Haploinsufficiency, de novo and biallelic missense variants in CSMD2 are associated with neurodevelopmental disorders.

CUB and Sushi domains-containing CSMD proteins are involved in neuron development. Variants in the CSMD1-3 paralogs were GWAS-associated with cognition, attention-deficit hyperactivity disorder (ADHD), schizophrenia and brain structure, suggesting that these genes could be linked to neurodevelopmental disorders (NDD). Indeed, rare biallelic variants in all three CSMD paralogs were recently associated with epilepsy, while rare biallelic variants in CSMD1 were also linked with intellectual disability and cortical malformations. We identified nine individuals from eight families carrying CSMD2 bi

Infantile 22q11.2 deletion syndrome with secondary monogenic variants following inconclusive whole exome sequencing: a case report.

BACKGROUND: Chromosome 22q11.2 deletion syndrome (22q11.2DS), historically known as DiGeorge syndrome, is a common microdeletion disorder characterized by a wide range of multisystem manifestations. While Whole Exome Sequencing (WES) is increasingly used as a first-tier diagnostic tool for complex infant presentations, it has limited sensitivity for detecting copy-number variants such as microdeletions. Parental consanguinity adds further diagnostic complexity by increasing the likelihood of co-occurring single-gene variants. CASE PRESENTATION: We present the case of a 12-month-old female bor

A multi-omics approach elucidates the link between artificial food colorings and common cancers.

BACKGROUND: Artificial food colorings (AFCs) are widely used, yet their potential links to cancer remain unclear. We investigated associations between commonly used AFCs and cancer-related molecular networks and prognosis. METHODS: AFCs-related targets were collected from CTD, ChEMBL, SEA, and TargetNet, and cancer-related targets from GeneCards, OMIM, and CTD. Overlapping targets were subjected to STRING-based PPI analysis and Cytoscape visualization, followed by GO/KEGG enrichment. Core targets were evaluated for differential expression in GEO datasets of non-small cell lung cancer (NSCLC),

mTOR and STING Modulation Restores Hydrolase Activities and Decreases Alpha-Synuclein Pathology in the Cells from the Patients with GBA1-Associated Parkinson's Disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by dopaminergic neuron loss and pathological accumulation of alpha-synuclein. Aberrant activation of the mechanistic target of rapamycin (mTOR) and of the stimulator of interferon genes (STING) signaling pathways are increasingly recognized as central contributors to PD pathogenesis. The GBA1 variants are the most common genetic risk factor for PD. However, currently there are no disease-modifying therapies for treatment of the GBA1-associated PD (GBA1-PD). Here, we investigated the effects of selective inhibito

Network Pharmacology and Experimental Validation to Explore the Potential Mechanism of Salvianolic Acid B in Reversing Oxaliplatin Resistance of Colorectal Cancer Cells.

Chemotherapy resistance of colorectal cancer (CRC) seriously affects the therapeutic effect. Salvianolic acid B (SalB) is a primary active compound found in the traditional Chinese medicine (TCM) Danshen, known for its antitumour properties and ability to reverse chemoresistance. However, the underlying mechanism remains to be elucidated. The present study aims to explore the chemoresistance-reversal effects of SalB on colon cancer cells and elucidate its potential mechanisms using both network pharmacology and experimental approaches. Accordingly, SalB- and disease-related targets were extrac

[Overview of cytoskeletal actin mutations associated with rare diseases].

Dynamics of the actin cytoskeleton play an important role in many physiological processes. Mutations in actin and actin-binding proteins cover a wide spectrum of diseases, called actinopathies. Of the six actin isoforms, the cytoskeletal β- and γ-actin isoforms differ from each other by only four amino acids. This results in overlapping and isoform-specific functions. Mutations in the ACTB and ACTG1 genes have been associated with rare diseases, so-called non-muscle actinopathies (NMA). The phenotype of affected individuals covers a wide clinical spectrum - from asymptomatic variants to mutati

BMPR2 Splice-Site Variant in a Patient With Pulmonary Arteriovenous Malformation and Delayed-Onset Pulmonary Arterial Hypertension: A Case Report and Mechanistic Phenocopy Hypothesis.

Pulmonary arteriovenous malformations (PAVMs) are rare vascular anomalies most commonly seen in hereditary hemorrhagic telangiectasia (HHT), a condition associated with mutations in ENG, ACVRL1, SMAD4, or GDF2. In contrast, BMPR2 variants are well-established in heritable pulmonary arterial hypertension (PAH), but their relationship to PAVMs remains poorly understood. We report the case of a 41-year-old woman with an incidentally discovered PAVM, initially treated with embolization and subsequent surgical resection. She remained asymptomatic for several years until progressive exertional dyspn

BBP acts as an exogenous modulator of MMP9 to rewire vascular immunity and promote atherosclerosis.

Environmental exposure to endocrine-disrupting chemicals has been increasingly implicated in cardiovascular disease, yet the causal molecular mechanisms linking such exposures to atherosclerosis remain incompletely defined. Benzyl butyl phthalate (BBP), a widely used plasticizer, has been associated with vascular dysfunction, but its mechanistic contribution to atherogenesis is unclear. Here, we employed an integrative systems biology framework combining network toxicology, transcriptomic profiling, protein-protein interaction network analysis, molecular docking, molecular dynamics simulations

A Tiered Genetic Diagnostic Approach in Newborns With Major Congenital Anomalies: Experience From a Tertiary NICU.

BACKGROUND: Major congenital anomalies (MCAs) are a leading cause of neonatal morbidity and mortality and frequently reflect underlying chromosomal, copy number, or single-gene defects. In neonates with multiple MCAs, early molecular diagnosis is critical but remains challenging due to phenotypic heterogeneity, overlapping developmental pathways, and limitations in selecting the most appropriate genetic test. METHODS: This prospective study included 146 neonates with congenital anomalies who were referred for genetic consultation from a tertiary neonatal intensive care unit between April 2022

DNA methylation marks associated with body composition in children from India and the Gambia: findings from the EMPHASIS study.

BACKGROUND: Differences in body composition during childhood can influence long-term health, with notable links to cardiometabolic disorders in later life. While genetic associations with body composition traits are well-studied, less is known about the role of epigenetic mechanisms, particularly in low- and middle-income countries where the burden of cardiometabolic disease is high. We investigated links between DNA methylation and three compartments of body composition: fat mass, lean mass, and bone measures using data from children enrolled in the Epigenetic Mechanisms linking Pre-conceptio

Mendelian Randomization and Bioinformatics Identify IFI27 as a Candidate Biomarker for Recurrent Pregnancy Loss in Systemic Lupus Erythematosus.

Systemic lupus erythematosus (SLE) is associated with adverse pregnancy outcomes, but its causal relationship with recurrent pregnancy loss (RPL) and their shared molecular characteristics remain unclear. This study integrated bidirectional two-sample Mendelian randomization (MR) and transcriptomic bioinformatics analyses to investigate this relationship and identify candidate shared biomarkers. FinnGen and UK Biobank were selected because they provide large, non-overlapping, European-ancestry genome-wide association study (GWAS) summary statistics.
