11, which has been previously implicated in defecation and osmotic resistance in C. elegans. We find that complete loss of OSM-11 causes defects in vulval precursor cell (VPC) fate specification during vulval development consistent with decreased Notch signaling. OSM-11 is a secreted, diffusible protein th

The insulin superfamily constitutes a phylogenetically conserved hormonal system that critically regulates growth, metabolism, and reproduction in Metazoa. However, the functional diversification and the architecture of its regulatory networks of this superfamily remains poorly characterized in crustaceans. Here, we identify and characterize two little-studied insulin superfamily genes in the Pacific white shrimp Litopenaeus vannamei, gonadulin (LvGon) and insulin-related peptide (LvIRP). LvGon is predominantly expressed in neural tissues (brain and thoracic ganglia), while LvIRP is specifical

Indirect flight muscles (IFMs) of Drosophila melanogaster are among the most structurally ordered and metabolically demanding tissues in the animal kingdom. Importantly, IFMs comprise two anatomically and mechanistically distinct muscle groups: the dorsal longitudinal muscles (DLMs) and the dorsoventral muscles (DVMs). Their ability to sustain high-frequency, stretch-activated oscillations depends not only on specialised sarcomeric architecture but on multilayered signalling networks that coordinate lineage specification, myoblast proliferation, fibre patterning, mechanotransduction, metabolic

Prenatal valproic acid (VPA) exposure is strongly linked to developmental cardiotoxicity, yet no targeted pharmacological countermeasure exists. Converging mechanistic evidence indicates that VPA cardiotoxicity centers on AMPK/PGC-1α suppression, positioning the AMPK activator metformin (MET) as a rational candidate for in vivo protection. We examined MET's protective effects against VPA-induced developmental cardiotoxicity in zebrafish embryos. Embryos were randomly allocated to four groups (Control, VPA 0.1 mM, MET 10 mM, VPA + MET) immediately after fertilization and chronically exposed unt

BACKGROUND AND AIMS: Phosphorus (P) deficiency severely limits rice productivity, yet the role of root developmental regulators in P homeostasis remains unclear. Here, we show that the rice PLETHORA transcription factor OsPLT1 suppresses rice tolerance to P-deficient conditions. Under P-starved conditions, osplt1 mutants accumulated more biomass and exhibited higher soluble and total P concentrations, whereas OsPLT1-overexpressing lines were hypersensitive to P deficiency.
KEY RESULTS: Mechanistically, osplt1 mutants displayed reduced P retention in root cell walls, particularly in the water-

Gestational diabetes mellitus (GDM) exposes the fetus to chronic hyperglycemia and increases the risk of later metabolic disease. The mechanisms by which early hyperglycemia reprograms lipid and glucose metabolism remain unclear, largely due to maternal and placental confounders in mammalian models. To overcome these limitations, we developed a zebrafish model that isolates fetal hyperglycemia and enables direct investigation of its developmental and long-term metabolic effects. To mimic late-onset GDM, zebrafish embryos were exposed at 4 days post-fertilization to glucose concentrations compa

Vanillic acid (VA), widely used in industrial and therapeutic applications, also presents potential ecological and biological risks. This study systematically investigated the developmental and neurotoxic effects of VA in zebrafish and explored its mechanistic pathways within an Adverse Outcome Pathway (AOP) framework. VA exposure resulted in concentration-dependent toxicity, with an LC50 of 638.3 μM. High VA concentrations induced developmental abnormalities, including malformations of the eyes, otic vesicles, craniofacial structures, and tails. Behavioral assessments revealed a significant r

Tetralogy of Fallot (TOF) is the commonest cyanotic congenital heart disease, arising from antero-cephalad deviation of the outlet septum during outflow tract (OFT) morphogenesis, producing a ventricular septal defect, overriding aorta, pulmonary stenosis, and right ventricular hypertrophy. Despite advances in surgical correction, the developmental origins of TOF remain incompletely understood, and the genetic architecture of the majority of non-syndromic cases is yet to be resolved. Experimental models have been central to progress in this field, yet each model may capture only part of the di

PurposeAnterior Segment Dysgenesis (ASD) encompasses a heterogeneous group of congenital ocular malformations resulting from disrupted differentiation of neural crest-derived tissues. These disorders frequently lead to developmental glaucoma, a major cause of childhood blindness. This review summarizes the current clinical, genetic, and management perspectives on ASD associated with glaucoma.MethodsA comprehensive literature search of PubMed, Scopus, and Web of Science (January 2000-August 2025) identified 186 relevant studies reporting clinical, molecular, and therapeutic findings. BACKGROUND AND AIMS: Maternal obesity increases the risk of the paediatric form of metabolic dysfunction-associated steatotic liver disease (MASLD), affecting up to 30% of youth, but the developmental origins remain poorly understood.
METHODS: Using a Japanese macaque model, we investigated the impact of maternal Western-style diet (mWSD) or chow diet followed by postweaning WSD (pwWSD) or chow diet focusing on bile acid (BA) homeostasis and hepatic fibrosis in livers from third-trimester fetuses and 3-year-old juvenile offspring.
RESULTS: Juveniles exposed to mWSD had increased hepatic coll

INTRODUCTION: The development of novel scaffold incorporating Cissus quadrangularis extract, bioceramics and tendon extracellular matrix (ECM) for periodontal bone regeneration necessitates a thorough assessment of their embryotoxicity to ensure biocompatibility and safety. This study evaluates the embryonic toxicology of these innovative scaffold using the zebrafish model, which provides a rapid transparent and highly sensitive system for assessing development toxicity.
MATERIALS AND METHODS: Zebrafish embryos were exposed to scaffold containing Cissus quadrangularis extract (80 % ethanol),

Intrauterine growth restriction (IUGR) is associated with an elevated risk of long-term metabolic disorders, including obesity and type 2 diabetes, aligning with the Developmental Origins of Health and Disease hypothesis. However, the mechanisms underlying metabolic programming remain elusive. This study investigates the impact of gestational protein restriction (PR) on fetal hypothalamic development, focusing on the formation of neuronal populations regulating appetite and energy balance. Using a rat isocaloric PR model (8% protein preconception, 4% during gestation), we examined hypothalamic

Constitutively active mutants of BRAF cause cardio-facio-cutaneous (CFC) syndrome, characterized by growth and developmental defects, cardiac malformations, facial features, cutaneous manifestations, and mental retardation. An animal model of human CFC syndrome, the systemic BrafQ241R/+ mutant mouse, has been reported to exhibit multiple CFC syndrome-like phenotypes. In this study, we analyzed the effects of Braf mutations on neural function, separately from their effects on developmental processes. To this end, we generated Braf mutant mice expressing BRAFQ241R specifically in mature excitato

Colchicine is a type of alkaloid commonly used clinically for treating diseases such as gouty arthritis. Studies suggest that colchicine is toxic to the kidneys. However, the specific mechanisms are unclear. Herein, we used zebrafish to validate the renal toxicity of colchicine and investigate the underlying mechanisms. Zebrafish larvae at 3 days post-fertilization were exposed to various concentrations (15, 20, and 25 mg/L) of colchicine solution for 72 h. Results showed that colchicine caused death, slowed heart rate, slowed growth, increased interpupillary distance, caused periorbital and r
